Nano Pt particles were dispersed on carbon-based supports by a polyol process for a catalyst application in a polymer electrolyte fuel cell. We tried to optimize the effect of pH on the electrostatic forces between the support and the Pt colloids. We investigated the relationship among the surface charges on the carbon support, the solution pH, and the concentration of a glycolate, and the Pt particle size. The produced catalyst with nano Pt particles on the support was evaluated by the long-term cyclic voltammetry (CV) performance test and compared with the results from a commercial catalyst. Our experimental results reveal that the pH-control can modify the particle size distribution and the dispersion of the nano Pt particles. This resulted in a cost-effective method for the synthesis of highly Pt loaded Pt/C catalysts for fuel cells better than a commercial catalyst system.
본 연구는 표면에 세포를 부착하는데 있어서, 다양한 기판 표면에 보편적인 플랫폼으로써 적용될 수 있는 세포 부착을 위한 기능성 표면의 제작 기술 및 이를 이용한 세포의 선택적인 고정과 편리한 세포 패터닝의 방법을 보여주었다. 세포 부착에 적합한 기능성 표면의 제작은 산소 플라즈마 처리를 이용한 다양한 기판의(유리, PMMA, PS, PDMS) 표면 활성화 및 상반되는 고분자 전해질의(PAH, PDAC, PSS, PAA) 정전기적 인력을 통한 증착으로 이루어진 다층의 고분자 전해질 층을 통해 제작될 수 있었다. 또한, 고분자 전해질로 증착된 표면 위로 마이크로 몰딩 인 케필러리 방법을 사용하여 PEG 마이크로 구조물을 제작함으로써 세포의 선택적인 고정이 이루어질 수 있었다. 다층의 고분자 전해질로 증착된 표면은 세포와의 강한 정전기적 인력으로 세포 부착에 유리한 표면을 제공하였다. 반면에, 제작된 PEG 마이크로 구조물은 물리적, 생물학적인 장애물의 역할로써 세포의 비 특이적인 흡착을 방지하였다. 세포 부착을 위한 기능성 표면을 제작하는 동안 표면의 특성은 접촉각 측정을 통해 이루어 졌다. 다양한 기판 상에서 개질된 표면은 세포 부착을 위한 적합한 환경의 제공과 함께 세포의 마이크로 패터닝 기술에서 높은 수율의 세포 패터닝을 제공한다. 상기의 제안된 세포 부착을 위한 기능성 표면 제작 기술 방법은 제작 과정이 매우 간단하고, 편리하여 손쉽게 구현이 가능하며, 제작 공정에서 어떠한 해로운 용매도 사용하지 않기 때문에 친환경적이다. 또한, 이를 이용하여 세포를 이용하는 바이오 칩 및 바이오 센서, 세포를 기반으로 하는 시스템 등에서 기본이 되는 기술로 사용될 수 있는 넓은 응용 범위를 갖는다.

Genistein (GT), a major isoflavone found in soybeans, has a potent antioxidant effect that protects the skin from UV-induced damages and malignant melanoma. In order to enhance the cellular uptake of GT, liposome/Tat complexes were prepared by an electrostatic interaction of anionic liposome (DMPC/DCP, 9:1 in molar ratio) with Tat peptide (0.02 to 0.08 mole), one of the well-known cell penetrating peptide (CPP). As the amount of Tat increased, the size increased but the zeta potential decreased. In vitro release study with dialysis membrane elicited GT release from liposomal preparations in a controlled manner. The addition of Tat increased GT release, especially for the initial period. In the cellular uptake study by incubating B16 melanoma cells with various liposomal preparations containing GT, B16 melanoma cells demonstrated a time-dependent increase of drug accumulation. Compared to the aqueous GT suspension, intracellular uptake was substantially enhanced by anionic liposomal formulation and further increased by the complex formulation. Therefore, liposome/ Tat complex might be a good candidate for facilitating intracellular drug delivery.
CA(1-8)-ME(1-12) [CA-ME], composed of cecropin A(1-8) and melittin(1-12), is a synthetic antimicrobial peptide having potent antibacterial and antitumor activities with minimal hemolytic activity. In order to investigate the effects of the flexible hinge sequence, Gly-Ile-Gly, of CA-ME on antibiotic activity, CA-ME and three analogues, CA-ME1, CA-ME2, and CA-ME3, were synthesized. The Gly-Ile-Gly sequence of Ca-ME was deleted in CA-ME1 and replaced with Pro and Gly-Pro-Gly in CA-ME2 and CA-ME3, respectively. CA-ME1 and CA-ME3 showed a significant decrease in antitumor activity and phospholipid vesicle-disrupting ability. However, CA-ME2 showed similar antitumor and vesicle-disrupting activities, as compared with CA-ME. These results suggest that the flexibility or ${\beta}$-turn induced by Gly-Ile-Gly or Pro in the central part of CA-ME may be important in the electrostatic interaction of the N-terminus cationic ${\alpha}$-helical region with the cell membrane surface and the hydrophobic interaction of the C-terminus amphipathic ${\alpha}$-helical region with the hydrophobic acyl chains in the cell membrane. CA-ME3 exhibited lower antitumor and vesicle-disrupting activities than CA-ME and CA-ME2. This result suggests that the excessive ${\beta}$-turn structure caused by the Gly-Pro-Gly sequence in CA-ME3 seems to interrupt ion channel/pore formation in the lipid bilayer. We concluded that the appropriate flexibility or bilayer. 암을 치료하기 위한 양이온성 리포솜은 화학치료요법의 분야에서 발전되어 왔다. 양이온성 리포솜은 음이온성의 세포 표면에 정전기적 상호작용에 의해 결합이 된다. 본 연구의 목적은 음이온성 세포와 이온결합을 할 수 있는 양이온성 리포솜을 제조하는 것이다. 양이온성 리포솜은 지질인 1,2-디스테로일-sn-글리콜-3-포스포에탄올아민 (DSPE)과 양이온성 고분자인 폴리에틸렌이민 (PEI)을 그라프트 중합으로 합성된 물질로부터 제조하였다. 리포솜 표면의 이온 특성은 제타 포텐셜을 측정하여 확인하였다. 소 혈청 수용액에서 리포솜의 혈장 단백질 흡착 특성은 입자 크기와 탁도 변화를 측정하여 확인하였다. 완충 용액 속에서 리포솜 안정성을 예측하기 위하여 입자 크기를 7일 동안 상온에서 측정하였다. 양이온성 리포솜은 소 혈청 수용액에서 많은 양의 혈장 단백질이 흡착되었다. 이는 혈장 단백은 음전하를 가지고 있어서 양이온성 리포솜의 표면과 잘 붙기 때문이다. 양이온성 폴리에틸렌이민을 이용한 리포솜의 표면 변형은 소 혈청 수용액 내에서 단백질 흡착을 강화시킨다는 것을 예상하게 한다. 또한, 제조된 리포솜의 완충 용액 내에서 7일 동안 안정한 상태임을 관찰하였다.
This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.
초고온성 고세균 Thermococcus onnurineus NA1은 개미산, 일산화탄소, 또는 전분 등을 이용해서 수소를 생산하는 것으로 알려져 있다. 본 연구에서는 T. onnurineus NA1의 고정화 세포를 이용한 수소생산을 고찰하였다. 고정화 실험결과, T. onnurineus NA1은 표면에 아민기가 코팅된 규조토 담체에 정전기적 인력에 의해 효과적으로 고정화되었고, 1 g의 담체에 고정화 될 수 있는 최대 세포의 양은 71.7 mg-dcw로 확인되었다. 고정화 세포를 이용한 세 번의 반복회분식 배양을 통해 개미산으로부터 수소생산 특성을 고찰하였고, 그 결과 배양이 반복됨에 따라 고정화 세포 농도의 증가에 기인하여 초기수소생산속도가 2.3 에서 4.0 mmol l−1 h−1로 상당량 증가됨이 관찰되었다. 따라서, T. onnurineus NA1의 고정화세포 시스템은 수소생산을 위한 좋은 대안이 될 수 있을 것으로 사료된다. 본 연구는 초고온성 고세균의 고정화세포를 수소생산에 적용한 첫 번째 사례이다.
최근 마이크로 및 나노테크놀러지 (nanotechnology) 분야에서 가장 범용적으로 사용되고 있는 폴리머인 Poly-(dimethylsiloxane) (PDMS)의 표면을 다층의 고분자 전해질을 이용하여 표면 개질 및 그 특성을 보였다. 서로 상반되는 전하를 나타내는 고분자 전해질의 정전기적 인력을 통해 개질된 PDMS의 표면 성질은 접촉각 분석기를 이용한 접촉각의 측정 및 Fourier transform infrared (FT-lR) spectroscopy를 이용해 측정함으로써 확인할 수 있었다. 상기의 표면 개질 방법을 통하여 원하는 표면의 성질을 구현 할 수 있고 생체 물질의 부착을 위한 표면 또한 쉽게 만들 수 있다. 다층의 고분자 전해질로 개질된 PDMS 표면에 부착된 박테리아는 표면이 개질 되지 않은 PDMS 표면과 매우 높은 대조를 이루는 것을 확인 할 수 있었다. PDMS 표면에 세포 부착을 위한 경우 그것의 소수성인 표면 성질로 인한 제약을 본 연구에서 제안한 표면 개질 방법을 이용하여 해결 할 수 있었다. 상기 방법의 가장 큰 장점은 간단하고, 빠르게 표면을 개질 할 수 있는 방법이라는 점에 있으며, 고분자 전해질의 여러 조합을 통해 원하는 표면의 성질을 조절 할 수 있으므로 매우 중요한 기술로 생각된다. 본 연구에서 제안된 방법은 간단하고, 편리하며, 매우 재현성이 높고, 빠르게 구현할 수 있어서 이것을 이용하여 바이오 센서 및 바이오 칩, 랩온어 칩 분야, 패터닝, 세포와 표면 간의 상호작용 연구를 위한 응용 분야로의 적용이 될 것으로 기대된다.

The accumulation of ${\beta}$-amyloid (A${\beta}$) aggregates is a characteristic of Alzheimer's disease (AD). Furthermore, these aggregates have neurotoxic effects on cells, and thus, molecules that inhibit A${\beta}$ aggregate formation could be valuable therapeutics for AD. It is well known that aggregation of A${\beta}$ depends on its hydrophobicity, and thus, in order to increase the hydrophilicity of A${\beta}$, we considered using citrate, an anionic surfactant with three carboxylic acid groups. We hypothesized that citrate could reduce hydrophobicity and increase hydrophilicity of A${\beta}_{1-40}$ molecules via hydrophilic/electrostatic interactions. We found that citrate significantly inhibited A${\beta}_{1-40}$ aggregation and significantly protected SH-SY5Y cell line against A${\beta}_{1-40}$ aggregates-induced neurotoxicity. In details, we examined the effects of citrate on A${\beta}_{1-40}$ aggregation and on A${\beta}_{1-40}$ aggregates-induced cytotoxicity, cell viability, and apoptosis. Th-T assays showed that citrate significantly inhibited A${\beta}_{1-40}$ aggregation in a concentration-dependent manner (Th-T intensity: from 91.3% in 0.01 mM citrate to 82.1% in 1.0 mM citrate vs. 100.0% in A${\beta}_{1-40}$ alone). In cytotoxicity and viability assays, citrate reduced the toxicity of A${\beta}_{1-40}$ in a concentration-dependent manner, in which the cytotoxicity decreased from 107.5 to 102.3% as compared with A${\beta}_{1-40}$ aggregates alone treated cells (127.3%) and the cell viability increased from 84.6 to 93.8% as compared with the A${\beta}_{1-40}$ aggregates alone treated cells (65.3%). Furthermore, Hoechst 33342 staining showed that citrate (1.0 mM) suppressed A${\beta}_{1-40}$ aggregates-induced apoptosis in the cells. This study suggests that citrate can inhibit A${\beta}_{1-40}$ aggregation and protect neurons from the apoptotic effects of A${\beta}_{1-40}$ aggregates. Accordingly, our findings suggest that citrate administration should be viewed as a novel neuroprotective strategy for AD.
실리콘계 태양전지 제조과정에서 발생하는 불량품에서 실리콘웨이퍼를 회수하는 연구를 수행하였다. 상온($25^{\circ}C$)에서 인산용액 농도, 산성불화암모늄 농도, 킬레이트제 종류 및 농도를 변화시키면서 폐태양전지의 반사방지막 및 N층의 제거 효율을 조사하였다. 10 wt% 인산, 2.0 wt% 산성불화암모늄, 1.5 wt% Hydantoin 사용 시 제거 효율이 가장 우수 하였다. 인산농도가 증가할수록 미세입자의 표면전위가 (+)로 변하여 정전기적 인력에 의해 실리콘웨이퍼 표면에 재흡착하여 표면처리 전보다 두께가 두꺼워졌으며, 표면의 오염도도 증가하였다. 인산-산성불화암모늄-킬레이트제 용액에 의한 표면처리방법은 모든 공정이 상온에서 수행되며, 공정이 단순하고, 폐수 발생량이 적고, 표면제거 효율이 우수한 방법으로 폐 태양전지의 재활용 및 기존 RCA 세정법의 대안으로 가능성이 매우 클 것으로 판단되었다.
