HPLC-UVD를 이용하여 diflubenzuron이 다종농약다성분 분석법 제2법에 적합한지를 검증하고자 본 연구를 수행하였다. 유효성을 검증하기 위하여 농약이 검출되지 않은 시금치, 엇갈이배추, 가지, 애호박, 청피망, 오이, 참외 등 7종 농산물을 대상으로 직선성, 특이성, 검출한계, 정량한계, 정확성, 정밀성을 확인하였다. Diflubenzuron 표준용액을 0.05-5 mg/kg 농도로 희석하여 분석한 표준검량선의 직선성은($R^2=0.99999$)은 매우 만족할 만한 수준이었다. 검출한계는 0.008 mg/kg, 정량한계는 0.02 mg/kg 이며 회수율은 77.5-105.6%, 상대표준편차는 최대 20% 미만, 정밀도는 intra-day test에서 0.4-1.9%, inter-day test에서 0.7-1.9%를 나타내어 잔류농약분석법에서 허용하는 회수율 기준 70-120% 범위와 상대표준편차 20% 이하의 기준에 적합하였다. LC-MSD를 이용한 재확인 과정을 통해 잔류결과의 신뢰성을 확보하였다. 따라서 유통농산물 중 diflubenzuron의 일상적 검사법으로 빠르고 간편한 다종농약다성분 제2법을 적용할 수 있을 것으로 판단된다.
Background: With volatile organic compounds (VOCs) containing aromatic and halogenated hydrocarbons such as benzene, toluene, and xylene that can adversely affect the respiratory and cardiovascular systems when a certain concentration is reached, it is important to accurately evaluate the source and the corresponding health risk effects. Objectives: The purpose of this study is to provide scientific evidence for the city of Seoul's VOC reduction measures by confirming the risk of each VOC emission source. Methods: In 2020, 56 VOCs were measured and analyzed at one-hour intervals using an online flame ionization detector system (GC-FID) at two measuring stations in Seoul (Gangseo: GS, Bukhansan: BHS). The dominant emission source was identified using the Positive Matrix Factorization (PMF) model, and health risk assessment was performed on the main components of VOCs related to the emission source. Results: Gasoline vapor and vehicle combustion gas are the main sources of emissions in GS, a residential area in the city center, and the main sources are solvent usage and aged VOCs in BHS, a greenbelt area. The risk index ranged from 0.01 to 0.02, which is lower than the standard of 1 for both GS and BHS, and was an acceptable level of 5.71×10-7 to 2.58×10-6 for carcinogenic risk. Conclusions: In order to reduce the level of carcinogenic risk to an acceptable safe level, it is necessary to improve and reduce the emission sources of vehicle combustion and solvent usage, and eco-car policies are judged to contribute to the reduction of combustion gas as well as providing a response to climate change.
Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.
Recently, it has been a big issue to distinguish the dried roots of Cynanchum wilfordii and C. auriculatum in health functional food market. The original plant species of Cynanchi Wilfordii Radix belong to the Asclepiadaceae family is differentially described in the national pharmacopoeia of Korea, China and Japan. Owing to the morphological similarities of the dried roots of this plant to those of C. auriculatum, which is often misidentified in Korean herbal medicine marketplace, distinguishing these two species is exceedingly difficult. The purpose of this study was to compare the conventional-PCR with the real-time PCR for detection of C. wilfordii and C. auriculatum DNA. We also tried to realize a quantitative real-time PCR assay using species-specific matK primers, which allowed us to estimate the ratio of C. willfordii and C. auriculatum using varying ratios of mixed genomic DNA template from the two species. The differentiation of intentional and unintentional mixture in this study would be applied to food safety management and can be helpful for protection of consumer's right and cultivators.
Objectives: A great number of people visit forests for their bountiful healing factors. We investigated the quantity of terpene and analyzed the correlations with meteorological and environmental factors at Mt. Mudeung in order to support public health. Methods: The terpene amounts were investigated along 11 main trails using stainless steel tube packed by Tenax TA (150 mg) and Carbopack B (130 mg) during March to November 2012. Terpene amounts of 20 species (${\alpha}$-pinene, camphene, etc.), and meteorological and environmental factors were investigated in the field. Results: Terpene of 16 species was released from the forest and total terpene amounts were 2,080 pptv at the site of Chamaecyparis obtusa, the highest among 11 sites, nearby the first reservoir on Mt. Mudeung. Terpene concentrations in the forest were nine to 23 times higher than found in urban areas. Total terpene amounts had positive correlations with temperature, humidity, carbon dioxide and oxygen (p<0.01) with $R^2$ of 0.345, 0.369, 0.591, 0.145, respectively, from April to July. Wind speed and solar radiation in the forest had a negative correlation with terpene amounts and showed statistical insignificance with p-values of 0.118 and 0.233, respectively. Conclusions: This study suggests that the amounts of terpene around Mt. Mudeung are indeed higher, so visitors may enjoy a therapeutic walk in the forest with a healing effect. These results showed the forest was very effective for improving human health.
Objectives: The genus Legionella is common in aquatic environments. Some species of Legionella are recognized as potential opportunistic pathogens for human, notably Legionella pneumophila that causes, Legionellosis. Thus, we investigated the contamination of Legionella pneumophila on water supply systems in Seoul, including cooling towers, public baths, hospitals and fountains. Methods: The existence of 16S rRNA and mip gene of L. pneumophila was confirmed in the genome of the isolated strains by PCR. Results: During the summer season of 2010 and 2011, Legionella pneumophila were detected from 163 samples (21.1%) out of 772 samples collected. Among the 163 strains of L. pneumophila, eighty one isolates belonged to serogroup 1 (57.4%), 23 isolates were serogroup 5 (16.3%), 21 isolates were serogroup 6 (14.9%), 8 isolates were serogroup 2 (5.79%), and 8 isolates were identified in serogroup 3 (5.7%). Through PFGE (pulsed-field gel electrophoresis) analysis using Sfi I, genetic types of L. pneumophila were classified into five (A to E) patterns by the band similarity with excess of 70% from public baths. Conclusions: The PFGE patterns of the serotypes showed a tendency for diversity of L. pneumophila. Our results suggest the existence of serological and genetic diversity among the L. pneumophila isolates.
Eggs of parasite were found 73 of 1,459 in sand samples from public parks and playgrounds in Incheon for 5 years (from 2004 to 2008). Of the 1,459 sand samples examined, 17.8% of samples were found to be positive ova of Toxocara canis, 11.0% for Toxocaris leonina, 5.5% for Toxocara cati, 2.7% Ancylostoma caninum and 1.4% for Strongyloides spp. It was found mixed infestation with Toxocara canis and Toxocara cati 60.2%.
A total of 1,354 samples was collected from bovine and porcine carcass from January 2005 to December 2006 in a slaughter house. Twenty five strains(1.8%) of Listeria monocytogenes were isolated from 1,354 samples using selective media. Ten(1.4%) L monocytogenes were isolated from the 677 of bovine carcasses, and 15(2.2%) were isolated from the 677 of porcine carcasses. Among 15 L mono-cytogenes from porcine, 11 siolates were serovars 1/2c, followed by 1/2b (3 strains, 20.0%) and 1/2a(1 strain) Out of 10 bovine samples, positive cases in 1/2a were 9 strains (90.0%), 1/2b were 1 strains(10.0%). PCR primers were selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene (hlyA) of L mono-cytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested. A total of 25 L monocytogenes strains were analysed by PFGE after digestion with Apa I. PFGE analysis of genomic DNA showed the $14{\sim}18$ fragments ranging in size from 30 to 550 kb, resulting in 14 patterns.
LC-MS/MS를 이용하여 신속하고 정밀한 카벤다짐 분석법을 연구하였다. 농산물 중 카벤다짐은 acetonitrile로 추출한 후 SPE cartridge을 이용하여 전처리를 수행하였으며, 최소 검출한계 및 정량한계는 각각 0.001과 0.004 mg/kg이었다. 농산물에 각각 1.0 mg/kg 및 0.02 mg/kg 농도의 카벤다짐을 첨가하여 분석 시 회수율과 정밀성은 각각 상추가 84.9%, 1.0% 및 87.8%, 3.8% 얼갈이배추가 83.4%, 1.5% 및 86.8%, 2.7% 양배추가 86.4%, 2.0% 및 77.3%, 1.5% 근대가 83.3%, 0.2% 및 86.8%, 2.0 % 이었으며, 콩나물은 85.9%, 3.0% 및 90.1%, 1.3%, 그리고 숙주나물이 83.3%, 2.0% 및 85.9%, 1.9% 로 조사되었다. 본 연구의 시험법은 식품공전의 다성분분석법에 비해 신속하고 정밀한 분석법으로 농산물에 대한 카벤다짐 분석에 적합한 것으로 조사되었다.
This study investigated the mycotoxins (aflatoxin B1, B2, G1, G2, fumonisin B1, B2, ochratoxin A and zearalenone) contained in edible and medicinal plants in Seoul Yangnyeong market during 2020-2021. We analyzed contamination of mycotoxins using LC-MS/MS and evaluated risk assessment. The method was validated by assessing matrix effects, linearity, limit of detection (LOD), limit of quantification(LOQ) and recovery. Matrix-matched standard calibration was used for calibration curves showed good linearity (r2>0.999). The LOD, LOQ and recovery were 0.01-0.23 ㎍/kg, 0.04-0.71 ㎍/kg and 75.5-117.9% respectively. Mycotoxins were detected in 22 of 171 samples; aflatoxin B1 (6.66 ㎍/kg), fumonisin (7.54-64.68 ㎍/kg), ochratoxin A (4.21-10.56 ㎍/kg) and zearalenone (7.31-60.76 ㎍/kg). In the risk assessment, the MOE (Margine of Exposure) of aflatoxin B1 and ochratoxin A were in the range of 1.48×103-2.36×105. No items exceeded 100% in %TDI (Tolerable Daily Intake) of fumonisin (B1+B2) and zearalenone.
