Purification and Characterization of Protease Produced by Bacillus subtilis YG-95 (Bacillus subtilis YG-95가 생산하는 protease의 정제와 특성)

  • The protease produced by Bacillus subtilis YG-95 was purified by precipitating with ammonium sulfate, DEAE-sepharose 6B and Sephadex G-100 column chromatogtaphies and its purified enzymological characteritics were investigated. The molecular weight of purified protease was estimated about 43kilodalton by SDS PAGE The optimum pH and temperature for the purified protease activity were pH 10.0 and $55^{\circ}C$, respectively. The enzyme was stable in broad range of pH 5.0 to 12.0. and at the below $45^{\circ}C$. The purified enzyme activty was inhibited by $Fe^{3+}$ and $Al^{3+}$. The activity was significantly inhibited more than 80% by O-Phenanthroline, PMSF and SDS. The $K_m$ value of the purified enzyme against Soy Protein Isolate as a substrate was 1.28 mg/ml.

Riboflavin Status Influences the Biosynthesis of Flavin Peptides and Related Enzyme Activities in Rat Liver Mitochondria (리보플라빈 결핍이 쥐간의 미토콘드리아의 플라빈 펩티드와 관련된 효소 활성에 미치는 영향)

  • The effeds of riboflavin defidency on the biosynthesis of flavin pepddes and levels of flavoenzymes and catecholamines have been investigated. The percentage of 14C. riboflavin radioactivity formed in mitochondria appeared to increase up to 2 weeks but started to decline at 3 weeks. A significant increase of radioactivity incorporation into mitochondria and into trypsin-digestable plus trypsin-non-digestibie flavin peptides was detected in riboflavin-deficient animals. More than 35% of incorporation was observed at the end of the first week and 160% higher incorporation was observed in fiavin peptide after the second week. Activities of MAO and succinate dehydrogenase were affected markedly by riboflavin status whereas those of acetyichoilnesterase were not affected. Riboflavin defidency also brought about marked reductions in levels of epineplrrine and norepinephrine. it is concluded that the levels of flavin peptides, MAO and succinate dehydrogenase, and catecholamines were affected significanily by the availability of riboflavin and in particular the duration of its depiction.

Effeds of Malathion on the Development of the Chick Embryo Cerebrum (계배의 대뇌의 발생에 미치는 Malathion의 영향)

  • Chick embryos which have received a single injection of the organophosphate compound,malathion (0.1 mg, 0.5 mg, 1.0 mg or 2.0 mg in 0.05 ml of corn oil) via the yolk sac at certain times (2 daya, 4 days or 6 days after incubation) have been investigated. After 9 days of incubation, chick embryos were harvested to investigate the effects of malathion on the development of cerebrum morphologically and biochemically. The effects of simultaneous injection of malathion and nicotinamide were also compared. On ultrastructural examination, neurons in cerebral cortex showed to be inhibited in their differentiation by malathion; nuclear irregularity, swelling of endoplasmic reticulum, and cytoplasmic vacuoles were observed. On cytochemical study of acetylcholinesterase(AChE) by electron microscope, the positive reaction products of this enryme were localized at the membrane of nucleus and endoplasmic reticulum of neurons. Inhibition of AChE activty was severe in groups treated with relatively low doses, which was consistent with the results of spectrophotometric analysis. The activity of lactate dehydrogenase(LDH) of cerebrum in groups treated with malathion was higher than that of the control group. The nicotinamide adenine dinucleotide(NAD) content of chick embruo treated with malathioti decreased significantly, and nicotinamide coinjection raised the NAD level as compared with the control group, thus preventing malathion-induced momhological alteration. In conclusion, it is suggested that malathion changes the ultrastructure of differentiating neurons and alters some enzyme activities in chick embryo cerebrum, and the severity of which is consistently dose-or age-dependent.

Studies on the Antioxioative Character in the Etnyl Acetate Extractions of Rumex crispus (Rumex crispus의 에칠아세테이트 추출물의 항산화 성분에 관한 연구)

  • This study was undertaken to investigate the antioxidative substance and activity of ethyl acetate extracted from Rumex crispus. Sample extracted follow in proper course of a solvent. Material refinement was carried out using silicagel column and Sephadex LH-20 column chromatography. Material sorting was carried by Gas Chromatography(GC/MS). 1,1-Diphenyl-2-Picrylhydrazyl(DPPH) free radical scavenging and enzyme activity were measured for antioxidative activity. as result of testing by DPPH free radical scavenging activity, Antioxidative activity was shown as the highest in the root, then leaf and stem in order. Ethyl acetate extraction of root part were 50% inhibitory concentration (IC50) Rumex activty(6.1 ug/mL). Rumex nipponicus(9.8 ug/ml) and Rumex acetoceae(31.5 ug/mL) in leaf part. The highest antioxidative activity of sample refined through silicagel column chromatography of Rumex crispus was appealed Fraction 5(IC50;3.57 ug/mL) in root and Fraction 6(IC50;85.9 ug/mL) in leaf. Fraction 5 in roof & Fraction 6 in leaf were refined using Sephadex LH-20 column chromatography. The highest antioxidative activity were appeared Fraction 4 (IC50;3.57 ug/mL) and Fraction 4 (IC50;18.41 ug/mL)in leaf. As for main phenol compounds 2,6-Dichloro-4-nitropnenol and 2-Isopropyl-5-methyl Phenol were identified in root and leaf, While 4-Vinyl-2- methoxy-phenol and 2,3-Dihydro- benzofuran were identifica ted only in leaf. Enzyme activity was shown low both in peroxidase(PDD) Non-activate(IU/mg protein)and in Superoxide dismutase(SOD) non-activate(IU/mg protein). 2,6-Dichloro-4-nitrophenol, 2-Isopropyl-5-methyl phenol, 4-Vinyl-2-methoxy-phenol were obtained in this experiment and these compounds are phenolic compounds which have OH group in the structure. With the result of this study these phenolic compounds which are extracted from Rumex crispus have high antioxidative effect. This antioxidative effect of Rumex crispus can be applied for chromo-preventive and antioxidative supplements which can be used for anti-allegy, aging, anti-tumor, aging and other oxidative disease for health promotion.

Studies on the Microflora and Enzymes Influencing on Korea Native Kochuzang (Red Pepper Soybean Paste) Aging (재래식(在來式) 고추장 숙성(熟成)에 미치는 미생물(微生物) 및 그 효소(酵素)에 관(關)한 연구(硏究))

  • The study was carried out to investigate the changes of the various chemical components and the microflora during the aging period of Korean navive Kochuzang. (Red pepper soybean paste) Korean native maeju loaves were separated into surface and inner parts. Three kinds of Korean native Kochuzang were prepared from surface part, inner part, and ordinary of maeju. The selection and the indentification of the high enzyme producing strains from the microflora and characteristics of their enzymes were studied. I. The changes of the various chemical components during the aging period of Kochuzang. 1) The changes of pH in the 3 kinds of Kochuzang displayed rapid decrease for the first 10 days after preparing and gradual curve of decrease until 60 days, but slight increase for the next 30 days. The pH of the surface part Kochuzang was lower than that of inner part or ordinary Kochuzang. 2) The total acid contents in the 3 kinds of Kochuzang showed gradual increase until the 60 days but it slowly reduced after this time. 3) The total nitrogen contents in the 3 kind of Kochuzang showed gradual inerease up to the 60 days, but slight decrease after this time. 4) The changes of trichloroacetic acid soluble nitrogen in the 3 kinds of Kochuzang showed a remarkable increase for the first 10 days, however gradual increase after this time. 5) The increase of amino nitrogen contents in the 3 kinds of Kochuzang seemed to be remarkable until the first 30 days, however to be less remarkable after this time. 6) The contents of reducing sugar in the 3 kinds of Kochuzang showed remarkable increase until the first 50 days and it slowly reduced after this time. II. The changes of microflora during the aging period of Kochuzang. 1) Aerobic, anaerobic bacteria and mold in the 3 kinds of Kochuzang were increased until the first 30 to 40 days, but they were reduced after this time. 2) No yeast in the three kinds of Kochuzang appeared until the first 20 days. Yeast were proved to grow, when the pH value was decreased below 5.4 after the 30 days. Yeasts in the surface part and ordinary Kochuzang were gradually increased and those in the inner part Kochuzang were decreased as aging. III. The selection and identification of high amylase and protease producing strains from the microflora during the aging period of Kochuzang. 1) The amylase and protease highly producing strains from microflora were identified as Bacillus subtilis-P, Bacillus subtilis-G, Bacillus licheniformis-K, Aspergillus oryzae-B. 2) Amylase activity of Aspergillus oryzae-B was highest among the strains and the strains in order of the higher activity to the lower one were Bacillus subtilis-P Bacillus licheniformis-K, Bacillus subtilis-G. Protease activities of Aspergillus oryzae-B and Bacillus subtilis-P were about the same and the strains in order of the higher activity to the lower one were Bacillus licheniformis-K, Bacillus subtilis-G. 3) Amylase activity was inhibited more than protease activity was with NaCl concentration. Amylase activity was inhibited by 45 to 65 percent and protease activity by 40 to 46 percent at the concentration of 15 percent NaCl, which was the average concentration of NaCl in Kochuzang.
