Isozymic Variation of Local Populations in the 3 Species of Phyllostachys (왕대속 3종의 지역개체군간 동위효소변이)

  • Intraspecific and interspecific isozyme variations and their relationship of 16 local populations in 3 species of Phyllostachys, that is, P. bambusoides, P. nigra var. henonis and P. pubescens were investigated by multi-variate analysis. Leaf isozymes of Phyllostachys such as 6-PGD (6-phosphogluconate dehydrogenase), MDH (malate dehydrogenase), PGI (phosphoglucoisomerase), PRX (peroxidase), PGM (phosphoglutamase), IDH (isocitrate dehydrogenase) showed electrophoretic variations in the number of zymotypes (7, 6, 6, 9, 3 and 5, respectively). In the cluster analysis on the isozymic characteristics, 16 populations were classified into 3 species at the euclid genetic distance of 2.041. P. nigra var. henonis and P. bambusoides were clustered first at 2.813 and then P. pubescens at 3.001. So far, 3 local types of intraspecific ariation were found in P. nigra var. henonis and P. bambusoides.

Studies on the Varietal Differences of the Electrophoretic Protein Patterns in the Pupal Haemolymph of the Silkworm, Bombyx mori (가잠용에 있어서 체액단백질의 품종간 차이에 대하여)

  • In order to know the similarity of the electrophoretic protein patterns, distributional aspects of the frequency in each protein band, differences of protein patterns between female and male, and the number of the total protein bands of each variety in the haemolymph of the silkworm pupae according to their geographical origin, the silkworm varieties, and both the sexes, variable electrophoretic protein patterns were observed by polyacrylamide gel electrophoresis, using the silkworm strains which have been reared in the sericultural experiment station. 1. The total number of electrophoretic protein bands classified were twenty-eight, and nearly fourteen bands were shown by each variety on an average. 2. According to their geographical origin, the mean values of the similarity were 20.0%, 28.3% and 14.3% in Japanese, Chinese and European respectively, and the highest similarity was observed between Chinese varieties. 3. The similarity of 129 varieties were tested and it was 22.5%, 23.5% in female and male respectively. 4. The electrophoretic protein patterns in 129 varieties were tested and variation was found in 109 patterns in female and 112 in male. 5. The frequency of the each specified protein band in female and male having variation in protein bands in each variety was found averagely 6.6%. 6. The distribution frequency of the classified 28 protein bands in both the sexes within each geographical origin was variable from 0% to 100%. 7. Among the classified 28 protein bands, HP 11 protein band was found common in all the varieties tested. 8. HP 3 and HP 4 protein bands in female showed high density in electropherograms than in male.

  • We used the electrophoretic deposition (EPD) process to fabricate a glass composite and investigated the EPD parameters to find the optimum deposition time by understanding the relationship among the process parameters of zeta potential (ZP), pH, deposition yield and saturation point. A binder and a dispersing agent were mixed properly with glass frit ($0.2{\sim}25{\mu}m$, d50=$8.77{\mu}m$) in an ethyl alcohol medium for the preparation of the slurry. The pH and ZP were in an inverse relationship to each other due to the generation of $H_3O^+$ ions with the addition of the dispersing agent in the slurry. The acidic nature of the dispersing agent was resulted in a decrease of the pH and an increase of the ZP. Otherwise, the pH increased with the addition of the glass frit in the slurry because $H_3O^+$ ions were absorbed on the glass frit. Therefore, the $OH^-$ ions correspondingly increased. The saturation point of EPD was strongly correlated with the variation of the pH in the slurry; this is caused by a chemical reaction between the ethyl alcohol and the ions that make up the glass frit. An adjustment of the pH variation and the saturation point in the slurry can be established with respect to the optimum deposition time in the slurry.

  • This study examined carbon layer coating on silicon carbide (SiC) fibers by utilizing solid-state and wet chemistry routes to confer toughness to the fiber-reinforced ceramic matrix composites, as an alternative to the conventional pyrolytic carbon (PyC) interphase layer. Electrophoretic deposition (EPD) of carbon black nanoparticles using both AC and DC current sources, and the vacuum infiltration of phenolic resin followed by pyrolysis were tested. Because of the use of a liquid phase, the vacuum infiltration resulted in more uniform and denser carbon coating than the EPD routes with solid carbon black particles. Thereafter, vacuum infiltration with controlled variation in phenolic resin concentration, as well as the iterations of infiltration steps, was improvised to produce a homogeneous carbon coating having a thickness of several hundred nanometers on the SiC fiber. Conclusively, it was demonstrated that the carbon coating on the SiC fiber could be achieved using a simpler method than the conventional chemical vapor deposition technique.

Binder-free Sn/Graphene Nanocomposites Prepared by Electrophoretic Deposition for Anode Materials in Lithium Ion Batteries

  • Nanocomposites consisting of Sn nanoparticles and graphene oxide (GO) were electrophoretically deposited onto Cu current collectors that was used for anodes in Li ion batteries (LIBs). In order to optimize the electrochemical performance of nanocomposites as an anode material by controlling the oxygen functionality, the GO was subjected to $O_3$ treatment prior to electrophoretic deposition (EPD). During thermal reduction of the GO in the nanocomposites, the Sn nanoparticles were reduced in size, along with the formation of SnO and/or $SnO_2$ at a small fraction, relying on the oxygen functionalities of the GO. The variation in the duration of time for the $O_3$ irradiation resulted in a small change in total oxygen content, but in a significantly different fraction of each functional group in the GO, which influenced the Sn nanoparticle size and the amount of SnO (and/or $SnO_2$). As a result, the EPD films prepared with the GO that possessed the least amount of carboxylic groups (made by treating GO in an $O_3$ environment for 3 h) showed the best performance, when compared with the nanocomposites composed of untreated GO or GO that was $O_3$-treated for a duration of less than 3 h.

  • A basic study of the effect of the SDS on the behavior of the anionic surfact in electrophoresis was performed. Having been interacted with SDS micelle, long chain carboxylates and sulfonates which have more than 10 carbons have been suffered larger changes in their apparent mobilities. Based on the result of the comparison among apparent, electroosmotic and electrophoretic mobilities, it is demonstrated that the major cause for the variation of the apparent mobilities of the anions is the variation of the electrophoretic mobilities.

  • Four kinds of Nicotiana species and five varieties belonging to N. tabacum were used as materials for electrophoretic analysis of the tetrazolium oxidase isozyme to examine the taxonomic affinity among them based on the biochemical property. All the five verieties of N. tabacum showed same isozyme bands despite the fact that these varieties had notably varied characteristics including morphological traits. The band patterns were more or less different among the four species. Although N. rustica and N. tabacum were of the same genome group of AABB, their isozyme bands showed considerable difference. N. sylvestris, genome A donor of Nicotiana species, was found to be markedly different from N. tatacum in band pattern, including the absence of system 2 in N. sylvestris.

  • Acid mucopolysaccharides were isolated from nucleus pulposus of whale embryo. The separation of acid mucopolysaccharides was most excellent in $0.03{\%}$ hexamine cobaltic chloride in 0.05 M-sodium acetate buffer solution at pH 4.8. Changes in electrophoretic mobility of acid mucopolysaccharides were observed when the sample solution on top of spacer gel were covered with $40{\%}$ sucrose solution. This effect was not observed by the addition of hexamine cobaltic chloride to the buffer solution.

  • A carbon paste electrode was constructed with peroxidase extracted from Horseradish and the variation of the response of the sensor with pH was investigated. Current profiles showed two highest sensitivities at two pH values respectively. In addition, two bands were observed in the electrophoretic expansion. A coincidence of the two experimental results added support to the possibility that the biosensor has two different isozymes. Assuming that current profiles are the sum of two gaussians, we deconvoluted them and determined the optimum pH of peroxidase isozymes.

  • Bowman-Birk type protease inhibitors of soybeans were purified with gel filtration on Sephadex G-75. Significant differences were found in the content and in the electrophoretic patterns of the purified protease inhibitors. Ten among fourteen electrophoretic bands appeared as protease inhibitors. The chymotrypsin and trypsin inhibiting activities in soybeans showed three and four fold variation respectively. And the cultivars with higher chymotrypsin inhibiting activities seemed to have higher cysteine contents.
