Characterization of Molecular Composition of Bacterial Melanin Isolated from Streptomyces glaucescens Using Ultra-High-Resolution FT-ICR Mass Spectrometry

  • In this study, the chemical composition of bacterial melanin isolated from the Streptomyces glaucescens strain was elucidated by ultra-high-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry. Ultra-high-resolution mass profiles of the microbial melanin product were acquired using a 15 Tesla FT-ICR mass spectrometer in positive and negative ion modes via electrospray ionization to obtain more complete descriptions of the molecular compositions of melanin-derived organic constituents. A mass resolving power of 500,000 (at m/z 400) was achieved for all spectra while collecting 400 scans per sample with a 4 M transient. The results of this analysis revealed that the melanin pigment isolated from S. glaucescens predominantly exhibits CHON and CHO species, which belong to the proteins class of compounds, with the mean C/O and C/N ratios of 4.3 and 13.1, thus suggesting that the melanin could be eumelanin. This analytical approach could be utilized to investigate the molecular compositions of a variety of natural or synthetic melanins. The compositional features of melanins are important for understanding their formation mechanisms and physico-chemical properties.

  • Angiotensin I, a model decapeptide, was glycosylated and partially hydrolyzed with HCl (6 N, 80 $^{\circ}C$, 4 h), aminopeptidase, and carboxypeptidase Y. A single peptide mass map obtained from truncated peptides in the partial acid hydrolysate of angiotensin and its glycosylation product mixture by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry enabled sequencing of angiotensin by a combinatorial procedure. MALDI-TOF and electrospray ionization (ESI) tandem mass spectrometric results indicate that both the N-terminal amino group of aspartic acid and the guanidinium group of the second residue arginine are glycosylated.

Investigation of the Copper (Cu) Binding Site on the Amyloid beta 1-16 (Aβ16) Monomer and Dimer Using Collision-induced Dissociation with Electrospray Ionization Tandem Mass Spectrometry

  • The copper ion, Cu(II), binding sites for amyloid fragment Aβ1-16 (=Aβ16 ) were investigated to explain the biological activity difference in the Aβ16 aggregation process. The [M+Cu+(z-2)H]z+ (z = 2, 3 and 4, M = Aβ16 monomer) and [D+Cu+(z-2)H]z+ (z = 3 and 5, D = Aβ16 dimer) structures were investigated using electrospray ionization (ESI) mass spectrometry (MS) and tandem mass spectrometry (MS/MS). Fragment ions of the [M+Cu+(z-2)H]z+ and [D+Cu+(z-2)H]z+ complexes were observed using collision-induced dissociation MS/MS. Three different fragmentation patterns (fragment "a", "b", and "y" ion series) were observed in the MS/MS spectrum of the (Aβ16 monomer or dimer-Cu) complex, with the "b" and "y" ion series regularly observed. The "a" ion series was not observed in the MS/MS spectrum of the [M+Cu+2H]4+ complex. In the non-covalent bond dissociation process, the [D+Cu+3H]5+ complex separated into three components ([M+Cu+H]3+, M3+, and M2+), and the [M+Cu]2+ subunit was not observed. The {M + fragment ion of [M+Cu+H]3+} fragmentation pattern was observed during the covalent bond dissociation of the [D+Cu +3H]5+ complex. The {M + [M+Cu+H]3+} complex geometry was assumed to be stable in the [D+Cu+3H]5+ complex. The {M + fragment ion of [M+Cu]2+} fragmentation pattern was also observed in the MS/MS spectrum of the [D+Cu+H]3+ complex. The {M + [y9+Cu]1+} fragment ion was the characteristic fragment ion. The [D+Cu+H]3+ and [D+Cu+3H]5+ complexes were likely to form a monomer-monomer-Cu (M-M-Cu) structure instead of a monomer-Cu-monomer (M-Cu-M) structure.

  • 알킬페놀에톡실레이트 (APEO)는 주로 비이온성 계면활성제로 쓰이고 있으며 세제, 액체연료 및 농약에도 이용이 되고 있다. APEO 자체는 독성물질로 분류되어 있지 않지만, 그 대사생성물질인 단사슬 APEO, 알킬페놀 및 카르복실 유도체등은 폐수 또는 음용수를 염소처리하는 동안 mutagenic ring halogenated derivative를 생성한다. 이들 생성물들은 estrogenic effect를 나타내기 때문에 APEO를 제한 또는 규제하고 있는 추세이다. 본 연구에서는 APEO의 대사체로서 단사슬 APEO 인 4-nonylphenol-di-ethoxylate (NP2EO), 4-octylphenol-di-ethoxylate (OP2EO)를 분석하기 위해서 p-n-nonylphenol-di-ethoxylate-ring-$^{13}C_6$을 내부표준물질로 사용하여 HPLC/ESI/MS를 이용하여 분석하였다. 분석조건은 이온화 용매인 trifluoroacetic acid가 $10{\mu}M$이 되도록 각각 물과 메탄올에 첨가시켜 사용하였다. 물시료 1 L에 진한 황산을 이용하여 pH를 2이하로 조정한 후, 아세톤, 메탄올 및 물 (pH 2)로 활성화시킨 Sep-Pak $C_{18}$에 loading 시킨 후 아세톤으로 용출하여 시료용액으로 하였다. 이 방법으로 농도범위가 20 ~ 500 ng/L 내에서 검량선의 직선성은 r=0.999 (OP2EO)와 0.990 (NP2EO)였으며, 검출한계는 OP2EO는 20 ng/L, NP2EO는 50 ng/L 이었다. 정확도 및 정밀도는 85.8~122.1% 및 8.2~18.8 %로 좋은 결과를 나타냈다. 이 방법은 환경시료로부터 미량의 APEO를 분석하는데 사용될 수 있고, APEO 오염실태 조사에 응용될 수 있을 것으로 사료된다.

LC-MS/MS를 이용한 향사육군자탕의 주요성분의 함량분석 (Quantitative Analysis of Hyangsayukgunja-Tang Using an Ultra-Performance Liquid Chromatography Coupled to Electrospray Ionization Tandem Mass Spectrometry)

  • The aim of this study was to quantitatively analyze for quality assessment of eighteen marker compounds, including homogentisic acid, 3,4-dihydroxybenzaldehyde, spinosin, liquiritin, hesperidin, ginsenoside Rg1, liquiritigenin, ginsenoside Rb1, glycyrrhizin, 6-gingerol, atractylenolide III, honokiol, costunolide, dehydrocostuslactone, atractylenolide II, nootkatone, magnolol, and atractylenolide I, in Hyangsayukgunja-tang using an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer. The column for separation of eighteen marker components were used a UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}$) and kept at $45^{\circ}C$ by gradient elution with 0.1% (v/v) formic acid in water and acetonitrile as mobile phase. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}l$, respectively. The correlation coefficient of all marker compounds was ${\geq}0.9914$, which means good linearity, within the test ranges. The limits of detection and quantification values of the all analytes were in the ranges 0.04-1.11 and 0.13-3.33 ng/mL, respectively. As a result, five compounds, homogentisic acid, 3,4-dihydroxybenzaldehyde, spinosin, liquiritigenin, and atractylenolide I, in this sample were not detected and the amounts of the 13 compounds except for the 5 compounds were $8.10-6736.37{\mu}g/g$ in Hyangsayukgunja-tang extract.

  • 탄수화물 형태의 당뇨치료제인 acarbose 및 그 대사체를 LC/MS를 사용하는 간편하고 감도 좋은 분석방법을 개발하였다. 혈장시료를 주사기 필터로 거른후 다른 시료전처리과정 없이 LC/MS에 직접 주입하여 분석하였다. Acarbose에 대한 검정곡선은 $0.1{\sim}10{\mu}g/m{\ell}$ 농도범위에서 $r^2=0.9963$의 선형성을 나타내었다. ${\alpha}$-Amylase와 ${\beta}$-amylase를 사용한 in vitro 배양 시험을 통해서 생성된 대사체 component-I과 component-II도 정성적으로 확인 가능하였다. 이러한 LC/MS 분석법은 acarbose의 약동력학적 연구는 물론이며 다른 탄수화물 형태 약물의 연구에도 활용이 가능할 것이다.

  • A liquid chromatography-tandem mass spectrometric (LC/MS/MS) method was developed for the determination of a new anti-angiogenic drug KR-31831 in rat plasma. KR-31831 and internal standard, KR-31543 were extracted from rat plasma with dichloromethane at basic pH. A reverse-phase LC separation was performed on Luna C8 column with the mixture of acetonitrile-ammonium formate (10 mM, pH 4.5) (67:33, v/v) as mobile phase. The detection of analytes was performed using an electrospray ionization tandem mass spectrometry in the multiple-reaction-monitoring mode. (omitted)

Quantitation of CP4 5-Enolpyruvylshikimate-3-Phosphate Synthase in Soybean by Two-Dimensional Gel Electrophoresis

  • Changes of CP4 5-enolpyruvylshikimate-3-phosphate synthase (CP4 EPSPS) in the glyphosate-tolerant Roundup Ready soybean were examined using purified CP4 EPSPS produced in cloned Escherichia coli as a control. CP4 EPSPS in genetically modified soybean was detected by twodimensional gel electrophoresis (2-DE) and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and electrospray ionization tandem mass spectrometry (ESI-MS/MS) with databases. CP4 EPSPS in soybean products was resolved on 2-DE by first isoelectric focusing (IEF) based on its characteristic pI of 5.1, followed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) based on its molecular mass of 47.5 kDa. We quantified various percentages of soybean CP4 EPSPS. The quantitative analysis was performed using a 2D software program on artificial gels with spots varying in Gaussian volumes. These results suggested that 2-DE image analysis could be used for quantitative detection of GM soybean, unlike Western blotting.

Stabilization of Compact Protein Structures by Macrocyclic Hosts Cucurbit[n]urils in the Gas Phase

  • Characterization of intact protein structures in the gas phase using electrospray ionization combined with ion mobility mass spectrometry has become an important tool of research. However, the biophysical properties that govern the structures of protein ions in the gas phase remain to be understood. Here, we investigated the impact of host-guest complexation of ubiquitin (Ubq) with macrocyclic host molecules, cucurbit[n]urils (CB[n]s, n = 6, 7), on its structure in the gas phase. We found that CB[n] complexation induces the formation of compact Ubq ions. Both CB[6] and CB[7] exhibited similar effects despite differences in their binding properties in solution. In addition, CB[n] attachment prevented Ubq from unfolding by collisional activation. Based on the experimental results, we suggest that CB[n]s prevent unfolding of Ubq during transfer to the gas phase to promote the formation of compact protein ions. Furthermore, interaction with positively charged residues per se is suggested to be the most important factor for the host-guest complexation effect.

축산물 중 잔류 성장보조제 분석을 위한 액체크로마토그라피-질량분석법 개발 및 적용 (Development of Analytic Methods for Veterinary Drug Residue in Animal Products by Liquid Chromatography-Mass Spectrometry)

  • 본 연구에서는 동물식품 내 잔류하는 동물의약품 중 성장보조제인 에스트라디올(Estradiol-17��), 테스토스테론(Testosterone), 프로게스테론(Progesterone)에 대한 분석법을 개발하고자 하였다. 분석대상 물질은 액체크로마토그라피를 사용하여 분리하였고, electrospray ionization(ESI) 과정을 거쳐 질량분석기에 주입되어 multiple reaction monitoring(MRM) 모드로 검출하였다. 개발된 분석법은 CODEX CAC/GL 71-2009에 근거하여 유효성을 검증하였고, 분석의 실효성 허용범위를 충족함을 확인하였다. 국내 유통되는 소고기, 돼지고기, 닭고기에 대해 확립된 분석법으로 분석을 진행하여 실제시료 적용성을 확인하였다. 이를 통해 개발된 분석법이 국내 유통 축산물에서 확인되고 있는 성장보조제 일부에 대해 신속하고 신뢰성 높은 분석이 가능함을 확인하였다. 후속 연구를 통해 확립된 분석법을 기반으로 분석대상 성장보조제의 범위를 확장하고, 이를 모두 포함시켜 동시분석법을 확립한다면 활용성이 높은 분석법이 완성될 것으로 확신한다.
