Electron Microscopical Description on the Egg Stalk-like Structure of the Rockfish, Sebastes inermis (Teleostei: Scorpaenidae) (볼락 (Sebastes inermis) 난병 유사구조의 전자현미경적 기재)

  • The ovary of the rockfish, Sebastes inermis was cystovarian condition and consists of numerous ovarian lobules. Previtellogenic oocytes were embedded in the connective tissue of the ovarian lobules. As the oocytes grows, it protruded into the ovarian cavity and form grapes cluster-like structure with egg stalk-like structure. Yolk granules and zona radiata were not observed in the embedded early oocytes about 20 $\mu$m in diameter. In the oocytes about 80 $\mu$m in diameter, yolk granules with electron dense were abundant in the ooplasm near the egg stalk-like structure. However, pinocytosis was not obsened in the pore canal system of the zona radiata. In this stage, microfilaments, developed endoplasmic reticula, tubular mitochondria, granular materials with low electron density and active multivesicular bodies were observed in the egg stalk-like structure. This results suggest that the egg stalk-like structure of the rockfish are related with the oocyte attachment and exogenous vitellogenesis of the intial vitellogenic oocyte.

  • Reproductive biology of the female red tongue sole, Cynoglossus joyneri was investigated by means of histological methods. Gonadosomatic index (GSI) was the highest in June. Developmental pattern of oocytes was group-synchronous. Egg stalk and yolk nucleus were observed in the early growing oocytes. Reproductive cycle could be classified into the growing ($February{\~}May$), maturation ($May{\~}June$), ripe and spent ($June{\~}August$), and recovery and resting ($August{\~}February$). The absolute fecundity per individual ($TL 28.1{\~}30.8 cm$) was 2,197 and relative fecundity per g body weight was 18.0.

  • 수서동물 난소의 해부학적인 구조는 종 특이성, 각각의 생식생태 및 계통에 따라 다르다. 보통 난모세포의 난병 (egg stalk)은 이매패류 (Beams and Sekhon, 1966)와 전복류 (Lee, 1974)에서 관찰 할 수 있는 구조로 이들 난모세포의 난황축적과 밀접한 관계가 있다. 경골어류에서 난병구조는 불볼락, Sebastes thompsoni에서 보고된 적이 있으나 (Lee et al., 1998), 이들의 세부적인 구조는 언급되지 못했다. 본 연구는 볼락에서 관찰된 난병구조를 광학현미경과 전자현미경을 통하여 기재하였다. Microstructural Differentiation of the Oocyte in the Abalone Haliotis discus hannai (북방전복(Haliotis discus hannai) 난모세포의 미세구조적 분화)

  • The purpose of this study was to provide basic information on sexual maturity and reproductive biology for the management of biological resources in abalone Haliotis discus hannai. The nucleus of the oogonium occupied about 42% of the cytoplasm, and had a distinctive basophilic chromatin. The cytoplasm of previtellogenic oocytes was homogeneous and the size of nuclear pores increased. Fine granular and vacuolar yolk granules were observed in the cytoplasm of the initial vitellogenic oocyte. In this stage, the egg stalk and jelly membrane began to develop. The nucleus of the active vitellogenic oocyte was located near the animal pole. Yolk granules were strongly acidophilic. Lampbrush chromosomes were observed in the nucleus and rough endoplasmic reticulum. Annulate lamellae developed in the cytoplasm. The shape of the ripe oocyte was rounded polygonal. The size of ripe oocytes was 202.9±21.40×142.1±18.82 ㎛ and the thickness of the jelly membrane was 10.1±1.52 ㎛. These results show that yolk accumulation in H. discus hannai is based on two methods: exogenous accumulation, through the egg stalk, and endogenous accumulation, through intracellular organelles. Management of biological resources will be necessary when oocytes predominate after the active vitellogenic stage.

  • Gonad structure, germ cell development and reproductive cycle of the smallmouth scorpionfish, Scorpaena miostoma were investigated based on histological method. Samples were collected monthly in the vicinity of Suyoung Bay, Pusan, Korea from November 1995 to October 1996. The testis is seminiferous tubule type in internal structure. Seminiferous tubule consists of numerous testicular cysts which contain numerous germ cells in same developmental stage. The ovary consists of several ovarian lamellae originated from ovarian outer membrane. Oogonia originated from the inner surface of the ovarian lamella protrude to the ovarian cavity in oocyte stage, and they are suspended by the egg stalk. Biological minimum size of female and male were 12.5cm in total length. Gonadosomatic index (GSI) of female (3.81) and male (0.23) were the highest in October. Reproductive cycle was classified into the following successive stages: in female, growing stage $(May\~August)$, maturation stage $(September\~October)$, ripe and spawning stage $(November\~December)$, recovery and resting stage $(January\~April)$, and in male, growing stage $(June\~August)$, maturation stage $(September\~October)$, ripe and spent stage $(November\~January)$ and recovery and resting stage $(February\~May)$.

Ovarian Structure and Oogenesis of the Spiny Top Shell, Batillus cornutus (Lightfoot, 1786) (Gastropoda: Turbinidae) (소라, Batillus cornutus의 난소구조 및 난자형성과정)

  • The ovarian structure and ultrastructural changes in germ cells during oogenesis of the spiny top shell, Batillus cornutus, have been investigated by light and electron microscopic observations. Specimens of the spiny top shell were collected from the subtidal zone of Wando, south coast of Korea. Spiny top shell was dioecious. The ovary was located on the surface of the hepatopancreas in the spiral posterior region. The ovary had greenish color in the gonadal mature season. The ovary comprised many ovarian lobules, and the ovarian lobule consisted of connective mesenchymal tissue and epithelial cells. Oogonium had a large nucleus with nucleolus of high electron density. In previtellogenic oocyte, small yolk granules of low electron density were scattered in the cytoplasm. Oocytes in the initial vitellogenic stage were connected with the ovarian lobule by egg stalk. The result of TEM observations showed that initial vitellogenic oocyte contained well-developed Golgi complex, endoplasmic reticula, tubular mitochondria and numerous yolk granules of various electron densities and sizes. The electron density, size and quantity of yolk granules which were distributed in the active vitellogenic oocyte were increased compared to the previous stage. Thickness of egg envelope in the late active vitellogenic oocyte was approximately 4.4 $[\mu}m$. Cytoplasm of ripe oocyte was filled with proteid yolk globules of high electron density and lipid yolk globules of low electron density. In this stage, the thickness of egg envelope was approximately 6.5 ${\mu}m$.

  • The ultrastructural changes in germ cells during oogenesis of the melania snail, Semisulcospira libertina libertina have been investigated by light and electron microscopy. The ovary is located on the surface of the hepatopancreas in the spiral posterior region. The ovary exhibited greenish color in the gonadal mature season. The ovary was composed of a number of oogenic follicles. Oogenesis was divided into five stages with histological features: (1) oogonia, (2) previtellogenic, (3) initial vitellogenic, (4) active vitellogenic, and (5) mature stages. Oogonia were oval in shape, $4-6\;{\mu}m$ in diameter, and had a large nucleus. Previtellogenic oocytes were about $20\;{\mu}m$ in diameter and the cytoplasm reacted with hematoxylin in H-E satin. Initial vitellogenic stage, oocytes were $60-80\;{\mu}m$ in diameter, and small yolk granules of low electron density are scattered in the cytoplasm. Oocytes in the initial vitellogenic stage were connected with ovarian follicle by egg stalk. Active vitellogenic oocyte were $100-120\;{\mu}m$ in diameter. Electron density, size and quantity of yolk granules that are distributed in the cytoplasm have increased from the previous stage. Result of TEM observations, the oocyte contains well-developed Golgi complex, endoplasmic reticula and tubular mitochondria in the cytoplasm. Cytoplasm of mature oocyte was filled with proteinaceous yolk globules of high electron density. In this stage, the length of microvilli in the egg envelope was approximately $1.1\;{\mu}m$.

  • The developmental stage of germ cells during oogenesis can be categorized into six stages with histological features: (1) oogonium, (2) previtellogenic oocyte, (3) initial vitellogenic oocyte, (4) early active vitellogenic oocyte, (5) late active vitellogenic oocyte and (6) ripe oocyte. The size of oocyte, nucleus and nucleolus illustrated the increase tendency but size ratio of nucleolus to nucleus was decreased during oogenesis. During oogenesis the stainability in the cytoplasm of oocyte changes from basophilic to eosinophilic in H-E stain. And egg stalk and outer jelly membrane was developed in the oocyte. These histological changes are seemed to be yolk accumulation in the oocyte and preparation process for spawning.

Development of the Trichomes in Floating Leaves of Salvinia Species (생이가래속(Salvinia) 부유엽 모용의 분화발달)

  • Salvinia is an aquatic plant forming dimorphic leaves that have been modified into floating and submerged leaves. A air of floating leaves plays an important role for the floating and photosynthesis while the submerged leaves, which are lim and long, have the form and function of root. Many aquatic plants develop trichomes in the epidermis but in Salvinia, richomes grow densely in the epidermis of the dimorphic leaves. The present study examined the differentiation pattern of trichomes developing in the floating leaves of S. natans and S. molesta by scanning and transmission electron microscopy. Trichomes developing in the floating leaves of Salvinia showed very different patterns. In S. natans, they were arranged in a V-shape form, having 20${\sim}$25 rows at $18{\sim}25^{\circ}$ on both sides of the lamina divided by the midrib in the floating leaf. In each row, 8${\sim}$10 oval-shaped cells, $200{\sim}290{\mu}m$ in length, were arranged in a spiral fashion. Four trichomes of this form made a trichome unit, but their apical parts were separated from one another and developed into the so-called 'knuckle-crane' type. On the other hand, in S. molesta, trichomes differentiated in a unique pattern quite different from those of S. natans. At the early stage of differentiation, trichomes protruded from the epidermis and then 4${\sim}$6 cylindrical cells grew $400{\sim}600{\mu}m$ long and the four trichomes formed as an unit. The four grouped trichomes were interconnected through their apex and developed in the 'egg-beater' type. Then $300{\sim}600{\mu}m$ long multi-cellular stalk cells grew and protruded out of the epidermal surface from the basal part of the trichomes. Such a structural characteristic of trichomes is considered to play a very important role along with the aerenchyma tissue in the leaf mesophyll tissue for the floating of Salvinia on the water surface.

  • This study was carried out to investigate insect specles of Kiwi fruit, and ecological characteristicsof Stathmopoda m Chonnam province from 1991 to 1993 Twelve families and thirteen specieswere identified during the study.
