Free radical scavenging activity and protective effect from cellular oxidative stress of active compound from eggplant (Solanum melongena L.) (가지(Solanum melongena L.) 활성물질의 라디칼 소거능과 산화적 스트레스에 대한 세포 보호 효과)

  • To investigate the protective effect of eggplant (Solanum melongena L.) and its active compound, delphinidin, we used in vitro and cellular system. The active fraction from eggplant, BuOH fraction, showed protective effect from hydrogen peroxide-induced oxidative stress in WI-38 fibroblast cells. It suggests that eggplant would have the protective activity from radical-induced oxidative damage and its BuOH fraction would play the crucial role with antioxidative activity. In addition, delphinidin, the active compound from eggplant, exerted the strong 1,1-diphenyl-2-picrylhydrazyl scavenging effect with $IC_{50}$ value of 6.59 ${\mu}g/mL$. Furthermore, the cellular oxidative stress was induced by 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) in LLC-$PK_1$ cells, while treatment of delphinidin atteunated AAPH-induced oxidative stress as dose-dependent manner. The present study suggests the antioxidative activity of eggplant and delphinidin against free radical-induced oxidative stress.

Phenolic compounds from the leaves of eggplant (Solanum melongena L.) (가지(Solanum melongena L.) 잎으로부터 페놀 화합물의 분리 및 동정)

  • The leaves of eggplant (Solanum melongena L.) were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH, and water fractions. From the n-BuOH fraction, five compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. On the basis of physic-chemical and spectroscopic data including mass spectrometry, infrared spectroscopy, and nuclear magnetic resonance, they were identified to be caffeic acid (1), chlorogenic acid (2), cryptochlorogenic acid (3), panasenoside (4), and (6R,7E,9R)-4,7-megastigmadien-3-one-9-${\beta}$-${\small{D}}$-glucopyranoside (5). Compounds 3 and 4 were isolated for the first time from the leaves of S. melongena L. in this study.

Inhibitory Effect of Eggplant Extract on Adipocyte Differentiation in 3T3-L1 Cells (가지 물추출물의 3T3-L1 지방전구세포 분화 억제효능)

  • Abnormal growth of adipocyte characterized by increased cell numbers and differentiation is considered as an major pathological characteristic feature in obesity. Thus, inhibition of mitogenesis of preadipocytes and their differentiation to adipocytes would be beneficial for the prevention and inhibition of obesity. In the present study, we attempted to evaluate anti-adipogenic activity of eggplants (the fruits of Solanum melongena L.) employing preadipocytes cell line, 3T3-L1 as an in vitro assay system. Water extract of eggplants significantly inhibited adipocyte differentiation when treated during adipocyte differentiation process, as assessed by measuring fat accumulation using Oil Red O staining. Eggplant extract, however, showed little effects on fully differentiated adipocytes. Eggplant didn't show significant toxicity up to 500 ${\mu}g$/ml to the 3T3-L1 cells. Further studies with interval treatment demonstrated that eggplant exerted inhibitory activity on adipocyte differentiation via acting on early stage of adipogenesis. Conclusively, eggplants are suggested to be beneficial for the prevention of obesity.

  • Yellow spot or blotch symptoms were observed on the upper surface of eggplant (Solanum melongena. cv: Cheonryang) leaves in a commerical vinyl-house of Hapchon-gun, Kyongnam, Korea. We identified Leveillula taurica(L v.) Arnaud as a pathogen causing powdery mildew of eggplant which was observed newly in Korea. The fungal conidia from eggplant leaves were reinoculated to eggplant, tomato and pepper to confirm the same disease as the symptomatology and morphology of the pathogen.

Purification and Characterization of Tyrosinase from Solanum melongena

  • Tyrosinase was purified from Solanum melongena by ammonium sulfate precipitation, Sephadex G-150 and DEAE-Sephacel column chromatography. The molecular weight of the purified tyrosinase was approximately 88,600 daltons with 805 amino acid residues. The amino acid composition showed the characteristic high contents of glycine, glutamic acid and serine residues. The enzyme had high substrate specificity towards (+)-catechin. The $K_m$, value for L-DOPA was 20.8 mM. L-ascorbic acid, ${\beta}-mercapto-ethanol$, sodium diethyldithiocabamate, KCN and $NaN_3$ had strong inhibitory effects on enzyme activity. Sodium diethyldithiocabamate was a competitive inhibitor of the enzyme with a $K_i$ value of $5.2{\times}10^{-2}\;mM$. The optimum pH of the enzyme was 9.0 and the optimum temperature was $65^{\circ}C$ with L-DOPA as a substrate. In addition, the activity was enhanced by addition of $Ca^{+2}$ or $Cu^{+2}$, but decreased in the presence of $Fe^{2+},Fe^{3+}$ and $Zn^{2+}$ ions.

1-Methylcyclopropene and Carbon Dioxide Absorber Reduce Chilling Injury of Eggplant ($Solanum$ $melongena$ L.) during MAP Storage (1-Methylcyclopropene과 이산화탄소 제거제 처리에 의한 가지($Solanum$ $melongena$ L.) 과실의 MAP 저장 중 저온장해 경감)

  • This study was conducted to extend the postharvest life of eggplant ($Solanum$ $melongena$ L.) by modified atmosphere packaging using a OPP bag with 1-MCP to evaluate the alleviation effects of that treatment on external chilling injury during at low temperature. 1-MCP treatment at the level of $1.0{\mu}L{\cdot}L^{-1}$ were very effective in delaying the progress of surface bronzing of eggplant during 10 days of storage at $8^{\circ}C$, but not significant at $1^{\circ}C$. Also, the treatment of 1-MCP at the levels of $0.1{\sim}1.0{\mu}L{\cdot}L^{-1}$ are beneficial to complement low temperature storage and maintain quality indices such as higher firmness and lower weight loss than untreated control even at a critical low temperature 4 to $8^{\circ}C$. The treatment of carbon dioxide using dry ice (DI, 5 g/3 fruits) significantly promoted chilling injury of eggplant stored at $4^{\circ}C$, symptoms were appeared from 3 days after treatment and combination treatment of 1-MCP did not block the development of chilling injury symptom. Meanwhile, single treatment of $CO_2$ absorber (CA, 5 g/3 fruits) or combination treatment with 1-MCP at the level of $1.0{\mu}L{\cdot}L^{-1}$ prevented effectively the external and internal chilling injury at $4^{\circ}C$ during 6 days and 15 days of MAP storage, respectively. Results suggest that 1-MCP and carbon dioxide absorber treatments delay fruit deterioration and are beneficial to broaden storage temperature of eggplant fruits.

Growth Response of Eggplant (Solanum melongena L.) Using Balanced Manure Nutrients Supply by Fertigation Culture (가축분뇨 맞춤비료를 이용한 관비재배가 가지 (Solanum melongena L.) 재배에서 가지 작물의 생육 반응)

  • The present study was conducted to evaluate fertigation effects of balanced manure nutrients from combined fertilizers on the growth of eggplant (Solanum melongena L.) and soil chemical properties in greenhouse cultivation. There are 4 treatments for fertigation; animal liquid manure (LM), LM supplemented with chemical ferilizer (LM+CF) to make nutrient-balanced ferilizer, LM supplemented with chemical ferilizer and phosphorus acid (LM+CF+PA), and conventional chemical fertilzer (CCF). Fruit lenth, width, and weight for LM treatment were significantly lower than LM+CF, LM+CF+PA, and CCF treatments. There was no significant differences in fruit length, width, and weight among LM+CF, LM+CF+PA, and CCF treatments. The yield of eggplants per plant for LM treatment (26.2 kg) was decreased by 6% compared to CCF treatment (27.9 kg). The growth and yield of eggplants were not significant different among CCF, LM+CF, and LM+CF+PA treatments. The marketable yield for LM treatment was lowest among 4 treatments and that for LM+CF+PA treatment (26.6 kg) was increased by 8% compared to CCF treatment (24.1 kg). The chemical properties of the soil for CCF treatment, in general, tended to be higher compared to the other treatments. In conclusion, the present study demonstrates that the application of balanced liquid manure combined with mineral fertilizer to soil is considered as a good management practices because it improves, eggplant quality and soil properties.

  • 우리 나라에서의 연간 가지의 1인당 소비량은 100g 내외로 미비하지만, 일본에서는 1인당 약 2kg 내외로 소비하고 있다. 이러한 일본의 수요에 부응하여 우리나라에서는 계약재배 형태로 매년 일본에 수출이 증가되면서 재배면적이 증가되고 있다. 그러나 국내 가지의 생산성은 일본에 비해 크게 미치지 못하고 있는 실정이다. 이러한, 수출을 위한 가지는 재배시기가 동절기에 시설내에서 이루어져 저온과 일조부족과 같은 불량한 환경으로 인해 수량의 감소 및 품질의 저하를 가져온다. (중략)

The Skin Protecting Effects of Ethanolic Extracts of Eggplant Peels (가지 외피 에탄올 추출물의 피부보호효과)

  • We investigated the in vitro antioxidant and antimelanogenesis effects of ethanol extracts from eggplant (Solanum melongena L.) peels. The total phenolics and chlorogenic acid in ethanol extracts were 2,465 mg/100 g and 2.08 mg/100 g, respectively. ABTS radical scavenging activity, ferric reducing/antioxidant power assay, and malondialdehyde (MDA) inhibitory effect of the extracts increased in a dose-dependent manner. In addition, the extracts generally showed strong UV absorption in the range of UV-B (290-320 nm). The $IC_{50}$ of mushroom tyrosinase inhibitory activity of ethanol extracts from eggplant peels was 870 ${\mu}g/mL$. Importantly, the melanin syntheses of B16/F10 melanoma cells were decreased by extracts in a concentration-dependent manner. Overall, these results suggest that eggplant peels can be potentially applied as a anti-melanogenic agent as well as an antioxidant resource.
