Differentiation and upregulation of heat shock protein 70 induced by a subset of histone deacetylase inhibitors in mouse and human embryonic stem cells

  • Inhibiting histone deacetylase (HDAC) activity modulates the epigenetic status of cells, resulting in an alteration of gene expression and cellular function. Here, we investigated the effects of HDAC inhibitors on mouse embryonic stem (ES) cells. The HDAC inhibitors trichostatin A, suberoylanilide hydroxamic acid, sodium butyrate, and valproic acid induced early differentiation of mouse ES cells and triggered induction of heat-shock protein (HSP)70. In contrast, class III HDAC inhibitors failed to induce differentiation or HSP70 expression. Transcriptional upregulation of HSP70 was confirmed by mRNA expression analysis, an inhibitor study, and chromatin immunoprecipitation. HSP70 induction was dependent on the SAPK/JNK, p38, and PI3K/Akt pathways. Differentiation and induction of HSP70 by a subset of HDAC inhibitors was also examined in human ES cells, which suggests that the phenomenon generally occurs in ES cells. A better understanding of the effects of HDAC inhibitors may give more insight into their application in stem cell biology.

The Effect of Dammarane Glycosides of Panax ginseng on Primary Cultured Chicken Embryonic Muscle Cells (인삼의 dammarane계 glycosides 분획물이 일차 배양한 계배의 근육세포에 미치는 영향)

  • Effects of dammarane glycosides of Panax ginseng on primary cultured chicken embryonic skeletal muscle cells were studied by microscopic observation and determination of the activity of acetylcholinesterase. Muscle cells were prepared from the breast of 12-day-old chicken embryo and cultured with either a medium consisted of 87.5% Dulbecco's Modified Eagle Medium (DMEM), 10% horse serum and 2.5% chicken embryonic extract or a medium consisted of 90% DMEM and 10% horse serum. It was observed that dammarane glycosides of Panax ginseng seemed to show the tendency to stimulate the growth and the differentiation of the muscle cells cultured with a medium consisted of 90% DMEM and 10% horse serum under microscopic observation. The activity of acetylcholinesterase in the muscle cells cultured with a medium consisted of 90% DMEM and 10% horse serum was increased by dammarane glycosides of Panax ginseng.

  • The effect of protein constituents of six-year old fresh Panax ginseng root on chick embryonic brain, spinal cord and skeletal muscle dissociation cultures was studied. The protein constituents showed the enhancing effect on cultured brain, spinal cord and skeletal muscle cells. The neurite formation from brain and spinal cord cells and the outgrowth of neurite seemed to be enhanced by almost all of the protein constituents employed for this study. The maturation of skeletal muscle cells was stimulated by the protein constituents. This enhancing effect of the protein constituents was more vivid when brain, spinal cord and skeletal muscle cells were cultured with a medium which did not contain chick embryonic extracts known as an essential component for primary cell culture. The protein fraction having molecular weight range of 1,000 to 5,000 out of all the protein fractions employed for this study showed the most stimulatory effect on cultured brain, spinal cord and skeletal muscle cells.

The Replication of Parvovirus KBSH DNA in the Embryonic Swine Kidney Cells (돼지 태아 신장세포에 있어 parvovirus KBSH의 DNA 복제과정에 관하여)

  • To study the replication process of the single-stranded DNA parvovirus KBSH-isolated from normal human cell cultures-in actively dividing embryonic swine kidney cells, amount of the synthesized viral hemagglutination (HA) antigen and the overall rate of viral double-stranded replicative form(RF) DNA synthesis were wxamined. The initiation of viral RF KNA synthesis and the decrease of host DNA synthesis rate in viral infected cells occurred almost same time at 15-16 hour post infection(PI). And the release of viral HA antigen to media followed at 24 hour PI, concurrently the overall rate of viral RF DNA synthesis reaching its maximum. Evidence is presented which indicates that successful performance of viral RF DNA replication requires proteins synthesized in viral infected cells at 10-14 hour PI.
