Antioxidant Ability and Hair Growth Effect of Samhwang-Sasimtang in C57BL/6 Model (삼황사심탕의 항산화능 및 C57BL/6 마우스 모델에서의 발모 촉진효과)

  • The purpose of this study is to evaluate the antioxidant activity and hair growth effects of a natural herbal ethanol extract, Samhwang-Sasimtang(SS). In case of antioxidant ability of SS, the content of phenolic compounds was 28.44mg/g. The extract showed strong electron donating ability and free radical scavenging activity in a concentration-dependent manner. SOD-like activity also rose through increasing the concentrations of SS. In order to estimate the hair growth effects, the extract was applied to the back of seven-week-old C57BL/6 male mice ($150{\mu}{\ell}$ a day, five days a week, for four weeks) in four groups (C, control, saline; PC, positive control, 3% minoxidil; E1, experimental 1, 1% SS; E2, experimental 2, 2% SS). Ten mice were assigned to each group and five mice in each group were sacrificed at weeks 2 and 4, respectively. There was no a significant difference in body weight change among experimental groups. In macroscopic observation of hair growth at week 4, the scores of hair growth on the backs of mice were 80, 60, 40 and 20% in the PC, E2, E1 and C groups. In terms of histological observation, the ratio and thickness of hair follicles, the enzyme activities of ALP and ${\gamma}$-GT, immunohistological examination of IGF-1 and VEGF which are the hair growth factors, in each group were significantly high in this order: PC, E2, E1 and C group, at week 4. Meanwhile, hair growth inhibition factors, TGF-${\beta}1$ and Caspase-3, were reduced in PC, E1 and E2 groups compared with C group. These results indicate that SS extract may be effective in promoting hair growth, and suggest that it can be used practically as a superior natural agent for hair growth promotion.

  • In other previous in vitro tests, the hyphal growth of Raffaelea quercivorus was inhibited by the treatments of various concentration of nano-silver. In this field tests, treatment of different concentrations of nano-silver to oak trees in Cheolwon and Hongcheon sites showed the inhibition effects against wilt disease caused by Raffaelea quercivorus. However, nano-silver-treated oak tree in Chuncheon site showed mild wilt symptoms with no phytotoxicity. Scanning electron microscope (SEM) observation confirmed that the spore and hypha of Raffaelea sp. inside the vessels were damaged by nano-silver. This result indicates the application of nanosilver is effective for control of Raffaelea quercivorus in the field.

  • Electrochemical characteristics and kinetic parameters of copper ion reduction were investigated with a platinum rotating disk electrode (RDE) in a diffusion controlled region. Reduction of Cu(II) in sulfate had one-step two-xelectron process, while the reduction of Cu(II) in chloride solution was involved two one-electron processes. The transfer coefficient of Cu(II) in sulfate solution was lowest, and the transfer coefficient of Cu(I) in halide solutions had the value of nearly one. In chloride solutions, electrodeposition rate of Cu(II) was about one hundred times faster than Cu(I). Diffusion coefficient increased in the order of Cu(II) in chloride solution, Cu(I) in the iodide, bromide, chloride solution, Cu(II) in sulfate solution. The calculated ionic radii and activation energy for diffusion decreased in the same order as above. Morphological study on the copper electrodeposition indicated that the electrode surface became rougher as both concentration and reduction potential increases, and the roughness of the surface was analyzed with UV/VIS spectrophotometer.

  • Due to the attractive benefits with regard to bone health, digestion, and hydration, carbonated water consumption have rapidly grown over the past few years. However, the acidic drink has latent potential for enamel erosion. The most experimental studies about the enamel erosion have focused on the carbonated beverages with sugar and artificial sweeteners. Here, we determined the enamel erosion potential by commercially available carbonated waters with bovine teeth. The erosion was verified by pH value, calcium concentration, and scanning electron microscope. Then plaque accumulation by bacterial adhesion was determined on the enamel erosion surface to measure roughness. In the present study, we observed that the increased calcium content after being immersed in carbonated waters result from the overall enamel erosion. There were no significant differences between general carbonated waters and mineral waters for erosive capacity. Therefore, commercially available carbonated waters are potentially erosive. In addition, oral bacteria strongly adhered to the erosive enamel surfaces thereby facilitating the development of dental plaque. Thus, it is urgently necessary to provide food safety information on the carbonated water as acidic drink to prevent the enamel erosion.

The Effect on Anti-oxidative Activity and Increasing Extraction Yield of Aralia elata Cortex by Gamma Irradiation (감마선 조사에 의한 총목피(Aralia elata Cortex)의 추출수율 증대 및 항산화 효과)

  • This study was designed to investigate the effects on anti-oxidative activities and increasing extraction yield of Aralia elata Cortex by gamma irradiation. Electron spin resonance (ESR) analysis as physical techniques for irradiation identification of Aralia elata showed that a pair of peak appeared on a space of 6.0 mT at the left and right of symmetric unspecific central ESR spectrums, confirming that the plant was gamma-irradiated. The optimum extracting conditions for preparing gamma irradiated samples from Aralia elata Cortex were to extract with 50% ethanol for 15 hrs after 10 kGy irradiation. DPPH scavenging activity and ABTS radical cation inhibitory activity of the water and 50% ethanol extracts from non irradiated and irradiated Aralia elata Cortex was very high as over 80% and 98%, respectively, at tested low concentration of $50{\mu}g/mL$. Antioxidant protection factor (PF) as anti-oxidation indicator of lipophilic compounds showed a very high level of activity as 2.18~2.78 PF. As for TBARs, water and ethanol extracts showed high level. Increase of TBARs inhibitory activity of water extracts was not shown by gamma-ray irradiation but ethanol extracts showed slight increasement of TBARs inhibitory activity with 10 kGy gamma-ray irradiation. These results shown confirmed increasement of extraction yield for phenolic compounds and anti-oxidative activity from Aralia elata. Thus, the treatment of gamma-irradiation can be used a way to amplify a solubility for biological active compounds and anti-oxidative activity in plants.

Iron Phosphate Coating on Pyrite Surface for Reduction of Acid Rock Drainage (산성배수 발생저감을 위한 황철석 표면의 철인산염 피막형성 연구)

  • Acid drainage occurs when sulfide minerals are exposed to an oxidizing environment. The objective of this study was to examine the optimum condition for creating a phosphate coating on standard pyrite surfaces for reduction of pyrite oxidation. The solution of $10^{-2}M\;KH_2PO_4,\;10^{-2}M\;H_2O_2$ was identified as the best phosphate coating agent for the reduction of pyrite oxidation. The formation of an iron phosphate coating on pyrite surfaces was confirmed with ore microscope and scanning electron microscope equipped with energy dispersive spectroscopy. The temperature did not significantly affect the formation of phosphate coating on the surface of pyrite. However, the phosphate coating was less stable at higher temperature than at lower temperature. The phosphate coating was quitely stable at wide range of pH and $H_2O_2$ concentration. The less than $3.4\%$ of phosphate was dissolved at pH 2.79 and 10.64 and less than $1.0\%$ of phosphate was dissolved at 0.1M $H_2O_2$. On the basis of these results, the phosphate coating can effectively reduce the negative environmental impact of acid rock drainage.

Experimental Evaluation of an Analytical Method for Chlorofluorocarbons (CFSs) in Air and Water Using Gas Chromatography (가스 크로마토그래피를 이용한 대기와 물시료의 CFCs(chlorofluorocarbons) 분석법의 실험적 평가)

  • CFC(CFC-12, CFC-11 and CFC-113) analytical system for air and water was constructed using a customized purge and trap extraction device and a gas chromatograph with an electron capture detector. Sampling methods of air and water for CFCs were also established. The analytical system was experimentally optimized to result in reproducibilities of triplicates less than 2% for current air samples and less than 5% for groundwater samples with CFC-12 concentration of 160 to 180 pg/kg, and verified with respect to the CFC system in USGS, which showed analytical results were in agreement within 10%. CFCs in air were monitored at three sites over 19-month period in the central part of South Korea, and the result indicates no significant local sources of CFCs in those areas. For groundwater in Jeju Island, CFCs were measured over a year with a two-month interval. The time-series data showed seasonal fluctuations which could be interpreted by the effect of recharge pulse derived from large amount of rainfall during monsoon period with a few month delay, which indicates high permeability of basaltic rocks in Jeju Island.

Growth of GaAs/AlGaAs structure for photoelectric cathode (광전음극 소자용 GaAs/AlGaAs 구조의 LPE 성장)

  • In this paper, GaAs/AlGaAs multi-layer structure was grown by liquid phase epitaxy with graphite sliding boat, which can be used as a device structure of a photocathode image sensor. The multi-layer structure was grown on an n-type GaAs substrate in the sequence as follows: GaAs buffer layer, Zn-doped p-type AlGaAs layer as etching stop layer, Zn-doped p-type GaAs layer, and Zn-doped p-type AlGaAs layer. The Characteristics of GaAs/AlGaAs structures were analyzed by using scanning electron microscope (SEM), secondary ion mass spectrometer (SIMS) and hall measurement. The SEM images shows that the p-AlGaAs/p-GaAs/p-AlGaAs multi-layer structure was grown with a mirror-like surface on a whole ($1.25mm{\times}25mm$) substrate. The Al composition in the AlGaAs layer was approximately 80 %. Also, it was confirmed that the free carrier concentration in the p-GaAs layer can be adjusted to the range of $8{\times}10^{18}/cm^2$ by hall measurement. In the result, it is expected that the p-AlGaAs/p-GaAs/p-AlGaAs multi-layer structure grown by the LPE can be used as a device structure of a photoelectric cathode image sensor.

A Study on the Content Variation of Metals in Welding Fumes (용접흄 충 금속함량 변화에 관한 연구)

  • Concentration of welding fumes and their components is known to be hazardous to welder and adjacent worker. To determine the generation rates of metals in fumes, $CO_2$ flux cored arc welding on stainless steel was performed in well designed fume collection chamber. Variables were different products of flux cored wire(2 domestic products and 4 foreign products) and input energy(low-, optimal- , high input energy). Mass of welding fumes was determined by gravimetric method(NIOSH 0500 method), and 17 metals were analysed by inductively coupled plasm-atomic emission spectroscopy(NIOSH 7300 method). Flux cored wire tube and flux were analysed by scanning electron microscopy to determine their metal composition. 17 metals were classified by their generation rates. Generation rates of iron, manganese, potassium and sodium were all above 50mg/min at optimal input energy level. Generation rates of chromium and amorphous silica were 25~50mg/min. At 1~25mg/min level, nickel, titanium, molybdenum, and aluminum were included. Copper, zinc, calcium, lead, magnesium, lithium, and cobalt were generated below 1 mg/min. Generation rates of metal components in fumes were influenced by input energy, types of flux cored wire. Flux cored wire was consisted of outer shell tube and inner flux. Iron, chromium, and nickel were the major components of outer tube. Flux contained iron, chromium, nickel, potassium, sodium, silica, and manganese. The use of flux cored wire can increase the hazards by increasing the amounts of fumes formed relative to that of solid wire. The reason might be the direct transfer of elements from the flux, since the flux is fine power. Ratio of metals to the fume of flux cored wire was lower than that of solid wire because non-metal components of flux were transferred. Total metal content of fumes in flux cored arc welding was 47.4(24.3~57.2) percent that is much lower than that of solid wire, 75.9 percent. We found that generation rates of iron, manganese, chromium and nickel, all well known to cause work related disease to welder, increased more rapidly with increasing input energy than those of fumes. To reduce worker exposure to fumes and hazardous component at source, further research is needed to develop new welding filler materials that decrease both the amount of fumes and hazardous components.

Characteristics and Antioxidative Activity of Fermented Mixed Grain Beverages Produced by Different Microbial Species (잡곡발효물의 제조와 항산화 활성 비교)

  • The purpose of this study was to evaluate the functional characteristics of fermented mixed grain beverages obtained using different microbial species and to evaluate their suitability for consumption. Various fermented mixed grain beverages were prepared through fermentation with Aspergillus (A.) oryzae CF1003 (A), A. acidus KACC46420 (B), Rhizopus (R.) delemar KACC46149 (C), R. oryzae KACC45714 (D), R. oryzae KACC46148 (E), A-E mixed strains (F), A. oryzae CF1001 (G), A. acidus CF1005 (H) and A+H mixed strains (I). The visual appearance, flavor, taste, and the antioxidant capacity of each fermented beverage were then assessed. The chromaticity and aesthetic quality of the fermented beverage was measured and all fermented beverages appeared yellow. The C-, G-, H- and I-fermented beverages received scores of 3.319, 3.206, 3.170 and 3.025 points, respectively, following a sensory evaluation, while the others received less than 3 points. The polyphenol content of the different beverages were similar, while the flavonoid content significantly differed. In particular, the flavonoid content of the C- and E-fermented beverages was significantly higher than other beverages. Although the electron donating ability and reducing power of the fermented beverages was very low, the superoxide dismutase (SOD)-like activity of all beverages (except the E-fermented beverage) increased in a concentration-dependent manner. Specifically, the SOD-like activity from the F-fermented beverage at 10,000 ppm was more than 50%. Interestingly, the antioxidant activities of the beverages were unrelated to their polyphenol or flavonoid levels. This study also found that the aesthetic qualities of G- and H-fermented beverages were the highest and that this was completely independent of their antioxidant capacity. Therefore, our results suggest that further studies are required to develop mixed grain-derived fermented beverages that can also fulfill a useful functional purpose.
