The Effects of Enzyme Complex on Performance, Intestinal Health and Nutrient Digestibility of Weaned Pigs

  • Two experiments were conducted to evaluate the effect of supplementing a corn-soybean meal-based diet with an enzyme complex containing amylase, protease and xylanase on the performance, intestinal health, apparent ileal digestibility of amino acids and nutrient digestibility of weaned pigs. In Exp. 1, 108 piglets weaned at 28 d of age were fed one of three diets containing 0 (control), 100, or 150 ppm enzyme complex for 4 wks, based on a two-phase feeding program namely 1 to 7 d (phase 1) and 8 to 28 d (phase 2). At the end of the experiment, six pigs from the control group and the group supplemented with 150 ppm enzyme complex were chosen to collect digesta samples from intestine to measure viscosity and pH in the stomach, ileum, and cecum, as well as volatile fatty acid concentrations and composition of the microflora in the cecum and colon. There were linear increases (p<0.01) in weight gain, gain: feed ratio and digestibility of gross energy with the increasing dose rate of enzyme supplementation during the whole experiment. Supplementation with enzyme complex increased the digesta viscosity in the stomach (p<0.05) and significantly increased (p<0.01) the concentrations of acetic, propionic and butyric acid in the cecum and colon. Enzyme supplementation also significantly increased the population of Lactobacilli (p<0.01) in the cecum and decreased the population of E. coli (p<0.05) in the colon. In Exp. 2, six crossbred barrows (initial body weight: $18.26{\pm}1.21$ kg), fitted with a simple T-cannula at the distal ileum, were assigned to three dietary treatments according to a replicated $3{\times}3$ Latin Square design. The experimental diets were the same as the diets used in phase 2 in Exp. 1. Apparent ileal digestibility of isoleucine (p<0.01), valine (p<0.05) and aspartic acid (p<0.05) linearly increased with the increasing dose rate of enzyme supplementation. In conclusion, supplementation of the diet with an enzyme complex containing amylase, protease and xylanase improved piglet performance. This is likely a result of improvement in nutrient digestibility, volatile fatty acid concentrations and bacteria ratio in the large intestine.

The Slow and Tight Binding of MR-387A to Aminopeptidase N

  • MR-387A [(2S, 3R)-2-hydroxy-3-amino-4-phenylbutanoyl-L-valyl-L-prolyl-(2, 4-trans)- L-4-hydroxy-proline] reversibly inhibits aminopeptidase N (BC 3.4.11.2) in a process that is remarkable for its unusual degree of time dependence. The time required to inactivate the enzyme by 50$%$ ($t_{1/2}$) for establishing steady-state levels of $EI^*$complex was approximately 5 minutes. This indicates that the inhibition is a slow-binding process. In dissociation experiments of $EI^*$ complex, enzymic activity was regained slowly in a quadratic equation, indicating that the inhibition of aminopeptidase N by MR-387A is tight-binding and reversible. Thus, the binding of MR-387A by aminopeptidase N is slow and tight, with $K_{i}$ (for initial collision complex, EI) and $K_i{^*}$ (for final tightened complex, $EI^*$) of $2.2\times10^{-8}$ M (from Lineweaver-Burk plot) and $4.4\times10^{-10}$ M (from rate constants), respectively. These data indicate that MR-387A and aminopeptidase N are bound approximately 200-fold more tightly in the final $EI^*$complex than in the initial collision EI complex.

사료 내 복합효소제의 첨가가 육계 생산성 및 도체특성 및 저장성에 미치는 영향 (Effects of Dietary Supplementation of Enzyme complex on Growth Performance, Carcass Characteristics and Meat storability in Broiler Chickens)

  • 본 연구는 육계사료 내 금속단백질분해효소와 xylanase를 포함하는 복합 효소제 첨가 급여가 육계 생산성, 도체특성 및 계육 저장성에 미치는 영향을 조사하기 위하여 실시하였다. 실험 디자인은 2 (기초사료, 저영양사료) ${\times}$ 3 (0, 0.5, 1 g/kg 효소첨가) 요인 배치로 설계하였다. 실험동물은 ross broilers 수컷 360수를 공시하여 다음의 6개의 시험 그룹에 완전임의 배치하였다. BD-T0(기초사료+복합효소제 0 g/kg), BD-T1(기초사료+복합효소제 0.5 g/kg), BD-T2(기초사료+복합효소제 1 g/kg), LD-T0(저영양사료+복합효소제 0 g/kg), LD-T1(저영양사료+복합효소제 0.5 g/kg), LD-T2(저영양사료+복합효소제 1 g/kg). 사료와 물은 42일 동안 자유채식을 실시하였으며, 사료섭취량과 체중은 1주 간격으로 측정하였다. 사양시험 종료 후에 방혈로 도살하고 분석에 필요한 시료를 채취하였다. 사료 섭취량은 효소첨가 그룹이 효소 비첨가 그룹에 비해서 유의하게 감소하였으며, 체중과 일당증체량은 효소첨가 그룹이 효소 비첨가 그룹에 비하여 유의하게 증가하였다. 따라서 사료요구율은 복합효소 첨가에 의해 유의하게 개선되었다(p<0.05). 복합효소첨가 급여에 의하여 도체율과 생산지수 또한 유의하게 개선되었다(p<0.05). TBARS는 계육 저장 6일차에 효소첨가 그룹이 효소 비첨가 그룹보다 낮은 값을 나타내었다(p<0.05). 본 연구의 결과를 종합해보면, 사료 내 복합효소제의 첨가 급여는 생산성과 도체성적 그리고 저장성을 유의하게 개선시켰다.

An optimized cupric reducing antioxidant capacity (CUPRAC) method for assessment of xanthine oxidase inhibition activity

  • This protocol clarifies a simple and precise method for measuring the activity of xanthine oxidase (XO) enzyme inhibitor. XO enzyme, which accelerates oxidative stress-related disorders through its capacity to generate hydrogen peroxide and superoxide anion radicals (O2•-), has been found to be inhibited by several plant extracts. Enzyme samples were incubated with a suitable buffer containing adequate amounts of xanthine as a substrate to determine XO activity. The method depends on direct measurements of uric acid and hydrogen peroxide production to test XO with and without interference. The CUPRAC reagent (Cu(Nc)22+) was used to inhibit enzyme reaction after incubation was complete. The generated urate and peroxide reduced the Cu(II)-neocuproine complex (Cu(Nc)22+) to a brightly colored Cu(I)-neocuproine complex (Cu(Nc)2+), which was assessed with a spectrophotometer at 450 nm. XO activity was found to be directly related to the increased absorbance of the colored Cu(I)-neocuproine complex (Cu(Nc)2+). To eliminate catalase enzyme interference, the proposed method used sodium azide and was validated against XO activity using the UV method in matched samples with t-test analysis. The proposed assay can determine XO activity with high precision, as indicated by the correlation coefficient (R2 = 0.9935) from comparison with the reference protocol.

A SURVEY OF N-STRING TANGLE ANALYSES OF DNA-ENZYME SYNAPTIC COMPLEXES

  • In last 30 years, mathematical tangle theory is applied to molecular biology, especially to DNA topology. The recent issues and research results of this topic are reviewed in this paper. We introduce a tangle which models an enzyme-DNA complex. The studies of 2-string tangle equations related to Topoisomerase II action and site-specific recombination is discussed. And 3-string tangle analysis of Mu-DNA complex, n-string tangle analysis ($n{\geq}4$) of DNA-enzyme synaptic complexes are also discussed.

바이오 에탄올 부산물 DDGS와 복합 효소제 첨가급여가 돈육의 품질 특성에 미치는 영향 (Effects of Dietary Bio Ethanol By-product and Complex Enzyme on Meat Quality of Pork Loin)

  • 시험동물은 $68.77{\pm}0.10kg$인 3원 교잡종(Landrace${\times}$Yorkshire${\times}$Duroc) 비육돈 96두를 공시하였으며, 8주간 사양시험을 실시하였다. 시험설계는 1) corn-soybean meal diet, 2) corn-soybean meal diet + 0.05% enzyme complex, 3) cornsoybean meal diet with DDGS 및 4) corn-soybean meal diet with DDGS + 0.05% enzyme complex으로 4개 처리를 하여 처리당 6반복, 반복당 4두씩 완전임의 배치하였다. 등심의 pH는 DDGS를 함유한 두 처리구가 CON - Enzyme 처리구와 비교하여 유의적으로 증가하였다(p<0.01). 관능평가에 있어서 경도(Firmness)은 DDGS - Enzyme 처리구가 CON 처리구와 비교하여 유의적으로 증가하였으며(p<0.01), 육색에 있어서 $a^*$ 값은 DDGS를 함유한 두 처리구가 DDGS를 함유하지 않은 두 처리구와 비교하여 유의적으로 높게 나타났다(p<0.05). 등심 및 등지방의 지방산 조성에서 총 UFA와 총 UFA/SFA는 DDGS를 함유한 처리구가 CON 처리구와 비교하여 유의적으로 높은 효과를 나타냈다(p<0.001). 결론적으로 DDGS 급여는 등심의 pH, 경도, 육색, 불포화 지방산 조성을 증가시켰으나, 복합 효소제 첨가 급여에 따른 육질의 변화는 크게 나타나지 않았다.

토양내에 고정화되어 있는 Cellobiohydrolase의 효소학적 성질 (Enzymatic Properties of Cellobiohydrolase immobilized in Soil)

  • The enzymatic properties of soil cellobiohydrolase were examined and compared with those of cellobiohydrolase-active extracts from soil in the forms of enzyme-humic complex and humicfree enzyme, and cellobiohydrolase partially pruified from Aspergillus niger. The pH optima of soil cellobiohydrolase and cellobiohydrolase-humic complex were greater by 1.5-3.0 pH units than those of cellobiohydrolase in humic-free extract and from A. niger. Soil cellobiohydrolase and cellobiohydrolase-humic complex were remarkably resistant to thermal denaturation and proteolysis. These results confirm that cellobiohydrolase in soil is atable in conditions which rapidly inactivate microbial cellobiohydrolase and that its stability is due to the immobilization of this enzyme by association with humic substances. The Michaelis-Menten constants (Km) for soil, cellobiohydrolase-humic complex, humic free extract and cellobiohydrolase from A. niger were 22.1mg/ml, 11.3mg/ml, 10.6mg/ml and 4.5 mg/ml of Avicel, respectively.

효소복합체가 양돈슬러리 및 돈사 악취발생에 미치는 영향 (Effects of Enzyme Complex on Odor Emission from Swine Slurry and Swine Buildings)

  • 본 연구는 효소복합체 처리가 양돈분뇨의 액비화 과정의 악취제어 가능성을 조사하기 위하여 액비저장 탱크에 4주간 저장하면서 효소복합체 처리, 폭기 처리, 효소와 폭기 동시 처리 및 대조구로 나누어 비교하였다. 또한 효소복합체를 돈사에 살포하여 돈사내 공기의 질과 양돈장 부지경계선에서의 복합악취농도를 조사하였는데, 그 결과를 요약하면 다음과 같다. 효소복합체와 폭기 처리는 양돈분뇨의 액비화 과정의 암모니아와 황화수소 농도에 영향을 미치는 것으로 조사되었으며, 암모니아와 황화수소 농도는 4주간의 액비화 과정 동안 지속적으로 감소되었다. 효소복합체의 일정 수준 처리는 양돈분뇨 액비화 과정의 악취저감을 위한 효율적인 수단이 될 수 있을 것으로 판단된다. 효소복합체와 폭기 처리는 양돈분뇨의 액비화 과정의 총질소, 총인 및 암모니아성 질소 농도에 영향을 미치는 것으로 조사되었으며, 효소복합체 처리는 양돈분뇨 액비의 품질 유지와 악취저감을 위한 하나의 기술적 대안으로 충분한 것으로 보인다. 효소복합체 살포는 돈사내 공기의 질을 개선하고 양돈장의 부지경계선에서의 복합악취 저감에도 효과적으로 작용하는 것으로 나타났다. 이상의 결과를 요약하면, 효소복합체 처리는 양돈분뇨 액비화 과정의 악취제어와 고품질 액비제조 가능성이 인정되며, 돈사 내부 및 양돈장 환경개선과 악취저감 효과가 있는 것으로 판단된다.

Modulation of Branched-Chain Amino Acid Metaolism by Exercise in Rats

  • A variety of important roles for branched-chain amino acids in metabolic regulation has been suggested. Branched-chain $\alpha$-keto acid dehydrogenase(BCKAD) complex is a rate limiting enzyme in branched-chain amino acid metabolism. The purpose of this study was to examine the effects of exercise on the activity and activity state of branched-chain $\alpha$-keto acid dehydrogenase in rat hert and liver thssues. Forty-eight Sprague-Dawley rats were assigned into three experimental groups : sedentary control, exercised, or exercised-rested. Submaximal exercise(running) for two hours significantly increased basal activity without a change in total activity in both tissues, with a concomitiant increase in activity state of the enzyme complex. At 10 min post-exercise, heart enzyme activity significantly decreased, though not to the control level, while liver enzyme activity remained unchanged. These data suggested that the exercise-induced increase in branched-chain $\alpha$-keto acid decarboxylation in rat tissues may not be the result of enzyme synthesis, but rather is due to increased activity of the BCKAD.

Comparison study between single enzyme and multienzyme complex in distiller's dred grains with soluble supplemented diet in broiler chicken

  • Upregulation of the nutritional value of feed is the major target of various studies in the livestock industry, and dietary enzyme supplementation could aid in digesting the nondegrading nutrients of grains in feed ingredients. Dried distillers' grains with solubles (DDGS) is a byproduct of the fermentation process in the beverage industry and can be used as a large supply source of fiber in feed. Therefore, we conducted an experiment with male broiler chickens to investigate the effect of various types of enzymes on DDGS and compare the efficacy of single enzyme and multienzyme complexes on growth performance and gut environments in broiler chickens. We used 420 1-day-old broiler chickens (Ross 308), and they were allotted into 4 dietary treatments with seven replications (CON, corn-soybean meal [SBM] diet; NC, DDGS supplemented diet; SE, 0.05 % of mannanase supplemented DDGS-based diet; MC, 0.10% of multienzyme complex (mannanase and xylanase, glucanase) supplemented DDGS-based diet. The dietary exogenous enzyme in the DDGS-supplemented diet could improve growth performance as much as the growth of the control group, and digestibility of dry matter, crude protein, and gross energy were significantly increased by enzyme addition in groups of chicks fed DDGS-supplementation diet. Moreover, the populations of pathogenic bacteria, coliforms, and Bacteroidetes were significantly decreased by enzyme supplementation, which might lead to improved gut mucus-secreting cells and inflammatory cytokines in the jejunum. Collectively, dietary single enzyme and multienzyme complexes could improve gut environments, including intestinal immune responses and gut microbial population, and lead to improvement of growth performance in broiler chickens.
