  • In association with the international validation project to establish a test protocol for the "Enhanced OECD Test Guideline 407", we performed a 28-day repeated-dose toxicity study of vinclozolin (VCZ), an androgen antagonist, and ketoconazole (KCZ), a biosynthesis inhibitor of testosterone (T), and assessed the sensitivity of new parameters for detecting endocrine-related effects of endocrine-disrupting chemicals.(omitted)

Monitoring of Endocrine Disruptor-suspected Pesticide Residues in Greenhouse Soils and Evaluation of Their Leachability to Groundwater (시설재배 토양 중 내분비계장애 추정농약의 잔류 모니터링 및 지하수 용탈 가능성)

  • This study was carried out to survey the residual characteristics of endocrine disruptor (ED)-suspected pesticides in greenhouse soils and assess their leachabilites to groundwater. Greenhouse soils were collected from 40 sites of greenhouse in 2008 in Korea. Sixteen ED-suspected pesticides which had been using in Korea, such as alachlor, benomyl, carbaryl, cypermethrin, 2,4-D, dicofol, endosulfan, fenvalerate, malathion, mancozeb, metribuzin, metiram, methomyl, parathion, trifluralin, and vinclozolin, in the soils, were analyzed by chromatographic methods using GLC-ECD and HPLC-DAD/FLD. Limits of detection (LODs) of the test pesticides ranged from 0.0004 to 0.005 mg/kg. Recoveries of the target pesticides from soil ranged from 72.69 to 115.28%. Four pesticides including cypermethrin were detected in the range of from 0.001 to 2.019 mg/kg, representing that their detection rate from greenhouse soils was 37.5%. The highest detection rate was observed from endosulfan which was detected from 16 site soils of the total samples, indicating that endosulfan is persistent in soil because of its very low mobility and high adsorption characteristics in soil. Based on the groundwater ubiquity scores (GUSs) of the pesticides detected from greenhouse soils, most of them have little possibilities of groundwater contamination except the fungicide vinclozolin with some leaching potential because of high water solubility and very low soil adsorption property.

  • To evaluate the arsenic effect of fish endocrine disruption, crucian carp (Carassius auratus) were treated with 0.5 mg/L and 3.0 mg/L of As$_2$O$_3$ for 14 days and gonadal development was examined by histological analysis. In ovaries from female crucial carps exposed to 3.0 mg/L, immature follicles and atretic follicles were appeared while various normal developmental stages of oocytes were shown in control group. Body weight and Gonadosomatic Index (GSI) of treated carp were also decreased compared to control group. However, no other significant histological changes in liver or kidney were shown in this exposure scheme. This means that reproductive organs are more sensitive than other organs and arsenic may exert endocrine disrupting effect through inhibiting the development of reproductive organ in fish.

Leaching Characteristics of the Endocrine Disruptor-suspected Pesticides in Upland Soil (내분비계장애추정농약의 밭토양 중 용탈 특성)

  • This experiment was carried out to estimate leaching potential of thirteen endocrine disruptor-suspected pesticides in upland soils using soil columns (5 cm I.D. ${\times}$ 35 cm H.) packed with soil A (sandy loam) and soil B (loam). When 12.6 mL of water, average precipitation in Cheongju area during the period from June to August, 2001-2010, was percolated through soil column packed with soil A every day for 21 days, no pesticides were detected from leachate, with the exception of metribuzin which was detected with negligible. Also, when 2 L of water was percolated consecutively five times through soil columns packed with soil A and B, irrespective of soil types, cypermethrin, endosulfan, fenvalerate, parathion and trifluralin, which were very low water solubilities and high soil $K_{oc}s$, were not detected from leachate and were distributed mostly in the depth of 0-5 cm, representing that water solubility and soil $K_{oc}$ are major contributing factors to their leaching behavior. Despite high average leaching rates in carbaryl and methomyl, actual possibilities of ground water contamination in the agricultural environment by them would be very low, considering that the negligible amount of pesticide was percolated through a lysimeter with an undisturbed soil core simulating the field conditions, while most of pesticide was percolated through a soil column with the disturbed soil profile.

  • 1,2-Dibromo-3-chloropropane (DBCP), a soil fumigant against nematodes, is a genotoxic carcinogen and also is classified by World Wildlife Fund as endocrine disruptors. DBCP has been extensively studied on genotoxicity, carcinogenicity, and damage in male reproductive-related organs. However, information on precise mechanism of mutagenesis and carcinogenesis of DBCP is yet unknown. (omitted)

  • Nonylphenol (NP) is a widespread environmental pollutant that has been shown to exert both toxic and estrogenic effects on mammalian cells. As the effects of NP on the reproductive system of adult male vertebrates are virtually unknown, we investigated not only the changes of reproductive hormone secretion in serum after chronic exposure to NP but also, in order to identify the site of its action, the reproductive hormone secretion in serum 48 hours after microinfusion of NP within hypothalamic preoptic area (POA). In the chronic exposure, the luteinizing hormone (LH), follicle stimulating hormone (FSH), and testosterone in serum were decreased but prolactin (PRL) concentrations were increased. The LH, FSH, and testosterone in serum were decreased through the direct infusion of NP into POA, while there was no difference in mean serum prolactin between NP and control groups. These observations suggest that NP as endocrine disruptor has modulatory effects on hypothalamo-pituitary-gonadal axis and that the site of action of NP could be hypothalamic POA.

  • Sexual behavior and reproductivity of male fIsh were studied as an in vivo screening method of endocrine disruptors. Male medaka (Oryzias latipes) were exposed to 17$\beta$-estradiol at nominal concentrations of 2 and 20 $\mu\textrm{g}$/l for 14 days. After exposure of the chemical, sexual behavior between male medaka and normal female which were injected with prostaglandin $F_{2\alpha}$ just before the test, was analysed by using video camera for one hour. Normal control male showed courtship dancing such as following, guarding, dancing and crossing while 17$\beta$-estradiol treated male did not show any type oj courtship dancing. Furthermore, fecundity and fertility were significantly decreased in the treated group. It was suggested that analysis of sexual behavior could be a useful endpoint for the screening of the endocrine disruptors.

  • We were carried out to monitor pesticide residues in herbal medicine. In Korea Pharmacy, these are no critics for pesticides except 5 kinds of organic chlorines (BHC, DDT, Aldrin, Endrin, Dieldrin) and heavy metals. We analysed 53 kinds of pesticides consisted of 14 kinds of organochlorines, 20 kinds of organophosphoruses, and 19 kinds of pesticides estimated endocrine disruptor on 373 samples consisted with 30kinds of herbal medicine. In this study, Domestic, Chinese and Japanese herbal medicines were tested with GC/ECD, GC/NPD and then confirmed with GC/MSD. Recoveries were 75$\sim$l10% in ECD detector and 76$\sim$97% in NPD detector. Detection limits were 0.004$\sim$0.064ppm in ECD detector and 0.006$\sim$0.094ppm in NPD detector. Organochlorines and others were not detected in samples.
