일부 자원 여성에서 모발염색 후 림프구의 DNA손상과 관련 요인 (DNA Damage in Lymphocytes after Hair Dyeing and Related Factors among Women Volunteers)

  • Objectives : To evaluate the DNA damage by hair dyeing in human lymphocytes Methods : Comet assays were carried out to evaluate the DNA damage in lymphocytes by hair dyeing. Twenty subjects were selected from women volunteers whose age ranged from 55 to 67 year old. All subjects had no smoking history. Blood samples were collected before and 6 hours after hair dyeing. DNA damage was evaluated by means of the tail moments, which were quantified by a KOMET 4.0 image analysis system. Results : The tail moments before hair dyeing showed no significant differences among subjects except for the high frequency group. The mean values of the tail moments in subjects with low and high frequencies of hair dyeing were 1.39 and 1.77, respectively (p<0.05). The tail moments after hair dyeing increased significantly, The mean values of tail moments in subjects before and after hair dyeing were 1.45 and 1.79, respectively (p<0.01). However, the difference levels of DNA damage in lymphocytes before and after hair dyeing were found to be slightly lower in both the dietary supplement taking group and high frequency group. Conclusions : The high frequency group appears to have a higher level of DNA damage than the low frequency group before hair dyeing. DNA damage in lymphocytes was found to be significantly higher in the volunteers after hair dyeing. In this study, the related factors such as high frequency and taking dietary supplements appeard to reduce DNA damage in lymphocytes after hair dyeing.

  • DNA damage such as genotoxicity was identified with comet assay, which blood cell of a marine parrot fish (Oplegnathus fasciatus) was exposed to an acidified seawater, lowered pH gradient making of $CO_2$ gas. The gradient of pH were 8.22, 8.03, 7.81, 7.55 with control as HBSS solution with pH 7.4. DNA tail moment of fish blood cell was $0.548{\pm}0.071$ exposed seawater of pH 8.22 condition, on the other hand, DNA tail moment $1.601{\pm}0.197$ exposed acidified seawater of pH 7.55 lowest condition. The approximate difference with level of DNA damage was 2.9 times between highest and lowest of pH. DNA damage with decreasing pH was significantly increased with DNA tail moment on blood cell of marine fish (ANOVA, p < 0.001). Ocean acidification, especially inducing the leakage of sequestered $CO_2$ in geological structure is a consequence from the burning of fossil fuels, and long term effects on marine habitats and organisms are not fully investigated. The physiological effects on adult fish species are even less known. This result shown that the potential of dissolved $CO_2$ in seawater was revealed to induce the toxic effect on genotoxicity such as DNA breakage.

A novel computer vision-based vibration measurement and coarse-to-fine damage assessment method for truss bridges

  • To assess structural condition in a non-destructive manner, computer vision-based structural health monitoring (SHM) has become a focus. Compared to traditional contact-type sensors, the advantages of computer vision-based measurement systems include lower installation costs and broader measurement areas. In this study, we propose a novel computer vision-based vibration measurement and coarse-to-fine damage assessment method for truss bridges. First, a deep learning model FairMOT is introduced to track the regions of interest (ROIs) that include joints to enhance the automation performance compared with traditional target tracking algorithms. To calculate the displacement of the tracked ROIs accurately, a normalized cross-correlation method is adopted to fine-tune the offset, while the Harris corner matching is utilized to correct the vibration displacement errors caused by the non-parallel between the truss plane and the image plane. Then, based on the advantages of the stochastic damage locating vector (SDLV) and Bayesian inference-based stochastic model updating (BI-SMU), they are combined to achieve the coarse-to-fine localization of the truss bridge's damaged elements. Finally, the severity quantification of the damaged components is performed by the BI-SMU. The experiment results show that the proposed method can accurately recognize the vibration displacement and evaluate the structural damage.

DNA damage in T- and B-lymphocytes of rats exposed to benzene

  • Single cell gel electrophoresis assay was carried out to evaluate DNA damage in T-and B-lymphocytes from rats exposed to benzene and the correlation between DNA damage and the level of t,t-muconic acids, which are urinary benzene metabolites, was investigated. In control rats, the mean values of Olive tail moments in T-and B-lymphocytes were 1.507$\pm$0.187 and 1.579$\pm$0.206 respectively. DNA damages of T-lymphocytes in rats exposed for 4 weeks showed the highest Olive tail moments at each benzene concentration examined (2.72-4.351). However this DNA damage was decreased after 6 weeks of exposure (1.74-2.09). DNA damages of B-lymphocytes did not show such differences with exposure time or benzene concentration (1.49-2.07) except at 200 ppm at 4 weeks. T-lymphocytes show significantly more damages than B-lymphocyte upon acute exposure to benzene.

Geno- and Ecotoxicity Evaluation of Silver Nanoparticles in Freshwater Crustacean Daphnia magna

  • Genotoxic- and ecotoxic assessments of silver nanoparticles (AgNPs) were conducted on the freshwater crustacean Daphnia magna. AgNPs may have genotoxic effects on D. magna, given that the DNA strand breaks increased when exposed to this nanoparticle. Increased mortality was concomitantly observed with DNA damage in the AgNPs-exposed D. magna, which suggests AgNPs-induced DNA damage might provoke higher-level consequences. The results of the comparative toxicities of AgNPs and Ag ions suggest that AgNPs are slightly more toxic than Ag ions. Overall, these results suggest that AgNPs may be genotoxic toward D. magna, which may contribute to the knowledge relating to the aquatic toxicity of AgNPs on aquatic ecosystems, for which little data are available.

Adaptive Responses of Escherichia coli for Oxidative and Protein Damage Using Bioluminescence Reporters

  • The recombinant bioluminescent Escherichia coli strains, DPD2511 and TV 1061 containing the katG and grpE promoters, respectively, from Vibrio fischeri fused to luxCDABE, were used to detect the adaptive and repair responses to oxidative damage caused by hydrogen peroxide $(H_2O_2)$, and protein damage due to phenol. The response ratio, represented as the bioluminescence induced in subsequent inductions of DPD2511 and TV1061 with the mother cells previously induced by each chemical, i.e., $H_2O_2$ and phenol during the previous induction stage, decreased suddenly compared with the ratio of the control culture of each strain, meaning there is a possible adaptive response to stress caused by chemicals. Protein damage due to phenol was completely repaired by the second culturing after the initial induction, as was oxidative damage caused by $H_2O_2$ which was also rapidly repaired, as detected by the recovery of bioluminescence level. This result suggests that E. coli promptly adapt and repair oxidative and protein damage by $H_2O_2$ and phenol completely.

  • Based on the reconnaissance of buildings in Dujiangyan City during 2008 Wenchuan earthquake, China, structural damage characteristics and the spatial distribution of structural damage are investigated, and the possible reasons for the extraordinary features are discussed with consideration of the influence of urban historical evolution and spatial variation of earthquake motions. Firstly, the urban plan and typical characteristics of structural seismic damage are briefly presented and summarized. Spatial distribution of structural damage is then comparatively analyzed by classifying all surveyed buildings in accordance with different construction age, considering the influence of seismic design code on urban buildings. Finally, the influences of evolution of seismic design code, topographic condition, local site and distance from fault rupture on spatial distribution of structural damage are comprehensively discussed. It is concluded that spatial variation of earthquake motions, resulting from topography, local site effect and fault rupture, are very important factor leading to the extraordinary spatial distribution of building damage except the evolution of seismic design codes. It is necessary that the spatial distribution of earthquake motions should be considered in seismic design of structures located in complicated topography area and near active faults.

Acceleration-based neural networks algorithm for damage detection in structures

  • In this study, a real-time damage detection method using output-only acceleration signals and artificial neural networks (ANN) is developed to monitor the occurrence of damage and the location of damage in structures. A theoretical approach of an ANN algorithm that uses acceleration signals to detect changes in structural parameters in real-time is newly designed. Cross-covariance functions of two acceleration responses measured before and after damage at two different sensor locations are selected as the features representing the structural conditions. By means of the acceleration features, multiple neural networks are trained for a series of potential loading patterns and damage scenarios of the target structure for which its actual loading history and structural conditions are unknown. The feasibility of the proposed method is evaluated using a numerical beam model under the effect of model uncertainty due to the variability of impulse excitation patterns used for training neural networks. The practicality of the method is also evaluated from laboratory-model tests on free-free beams for which acceleration responses were measured for several damage cases.

Improvement of MFL sensing-based damage detection and quantification for steel bar NDE

  • A magnetic flux leakage (MFL) method was applied to detect and quantify defects in a steel bar. A multi-channel MFL sensor head was fabricated using Hall sensors and magnetization yokes with permanent magnets. The MFL sensor head scanned a damaged specimen with five levels of defects to measure the magnetic flux density. A series of signal processing procedures, including an enveloping process based on the Hilbert transform, was performed to clarify the flux leakage signal. The objective damage detection of the enveloped signals was then analyzed by comparing them to a threshold value. To quantitatively analyze the MFL signal according to the damage level, five kinds of damage indices based on the relationship between the enveloped MFL signal and the threshold value were applied. Using the proposed damage indices and the general damage index for the MFL method, the detected MFL signals were quantified and analyzed relative to the magnitude of the damage increase.

Effects of tendon damage on static and dynamic behavior of CFTA girder

  • Experimental studies and finite element analyses have been carried out to establish the effect of tendon damage on the structural behavior of concrete filled tubular tied arch girder (CFTA girder). The damage of tendon is considered in different stages by varying the number of damaged cables in the tendon. Static and dynamic structural parameters are observed at each stage. The results obtained from the experiments and numerical studies have been compared to validate the studies. The tendons whose damage can significantly affect the stiffness of the CFTA girder are identified by performing the sensitivity analysis. The locations in the girder which are sensitive to the tendon damage are also identified.
