This study was conducted to evaluate the effects of different volume ($100{\mu}l$ vs. 2 ml) of microdrop culture on B6D2F1 mice oogenesis. In the present study, B6D2F1/CrljOri $F_1$ mice were utilized in order to maximize oogenesis. Also we used TCM-199, Dulbecco's medified Eagle's medium (DMEM), embryo culture medium (Fertilization medium, Cleavage medium, Blastocyst medium), G series medium and One step medium. Blastulation rate was not different between groups ($58.4{\pm}2.9%$ vs. $61.2{\pm}4.8%$). Zona hatched rate ($38{\pm}15.4%$ vs. $27{\pm}3.4%$) and attached rate ($55{\pm}13.9%$ vs. $46{\pm}3.9%$) did not differ by the volume of culture media. Total cell numbers ($59.8{\pm}9.7$ vs. $70.3{\pm}8.7$), ICM cell numbers ($15.8{\pm}0.6$ vs. $16.8{\pm}1.5$), TE cell numbers ($44.0{\pm}9.7$ vs. $53.6{\pm}7.3$), % ICM ($26.4{\pm}2.9%$ vs. $23.8{\pm}3.3%$) and ICM:TE ratio ($1:2.8{\pm}0.4$ vs. $1:3.2{\pm}0.6$) were not different between groups (i.e., $100{\mu}l$ vs. 2 ml). These results show that the capacity of the culture medium did not effect the cell numbers of B6D2F1 mice blastocysts. In summary, these results can provide fundamental data to maximize culture condition for in vitro fertilization on B6D2F1 mice.
The purpose of this study was to determine the effect of rbST treatment on progesterone concentration, volume of luteal tissue and pregnancy rate following embryo transfer. Recipient cows were assigned to control and rbST group, of which was given a single injection of rbST (500 mg. sc) at estrus detection. The concentration of progesterone was not significantly different between control and at 0, 3, 6 days after rbST treatment. However, the concentration of progesterone at 9, 12 days was significantly higher than in control group (4.6 and 6.8 vs. 3.9 and 4.5 ng/ml P4). The pregnancy rate after embryo transfer in rbST treatment was significantly higher than in control group (64.0 vs. 47.1 %; p<0.05). The results indicated that rbST treatment in recipient cows could be improved the efficiency of pregnancy rate after embryo transfer.
This experiment was carried out to investigate the major characters affecting the popping volume of popcorn. Tuygimok 1 (Kp1 ${\times}$ Kp2) and 8 popcorn hybrids' agronomic characters were tested to evaluate a certain extent how much they affect on the popping volume. Moisture con-tent was considered as the most important factor, but failed to evaluate the optimum moisture con-tent level in this experiment moisture range (12.2-14.4%) because popping volume increased as moisture content of kernels increased. The maximum popping volume was obtained at 55-60kg of kernel hardness, 80-90,um of pericarp thickness and 45-50% of S/H (Soft/Hard starch). But the Em/En(Embryo/Endosperm) ratio was negatively associated with the popping volume. Therefore the minimum popping volume was observed at the 10-11 % of Em/En ratio. Moisture content, hardness, pericarp thickness, Em/En and S/H ratio were selected as the appropriate variables for the maxi-mum popping volume using the stepwise forward regression method and the expecting popping volume was estimated by the multiple linear regression formular. The mean popping volume of ninepopcorn hybrids was about 29.2cm3/g.
Rice is a staple food for more than 50% of the world's population. Embryo comprises only 2 to 3% of the weight distribution of the entire pericarp but has higher concentration of vitamins, proteins, and essential fatty acids than the other parts of grains. Moreover, ${\alpha}$-tocoperol, ${\gamma}$-oryzanol, phytic acid and ${\gamma}$-aminobutric acid that have nutraceutical value are abundant. Increasing the volume of embryo assures the fortification of nutritional value of rice grain. We developed new black waxy giant embryo rice, Milyang 263 by crossing Josaengheugchal, a black waxy rice variety, and $ge^t$, a giant embryo mutant generated by tissue culture. The nutrient contents and physical properties of Milyang 263 were compared with several giant embryo mutants and normal embryo rice varieties. Changes in the nutrient properties after germination were also observed. Results indicated that this new black waxy giant embryo rice, Milyang 263, offers a promising source for improving nutritional quality of rice especially anthocyanin, essential minerals, and GABA.
Among proteins separated from methanol extract of follicular fluid with superose column, the components inducing sperm swim-up separation through sucrose layer were analysed with superose column in Smart system and SDS-PAGE. And the results obtained were as follows; The fractions of retention volume (RV) 0.83ml and RV 1.36ml separated with superose column should stimulate sperm migration and movement. However, RV 0.83 fraction was consisted of complex materials containing RV 1.36 component. RV 1.36 fraction contained a BSA analogue of 67 kilodaltons (Kd) and showed identical peak pattern with BSA fraction V. In conclusion, the protein of 67 Kd in follicular fluid should stimulate sperm migration and movement.
This study was carried out to investage the effect of cryoprotectant concentration and equilibration time on volume change and in vitro development of intact and bisected mouse embryos by rapid freezing. When compacted morulae were rapidly frozen in 3.0 to 4.0 glycerol or DMSO with 0.25M sucrose solution, the superior(P<0.05) post-thaw survival rate was obtained at the glycerol concentration of 4.0M(89.4%) than 3.0M(71.4%) or 5.0M(42.4%), but at the DMSO concentration of 3.0M(84.5%) than 4.0M(51.1%) or 5.0M(0.0%). The optimal equilibraton time for rapid freezing of ZP-free or bisected morulae in 4.0M glycerol with 0.25M sucrose was found tobe 3 minutes. The minimal volume of compacted morulaewhich corresponded with 61 to 62% of pre-equilibrated embryo volume was obtained from equilibration for 3 minutes in both 3.0 and 4.0M glycerol solutions with 0.25M sucrose.
This study was conducted to examine suspended embryogenic cells growth with days of culture, effects of various kinds/concentrations of osmoticum for induction of somatic embryos (SEs), following somatic embryos germination or plantlet regeneration. The proliferation pattern of embryogenic cells in suspension culture is characterized by settled cells volume (SCV) increased with the duration of culture with marked the maxium of SCV (10.1 ml) in 18 days of culture, however the SCV of cells gradually decreased after that. In comparison of kinds/concentrations of osmoticum on somatic embryo induction, the highest induction number (352.3/g FW) of the SE was showed in 0.2 M sucrose, in addition, we also observed some effects with treatments of 0.2 M maltose (203.7) and 0.3 M maltose (193.7), respectively. However, no somatic embryos produced in treatments of 7.5% PEG plus 0.15 M sucrose or maltose. In comparison of germination efficiency of SEs which occurred from the treatments of various kinds/ concentrations of osmoticum, the highest induction frequency of cotyledon (25.2%) was obtained from SEs that produced 0.3 M maltose, however, the best occurrence rates of hypocotyl (39%), radicle (30.3%) and plantlet regeneration (3.5%) were observed from the 0.2 M sucrose treatment, respectively.
The purpose of this study was to examine the effect of growing stages of the Korean Native Striped Bull (KNSB) on the freezability and fertility of frozen-thawed semen. First, we investigated the total motility (TM) and progressive motility (PM) according to the diluent used for semen freezing. Second, we examined the effect of the age of KNSB on semen volume, TM and PM of fresh and frozen-thawed semen. Third, we examined the effect of frozen semen from the different age of KNSB on the $in-vitro$ fertilization rate, and the artificial insemination pregnancy rate. The diluents used in this experiment were Triladyl$^{(R)}$ and Tris-egg yolk extender (EYE). Semen was collected from 5 KNSB in the growing stage (15 months) and 5 adult KNSB (36 months). When Triladyl or Tris-EYE extender was used for semen freezing, there was no difference of the mean TM and the mean PM. However, the mean TM was significantly higher in Bull No. 1885 than Bull No. 4283 ($p$ <0.05). The mean volume of semen collected from the 15-month-old bulls (2.3 ml) was significantly lower ($p$ <0.05) than that from the 36-month-old bulls (5.0 ml). The mean semen concentration was similar for the 15-month-old ($2.1{\times}10^9$ spermatozoa/ml) and 36-month-old ($1.8{\times}10^9$ spermatozoa/ml) bulls. For the 15-month-old and 36-month-old bulls, the mean TM of fresh semen were 93.7% and 88.3%, respectively, and the mean PM were 97.0% and 88.3%, respectively; the 15-month-old bulls showed a particularly high PM ($p$ <0.05). For the 15-month-old and 36-month-old bulls, the mean TM (56.0% and 58.0%, respectively) and the mean PM (64.0% and 70.7%, respectively) of frozen-thawed semen did not differ. The development rates of embryos after $in-vitro$ fertilization and the pregnancy rate after artificial insemination using frozen-thawed semen did not differ according to the bull's age. In summary, semen volume differed according to the bull's age, but semen concentration and survival rate, the $in-vitro$ fertilization rate, and the pregnancy rate did not differ according to the stripe bull's age. Accordingly, semen from bulls in the growing stage can be collected and frozen for the preservation and multiplication of rare livestock.
Larvae of Palaemon serrifer were reared in the laboratory under three different temperature regimes ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$) to study the effects of rearing temperature on larval survival and growth, as well as other traits such as embryo volume, number of embryos (fecundity), incubation period, development. Mode and development period. Growth pattern was analyzed by measuring the molt increment and intermolt period. The intermolt period consistently increased with size and instar number and was shortest at $25^{\circ}C$. However, molt increments generally decreased with instar number. Rapid loss in viability of neem (Azadirachta indica A. Juss.) seed is a major problem. Present effort was undertaken for developing a set pattern for assessing of viability and vigour in seed of various mother tree age of neem (Age I-06 years, Age II-15 years, Age III-25 years and Age IV->30 years old). Various viability test viz. triphenyle tetrazolium chloride test, electrical conductivity, excised embryo test, and germination test have been performed on seeds obtained from mother tree age classes. Inconsistency was observed with the TTC and EC test in germination of seed in laboratory as well as nursery. While various vigour tests viz. cold test, chemical stress test (methanol stress test), and accelerated ageing test alongwith ageing index, germination test (G%, MGT and GV) and various seedling growth parameters like seedling length (cm), number of leaves, collar diameter (cm), total biomass (g) alongwith mathematical indices i.e. vigour index, sturdiness quotient, volume index, quality index, root shoot ratio in nursery as well have been taken for study and showed better consistency. On the basis present study results of various viability and vigour test indicated that mother tree age class II performed better in comparison to others and it can be recommended for seed collection. Further it is also recommended that viability of neem seed may be assessed using various laboratory tests like excise embryo test and germination test (G%, MGT and GV) and vigour test may be taken preferably by cold germination test, chemical (methanol) stress test, accelerated ageing test in laboratory and germination alongwith various seedling growth parameters seedling length (cm), number of leaves, collar diameter (cm), total biomass (g) alongwith mathematical indices like Vigour Index, Sturdiness quotient, Volume Index, Quality index, root shoot ratio in nursery as discussed in this study.
