functional Properties of Sesame Protein Concentrate as Degree of Hydrolysis by Enzyme Treatments (효소처리한 참깨박 농축단백질의 가수분해정도에 따른 기능성)

  • This study was carried out to investigate the effect of hydrolysis by proteolytic enzymes on the functional properties of sesame protein concentrate. Sesame protein concentrate was hydrolyzed with papain, pepsin and trypsin to obtain 10% and 20% degree of hydrolysis. The nirogen solubility in water was increased with increasing the degree of hydrolysis. Bulk density was increased by enzymatic hydrolysis but water absorption capacity was increased only in the case of pepsin-hydrolyzed SPC. Higher fat absorption capacity was found in SPC with 10% DH than SPC with 20% DH. Emulsifying activity was also increased by enzymatic hydrolysis except SPC with 10% DH by papain.

Biodegradation of Starch-Filled Acrylate Film by α-Amylase (전분 충전 아크릴레이트 필름의 α-Amylase에 의한 생분해)

  • The biodegradability of vinyl acetate acrylate resin and com starch blend was studied by determination of the reduced sugars produced after enzymatic hydrolysis. The starch hydrolysis reaction by $\alpha-amylase$ was achieved within 5 minutes. Optimal ranges of temperature and pH for the starch hydrolysis by $\alpha-amylase$ were around $80^{\circ}C$ and 6.5-7.2, respectively. The biodegradability of the starch-filled acrylate films increased as the content of starch increased. The biodegradation of starch in the starch-filled acrylate film by $\alpha-amylase$ was about 48.6% of that of pure starch. This value of biodegradable starch-filled acrylate film gave a good result with enzymatic shortcut test. The surface morphologies of the starch-filled acrylate film after enzymatic hydrolysis were investigated by scanning electron microscopy (SEM).

  • In this study, real-time and label-free methods for monitoring the enzymatic reaction of urease, which releases ammonia through the hydrolysis of urea in an aqueous solution, were developed using a liquid crystal (LC)-based pH sensor. Nematic liquid crystal 4-cyano-4'-pentylbiphenyl (5CB), doped with 4'-pentyl-biphenyl-4-carboxylic acid (PBA), exhibited a shift in optical appearance from bright to dark when it was in contact with ammonia generated from the enzymatic reaction between urease and urea. This optical change was attributed to the anchoring transitions of LCs caused by hydrophobic interactions between the tails of deprotonted PBA ($PBA^-$) molecules and the LCs at the aqueous/LC interface. This novel technique holds great promise for the sensitive detection of urease along with its substrates and inhibitors.

  • Since cellulose is the only organic material that is annually replenishable in very large quantities, we must explore ways to utilize it as a source of energy, food and chemicals. For the utilization of this resource, it is first enzymatic hydrolyzed to glucose, then the glucose can be used as a food, converted single cell protein by microorganism, fermented to clean burning fuel and other chemicals. Cellulolytic enzyme, cellulase, consists of two or three major components, $C_1-cellulase$, $C_x-cellulase$ and ${\beta}-glucosidase$. $C_x-cellulase$ are fairly common but $C_1-cellulase$ are quite rare. Trichoderma viride is the best source of active cellulose, especially $C_1-enzyme$. Saccharification rate of cellulose in greatly influenced by the degree of crystallinity and extent of lignification. But by the pretreatment the substrate with cellulose swelling agent, delignifying reagent and physical treatment, the degree of saccharification is enhanced. Thus, glucose syrups of 2 to 10% concentration are realized from milled newspaper. The enzymatic hydrolysis of such energy rich material, such as cellulose, to glucose is technically feasible and practically achievable on a very large scale.

  • Uridine diphosphate(UDP)-galactose-4-epimerase-less mutants of Salmonella pullorum were isolated after mutagenic treatment with ethidium bromide. When isolated gal E mutants of S. pullorum A2 and D1 were grown in the presence of galactose(0.1 W/V), they exhibited marked bacteriolysis in heart infusion broth. The mutant strains were further investigated the characteristics of enzymatic activities in the Leoloir galactose pathway. Isolated A2 and D1 strains were completely deficient in UDP-galactose-4-epimerase activity. And the activity of other enzymes involved in galactose metabolism were reduced significantly.

  • Pesticide waste and chemical stockpiles are posing a potential threat to both Vie environment and human health. There is currently a great effort toward developing effective and economical methods for the detoxification of these toxic organophosphates. In terms of safety and economy, enzymatic biodegradation has been recommended as the most promising tool to detoxify these toxic materials. To develop an enzymatic degradation method to detoxify such toxic organophosphorus compounds, a gene encoding organophosphorus acid anhydrolase (OPAA) from genomic DNA of Alteromonas haloplanktis C was subcloned and expressed. The enzyme consists of a single polypeptide chain with a molecular weight of 48 kDa. It demonstrates strong hydrolyzing activity on sarin, an acetylcholinesterase inhibitor. Moreover, its high activity is sustained for a considerable length of time. It is projected that the recombinant OPAA can be applied as an enzymatic tool that can be used not only for the detoxification of pesticide wastes, but also for the demilitarization of chemical stockpiles.

Antioxidant Effect of Enzymatic Hydrolyzate from a Kelp, Ecklonia cava

  • The potential antioxidative activity of water-soluble enzymatic hydrolyzates from a kelp, Ecklonia cava was evaluated by free radical scavenging and lipid peroxidation assays. To prepare water-soluble hydrolyzates from E. cava the seaweed was enzymatically hydrolyzed by five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase). Among all the hydrolyzates, Celluclast hydrolyzate effectively scavenged free radicals released from DPPH (1,1-diphenyl-2- pricrylhydrazyl) and recorded around 73% scavenging activity at the concentration of 4 mg ${\cdot}ml^{-1}$. This hydrolyzate was thermally stable and DPPH radical scavenging activity remained 80% or higher at heating temperatures of 40 and 60$^{\circ}C$ up to 12 h and around 80% at 100$^{\circ}C$ up to 8 h. AMG and Ultraflo hydrolyzate inhibited the lipid peroxidation of fish oil as that of $\alpha$-tocopherol. These results suggested that an enzymatic extraction will be an effective way for the production of a potential antioxidant from seaweeds.

Studies on the Enzymatic Properties of Rice Bran Lipase (미강 Lipase의 효소 화학적 성질에 대한 연구)

  • Some enzymatic properties of the lipase extracted from rice bran were studied. The rate of free fatty acid formation as a function of incubation time was maximum after 30 min. at $35^{\circ}C$. The activation energy of the enzyme was 1.83 Kcal/mol and using a olive oil emulsion as substrate, the Km value was $0.8{\times}10^{-3}M$. The enzyme activity was more stimulated by calcium ion. Application of these enzymatic properties onto the effective utilization of rice bran and preservation of rice were discussed.

Enzymatic Activities in Petroleum Wastewater Purification System by an Activated Sludge Process

  • The enzymology of an activated sludge system for a petroleum wastewater purification process was investigated. Leucine-aminopeptidase (L-AMP), ${\beta}$-glucosidase (${\beta}-GLC$), and lipase (LIP) were selected for the study. It was found that more than 81.7% of enzymatic activity was associated with microbial cells in the activated sludge floc. The metabolic response of a mixed microbial population to increased phenol concentration showed that L-AMP activity increased in the activated sludge, whereas activities of ${\beta}-GLC$ and LIP decreased, due to the inhibitory effect of the phenol which varied from 100 mg/l to 500 mg/l.

Comparison of enzymatic hydrolysis characteristics of mushroom culutured waste (MCW) and Cork oak by alkali treatment (알칼리 처리에 따른 폐골목 및 굴참나무의 효소당화 특성 비교)

  • The mushroom cultured waste(MCW) from cork oak was evaluated as the raw material for bioethanol production. For enzymatic hydrolysis, cellulase cocktails (Celluclast 1.5L and Novozym 188) was used for polysaccharides to monosaccharides conversion. Compared with sound cork oak woodmeal, woodmeal from MCW showed higher cellulose to glucose conversion. To improve polysaccharides to monosaccharides conversion, pretreatment by sodium hydroxide was applied. Even though more xylan and lignin were removed in woodmeal of MCW than that of cork oak, concentration of glucose was higher from sodium hydroxide treated cork oak woodmeal (51.3 g/L) than treated MCW woodmeal (41.6 g/L).
