홍게 가공부산물을 고부가가치 식품소재로 이용하기 위하여 단백질 분해효소를 이용하여 가수분해하고 반응표면분석법으로 가수분해 조건을 최적화 하였다. 홍게 가공부산물을 단백질 분해 효소인 Flavourzyme으로 가수분해한 결과 효소반응곡선은 반응 초기 빠른 반응속도를 나타내다가 이 후에 느려지는 전형적인 형태를 나타내었다. 반응초기 90분까지 가수분해도는 30%까지 증가하다가, 이후 최종적으로 32-36%를 나타내었다. 최적화를 하기 위한 가수분해 요인변수로는 반응온도, 반응시간 및 홍게 가공부산물에 대한 Flavourzyme의 양을 선정하였고, 5개의 수준에서 부호화하여 이들을 중심합성설계법을 이용하여 반응표면분석을 실시하였다. 홍게 가공부산물을 Flavourzyme을 이용하여 반응표면 분석법으로 가수분해 조건을 최적화한 결과, 온도 $51.8^{\circ}C$, 반응시간 4시간 45분, 홍게 가공부산물에 대한 Flavourzyme의 양 3.8%로 나타났다. 홍게 가공부산물 효소분해물은 향미소재 및 반응향 제조의 전구물질로서 이용할 수 있을 것이다.
다양한 전처리 방법 중에서 묽은 황산법으로 전처리한 폐옥수수대가 효소적 당화 공정을 거쳤을 때 가장 높은 포도당 수율을 보였다. 이 효소적 당화공정을 통계적으로 분석한 결과 효소량, 고액비, 반응시간 모두 당화효율과 비례적인 관계를 보였으며 그 중에서 효소량이 가장 큰 영향을 주는 인자로 파악되었으며 최적 조건에서 76.1%의 당화 효율을 보일 것으로 예측되었다. 분리당화 발효공정에서, Saccharomyces cerevisiae는 효소 당화를 거쳐 얻은 포도당의 80%이상을 에탄올 수율 37%, 생산성 0.42 $g/L{\cdot}hr$로 바이오에탄올을 생산하였다. 동시당화 발효공정에서는 전처리된 시료가 가진 글루칸 59.5%가 0.20 $g/L{\cdot}hr$의 생산성으로 에탄올로 전환되었다. 두 공정을 통해서 얻을 수 있는 폐옥수수대 1 kg 당 바이오에탄올 양은 88 g 정도로 거의 같은 것으로 나타났다. 분리당화 발효공정과 동시당화 발효공정을 통해 강원도 폐옥수수대로부터 생산할 수 있는 바이오에탄올은 수거율 50% 기준으로 약 190만 리터로 예측되었다.
The mushroom cultured waste(MCW) from cork oak was evaluated as the raw material for bioethanol production. For enzymatic hydrolysis, cellulase cocktails (Celluclast 1.5L and Novozym 188) was used for polysaccharides to monosaccharides conversion. Compared with sound cork oak woodmeal, woodmeal from MCW showed higher cellulose to glucose conversion. To improve polysaccharides to monosaccharides conversion, pretreatment by sodium hydroxide was applied. Even though more xylan and lignin were removed in woodmeal of MCW than that of cork oak, concentration of glucose was higher from sodium hydroxide treated cork oak woodmeal (51.3 g/L) than treated MCW woodmeal (41.6 g/L).
3가지의 다당류 분해효소를 사용한 가수분해가 미역추출액의 상징액율, 고형분 수율, 점도에 미치는 영향을 조사하였다. 그 결과 추출전에 효소분해할 때 고형분 농도를 27.31%, 고형분 수율을 14.04%까지 증가시켰다. 그러나 미역 분산액을 원심분리 후의 상징액율은 약간 낮았다. 추출액의 점도는 효소분해 초기에 유의적으로 증가하였다.
Alnus sibirica Fisch. ex Turcz.(AS) geographically distributes in Korea, Japan, Northeast China and Russia. The bark of Alnus species have been used as traditional medicine for the treatment of allergic skin diseases. The content evaluation of compounds which were isolated from the barks of A. sibirica processed enzymatic hydrolysis(EAS); hirsutanonol(1), 5-O-methylhirsutanonol(2), rubranol(3), hirsutenone(4) and muricarpone B(5) was conducted using HPLC. Consequently, in EAS, the content of glycoside was decreased considerably. But the aglycones(1, 3 and 4) were increased highly and 2, 5 were formed newly.
The structural properties of cellulose are significantly changed with the progress of hydrolysis reaction. The effects of changes on such properties of cellulosic substrate as crystallinity, amicessibility of enzyme to the active site of cellulose surface, and particle size on the kinetics of enzymatic hydrolysis have been studied. Among those physical studies, the apparent surface active site of cellulose particle was found to have the most significant effect on the hydrolysis kinetics. Based on the experimental results, the adsorption affinity of enzyme and hydrolysis rate were mainly influenced by the surface roughness of cellulose particle. The extent of accesssible active site may be expressed as the change of particle diameter. The Langmuir isotherm was proposed in terms of enzyme activity to explain the actual action of enzyme protein.
Background: Minor saponins or human intestinal bacterial metabolites, such as ginsenosides Rg3, F2, Rh2, and compound K, are more pharmacologically active than major saponins, such as ginsenosides Rb1, Rb2, and Rc. In this work, enzymatic hydrolysis of ginsenoside Rb1 was studied using enzyme preparations from cultured mycelia of mushrooms. Methods: Mycelia of Armillaria mellea, Ganoderma lucidum, Phellinus linteus, Elfvingia applanata, and Pleurotus ostreatus were cultivated in liquid media at $25^{\circ}C$ for 2 wk. Enzyme preparations from cultured mycelia of five mushrooms were obtained by mycelia separation from cultured broth, enzyme extraction, ammonium sulfate (30-80%) precipitation, dialysis, and freeze drying, respectively. The enzyme preparations were used for enzymatic hydrolysis of ginsenoside Rb1. Results: Among the mushrooms used in this study, the enzyme preparation from cultured mycelia of A. mellea (AMMEP) was found to convert ginsenoside Rb1 into compound K with a high yield, while those from G. lucidum, P. linteus, E. applanata, and P. ostreatus produced remarkable amounts of ginsenoside Rd from ginsenoside Rb1. The enzymatic hydrolysis pathway of ginsenoside Rb1 by AMMEP was $Rb1{\rightarrow}Rd{\rightarrow}F2{\rightarrow}$ compound K. The optimum reaction conditions for compound K formation from ginsenoside Rb1 were as follows: reaction time 72-96 h, pH 4.0-4.5, and temperature $45-55^{\circ}C$. Conclusion: AMMEP can be used to produce the human intestinal bacterial metabolite, compound K, from ginsenoside Rb1 with a high yield and without food safety issues.
This study was carried out to know the possibility of simultaneous utilization of laccase from white-rot fungus with cellulase on enzymatic hydrolysis of cellulosic substrate from autohydrolyzed oak wood. Laccases from 3 white-rot fungi, Pleurotus ostreatus. Ganoderma lucidum, and Phanerochaete chrysosporium, were isolated, purified and measured their activities. The highest activity was shown in Pleurotus ostreatus and the lowest in Phanerochaete chrysosporium. Laccase from Pleurotus ostreatus has optimum pH of 5.94, Km value of 3.209 mM and appeared to be stable at relatively wide pH range, 4.7-8.72. Temperature stability showed that 60% activity was preserved after 40 minutes at $50^{\circ}C$. Laccase from Ganoderma lucidum reached to the maximum activity during 15-20 day incubation. This enzyme has optimum pH of 6.45, Km value of 6.71 mM and pH range of 5.0-9.0 for stabilization. 95% activity was preserved at $30^{\circ}C$ and 58% activity at $50^{\circ}C$. Concerned to the enzymatic hydrolysis of cellulosic substrate with both enzymes, cellulase and laccase, simultaneously, mixed culture filtrates and mycellium extracts were shown higher hydrolysis rates than those of Trichoderma viride. There were no significant differences in the extent of hydrolysis among various mixed culture filtrates and mycellium extracts.
본 연구는 농업 부산물인 옥수숫대를 이용하여 옥살산 전처리와 효소가수분해를 통한 에탄올 발효 효율 향상조건을 탐색하였다. 옥살산 전처리 옥수숫대의 효소가수분해는 Accellerase 1000을 이용하였으며, $50^{\circ}C$ 온도조건과 pH 4.5에서 96시간 가수분해하여 가장 높은 단당류 수율인 $64.8g/{\ell}$의 단당류 수율을 나타냈다. 옥수숫대에서 생산된 단당류의 발효에는 Pichia stipitis CBS 6054를 공시균주로 사용하였고, 전처리 옥수숫대 및 효소 투입량이 각각 10~14%와 15 FPU 이었을 때 효율적인 에탄올 생산에 가장 적합한 것으로 판명되었다. 이러한 조건에서 24시간 발효 후에 약 88.2%의 에탄올 전환율에 해당되는 0.45의 에탄올 수율을 얻었다.
분쇄마찰매체효소반응계에서 glucoamylase와 Alpha-amylase의 보완작용에 의한 옥수수 생전분의 효소당화 mechanism을 규명코자, 분리.정제된 상기 효소의 혼합 비율에 따른 당화양상 및 생성당 조성, 전분입자 구조, 생전분입자의 particle size 분포, X-ray 회절양샹의 변화 등을 관찰하였다. 분쇄마찰매체 효소반응계에서도 효소를 분리하여 사용하였을 때보다 glucoamylase와 $\alpha$-amylase를 혼합사용할 경우 당화율이 크게 향상되었으며, 고순도의 glucose 생산에 적합한 두 효소의 혼합비율은 약 1:1-1:2(unit) 로 판명되었다.
