Production and Characteristics of Cello- and Xylo-oligosaccharides by Enzymatic Hydrolysis of Buckwheat Hulls (메밀껍질의 효소분해에 의한 기능성 올리고당의 생산 및 특성)

  • This study was conducted to produce oligosaccharides from buckwheat hull by using commercial enzymes. Yields of oligosaccharides obtained by enzymatic hydrolysis of the cellulose and hemicellulose fractions were 132.37 and 393.04 g/kg, respectively. Xylose, glucose, fructose, xylobiose, xylotriose, cellobiose, and cellotriose were detected in the hydrolysate produced from buckwheat hull. Antioxidant activity of oligosaccharide from cellulose fraction (OSC) reduced with increasing hydrolysis time; however, the antioxidant activity of oligosaccharide from hemicellulose fraction (OSF) increased as the hydrolysis time was prolonged. OSF and OSC showed higher increase in viable counts compared to the control. As a result, oligosaccharides produced from buckwheat hull by enzymatic hydrolysis showed antioxidant activity and prebiotic effects. It is suggested that utilization of oligosaccharides produced from buckwheat hull as functional food materials may be improved when hydrolysis time and conditions are controlled for this purpose.

  • Lactic acid production from ${\alpha}$-cellulose by simultaneous saccharification and fermentation (SSF) was studied. The cellulose was converted in a batch SSF using cellulase enzyme Cytolase CL to produce glucose sugar and Lactobacillus delbrueckii to ferment the glucose to lactic acid. The effects of temperature, PH, yeast extract loading, and lactic acid inhibition were studied to determine the optimum conditions for the batch processing. Cellulose was converted efficiently to lactic acid, and enzymatic hydrolysis was the rate controlling step in the SSF. The highest conversion rate was obtained at 46$^{\circ}C$ and pH 5.0. The observed yield of lactic acid from ${\alpha}$-cellulose was 0.90 at 72 hours. The optimum pH of the SSF was coincident with that of enzymatic hydrolysis. The optimum temperature of the SSF was chosen as the highest temperature the microoraganism could withstand. The optimum yeast extract loading was found to be 2.5g/L. Lactic acid was observed to be inhibitory to the microorganisms' activity.

  • A novel pretreatment of rice straw has been developed to increase the reactivity of cellulose, in particular to increase the rate and extent of cellulose enzymatic hydrolysis. This technique is called ammonia-freeze-explosion method and relies on treatment of the lignocellulosic material with a volatile liquid under pressure followed by pressure release to evaporate the liquid and reduce the temperature by Bme E. Dale of Texas A & M University. Volatile liquids which also chemically explosion and swell lignocellulosic materials are particularly effective when used in this technique. Above four times of conversion of cellulose to glucose has been achived by enzymatic hydrolysis of rice straw with this method.

  • Tonin(Persicae Semen)is the herb medicine that cntains amygdalin as a major ingredient. Amygdalin in water is decomposed into benzaldehyde. HCN. and glucoseby emulsin. a hydrolysis enzyme intonin. A useful and practical method for the optimum extraction condition of amygdalin without enzymatic hydrolysis is required. The extraction yield of amygdalin of natural formula tonin was 0.1 % from crude powers. 1.4% from small pieces. 3.5% from half pieces and 2.4% from whole pieces. (omitted)

Recent Developments and Prospects in the Enzymatic Acylations (효소를 이용한 아실화 반응의 최근 동향과 전망)

  • Enzymatic acylations catalyzed by hydrolytic enzymes, along with enzymatic hydrolysis, are established reactions in the synthesis of fine chemicals such as chiral intermediates and polymerizations in the industry. Those reactions have been carried out mostly in organic media due to the thermodynamic limitations. Recently, there have been reports on enzymatic acylations in aqueous media. They have dealt with the elucidation of reaction mechanisms of hydrolases and acyl transferases based on their X-ray structures, homology comparison of the two kinds of enzymes, substrate engineering of acyl donors and computational design of acyl transferases for enzymatic acylations in aqueous media. Enzymatic acylations play an important role in the combinatorial synthesis of natural products such as polyketides and nonribosomal peptides. In this review, the historic developments of enzymatic acylations and industrial examples are described briefly. In addition, recent developments of enzymatic acylations in the modification of natural products and their prospects will be discussed.

Cholesterol Lowering Effect of Enzymatic Hydrolysates of Squid in Rats

  • This study evaluated effects of enzymatic hydrolysates of squid on cholesterol lowering in rats. Thirty male rats were blocked into 3 groups [high cholesterol diet (control), 5% normal squid, and 5% enzymatic hydrolysates of squid] and were raised for 10 weeks. Triglyceride level in enzymatic hydrolysates of squid-fed rats was lower than that in the control. Serum low density lipoprotein-cholesterol level followed in the order of control>normal squid>enzymatic hydrolysates. Serum high density lipoprotein-cholesterol level in enzymatic hydrolysates of squid-fed rats was higher than that in control rats. Liver cholesterol level in enzymatic hydrolysates of squid-fed rats was lower than that in control rats.

  • Optimum conditions for enzymatic hydrolysis of purified rapeseed(Brassica napus var. Youngsan) protein were investigated. Pronase showed higher activity in the hydrolysis of rapeseed protein than that of alcalase and neutrase. Preheated treatment of the rapeseed protein decreased the activity of pronase. The degree of hydrolysis of rapeseed protein was greater in distilled water than in phosphate buffer solution. Degree of hydrolysis was reached in steady state after 1 hr. Optimum conditions of the hydrolysis of the rapeseed protein were $40^{\circ}C$ in reaction temperaturem pH 8.0 in substrate solution, 1/100 (w/w) in the ratio of enzyme to substrate and 1% (w/v) in substrate concentration for pronase, respectively. At the optimum hydrolysis conditions, Km value was 3.48% (w/v).

  • Present experiment in vivo shows that some conversions of active groups in chloramphenicol residue of ester, that is hydrolytic cleavage of dichloracetamide and glucuronide formation, seem to take place prior to hydrolysis. This result suggest that the enzymatic hydrolysis rate in vitro, is not available as an index for the evaluation of the chlorampherical ester potency.

Effectiveness of Enzymatic Hydrolysis on Polyamide Fabric

  • We compared the effectiveness of amidase (amano acylase, AA) and an endopeptidase, (trypsin, TR) in modifying the hydrophobicity of polyamide fabric. We evaluated the number of amino groups released into the reaction mixture in order to optimize the treatment conditions. We found that a large number of amino groups were released into the reaction mixture due to the cleavage of amide bonds by AA hydrolysis; however, the TR hydrolysis exhibited a relatively lower activity compared to AA hydrolysis. In AA and TR hydrolysis, significant differences were observed in the K/S values and moisture regain. Amide bonds in polyamide fabric were hydrolyzed by AA hydrolysis effectively. Compared to TR, AA formed more hydrolysis product (amino groups) on the fabric surface. Thus, the hydrophobicity of polyamide fabric was modified using AA hydrolysis (as verified by the wettability test) without any deterioration of fiber strength.

Physicochemical Properties of Modified Rice Powder for Rice-Based Infant Foods - Thermal-enzymatic Treatment on Rice Powder (반고형 이유식의 개발을 위한 변형 쌀가루 제조 및 이화학적 특성 I - 열-효소 처리한 쌀가루)

  • The establishment of optimal producing condition for rice-based infant foods using modified rice powder was attempted. The modification of rice powder was prepared by microwave heating as well as partial enzymatic hydrolysis. Not only thermal treatment but also enzymatic hydrolysis on rice powder increased D.E. value from 1.25 to 3.81. The water binding capacity, swelling power, solubility, digestibility by ${\alpha}-amylase$, light transparency, and paste clarity of the modified rice powder compared to unmodified rice powder were increased from 107% to 249%, from 7.80 to 42.52, from 0.04% to 0.89ft, from 9.19% to 23.01, from 33% to 42%, and from 2.2% to 3.9%, respectively. On the other hand, gelatinization temperature, apparent viscosity, and degree of retrogradation of modified rice powder showed negative correlation with D.E. value. The 「results suggested that the thermal and enzymatic treatment on rice powder improved the physicochemical properties of rice based infant food by enhancing carbohydrate absorptionability and lowering the viscosity and opacity.
