Application of Capillary Electrophoresis for Quality Control Analysis of Complex Medicine (모세관 전기영동 분석법의 복합약물제제의 품질관리 분석에 응용을 위한 연구)

  • Capillary electrophoresis (CE) is perceived as an attractive tool for the analysis of pharmaceuticals and biological materials because of their high separation efficiency, easy separation and low running cost. New concept of micellar electrokinetic capillary chromatography (MECC) expanded the application of CE to the separation of neutral molecules. Validation of CE as an analytical technique for quality control of pharmaceuticals should be confirmed by quantitative analysis and the peak confirmation. In this study, the quantitative analyses of various types of neutral, acidic and basic components (acetaminophen, caffeine, ascorbic acid, riboflavin, thiamine, chlorpheniramine, phenylpropanolamine, dl-methylephedrine and dextromethorphan) in complex cold medicines have been accomplished using CE. Combined methods of MECC using SDS and capillary zone electrophoresis lowering the pH of running buffer were adopted to determine the ingredients in capsule type or liquid formula complex medicines without particular sample pretreatment. The results indicate that CE is a promising technique for quality control analysis of pharmaceuticals as a validation method.

  • Lab-on-a-chip is a miniaturized analytical device in which all of the procedures for the analysis of molecules are carried out, such as pretreatment, reaction, separation, detection, etc. Lab-on-a-chip has increasing concern as a device not only for rapid detection of molecules but also for high throughput screening and point of care, because conventional laborious and time consuming analytical procedures can be substituted. Thus, a lot of microfabrication and analytical techniques for lab-on-a-chip have been developed with microstructures smaller than a few hundreds of micrometers. Separation of the molecules is one of the most important components of lab-on-a-chip, because effective separation method can simplify the design and can provide better sensitivity. The electrokinetic separation based on capillary electrophoresis is most widely employed technique in lab-on-a-chip for the control of fluids and the separation of molecules. In this article, bioseparation techniques and its applications realized in lab-on-a-chip are reviewed.

  • 최근 에너지에 대한 관심의 증대 및 센서 노드로의 개발을 위해 무전원 동력 장치(sustainable energy conversion system)에 대한 관심이 크게 증대되고 있다. 본 연구에서는 수압(hydraulic pressure)을 이용하여 전기를 발생시키는 새로운 개념의 나노유체 에너지 변환 시스템에 대한 연구를 진행하였다. 표면 패터닝 기법을 통해 제작된 나노 채널 및 일차원 마이크로 유체 기반의 플루이딕 소자를 이용하여 외부저항, 버퍼용액의 농도, 압력에 따른 streaming potential을 구하였다. electrokinetic 현상과 이에 따른 유체의 streaming potential을 이용하여 압력(pressure)을 전기적으로 변환시키는 에너지 변환용 나노 유체시스템을 본 논문을 통해 제안하고자 한다.

Electrohydrodynamic Flow around a Circular-Cylindrical Rod Submerged in a Dielectric Liquid (비전도성 액체에서의 실린더 막대 주변에 생기는 전기동역학적 유체흐름)

  • The induced-charge electroosmosis (ICEO) is a kind of electroosmotic flow which is generated by the electrical charge induced by an externally-applied electric field. That kind of electrokinetic phenomenon provides a nonmechanical technique to handle microscale flows and particles. In this work, we report that the ICEO-like flow is observed around two kinds of circular-cylindrical rod submerged in a dielectric liquid. The conductivity of the solution is varied by adding a surfactant. The flow field is visualized by the PIV method, and average flow speed shows a remarkable dependence on electrical input frequency. Interestingly, the characteristics of the flow are quite different from the conventional ICEO with respect to the flow direction and the locations of center of vortices.

  • In the present study, we separated the marker particles from the suspending particle mixture solution using isotacho-electrophoresis technique, a novel quantitative ionic particle separation method, in the microchannel. A multiple stacking zone of the suspending particle was visualized with variations in electric field strength, pH value and concentration of the ionic solution. In particular, the electrophoretic mobility of ionic particle (fluorescein) was estimated based on the electrophoretic velocity value measured by the particle image velocimetry. As a result, isotacho-electrophoresis zones were clearly visualized as going downstream in the electric field. The particle migration velocity increased proportional to the applied voltage increase; it was also affected by the pH value variations in the ionic solution.

  • AC-electroosmosis is one of the electrokinetic forces leading to phenomena peculiar in the microfluidics. This paper shows particle deformation in the microchannel with rectangular electrodes on the bottom wall for the AC-electroosmotic flows. We make a PDMS microchannnel with ITO electrodes To measure velocity distributions of the particles we used a three-dimensional particle tracking velocimetry (micro-PTV) technique this method is Particle tracking by interpolation the diffraction pattern ring diameter variations with the defocusing distances of base particle locations. we induce a function of frequency at the electrode. We find the velocity of particles is the most at the edge of the electrodes and Particles move to side wall or center of the channel for the bottom and middle.

Analysis of Electroosmosis Drainage in Clayey Soil (점성토의 전기삼투 배수에 관한 분석)

  • The characteristics of electroosmosis drainage in clayey soil were investigated, when an electrokinetic technique was applied for the purpose of separating heavy metals in contaminated ground. A series of laboratory tests, considering voltage, zeta potential, pH distribution, and current, were performed for a lead-contaminated kaolin. The results of laboratory tests were compared with numerical analysis of finite difference method. The 1311owing conclusions were obtained: The flow velocity in electroosmosis was very sensitive to the chemical and electrical characteristics of the clay. As the concentration of ion increases, the flow rate decreases and the amount of drainage also decreases as time elapses.

A Field Study on Electrokinetic Removal of Salts from Greenhouse Soil (전기동력학 기술을 이용한 시설재배지 토양 염류제거 실증 연구)

  • A pilot-scale electrokinetic (EK) separation field test ($2{\times}3{\times}0.2m^3$, $W{\times}L{\times}D$) was performed in a greenhouse to remove salts from saline soil. Initially, the greenhouse soil had high electrical conductivity (EC), about 9 dS/m, and contained mainly $Ca^{2+}$, $Cl^-$ and $SO_4^{2-}$ ions. After 2 weeks of EK treatment, the soil EC was reduced to 52% compared with its initial value. The EC reduction was mostly achieved within the first week (47%) due to removal of $Na^+$ and $Cl^-$ ions, but ions with a high adsorption capacity such as $Ca^{2+}$ and $SO_4^{2-}$ ions were difficult to be removed. During the EK test, the soil temperature increased and it reached around $50^{\circ}C$ at some regions. For in situ application to soils in cultivation, the current should be controlled to limit increases in temperature, especially near the cathodes. In conclusion, the in situ EK technique is feasible for the restoration of saline greenhouse soils in or no cultivation and an appropriate strategy is necessary for more effective remediation.

A Study on Salt Removal in Controlled Cultivation Soil Using Electrokinetic Technology (전기동력학 기술을 이용한 시설재배지 토양의 염류제거 효과연구)

  • To verify that the electrokinetic remediation is effective for decreasing salinity of fields of the plastic-film house, field tests for physical property, chemical property, and crop productivity of soils have been conducted. The abridged result of those tests is as follows. In the EK treatment, the electrokinetic remediation has been treated at the constant voltage (about 0.8 V $cm^{-1}$) for fields of the farm household. At this time, an alternating current (AC) 220 V of the farm household was transformed a direct current. The HSCI (High Silicon Cast Iron) that the length of the stick for a cation is 20cm, and the Fe Plate for an anion have been spread out on the ground. As the PVC pipe that is 10 cm in diameter was laid in the bottom of soils, cations descend on the cathode were discharged together. For soil physical properties according to the EK treatment, the destruction effect of soil aggregate was large, and the infiltration rate of water was increased. However, variations of bulk density and porosity were not considerable. Meanwhile, in chemical properties of soils, principal ions of such as EC, $NO_3{^-}$-N, $K^+$, and $Na^+$ were better rapidly reduced in the EK treated control plot than in the untreated control plot. And properties such as pH, $P_2O_5$ and $Ca^{2+}$ had a small impact on the EK. For cropping season of crop cultivation according to the EK treatment, decreasing rates of chemical properties of soils were as follows; $NO_3{^-}$-N 78.3% > $K^+$ 72.3% > EC 71.6% $$\geq_-$$ $Na^+$ 71.5% > $Mg^{2+}$ 36.8%. As results of comparing the experimental plot that EK was treated before crop cultivation with it that EK was treated during crop cultivation, the decreasing effect of chemical properties was higher in the case that EK was treated during crop cultivation. After the EK treatment, treatment effects were distinct for $NO_3{^-}$-N and EC that a decrease of nutrients is clear. However, because the lasting effect of decreasing salinity were not distinct for the single EK treatment, fertilization for soil testing was desirable carrying on testing for chemical properties of soils after EK treatments more than two times. In the growth of cabbages according to the EK treatment, the rate of yield increase was 225.5% for the primary treatment, 181.0% for the secondary treatment, and 124.2% for third treatment compared with the untreated control plot. The yield was increased by a factor of 130.0% for the hot pepper at the primary treatment (Apr. 2011), 248.1% for the lettuce at the secondary treatment (Nov.2011), and 125.4% for the young radish at the third treatment (Jul. 2012). In conclusion, the effect of yield increase was accepted officially for all announced crops.

Emerging Remediation Technologies for the Contaminated Soil/Groundwater in the Metal Mining Areas (금속광산지역 오염 토양/지하수의 복원기술 동향)

  • Pollution reduction and/or control technology becomes one of the pressing post-semiconductor research field to lead an advanced industrial structure. Soil/groundwater remediation techniques may act as a core technology which will create many demands on pollution reduction areas. A plenty numbers of abandoned metal mines were left without any remediation action in Korea, and it may be potential sources of heavy metal and As contamination in the ecosystem. In order to bring this soil contamination to a settlement, the emerging soil/groundwater remediation techniques should be introduced. Main research topics in the United States and Europe move towards the clean remediation technology without any secondary impact and the feasible application of developing technique into the field scale study. With these advantages, several soil/groundwater techniques such as electrokinetic soil processing, permeable reactive barrier, stabilization/solidification, biosorption, soil flushing with biosurfactant, bioleaching and phytoremediation will be summarized in this paper.
