A Drug Efflux Pump for Cationic Drugs including Disinfectants in Bacillus subtilis (양이온약제내성을 유도하는 Bacillus subtilis의 Drug Efflux Pump)

  • The Bacillus subtilis YvaE protein, the small multidrug resistance (SMR) family (TC #2.A. 7.1), is shown to catalyze efflux of multiple cationic drugs including many disinfectants, when it was cloned and expressed in Escherichia coli. When the yvaD gene was coexpressed with yvaE gene, the yvaD protein, encoded within a single operon with the yvaE gene, is shown to counteract the action ofYvaE. By ethidium efflux analysis, the cells harvoring a vector with yvaE gene showed a rapid ethidium efflux, compared with the control cells. These results clearly suggest that YvaE mediates drug export from the cell cytoplasm.

Putative Negative Regulation of Novel MarB along with MarA upon the Function of AcrAB/TolC Efflux Pump of Escherichia coli K-12 (대장균 K-12의 AcrAB/TolC Efflux Pump의 기능에 대한 MarB와 MarA의 추정적 억제조절)

  • This study was focused on the evaluation of MarB alongside with MarA for its regulatory effects upon the efflux function of AcrAB pump, which were induced or not, perhaps as a target. Transductions of marR and/or acrAB mutation which were derived from Mar and/or AcrAB mutants of wild type E. coli K-12, respectively, into the multicopy plasmid in wild type E. coli backgrounds or into the chromosome of isogenic parents were done. Minimal inhibitory concentration (MIC) of transduced mutants was compared with their original mutants. This study reports the indirect evidences that suggests a model in which MarB along with MarA have a putative negative regulatory effect upon the efflux function of AcrAB/TolC pump while MarA alone have a positive regulatory effect to the expression of acrRAB operon at transcription level. The target of MarB with MarA for its putative negative regulator might be the AcrAB efflux pump. Another efflux system (s) might be negatively regulated by MarB with MarA, and be involved in the efflux of antibiotics which were otherwise extruded preferentially by AcrAB efflux pump.

  • The purpose of this study is to examine whether an brain to blood efflux system for taurine is present on the blood-brain barrier (BBB) or not and this efflux transport system is regulated by CNS cell damage with oxidative stress agent such as diethyl maleate (DEM) or tumor necrosis factor-a (TNF-${\alpha}$), by using the brain efflux index (BEI) method. The brain efflux index value is defined as the relative amount of test compound efflux from cerebrum compared with that of a reference compound, [$\^$14/C] carboxyinulin, which has limited BBB permeability. Soil CO2 efflux in a warm-temperature and sub-alpine forest in Jeju, South Korea

  • Background: This study investigated the temporal variation in soil $CO_2$ efflux and its relationship with soil temperature and precipitation in the Quercus glauca and Abies koreana forests in Jeju Island, South Korea, from August 2010 to December 2012. Q. glauca and A. koreana forests are typical vegetation of warm-temperate evergreen forest zone and sub-alpine coniferous forest zone, respectively, in Jeju island. Results: The mean soil $CO_2$ efflux of Q. glauca forest was $0.7g\;CO_2\;m^{-2}\;h^{-1}$ at $14.3^{\circ}C$ and that of A. koreana forest was $0.4g\;CO_2\;m^{-2}\;h^{-1}$ at $6.8^{\circ}C$. The cumulative annual soil $CO_2$ efflux of Q. glauca and A. koreana forests was 54.2 and $34.2t\;CO_2\;ha^{-1}$, respectively. Total accumulated soil carbon efflux in Q. glauca and A. koreana forests was 29.5 and $18.7t\;C\;ha^{-1}$ for 2 years, respectively. The relationship between soil $CO_2$ efflux and soil temperate at 10 cm depth was highly significant in the Q. glauca ($r^2=0.853$) and A. koreana forests ($r^2=0.842$). Soil temperature was the main controlling factor over $CO_2$ efflux during most of the study period. Also, precipitation may affect soil $CO_2$ efflux that appeared to be an important factor controlling the efflux rate. Conclusions: Soil $CO_2$ efflux was affected by soil temperature as the dominant control and moisture as the limiting factor. The difference of soil $CO_2$ efflux between of Q. glauca and A. koreana forests was induced by soil temperature to altitude and regional precipitation.

Concentration of CCCP Should Be Optimized to Detect the Efflux System in Quinolone-Susceptible Escherichia coli

  • Unlike eukaryotic efflux pumps energized by ATPase bacterial efflux pumps are energized by the proton motive force. That is the reason why CCCP, an inhibitor of proton motive forcer is widely used to study the bacterial efflux pump. In many cases, efflux systems have been observed only in quinolone-resistant bacteria. Most of the quinolone-susceptible strains have been found to maintain little efflux pump. However some susceptible bacteria skewed the increased intracellular quinolone concentration only at a low concentration (0.01 or 0.1 mM) but net at a high concentration (1 mM) of CCCP. If bacterial cells were killed at high concentrations of CCCP and lost the integrity of their membranes, the intracellular quinolone would leak out from cells with no efflux system. The efflux pump system in the quinolone-susceptible strains could net be detected at the same concentration used for resistant bacteria. To test this hypothesist the intracellular quinolone concentration in the quinolone-susceptible and -resistant strains of Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus was assayed at various concentrations of CCCP. Since the effect of CCCP is very rapid, the survival of bacteria was observed by assaying the DNA synthesis in 5 min. In the case of E. coli, but not P. aeruginosa or S. aureus, the quinolone susceptible strain was more susceptible to CCCP than the quinolone resistant ones, especially when the incubation with CCCP was extended. Decrease of the intracellular quinolone concentration resulted in a false result-no or weak efflux system in the quinolone susceptible strains. Results suggested that the concentration of CCCP should be optimized in order to detect the efflux system in the quinolone susceptible strains of E. coli.

In vivo Brain-to-blood Efflux Transport of Choline at the Blood-brain Barrier

  • The purpose of this study was to clarify the efflux transport system of choline from brain to blood across the blood-brain barrier (BBB) in rats using the brain efflux index (BEI) method. $[^3H]$Choline was micro-injected into parietal cortex area 2 (Par2) of the rat brain, and was eliminated from the brain with elimination halflife of 45 min. The BBB efflux clearance of $[^3H]$choline was about 124 mL/min/g brain, which was determined from combination of an elimination rate constant $(1.54X10^{-2}min^{-1})$ and the distribution volume in the brain (8.05 mL/g brain). The efflux of $[^3H]$choline was inhibited by unlabeled choline in a dose-dependent manner and was significantly inhibited by cationic substrates, such as hemicholinium-3 and tetraethylammonium (TEA). These results suggest that the BBB may act as an efflux pump for choline to reduce the excessive choline concentration in the brain interstitial fluid.

Relationship between AdeABC Efflux Pump Genes and Carbapenem in Multidrug-resistant Acinetobacter baumannii

  • Multidrug-resistant strain of Acinetobacter baumannii (MDRAB) is an emerging pathogen in health care facilities, preventing MDRAB is a public health concern. We conducted this experiment on a clinical isolate of A. baumannii with two main goals: the role of the efflux pump system in the stress provision of carbapenem and the response to the transcription level of the efflux pump gene. A total of 34 strains of A. baumannii was isolated from the Yangsan Hospital of Pusan National University. First, when we compared and observed the expression of the efflux pump gene and antibacterial resistance to carbapenem, a strong correlation was observed between carbapenem resistance and overexpression of adeB (P=0.0056). Second, a correlation between the efflux pump and concentration gradient and tolerance to carbapenem stress at the AdeABC efflux pump genes transcription level was confirmed. Our results revealed that the expression of the AdeABC efflux pump is an important resistance determinant in obtaining antibiotic resistance of the carbapenem group in A. baumannii.

Kinetic Analysis about the Bidirectional Transport of 1-Anilino-8-naphthalene Sulfonate (ANS) by Isolated Rat Hepatocytes

  • The purpose of the present study was to investigate the bidirectional transport of 1-anilino-8-naphthalene sulfonate (ANS) using isolated rat hepatocytes. The initial uptake rate of ANS by isolated hepatocytes was determined. The uptake process of ANS was saturable, with a $K_m of 29.1\pm3.2 \mu M and V_{max} of 2.9\pm0.1$ mmol/min/mg protein. Subsequently, the initial efflux rate of ANS from isolated hepatocytes was determined by resuspending preloaded cells to 3.0% (w/v) BSA buffer. The efflux process for total ANS revealed a little saturability. The mean value of the efflux clearance was $2.2\pm0.1 \mu$ L/min/mg protein. The efflux rate of ANS from hepatocytes was markedly decreased at $4^{\circ}C$, indicating that the apparent efflux of ANS might not be attributed to the release of ANS bound to the cell surface, but to the efflux of ANS from intracellular space. The efflux clearance was furthermore corrected for the unbound intracellular ANS concentration on the basis of its binding parameters to cytosol. The relation between efflux rate and unbound ANS concentration was fitted well to the Michaelis-Menten equation with a saturable and a nonsaturable components. The $V_{max} and K_m$ values were 0.54 mmol/min/mg protein, and 10.0 $\mu$ M, respectively. Based on the comparison of the ratios of $V_{max} to K_m (V_{max}/K_m)$ corresponding to the transport clearance, the influx clearance was two times higher than the efflux clearance. Together with our preliminary studies that ATP suppression in hepatocytes substantially inhibited ANS influx rate, we concluded that the hepatic uptake of ANS is actively taken up into hepatocytes via the carrier mediated transport system.

Soil CO2 Efflux and Leaf-Litter Decomposition of Quercus variabilis and Pinus densiflora Stands in the Southern Region of Korean Peninsular

  • It is necessary to determine the amount of carbon dioxide ($CO_2$) absorbed by plants and released from forest floor into atmosphere, to gain a better understanding how forests participate in the global carbon cycle. Soil $CO_2$ efflux, litter production, and decomposition were investigated in Q. variabilis and P. densiflora stands in the vicinity of Gwangju, Chonnam province. Soil $CO_2$ efflux was measured using Infrared Gas Analyzer (IRGA) at midday of the 10th day at every month over 12-month period, to quantify seasonal and annual budgets of soil $CO_2$ efflux. Soil temperature and soil moisture were measured at the same time. Seasonal soil $CO_2$ efflux in Q. variabilis and P. densiflora were the highest in summer season. In August, maximum soil $CO_2$ efflux in Q. variabilis and P. densiflora was 7.49, $4.61CO_2{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. Annual $CO_2$ efflux in each stand was 1.77, $1.67CO_2kg{\cdot}m^{-2}$ respectively. Soil $CO_2$ efflux increased exponentially with soil temperature and related strongly in Q. variabilis ($r^2$=0.96), and in P. densiflora ($r^2$=0.91). Litter production continued throughout the year, but showed a peak on November and December. Annual litter production in the Q. variabilis and P. densiflora stands were $613.7gdw{\cdot}m^{-2}{\cdot}yr^{-1}$ and $550.5gdw{\cdot}m^{-2}{\cdot}yr^{-1}$.$yr^{-1}$, respectively. After 1 year, % remaining mass of Q. variabilis and P. densiflora litter was 48.2, 57.1%, respectively. The soil $CO_2$ efflux rates in this study showed clear seasonal variations. In addition, the temporal variation in the $CO_2$ efflux rates was closely related to the soil temperature fluctuation rather than to variations in the soil moisture content. The range of fluctuation of soil $CO_2$ efflux and litter decomposition rate showed similar seasonal changes. The range of fluctuation of soil $CO_2$ efflux and litter decomposition rate was higher during summer and autumn than spring and winter.

The Efflux Transport of Choline through Blood-Brain Barrier is Inhibited by Alzheimer's Disease Therapeutics

  • In the present study, we examined the effects of several therapeutics of Alzheimer's disease, such as donepezil hydrochloride, tacrine and $\alpha$-phenyl-n-tert-butyl nitrone (PBN) on choline efflux from brain to circulating blood. The brain-to-blood efflux of [$^3H$]choline in rats was significantly inhibited by tacrine and PBN. Also the [$^3H$]choline efflux was reduced by tacrine and donepezil hydrochloride in the TR-BBB cells, in vitro the blood-brain barrier (BBB) model. These results suggest that these drugs may influence choline efflux transport from brain to blood and regulate the choline level in brain resulting in the increase of acetylcholine synthesis.
