A Systematic Study of the Theaceae 6 Species in Korea (한국산(韓國產) 차나무과(科) 6종(種)의 계통(系統) 분류학적(分類學的) 연구(硏究))

  • This study was carried out to clarify a taxonomical relationships of the Korean Theaceae using characters from morphological, anatomical, electrophoretic and numerical methods. The results are summarized as follows ; Morphological data were cluster analysis by Euclidean distance, the complete and average linkage cluster were most distinctly classified into subfamily level. At the principal components analysis(PCA), the commutative contribution rate of three principal components showed to 91.1% total variance. By the leaf venation were classified semicraspedromous type of Theoideae and brochidodromous type of Ternstroemioideae. The stomatal types were classified Paracytic of Theoideae and Anomocytic type of Ternstroemioideae ; the former has founded subsideary cell the latter has not found. All taxa possessed common isozyme bands did not found out of Theaceae banding patterns. But, the activity of Theoideae were existed in below No.5(Rf. 4.0-4.4), in contrast to Ternstroemioideae were existed in more than No.7(Rf. 5.7-6.2). The cluster analysis of leaf characters and peroxidase isozymes were similarity between two methods.

  • Prunus tomentosa Thunberg seed was investigated to evaluate its possibility for use as food resources of fats and proteins. The seed contained 40.38% of crude fat and 26.59% of crude protein. The lipid fractions obtained by silicic acid column chromatography were mainly composed of 95.49% of neutral lipids, whereas compound lipids were only 4.51%. Among the neutral lipid components by thin-layer chromatography, triglycerides were 89.86%, sterols, monoglycerides, sterol esters, free fatty acids and diglycerides were 4.14%, 2.98%, 1.77%, 1.07%, and 0.18%, respectively. Oleic acid (65.06-66.05%) and linoleic acid (26.56-28.40%) were the main fatty acids in the total lipid, neutral lipid and triglyceride fractions. In the glycolipid and phospholipid fractions, predominant fatty acids were oleic acid (40.55-51.46%), linoleic acid (20.26-30.89%) and palmitic acid (17.64-21.43%). The extractability of salt soluble protein of seed was 60%, and recovery rate of main protein fraction separated by Sephadex G-200 was about 46.5%. The electrophoretic analysis showed 7 bands in seed protein.

Effects of Ripening Temperature on Starch Structure and Storage Protein Characteristics of Early Maturing Rice Varieties during Grain Filling (조생종 벼의 평야지 재배에 따른 등숙 온도 차이에 의한 전분구조 및 종자 저장단백질 특성 연구)

  • This study was performed to understand the effects of filling stage temperature on the characteristics of starch and storage protein and the quality of rice grains. Eight early maturing rice varieties were cultivated in Cheolweon (latitude $38^{\circ}15^{\prime}N$) and Suwon (latitude $37^{\circ}16^{\prime}N$) areas in Korea. Rice grown in Suwon, with relatively high ripening period temperatures, showed significantly reduced head rice ratio and eating qualities, higher protein and lower amylose contents than rice grown in Cheolweon. In rice that ripened under high temperature conditions, the starch contained significantly less short-chain amylopectin (DP < 12) but more intermediate- (DP 13-24) and long- (DP > 25) chain amlylopectin compared to rice that ripened under normal conditions. In addition, the electrophoretic pattern of rice storage protein under high- temperature conditions revealed decreased prolamin and increased glutelin contents.

Fractionation and Physicochemical Characteristics of Caprine Casein by DEAE-Cellulose (DEAE-Cellulose에 의한 산양유 Casein의 분별 및 이화학적 성질에 관한 연구)

  • This experiment was carried out to study fractionation and physicochemical characteristics of caprine casein. Acid caseins obtained from caprine colostral and normal milk were analyzed by chymosin treatment and fractionated by DEAE-cellulose column chromatography with linear gradient and electrophoresis. Protein, fat and lactose of caprine normal milk were 2.70${\pm}$0.27%, 3.82${\pm}$0.51%, and 4.10${\pm}$0.29%, respectively. More non-protein nitrogen(NPN) was released by chymosin treatment from caprine colostral casein than normal casein. The electrophoretic pattern of caprine casein was not similar to that of bovine casein, Caprine normal casein was fractionated by DEAE-cellulose column chromatography with a 0.08${\sim}$0.18 M NaCl linear gradient into 5 pes with the proportion of 5.27%, 26.07%, 25.97%, 30.40% and 12.29%, respectively. In order to identify the pure fraction, the chymosin-treated caprine normal casein was fractionated by DEAE-cellulosecolumn chromatography with a 0.08${\sim}$0.18 M NaCl linear gradient into 6 peaks with the proportion of 17.06%, 9.10%, 17.85%, 20.11%, 27.03% and 8.85%, respectively.

  • The multiple 7S globulins composed of two fractions (A and B) in the electrophoresis with Davis' method were isolated at different stages of the soybean seed development. Electrophoresis of their subunits liberated in PAWU solvent [phenol-acetic acid-water (2 : 1 : 1) solution plus 5M urea] yielded 4 major bands. Observation of both the electrophoretic bands of the multiple 7S fractions(7S-A and 7S-B) and those of their subunits was suggestive of a similarity of the subunit pattern between two 7S fractions. The two fractions in multiple 7S globulins were isolated with DEAE-Sephadex A-50 column$(2.0{\sim}100cm)$ chromatography. They were separated into 2 fractions in a linear gradient concentration of 0.28 to 0.40M NaCl with phosphate buffer (pH 7.8) containing 10mM ${\beta}-mercaptoethanol$(ME). The isolated protein was dissociated into subunits with two different solvent systems; in PAWU solvent and in Tris-HCl buffer(pH 8.0) containing 1% sodium dodecyl sulfate (SDS) and 40mM ME. The dissociated subunits were subjected to electrophoresis in PAWU-treated 7.5% acrylamide gel and in 1% SDS-treated 5.6% acrylamide gel. In PAWU gel electrophoresis, total 7S globulin was separated into 5 major bands, two of which were occupied in common by two 7S fractions(7S-A and 7S-B). In SDS gel electrophoresis, total 7S globulin was separated into 7 major bands, three of which were overlapped with the subunit of the two 7S fractions. The above results alluded us to the presence of a common and/or similar subunit between the multiple 7S globulins.

  • The composition of four rice protein groups is greatly affected by the extraction conditions. The extraction amounts of albumins and glubulines primarily depended on the temperature rather than the method of extraction. The total amount of glutelins, the major components of rice storage proteins, could be extracted by a successive extraction processes, extraction with 0.5% SDS-0.1M borate buffer(pH 8.3) followed by extraction with 0.5% SDS-0.6% ${\beta}-mercaptoethanol-0.1M$ borate buffer(pH 8.3). The extracted amounts of glutelin with these solvents were 54.1 and 45% respectively. The further purification of SDS soluble glutelins was achieved by Sephadex G-150 gel column chromatography. The molecular weight of the components in four protein groups has been estimated by SDS-polyacrylamide gel electrophoresis with or without ${\beta}-mercaptoethanol.$ The comparison of albumins and globulins by starch gel electrophoresis at pH 3.1 permitted us to identify seven rice varieties. However, at pH 8.95, the specific bands for Japonica type rice varieties were observed.

Genetic variation in five species of Korean Orostachys (Crassulaceae) (한국산 바위솔속(돌나물과) 5종에 대한 유전적 변이)

  • Starch gel electrophoretic studies using 24 populations of five Korean Orostachys species were conducted to investigate allozyme variation and to test hypotheses of systematic relationships among species. The resulting phenogram showed that the populations of five Korean Orostachys species were divided into two major groups. And they were concordant with molecular and morphological data in suggesting that Orostachys was divided into two groups corresponding to the subsect. Appendiculatae and subsect. Orostachys. The low genetic identities among Korean Orostachys species indicated that the species of Orostachys have diverged gradually through the model of geographical species. Comparing the previous genetic data from the species with similar life history and mode of reproduction, most of Korean Orostachys species revealed a significant low genetic variation, while the widespread O. japonicus showed a relatively high genetic variation among the Korean species. This kind of genetic variation pattern might be the results of the isolated habitats, limited numbers of individuals within the populations, destruction of habitats, inbreeding and asexual reproduction in Korean Orostachys populations. The Jungdongjin population (POP 21) of O. malocophyllus was genetically unrelated with remaining populations of the same species, and this interpretation was consistent to the results from the previous palynological and morphological studies. Our allozyme data supported the taxonomic treatment of recently proposed taxa, O. iwarenge (Makino) Hara for. magnus and O. margaritifolius.

  • To study affinity of proteolytic enzymes to soy proteins, the physicochemical and functional properties of enzymatically modified protein products, kinetic parameters and degree of hydrolysis were measured using trypsin, alcalase (serine type protease) and pronase. Bacterial alcalase and pronase showed much greater affinity to soy protein than animal intestinal trypsin. This effect was very significant when unheated soy isolate was used as a substrate. Specific activities of these enzymes decreased with the increment of substrate concentration (over 2.0%, w/v) when heat denatured soy protein was used as a substrate. However, the decrease in specific activity was negligible at substrate concentrations lower than 2.0%. Polyacrylamide gel electrophoretic results showed that the pattern of 2S protein band changed distinctly in alcalase hydrolysis as compared with those of trypsin and pronase. Protein solubilities of alcalase and pronase hydrolyzates increased by 25-30%, at their pI (pH 5.0) over the control. Virtually no change was observed in solubility by trypsin hydrolysis. Heat coagulability and calcium-tolerance of the protein increased by enzymatic hydrolysis. No clear tendency, however, was observed for emulsion properties, foam expansion and the amount of free -SH groups. The enzyme treatment considerably decreased foam stability.

  • The purpose of the present study is to investigate the electrophoretic patterns of ADH isozymes and activity at various developmental stages of Oregon-R and Kwangju strains of D. melanogaster. The ADH isozymes were seperated by agarose gel plate eletrophoresis and they were stained for ADH activity study. The optical density was measured by using densitometer and the total activity was determined by measuring the enzyme solution's absorbance of 340 nm by spectrophotometer. The results are as follows: 1. At egg stages of Oregon-R and Kwangju strains ADH 2, ADH 3, ADH 4 were observed in te zymograms, but all of ADH 1, ADH 2, ADH 3, ADH 4 and ADH 5 were detected in larval, pupal and imaginal stages. 2. The amount of ADH 1+ADH 2 (anion) was 20-40% and that of ADH 3+ADH 4+ADH 5 (cation) was 60-805. In the developmental stages other, than egg stages, ADH 3 of both strains showed the largest amount among all ADH isozymes, and especially ADH 4 of Kwangju strain showed less amount than that of Oregon-R strain. 3. The total activity of the ADH at the egg stage was found to be the lowest, at larval stage became to be higher and to be lower at pupal stage and then became to be the highest at the imaginal stage. It is noticeable that the activity of the ADH in Kwangju strain was higher than that in Oregon-R strain.

A comparative Study on the Activities of Glutamic Dehydrogenase and Glutamic Transaminase in Livers of the Crucian carp, Pigeon, and Rat (붕어, 비둘기, 흰쥐의 肝臟의 Glutamic Dehydrogenase 및 Glutamic Transaminase 의 活性에 관한 比較 硏究)

  • The present investigation involves a comparative study of enzymatic activities in various animals. The levels of the liver protein of rat (22.0$\pm$0.01mg/ml and pigeon (22.0$\pm$ 0.16) are twice as high as that of crucian carp (13.0$\pm$ 0.09)(p < 0.01). Generally, the specific acitivity (3.77 $\pm$0.18 unit/mg) of rat glutamic pyruvic transaminase (GPT) is highest, pigeon intermediate (1.93 $\pm$0.01), and crucian carp lowest (0.71$\pm$0.07). On the other hand, the specific activity (8.23$\pm$0.09 unit/mg)of rat glutamic oxaloacetic transaminase (GOT) is highest, pigeon intermediate (3.95$\pm$0.09), and crucian carp lowest (0.92$\pm$0.01) (p < 0.01). Ratios of GOT activity to GPT activity appear no remarkable difference from the levels of various animals. Specific activity of glutamic dehydrogenase(GDH) in pigeon tissue exceeds those of rat and crucian carp. In liver, rat GOT specific activity is greater than crucian carp and pigeon. On the other hand, pigeon GDH specific activity is greater than those of rat and crucian carp. This wouls seem to be in accord with protein metabolic intensity. The patterns for GDH isozyme were remarkably appeared in various animals. Glutamic dehydrogenase isozymes gave different electrophoretic mobilities in various animals. It is interesting that crucian carp, pigeon, and rat would show this difference, which may be indicative of an evolutional pattern. The fact that livers in various animals show quite different enzyme activities would suggest the existence of such a general phylogenetic relationship.
