Developmental characterization of embryo size mutant in rice (Oryza sativa L.)

  • In this experiment, three kinds of mutations(ge, re, and eml )relating to the size of embryos were used to study their generation, genetic mechanism and developmental characteristics, and the interactions between embryo and endosperm were also examined. Giant embryo mutation comprises 7 kinds including the already isolated ge, and ge-2, which share an identical gene site. The SAM and the size of radicule for the ge showed little difference compared to a normal type. The number of embryo cells did not increased as much as it would affect the size of embryo. Therefore, the enlargement of embryo was due to the enlargement of scutellum that originated from the corpulence of each cell. Both F$_1$' s of re ]and odm 49 formed reduce embryos, and other combinations of hybridization showed all wild type of embryo sizes. Accordingly, the odm 49 must have an identical gene site of re 1, while odm 48 and odm 62 have different gene sites. Their shoots and radicules also shrank by the same ratio, however no sign of physical change was noticed. The size of embryo cell showed no change, while the number of cells was the half of that of wild types. The three gene sites of re represent all of them control the size of the entire embryo forming organs. The eml 1 was defined to have temperature sensibilities that the generation of endosperms was active at a high temperature while that was hampered at a low temperature.

Inheritance Analysis of Giant Embryo Mutation Induced by T-DNA Insertion in Rice

  • Recently, giant embryonic rice and functional rice food are preferred by more consumers, which are attributed to the fact that the embryo has high concentrations of essential amino acids, fatty acids, and vitamins relative to other parts of rice grains. In this report, the heredity and stability of giant embryo mutations in successive generations were analyzed regarding a giant embryonic line, 'P47', induced by T-DNA insertion and a $F_2$ population from a cross between 'P47' and 'Junam'. The mutant lines with increases of 1.5, 1.7 and 1.8 times on embryo length, width and 100-embryo weight to those of the control showed stable inheritance across three generations. The continuous frequency distributions of embryo size in the $F_2$ population showed that the embryo size is a quantitative trait of polygene controlled. In addition, wide range of transgressive segregations of six traits affecting embryo size confirmed exchange of genetic materials and recombination between genes controlling embryo size. Five giant embryo mutant lines selected from the $F_2$ population will be used for artificial selection and improvement of giant embryonic varieties.

Analysis and mapping of the re-1 gene for reduced embryo size in rice

  • The objective of this study was to map the gene for reduced embryo size in rice using DNA markers. The reduced embryo size mutant was induced from N-methyl-N-nitrosourea (MNU) treated Taichung 65. Genetic analysis revealed that the phenotype of the reduced embryo was controlled by a single recessive gene, designated as re-1. For mapping the gene controlling embryo size, an $F_2$ population was developed from a cross between the Korean Tongil-type, Milyang 23 (Oryza sativa ssp. indica) and the mutant. The ratio of $F_2$ seeds nearly fitted to 3:1 ratio, indicating that this phenotype was controlled by a single recessive gene. Bulked sergeant analysis was performed with SSR markers. The gene for the reduced embryo size was detected on chromosome 1. The gene was further mapped between two SSR markers, RM315 and RM265 on chromosome 1 (approximately 1.5 Mb interval). The linked markers will facilitate selection of this grain character in a breeding program and provide the foundation for positional cloning of this gene.

Influence of pretreatment medium, fresh medium addition, and culture plate size on the production of embryos in isolated microspore culture of hot pepper (Capsicum annuum L.) (고추의 소포자 배양 시 전처리 배지, 새 배지의 첨가, 및 배양 용기의 크기가 배의 생산에 미치는 영향)

  • The influences of pretreatment medium, the addition of fresh medium, and the size of culture plate on the production of embryos were investigated in isolated microspore culture of hot pepper (Capsicum annuum L.). Among the media used for heat shock pretreatment ($32{\pm}1{^{\circ}C}$), high frequency embryo production was obtained when the sucrosestarvation medium A was used. On the other hand, neither 0.37 M mannitol solution nor NLNS medium supplemented with sucrose was not efficient for embryo production. The addition of culture medium to pretreatment media considerably decreased the embryo production even though embryo development proceeded further. The embryo production was not improved by the addition of fresh medium after 2 or 3 weeks from starting culture. Increase in the size of the culture plate from $3.5{\times}1.0$ cm to $6.0{\times}1.5$ cm improved embryo quality. These results will provide valuable information for developing an efficient microspore culture system of hot pepper for high frequency embryo production.

Embryo Collection, Transfer and Pregnancy of Riding Horses : First Successful Case in Korea

  • Embryo transfer (ET) could be a relevant tool for genetic improvement programs in horses similar to those already underway in other species and produce multiple foals from the same mare in one breeding season. However, there have been no reports describing equine embryo transfer performed in Korea. In the present study, we performed an equine embryo collection and transfer procedure for the first time. We examined the embryo collection and pregnancy, size of embryo during the incubation period after collection, and progesterone (P4) and estradiol-$17{\beta}$ (E2) concentrations in mare's serum at embryo collection and transfer. A total of 16 donors responded to estrus synchronization; estrus was induced in 12 donors and 4 recipients, and artificial insemination was successful in 10 donors and six blastocysts were collected from donors. Of these blastocysts, we monitored the size of blastocysts for 3 day during incubation and transferred 2 blastocysts to a recipient, with 1 successful pregnancy and foal achieved. The dimensions of equine embryo at day 7 to day 9 were $409{\mu}m$, $814{\mu}m$ and $1,200{\mu}m$. The serum P4 and E2 concentrations were $7.91{\pm}0.37ng/{\mu}L$ and $45.45{\pm}12.65ng/{\mu}L$ in the donor mare, and 1$6.06{\pm}3.27ng/{\mu}L$ and $49.13{\pm}10.09ng/{\mu}L$ in the recipient mare.

Optimum Chilling Terms for Germination of the Dehisced Ginseng(Panax ginseng C. A. Meyer) Seed (개갑 인삼종자의 발아 적정 저온감응기간)

  • Experiments were conducted to study the optimum chilling period for breaking physiological dormancy of dehisced ginseng(Panax ginseng C. A. Meyer) seed. Embryo of ginseng seed is too small to be noticed with naked eyed on harvesting time. Embryo grew to half size of endosperm after seeds were stratified for 3 months. It takes 6 months for this embryo to reach the size enough for germination. And it grew faster indoors than outdoors. Dehisced ginseng seed with full-size embryo did not germinate at room temperature and required chilling treatment for 75days in outdoors and 90 days in cold chamber. While seed receiving sufficient chilling treatment were left to be in room temperature, the chilling effect decreased remarkably.

  • The development potential of bovine somatic cells was evaluated using nuclear transfer. A single donor cell derived from fetus of HanWoo(Korean Native Cattle) was selected and deposited into perivitelline space of each enucleated oocyte before electrical fusion and activation. Nuclei of donor cells starved for 7 days (37%) tended to support the development of reconstitute embryo the blastocyst stage better than those of donor cells starved 3, 14 and 30 days. The cleavage rate was significantly lower(P<0.05) in reconstitute embryos derived from large size donor cells(51.2%), than those from small medium size donor cells(76.6 and 73.5, respectively). The developmental rate to blastocyst of reconstructed embryos from medium size donor cells was higher than those from small and medium size donor cells. This study demonstrates that an appropriate culture period for induction into quiescent stage and the size of donor cells effect on the efficiency of nuclear transfer using cultured bovine cells.

  • This study was carried out to produce twin calves by embryo transfer in Hanwoo and investigate the pregnancy and twin rate by recipient's conditions. All recipients were bred at estrus by artificial insemination with Hanwoo semen and then transfered an additional embryo produced in vivo or in vitro to tbe uterine horn contralateral to the corpus luteum on Day 7. The results obtained were as follows ; 1. The pregnancy rate was higher in young recipients of 3 years (68.8%) than in old ones of 10 years and greater(36.4%). And for CL size pregnancy rate was 57.9, 45.4 and 60.1% in large, medium and small size of CL of recipients, respectively. 2. 447recipients were transferred an additional embryos at 7th day after Al and average pregnancy rate was 57.5% and twin production rate was 22.2%. 3. Average pregnancy and twin production rate by direct transfer methods of frozenthawed IVF embryos was 56.0 and 16.7%. 4. The ratio of male to female twin in a total of 55 twin pairs was 54.6%, and average gestation lengths of male to female and female to female twin were 280.6$\pm$5.4 and 279.715.4 days, respectively. Average birth weight of twins was beavior in male and male twin(23.2i5.8kg) than in male and female twin(20.5$\pm$2.6kg).

Winter Occurrence Pattern of Chionoecetes opilio around Wangdol Reef in the East Sea near Uljin (겨울철 동해 왕돌토 지역에 서식하는 대게자원 (Chionoecetes opilio) 출현 양상)

  • An ecological investigation was conducted in winter from November 2007 to February 2008, around Wangdol Reef in the East Sea to analyze the population structure and fecundity of Chinoecetes opilio. The sex ratio was 30.11:69.89% (female:male) in November, 62.03:37.97% in December, 37.59:62.41% in January and 14.69:85.31% in February. Regression equations indicated highly positive relationships between size and weight parameters (carapace length, width, total weight). During the sampling period ovigerous females were continuously collected. The percentage of ovigerous females was 96% in November, 90.57% in December 89.80% in January, and 88.46% in February. The average number of embryos was 64,800. The regression equation between carapace width and the number of embryos was y = 2.6805x - 0.9182 ($R^2$ = 0.7166). Embryo volume increased as embryo development proceeded. The mean size of an embryo was 0.72 mm. Embryo volume ranged from 0.42 to $0.84\;m^3$ during embryo developmental stages 1 to 3.

Studies on the Rapid Freezing of Mouse Embryo II. Effects of the Development Stage and Seeding on the Mouse Embryo Survival of Rapid Freezing (Mouse 수정란의 급속동결에 관한 연구 제II보 Mouse수정란 급속동결에 있어서 수정란의 발육단계와 식빙(seeding)이 생존율에 미치는 영향)

  • The effects of seeding method and optimum time for freezing embryos according to the developmental stages on embryo survival rates after rapid freezing were determined using the FDA-test. The summarized results are as follows : 1. In the rapid freezing of embryos, the sucrose added medium together with Co-seeding or non-seeding showed the FDA scores of 4.67 and 4.20, respectively, but, raffinose addition obtained FDA scores of 4.27 and 3.97. 2. The developmental stage of embryos at freezing was most critical on the survival of embryos after thawing. Higher FDA scores were obtained in the order of blastocyst stage(4.94), morula stage(3.82) and ealy stage(2.65) in sucrose added medium. The same trend was observed in the raffinose added medium with an order or 4.91, 4.47 and 2.32. 3. Microscopic study of embryo before freezing and post-thawing indicated that the embryo showed shrinkage within 5 minutes after the embryo was transfer to the freezing medium. When thawed embryo was tranfered to the dilution medium, swelling of the embryo was observed and there after it reshrank indicating the removal of cryoprotectant from the embryo. The size of the embryo recovered to the original state when it was moved into a PBS-solution.
