Monitoring soil respiration using an automatic operating chamber in a Gwangneung temperate deciduous forest

  • This study was conducted to quantify soil $CO_2$ efflux using the continuous measurement method and to examine the applicability of an automatic continuous measurement system in a Korean deciduous broad-leaved forest. Soil respiration rate (Rs) was assessed through continuous measurements during the 2004-2005 full growing seasons using an automatic opening/closing chamber system in sections of a Gwangneung temperate deciduous forest, Korea. The study site was an old-growth natural mixed deciduous forest approximately 80 years old. For each full growth season, the annual Rs, which had a gap that was filled with data using an exponential function derived from soil temperature (Ts) at 5-cm depth, and Rs values collected in each season were 2,738.1 g $CO_2$ $m^{-2}y^{-1}$ in 2004 and 3,355.1 g $CO_2$ $m^{-2}y^{-1}$ in 2005. However, the diurnal variation in Rs showed stronger correlations with Ts (r = 0.91, P < 0.001 in 2004, r = 0.87, P < 0.001 in 2005) and air temperature (Ta) (r = 0.84, P < 0.001 in 2004, r = 0.79, P < 0.001 in 2005) than with deep Ts during the spring season. However, the temperature functions derived from the Ts at various depths of 0, -2, -5, -10, and -20 cm revealed that the correlation coefficient decreased with increasing soil depth in the spring season, whereas it increased in the summer. Rs showed a weak correlation with precipitation (r = 0.25, P < 0.01) and soil water content (r = 0.28, P < 0.05). Additionally, the diurnal change in Rs revealed a higher correlation with Ta than that of Ts. The $Q_{10}$ values from spring to winter were calculated from each season's dataset and were 3.2, 1.5, 7.4, and 2.7 in 2004 and 6.0, 3.1, 3.0, and 2.6 in 2005; thus, showing high fluctuation within each season. The applicability of an automatic continuous system was demonstrated for collecting a high resolution soil $CO_2$ efflux dataset under various environmental conditions.

소나무와 굴참나무 임분의 시비에 따른 토양 CO2 방출 동태 (Soil CO2 Efflux Dynamics in Response to Fertilization in Pinus densiflora and Quercus variabilis Stands)

  • 본 연구는 유사한 입지에서 생육한 소나무와 굴참나무 임분을 대상으로 시비에 따른 토양 CO2 방출량을 비교하기 위해 수행하였다. 성숙한 소나무와 굴참나무 임분을 대상으로 시비처리 전 캘리브레이션 기간(2015년 3월~2016년 2월)을 가진 후, 2016년 3월에 복합비료를 표면 시비하였다. 토양 CO2 방출량은 2015년 3월부터 2017년 2월까지 2년 동안 매월 측정하였다. 토양 CO2 방출량의 월별 변동은 굴참나무와 소나무 임분 모두 시비 전(2015~2016년)과 시비 후(2016~2017년) 유사하였다. 연평균 토양 CO2 방출량의 경우 소나무 임분의 시비구는 2015~2016년 1.620 μmol m-2 s-1였으나 2016~2017년 2.180 μmol m-2 s-1로 유의적으로 증가(P < 0.05)하였으며, 굴참나무 임분의 시비구도 2015~2016년 1.557 μmol m-2 s-1에서 2016~2017년 1.977 μmol m-2 s-1로 약 0.420 μmol m-2 s-1 정도 증가하였다. 그러나 비시비구는 두 임분 모두 2015~2016년과 2016~2017년 사이 토양 CO2 방출량에 유의적인 차이가 없었다. Q10 값은 굴참나무 임분의 시비구가 2015~2016년 3.14에서, 2016~2017년 3.41로 증가하였으나 소나무 임분의 시비구는 Q10 값에 변화가 없었다. 비시비구의 Q10 값의 경우 굴참나무 임분은 2015~2016년에 3.85였으나, 2016~2017년은 3.69로 감소하였으며, 소나무 임분은 2015~2016년은 3.15였으나, 2016~2017년은 3.65로 증가하였다. 본 연구 결과에 따르면 유사한 입지환경에서 생육한 소나무와 굴참나무 임분의 토양 CO2 방출량은 시비 후 증가하였으며, 시비에 따른 토양 CO2 방출은 소나무 임분이 굴참나무 임분에 비해 반응이 크게 나타났다.

Regulation of Choline Transport by Oxidative Stress at the Blood-Brain Barrier In Vitro Model

  • In the present study, we examined how the transport of choline is regulated at the blood-brain barrier (BBB) under the central nervous system (CNS) cellular damages by oxidative stress using a conditionally immortalized rat brain capillary endothelial cells (TR-BBB), in vitro the BBB model. It was also tested whether the choline uptake is influenced by membrane potential, extracellular pH, protonophore (FCCP) and amiloride in TR-BBB cells. In result, $[^3H]choline$ uptake was inhibited by FCCP and dependent on extracellular pH. The treatment of TR-BBB cells with 20 ng/mL tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$, 10 ng/mL lipopolysaccharide (LPS), 100 ${\mu}M$ diethyl maleate (DEM) and 100 ${\mu}M$ glutamate resulted in 3.0-fold, 2.6-fold, 1.8-fold and 2.0-fold increases of $[^3H]choline$ uptake at the respective peak time, respectively. In contrast, hydrogen peroxide and raffinose did not show any significant effects on choline uptake. In addition, choline efflux was significantly inhibited by $TNF-{\alpha}$, LPS and DEM producing cell damage states. In conclusion, the influx and efflux transport system for choline existed in TR-BBB cell line and this process was affected by several oxidative stress inducing agents.

급속액상부숙기술(TAO system)을 이용한 양돈분뇨의 연속처리 (Continuous Treatment of Piggery Slurry using the Thermophilic Aerobic Oxidation (TAO) System)

  • TAO system was applied to treatment of piiggery slurry. In the experiments, the volume of influx was differentiated in four types. The features of operation, evaporation rate, and efficiency of successive treatment were drawn. The results are; 1. During the operation, the inside temperature was maintained at over $60^{\circ}C$ with the highest $70.2^{\circ}C$. 2. The evaporation rate of influx volume was 31.4%. And the evaporation volume of per square meter was $108{\;}\ell/\textrm{m}^2$; slightly different from $120{\;}\ell/\textrm{m}^2$ of batch type. 3. The VFAs was decreased by 95% (from $1,538lmg/{\ell}$ in influx to $72.9mg/{\ell}$ in Efflux). Thus, successive operation of TAO system is considered to be possible.

Evaluation of Adverse Drug Properties with Cryopreserved Human Hepatocytes and the Integrated Discrete Multiple Organ Co-culture (IdMOCTM) System

  • Human hepatocytes, with complete hepatic metabolizing enzymes, transporters and cofactors, represent the gold standard for in vitro evaluation of drug metabolism, drug-drug interactions, and hepatotoxicity. Successful cryopreservation of human hepatocytes enables this experimental system to be used routinely. The use of human hepatocytes to evaluate two major adverse drug properties: drug-drug interactions and hepatotoxicity, are summarized in this review. The application of human hepatocytes in metabolism-based drug-drug interaction includes metabolite profiling, pathway identification, P450 inhibition, P450 induction, and uptake and efflux transporter inhibition. The application of human hepatocytes in toxicity evaluation includes in vitro hepatotoxicity and metabolism-based drug toxicity determination. A novel system, the Integrated Discrete Multiple Organ Co-culture (IdMOC) which allows the evaluation of nonhepatic toxicity in the presence of hepatic metabolism, is described.

연안 조류에서 Cd와 Zn 독성의 반대 작용 분석 (Analysis of Oppositive Interaction between Cd and Zn Toxicity in Coastal Algae)

  • The growth and heavy metal experiments revealed oppositive interactions between toxic metals(Zn and Cd) and Mn when the coastal diatom T. pseudonana were used. Cd and Zn inhibited the algal growth rate only at low Mn ion concentrations and this effect could be accounted for an inhibition of cellular Mn take by the toxic metals. Mn and Zn inhibited cellular Cd take and this indicated a reciprocal effects among the metals with respect to metal take. Saturation kinetics modeling of the take data was consistent with two metals competing with each other for binding to the Mn take system and with both Cd and Mn being transported into the cell by that system. Mathematical modeling of Mn and Cd take data revealed evidence fur a Cd efflux system.

  • Several recent studies demonstrate that receptor-mediated cAMP (adenosine 3',5'-monophosphate) production evokes marked change in magnesium ($Mg^{2+}$) homeostasis. The effects of dimaprit or/and phosphodiesterase (PDE) inhibitors on the $Mg^{2+}$ release from perfused guinea pig heart and collagenase-dispersed myocytes was studied to clarify an association of $H_2-histaminergic$ receptor-mediated $Mg^{2+}$ regulation with intracellular cAMP-degradation system. $Mg^{2+}$ efflux was stimulated in perfused hearts and myocytes by IBMX (3-isobutyl-1-methylxanthine), a calmodulin-sensitive PDE inhibitor, but not by RO 20-1724(4-(3-butoxy-4-methoxybenzyl)-2-imidazolidinone) or papaverine, cAMP-specific PDE inhibitors. $Mg^{2+}$ efflux was also be induced by dimaprit, a H-2-agonist. $Mg^{2+}$ effluxes induced by dimaprit were augmented by the presence of the PDE inhibitors. The augmentation of dimaprit-induced $Mg^{2+}$ effluxes by the PDE inhibitors were inhibited by ranitidine, a $H_2-antagonist$, and imipramine, a $Na^{+}-Mg^{2+}$ exchange inhibitor, in perfused hearts and myocytes and were also inhibited by amiloride in perfused hearts. These results suggest that the $H_2$-stimulated $Mg^{2+}$ effluxes from guinea pig heart can be regulated by the cytosolic nonspecific-dependent PDE systems and that it is induced by the $Na^{+}-Mg^{2+}$ exchanger stimulation.

  • 토양으로부터의 $CO_2$ 방출은 생태계의 탄소 순환에 중요한 위치를 차지한다. 주요 생태계의 순 $CO_2$ 교환과 일차 생산량 등을 정확히 산출하려면 군락 $CO_2$ 플럭스와 더불어 토양 $CO_2$ 플럭스의 관측이 함께 이루어져야 한다. 본 논문에서는 닫힌 역학 챔버 시스템을 활용한 토양 이산화탄소 플럭스의 관측 방법을 간략히 검토하고, 한반도 주요 생태계에 구축된 한국 타워 플럭스 관측 지역망(KoFlux)의 거점 관측소에서 예비 관측된 결과와 서로 다른 관측 시스템간의 상호비교 결과를 보고하고자 한다.

기관의 비정상 배기배출에 의해 생성되는 소음에 관한 연구 (A Study on the Noise Produced by Unsteady Exhaust Efflux of Engine)

  • This paper discusses prediction of the sound pressure level produced by simple engine exhaust systems(plain pipe, plain expansion chamber pipe, plain expansion chamber with internally extended inlet and outlet pipe, perforated pipe enclosed in a plain expansion chamber) and a computer program has been developed which predicts the sound pressure level and the frequency spectrum. The program utilizes unsteady flow gas dynamic theory and acoustic theory to predict the pressure-time history in the exhaust system and the mass flow rate-time history at the open end of the system and the sound pressure levels(1/3 Octave band levels) and the frequency spectrum in semi-anechoic room. The predictions are compared with measured levels and show a high degree of correlation.

Piperine이 적출 백서 정관내의 교감신경계에 미치는 영향 (Effect of Piperine on Peripheral Sympathetic Nervous System in Isolated Vas deferens of Rat)

  • To elucidate one of the effect of piperine on the peripheral sympathetic nervous system, influence of piperine upon the contractile action of norepinephrine, methoxamine and tyramine as well as uptake and release of $[^{3}H]-norepinephrine$ has been investigated in naive and chronic piperine-treated vas deferens of rats. $pA_2$ value for ${\alpha}_1-adrenoceptor$ of phentolamine was significantly increased. Chronic piperine-treated group was markedly shown increased efflux of $[^{3}H]-norepinephrine$ and muscular tension, but was not affected the neuronal up-take and release of $[^{3}H]-norepinephrine$. It can be concluded that potentiation of the effect of norepinephrine by acute and chronic piperine treated group may be due to the change of affinity of ${\alpha}_1-adrenoceptor$, and partly due to possible modification of storage mechanism.
