An outbreak of bovine coccidiosis in Korean native cattle (한우 콕시듐병의 발생)

  • Authors observed an outbreak of bovine coccidiosis at a farm stock in Gyeongnam, Korea, which were diagnosed on the ground of clinical and fecal examination with the following results. Twenty one of thirty four Korean native cattles evacuate feces containing blood and mucus or evacuate entirely of clots of fresh and red blood. On fecal examination showed eimeria zuernii. The patients were recovered with sulfamethazine and amprolium for 5 days. From these results the outbreak was considered to be typical bovine coccidiosis.

Epidemiological Survey on Eimeria spp. Associated with Diarrhea in Pre-weaned Native Korean Calves

  • Bovine coccidiosis is one of the most important parasitic diseases affecting calf productivity. Here, we investigated the prevalence of Eimeria spp. in pre-weaned native Korean calves and determined the correlation between diarrhea and Eimeria spp. Fecal samples were collected from individual calves (288 normal and 191 diarrheic) in 6 different farms. Of the 479 samples, Eimeria oocysts were detected in 124 calves (25.9%). Five Eimeria spp. were identified; E. zuernii (18.8%) was the most prevalent, followed by E. auburnensis (12.5%), E. bovis (7.5%), E. subspherica (5.8%), and E. bukidnonensis (1.0%). A significant correlation was observed between diarrhea and mixed infection with more than 2 Eimeria spp. (odds ratio [OR]=2.21; 95% confidence interval [CI]: 1.09-4.49; P=0.03) compared to single infection (OR=1.29; 95% CI: 0.77-2.15; P=0.33). Of the 5 Eimeria spp. identified, E. subspherica (95% CI: 1.24-5.61; P=0.01) and E. bukidnonensis (95% CI: 825.08-1,134.25; P=0.00) strongly increased the risk of diarrhea by 2.64-fold and 967.39-fold, respectively, compared to other species. Moreover, mixed infection with E. auburnensis and E. bukidnonensis was significantly associated with diarrhea (OR=2,388.48; 95% CI: 1,009.71-5,650.00; P<0.00) in pre-weaned native Korean calves. This is the first report to demonstrate the importance of E. bukidnonensis associated with diarrhea in pre-weaned native Korean calves. Further epidemiological studies should investigate the prevalence of E. bukidnonensis and the association between E. bukidnonensis and diarrhea.

Prevalence of internal parasites in housed cattle farms in Gangwon-do (강원지역에서 사육되는 소의 내부기생충 감염실태조사)

  • Parasites cause great economic loss in livestock in Korea, and can be categorized as either or indirect losses. In order to monitor the parasites from October 2005 to January 2006, 401 samples were randomly collected from cattle farms and examined the prevalence of gastro-intestinal (GI) parasites by using the sucrose flotation method and formalin-ether centrifugation method. The overall infection rate of GI parasites was about 43.4%. Of these parasites, nematodes were identified as Capillaria bovis (5.5%), Strongyloides papillosus (2.5%), Trichuris discolor (1.2%). Cestoda was observed only one species, Moniezia benedeni (2.2%). The rate of Eimeria spp was 39.2%. A total of five species of Eimeria was identified; among the species identified as above E bovis and E zuernii appeared with relatively higher infection rates. The combined infection of parasites was 35.2% in single species, 29.0% in double and 2.0% in triple infections. No cases of clinical symptoms were observed in this survey. The EPG levels of each species of parasites were very low in all infected cases.

  • Fecal samples collected from 1,424 cattle were examined for the presence of Coccidia species. A total of eleven species of Eimeria was identified; among the species identified as above E. bovis and E. zuernii appeared with relatively higher infection rates. The infection rate was most prevalent in Korean native cattle (29.07%), followed by the imported beef cattle (18.41%) and the dairy cattle (11. 99%). The number of species within positive samples ranged from one to four.

  • Fecal samples from beef and dairy calves less than 12-month-old of Chonnam area were examined for coccidial oocysts. The percentages of cattle and coccidian species found were: Eimeria alabamensis. 9.4; E. auburnensis, 12.5; E. bovis, 29.9; E. brasiliensis, 1.5; E. bukidnonensis, 2.1; E. canadensis, 10.4; E. cylindrica, 7.5; E. ellipsoidalis, 11.5; E. pellita, 1.7; E. subspherica, 10.4; E. wyomingensis, 2.9; and E. zuernii, 22.5. Fifty five percent of the fecal samples from 521 calves were positive for oocysts, and clinical coccidiosis was not found in any of these calves. The number of species in positive samples ranged from 1 to 7.

Diagnosis of bovine cryptosporidiosis by indirect immunofluorescence assay using monoclonal antibody (단세포군항체를 이용한 간접형광항체법에 의한 송아지 작은와포자충증의 진단)

  • Two hybridoma cell lines against Cwptosporinium possum oocysts nFRl-CN911 were produced. The isotype of these 2 monoclonal antibodies (mAbs) was IgG2b (lE7.2) and and IgM (C6). Enzyme immuno-transfer blotting analysis showed that 157.2 reacted specifically to 36 kDa protein and C6 reacted to 67 and 70 kDa proteins. C. pcnlum was bound specifically to the surface region of oocysts by these mobs. No cross-reactivity was observed with tachyzoites of ToxopLosma gonnii and oocysts of Eimeria zuernii,5. bouis and E. canadensis of bovine origin. The indirect immunofluorescence assay (IIF) using mAb C6 was successful with counterstain. With the IIF using mob C6, oocysts appeared as 3 to $5{\mu}m$ spherical objects fluorescing bright apple green against a reddish dark background. The IIF using mAb C6 was agreed in specificity and sensitivity with those of a commercial diagnostic kit. These results demonstrated that the produced mAbs were specific to C. parvum and that the mAb C6 could be used for diagnosis of cryptosporidiosis.
