The Analysis of Amplitude and Phase Image for Acoustic Microscope Using Quadrature Technique (쿼드러춰 방식에 의한 초음파현미경의 진폭과 위상영상 분석)

  • In this study, we have constructed the acoustic microscope using quadrature technique and analyzed the relative variation of image intensity and the quality of image by reconstructing the amplitude and phase image for surface defects with tiny hight variation. In this experiment, we have constructed the scanning acoustic microscope using the focused transducer with 3㎒ center frequency and the quadrature detector. And we have fabricated aluminum samples with round defects whose depth is different and reconstructed the amplitude and phase images for the samples. One sample has round defects with 2㎜ diameter and 100㎛ depth and the other has round defects with 4㎜ diameter and 5㎜ depth. In the result of line scanning for the sample with 100㎛ round defects, it has been shown that the variation rate of amplitude image intensity is 7% and the variation rate of phase image intensity is 89%. The phase image has better contrast than amplitude image for the sample. In contrast to this, the amplitude image has better contrast than phase image for the sample with 5㎜ depth's defects. Accordingly there is big difference between amplitude image and phase image for depth variation of defects whose boundary is 1 wavelength. Consequently the acoustic microscope using quadrature detector can be evaluated efficiently more than using envelope detector, for detecting defects which have height variation less than 1 wavelength. And also the phase image and the amplitude image can be used for detecting defects of tiny height variation with complimentary relation.

A Comprehensive Study on the Intake Patterns and Expenditures on Ramyun among Adults in Metropolitan Areas of Korea (수도권 거주 성인의 라면류 섭취 현황 및 실태에 관한 연구)

  • The purpose of this study is to identify instant noodle intake and consumption patterns of adults in metropolitan areas of Korea. There was a total of 702 subjects, 18 years of age and above, and data from the self-administrated questionnaires were collected and analyzed using comparative statistical analysis, including frequency, chi-square, t-test, and one-way ANOVA. According to the results, preference level for instant noodles among the adults was indicated to be $4.84{\pm}1.32$ and 54% of subjects either "liked" or "more than liked" noodles. People in 20s liked them the most. Noodles in envelope type package with soup are highly preferred. The most common reason why they chose to eat instant noodles was "convenience." At the same time, people avoid them because of their bad nutrition and calories. Half of the subjects consume instant noodles at least once a week mainly for lunch and dinner as a substitute for their meal. In the case of eating-together-with-family was the largest. 72.4% of subjects consumed entire soup additive content and 20.6% of them eat almost without soup because of being full and worrying that it might be harmful to health. Significant difference was shown in the preference level and intake patterns depending on respondents' general characteristics. Limitations and future research directions are also discussed.

Estimation to the Strength of Basalt in Jeju Island according to Rock Failure Criterions (암석의 파괴규준에 따른 제주도 현무암의 강도 산정)

  • In this study, a series of triaxial tests on Jeju basalt were carried out and then rock strength parameters were estimated by the Mohr-Coulomb failure criterion and the Hoek-Brown failure criterion using the test results. The characteristics of both failure criterions were investigated through comparing the estimated rock strength parameters. As the result of the Mohr-Coulomb criterion, the cohesions and the internal friction angles are determined as 5.35 MPa and $50.25^{\circ}$ of Pyoseonri basalt, 16.99 MPa and $60.66^{\circ}$ of Trachy-basalt, and 2.33 MPa and $37.05^{\circ}$ of Scoria, respectively. The cohesions and internal friction angles were estimated by the Hoek-Brown failure criterion in the basis of the results of regression analysis. The cohesions and the internal friction angles are determined as 4.77 MPa and $52.47^{\circ}$ of Pyoseonri basalt, 14.69 MPa and $60.70^{\circ}$ of Trachy-basalt, and 2.22 MPa and $47.60^{\circ}$ of Scoria, respectively. As the result of comparison between the Mohr-Coulomb failure criterion and the failure envelope predicted by the Hoek-Brown criterion, the cohesion estimated by the Hoek-Brown criterion is usually lower than that obtained from the Mohr-Coulomb criterion, whereas the friction angle estimated by the Hoek-Brown criterion is higher than that obtained from the Mohr-Coulomb criterion.

Study on Persistent Infection of Japanese Encephalitis Virus Beijing-l Strain in Serum-free Sf9 Cell Cultures

  • Sf9 cells have obvious advantages for the conventional production technology of vaccine. They are useful tools for high concentration and large-scale cultures. Sf9 cells were grown to maximal concentration, 8${\times}$l0$\^$6/ cells/$m\ell$ in a 500$m\ell$ spinner flask, with a doubling time at the exponentially growing phase of 24.5 hours, using serum-free media. To explore the ability of Sf9 cells to be infected by the Japanese encephalitis (JE) virus Beijing-l strain, Sf9 cells were infected with the virus. By 4-5 days post-infection, 10-15 % of the Sf9 cells showed cytopathic effect (CPE), from granularity to the formation of syncytia and multinucleated giant cells continuously observed over a period of 35 days. Positive fluorescent reactions were detected in 30-40% of cells infected with the JE virus Beijing-l strain, and the uninfected Sf9 cells were completely negative. Virus particles, propagated in Sf9 and Vero cells, were concentrated by sedimentation on 40% trehalose cushions by ultracentrifugation, and showed identical patterns of viral morphogenesis. Complete virus particles, 40 to 50 nm in diameter, were observed, and JE virus envelope (E) proteins, at 53 kDa, were found in the western blot analysis to the anti-JE virus E protein monoclonal antibody and reacted as a magenta band in the same position to the glycoprotein staining. To evaluate whether the infectious virus was produced in Sf9 cells inoculated with the JE virus Beijing-l stain, Sf9 cells were inoculated with the virus, and sample harvested every 5 days. The titers of the JE virus Beijing-l strain rose from 1.0${\times}$l0$\^$5/ to 1.5${\times}$l0$\^$6/ pfu/$m\ell$. The infected Sf9 cells could be subcultured in serum-free medium, with no change in the plaque sizes formed by the JE virus Beijing-l strain in the plaque assay. It is suggested that the ability of the JE virus Beijing-l strain to infect Sf9 cells in serum-free media will provide a useful insect cell system, where the JE virus replication, cytopathogenicity and vaccine immunogen can be studied.

Building Energy Savings due to Incorporated Daylight-Glazing Systems (통합 채광시스템의 건물 냉난방 에너지 성능평가)

  • The quantity of light available for a space can be translated in term of the amount of energy savings through a process of a building energy simulation. To get significant energy savings in general illumination, the electric lighting system must be incorporated with a daylight - activated dimmer control. A prototype configuration of an once interior has been established and the integration between the building envelope and lighting and HVAC systems is evaluated based on computer modeling of a lighting control facility. First of all, an energy-efficient luminaire system is designed and the lighting analysis program, Lumen-Micro 2000 predicts the optimal layout of a conventional fluorescent lighting future to meet the designed lighting level and calculates unit power density, which translates the demanded met of electric lighting energy. A dimming control system integrated with the contribution of daylighting has been applied to the operating of the artificial lighting. Annual cooling load due to lighting and the projecting saving amount of cooling load due to daylighting under overcast diffuse sky m evaluated by computer software ENER-Win. In brief, the results from building energy simulation with measured daylight illumination levels and the performance of lighting control system indicate that daylighting can save over 70 percent of the required energy for general illumination in the perimeter zones through the year A 25[%] of electric energy for cooling and almost all off heating energy may be saved by dimming and turning off the luminaires in the perimeter zones.

Shear behavior at the interface between particle and non-crushing surface by using PFC (PFC를 이용한 입자와 비파쇄 평면과의 접촉면에서의 전단 거동)

  • The shear behavior at the particle/surface interface such as rock joint can determine the mechanical behavior of whole structure. Therefore, a fundamental understanding of the mechanisms governing its behavior and accurately estimation of the interface strength is essential. In this paper, PFC, a numerical analysis program of discrete element method was used to investigate the effects of the surface roughness on interface strength. The surface roughness was characterized by smooth, intermediate, and rough surface, respectively. In order to investigate the effects of particle shape and crushing on particle/surface interface behavior, one ball, clump, and cluster models were created and their results were compared. The shape of particle was characterized by circle, triangle, square, and rectangle, respectively. The results showed that as the surface roughness increases, interface strength and friction angle increase and the void ratio increases. The one ball model with smooth surface shows lower interface strength and friction angle than the clump model with irregular surface. In addition, a cluster model has less interface strength and friction angle than the clump model. The failure envelope of the cluster model shows non-linear characteristic. From these findings, it is verified that the surface roughness and particle shape effect on the particle/surface interface shear behavior.

Vibration Analysis of SAR Antenna Reflectors During Satellite Maneuver (위성 기동 시 SAR 안테나 반사판에 발생하는 진동 분석)

  • Recently, there has been an increasing demand for SAR satellite as it can be operated regardless of the weather condition. In general, main reflector of the SAR is formed of multiple deployable panels to increase performance in the constrained payload envelope. By nature, deployable structure lacks structural stiffness and it is vulnerable to external disturbances and excitation. In particular, SAR satellites may have high levels of vibration occurring at the antenna reflecting surface due to higher angular rate requirements. During image capturing it is important to keep high surface accuracy of the reflector for the quality of images. In this research, a performance degradation of deployable SAR antenna due to structural deformation is analyzed. Panels for main reflectors are assumed to be flexible structures and multi-body simulation environment is established. Then, deflection of the panel is calculated while the satellite performs maneuvers. In addition, antenna gain and beam pointing error are analyzed to determine how these deflections affect antenna performance and mission.

The study of nondestructive evaluation method of paper records materials by NIR spectroscopy (근적외선 분광분석을 이용한 종이기록물의 비파괴 특성평가 연구)

  • Near Infrared Spectroscopy (NIRs) has been applied for rapid and nondestructive paper measurement by replacing the current destructive method to the property of paper. Current standard methods for the property of paper were pH, moisture, breaking length, and folding endurance, which data were compared with spectrum of FT-NIR spectrometer. Various paper products such as copy, envelope, white, newspaper, as well as old paper produced around 1960~1980 were used as the sample. The correlation ($R^2$) and standard error of prediction (SEP) results for breaking length, folding endurance, moisture and pH are $R^2$=0.914, SEP=0.508, $R^2$=0.926, SEP=0.281, $R^2$=0.941, SEP=0.931, pH $R^2$=0.949, SEP= -0.0631, respectively. This result show that NIRs can be applied to practical application for nondestructive analysis of paper records materials.

PERIOD CHANGES OF W UMa TYPE CONTACT BINARY SS ARIETIS (W UMa형 접촉쌍성 SS ARIETIS의 공전주기 변화)

  • The BVR CCD photometric observations of W UMa-type eclipsing binary SS Ari were made on ten nights from November 1996 to December 1996. Eight new times of minimum lights were derived. The analysis of times of minima of SS Ari confirms the suggestions of other previous investigators that the orbital period of SS Ari have been suffering from a sinusoidal varition. The amplitude and period for the cyclic period changes were calculated as about $58^{y}$ and $0.^{d}053$, respectively. The period variation has been discussed in terms of two potential mechanisms: 1) the light-time effect due to a hypothetical third body and 2) deformations in the convective envelope of a magnetically active component. In the earlier case, the third body has a mass of $1.3M_{\odot}$, if exist, in the form of a white dwarf or a binary system. It seems that the system velocities from the spectroscopic observations supports this interpretation. Meanwhile in the latter case, the primary component is mainly responsible for the magnetic activity of this system with a theoretical amplitude of $\pm0.^{m}08$. However, we cannot make a conclusion which is reasonable explanation at this point, due to lack of observational data. Moreover, the period variation of SS Ari shows duplication about $14^y$, cyclic period with an amplitude of about $0.^d001$ to the above periodic change. We also cannot make an acceptable conclusion for it at this time.

Generation and Characterization of a Neutralizing Human Monoclonal Antibody to Hepatitis B Virus PreS1 from a Phage-Displayed Human Synthetic Fab Library

  • The hepatitis B virus (HBV) envelope contains small (S), middle (M), and large (L) proteins. PreS1 of the L protein contains a receptor-binding motif crucial for HBV infection. This motif is highly conserved among 10 HBV genotypes (A-J), making it a potential target for the prevention of HBV infection. In this study, we successfully generated a neutralizing human monoclonal antibody (mAb), 1A8 (IgG1), that recognizes the receptor-binding motif of preS1 using a phage-displayed human synthetic Fab library. Analysis of the antigen-binding activity of 1A8 for different genotypes indicated that it can specifically bind to the preS1 of major HBV genotypes (A-D). Based on Bio-Layer interferometry, the affinity ($K_D$) of 1A8 for the preS1 of genotype C was 3.55 nM. 1A8 immunoprecipitated the hepatitis B virions of genotypes C and D. In an in vitro neutralization assay using HepG2 cells overexpressing the cellular receptor sodium taurocholate cotransporting polypeptide, 1A8 effectively neutralized HBV infection with genotype D. Taken together, the results suggest that 1A8 may neutralize the four HBV genotypes. Considering that genotypes A-D are most prevalent, 1A8 may be a neutralizing human mAb with promising potential in the prevention and treatment of HBV infection.
