Studies on Quality Changes and Antioxidant Activity During the Fermentation of the Salt Fermented Whangseoke (황석어(Collichthys nireatus Jordan et starks) 젓갈의 숙성과정 중 품질변화와 항산화작용에 관한 연구)

  • The effect of storage temperature on the quality and antioxidative activity Whangseoke sauce was studied over a period of 240 days. Fermented Whangseoke with $25\%$ salt were stored at $25^{\circ}C$. The quality change and antioxidant activity of Whangseoke in linoleic acid emulsion was evaluated with various parameters including acids values, peroxide values, TBA values, reducing sugar, brown color intensity, electron donating ability and reducing power at various time intervals for 240 days of storage. In general, it was observed, in all sample, that peroxide values, brown color intensity, electron donating ability and reducing power gradually increased, while reducing sugar decreased during storage at $25^{\circ}C$. The antioxidative activities of fermented Whangseoke were determined on tile linoleic acid emulsion system. The results showed that Whangseoke had antioxidant activity. These results suggest that antioxidant activity of Whangseoke seemed to influence by Maillard reaction products during the storage periods.

Effects of Supercritical Fluid Marc Extracts from Actinidia polygama Max. on Inflammation and Atherosclerosis (개다래 초임계 박추출물이 염증 및 동맥경화에 미치는 영향)

  • The fruit of Actinidia polygama, Mock-chun-ryo in Korea, has been used as traditional medicine for abdominal pain, rheumatic arthritis, and stroke. In a previous study, the ethanol extract of A. polygama Max. showed antiinflammatory activity in RAW 264.7 cells. In this study, we investigated the anti-inflammatory and anti-atherosclerosis effects of supercritical fluid marc extracts from A. polygama Max. Anti-inflammatory extracts were produced from supercritical fluid extraction of the silver vine under the following conditions; pressure, 1,500-4,500 psi, temperature $35-55^{\circ}C$ and extraction time 1-2 hr. To evaluate the anti-inflammatory and anti-atherosclerotic effects of the extracts, we studied nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-alpha (TNF-$\alpha$) levels in RAW 264.7 cells and MMP-9 activity in human aortic smooth muscle cells (HASMC). The Marc 11 extract inhibited the production of NO, $PGE_2$, and TNF-$\alpha$ by lipopolysaccharide in RAW 264.7 cells. Moreover, the marc 11 extract inhibited TNF-$\alpha$-induced MMP-9 activity in HASMC. These results indicate that the Marc 11 extract of A. polygama Max. has the potential for use as an anti-atherosclerosis agent.

Studies for Development of Freeze - Flow Process (Freeze - Flow Process 개발을 위한 기초연구)

  • To develope a process by which liquid foods can be stored in the liquid state at the frozen storage temperature, suitable cryoprotectants were selected. Orange juice, chosen as an example of liquid foods, was stored with combined cryoprotectants at $-15^{\circ}C$, and quality changes of orange juice during storage were evaluated. Among 7 cryoprotectants tested, NaCl solution had lower initial freezing point than others, and initial freezing points of glucose, fructose, glycerol, propylene glycol and citric acid were close to each other. Considering flavor quality of orange juice, cryoprotectants suitable for reducing freezing point of orange juice were glucose, fructose, glycerol, and citric acid. Combined cryoprotectants for reducing freezing point of 3 and 4 folds concentrated orange juice to $-15^{\circ}C$ consisted of 10% glucose, 8% frutose, 4.6% glycerol and 3% citric acid, and 5.5% glucose, 4.5% fructose, 4.6% glycerol, and 3% cirtric acid, respectively. When destruction of ascorbic acid, sedimentation volume and sensory flavor score of orange juice stored with combined cryoprotectants at $-15^{\circ}C$ and the control stored at $-18^{\circ}C$ were compared, there were no significant differences. These results indicated that liquid foods with suitable combined cryoprotectants could be stored at $-15^{\circ}C$ or below in the liquid state without adverse effect on quality of the stored products.

Effect of Soil Strength on Seedling Emergence of Rice and Barnyardgrasses in Direct Dry-Seeding (건답직파에서 토양경도가 벼와 피의 출아에 미치는 영향)

  • Seedling emergences of four rice varieties (Dongjinbyeo, Dadajo, Galsaekggarak-sharebyeo, and Italiconaverneco) and three barnyardgrass species(Echjnochloa oryzjcola, E. crus-gali var. crus gali E. crus-gali var. praticola) were evaluated in relation to soil strength. Soil strength was varied by compressing the entire volume of soil with a hydraulic jack so as to be 0.5, 1, 2, 3, 6kg /$\textrm{cm}^2$. Soil strength was measured with a penetrometer (Yamanaka type) and soil covering above the seed was 4cm deep. Experiments were conducted at two air temperature conditions of 17 and $25^{\circ}C$. At a soil strength of up to 2kg/$\textrm{cm}^2$, little or no decrease in seedling emergence occurred in all rice varieties and barnyardgrasses tested. Above that value, seedling emergence decreased progressively as the soil strength increased. The degree of decrease was greatest in Dongjinbyeo and smallest in Dadajo among tested rice varieties, and greatest in Echinochloa oryzicola among barnyardgrasses, being greater in barnyardgrasses than rice. Seedling emergence was delayed almost linearly as the soil strength increased. The delay was greatest in Dongjinbyeo among rice varieties and in Echinochloa oryzicola among barnyardgrasses. Mesocotyl length increased as soil strength increased up to 2 to 3kg / $\textrm{cm}^2$ in Dongjinbyeo and Dadajo in 17$^{\circ}C$ and $25^{\circ}C$, and up to 6kg/$\textrm{cm}^2$ in Galsaekggaraksharebyeo and Italiconaverneco in $25^{\circ}C$. Dongjinbyeo showed the least elongation of mesocotyl among rice varieties in any soil strength. The total length of mesotyl, first internode and incomplete leaf showed little variation with soil strength. The total length was longer than the 4cm covering depth in other varieties except Dongjinbyeo. This might have caused the lower emergence rate in Dongjinbyeo than other varieties in higher soil strength.

A Concentrated Onion Extract Lowers Serum Lipid Levels in Rats Fed a High-Fat Diet (흰쥐에서 양파 농축액의 고지혈 개선 작용)

  • It is known that onions, or bioactive compounds therein, providehealth benefits. The present study was designed to investigate whether a concentrated onion extract lowered blood lipid levels in rats fed a high-fat diet. Initially, male Sprague-Dawley rats were housed singly in an environment in which temperature and light duration were controlled, and had free access to a nutritionally adequate AIN-93G diet and deionized water. After an acclimatization period, rats were weight-matched and assigned to one of the following five groups: 1) a control group, fed the AIN-93G diet mixed with 10% (w/v) lard and 0.7% (w/v) cholesterol to induce hyperlipidemia (control); 2) three experimental groups, fed the AIN-93G diet mixed with a high-fat source plus concentrated onion extract at three different levels (termed the low-dose, medium-dose, and high-dose groups); and, 3) a placebo group, fed the AIN-93G diet with fats plus the same concentrated extract but devoid of onion-derived material. All five groups freely ingested their respective diets over 6 weeks. At 0, 3, and 6 weeks, blood samples were collected from the orbital sinus following overnight food deprivation. At 6 weeks, livers were collected. Both control and experimental groups continually gained body weight throughout the study. No significant differencein body weight gain was observed among groups. However, the serum concentrations of triglycerides, total cholesterol, and non HDL-cholesterol were significantly reduced by ingestion of concentrated onion extract. Also, the hepatic levels of total lipids and total fatty acids, especially C18:1 (oleic acid), were significantly decreased in rats fed a high level of concentrated onion extract, compared with the control and placebo groups. These results provide clear evidence that ingestion of a concentrated onion extract has a profound inhibitory effect on serum lipid levels in rats fed a high-fat diet. Our findings indicate that a concentrated onion extract may be used to alleviate hyperlipidemia by lowering serum cholesterol and triglyceride levels.

Operation of High Performance Elutriation-Type Sludge Fermenter and Feasibility for Its Application (고성능 세정식 슬러지 산발효조의 운전 및 적용성 평가)

  • The performance of a novel fermentation process, adopting a sludge blanket type configuration for higher hydrolysis/acidogenesis of the municipal primary sludge, was investigated under batch and semi-continuous conditions with various pH and temperature conditions. This acid elutriation slurry reactor provided higher system performance with a short HRT (5 days) and higher acidogenic effluent quality under pH 9 and thermophilic ($55^{\circ}C$) conditions. The hydrolysis of the sludge was revealed to be significantly dependent on seasonal effects for sludge characteristics but with little impact on acidogenesis. Based on the rainy season at the optimum conditions, VFA production and recovery fraction ($VFA_{COD}/COD$) were $0.18\;g\;VFA_{COD}\;g^{-1}\;VSS_{COD}$ and 63%. As byproducts, nitrogen and phosphorus releasing were $0.006\;g\;N\;g^{-1}\;VSS_{COD}$ and $0.003\;g\;P\;g^{-1}\;VSS_{COD}$, respectively. For the mass balance in a full-scale plant($Q=158,880\;m^3\;day^{-1}$) based on the rainy season, the VFA and non-VFA(as COD) production were $3,110\;kg\;VFA_{COD}\;day^{-1}$ and $1,800\;kg\;COD\;day^{-1}$, resulting in an increase of organics of $31\;mg\;COD\;L^{-1}$ and $20\;mg\;VFA_{COD}\;L^{-1}$ and nutrients of $0.7\;mg\;N\;L^{-1}$ and $0.3\;mg\;P\;L^{-1}$ in the influent sewage. The economical benefit from this process application was estimated to be about $67 per $1,000m^3$ of sewage except for energy requirements and also, better benefits can be expected during the dry season. Also, the results revealed that the process has various additional advantages such as pathogen-free stabilized solids production, excellent solids control and economical benefits.

Effect of Irrigation Water Depth on Greenhouse Gas Emission in Paddy Field (논물 담수심이 온난화 가스 배출에 미치는 영향)

  • The increasing emission of greenhouse gases may change agricultural environment. The agronomic productivity will depend upon change of temperature, precipitation, solar radiation and fertilization. This study was conducted to investigate greenhouse gas emission with irrigation water depth in paddy field. Area of each experiment plot is $70m^2$, Three treatments with three replications were carried out in this experiment, which was laid out as randomized complete block design. The treatments of irrigation water were maximum field water capacity and 4 and 8 cm depth. The application rate of fresh rice straw was $8,000kg\;ha^{-1}$ in combination with chemical fertilizers ($110kg\;N\;ha^{-1}$, $45kg\;P_2O_5\;ha^{-1}$ and $57kg\;K_2O\;ha^{-1}$). The $CH_4$ emission was highest at 32 days after rice transplanting with rice straw treatment. The $CH_4$ emission in the plot of maximum field water capacity was lower compared with 4 and 8 cm of irrigation depth. $CH_4$ and $N_2O$ emission under different water depth in the paddy field were 30 and $1.52kg\;ha^{-1}$ at 8 cm depth, 281 and $1.71kg\;ha^{-1}$ at 4 cm depth, and 219 and $2.01kg\;ha^{-1}$ at water saturated condition. The total emission of greenhouse gases equivalent to $CO_2$ emission with rice straw application were $6,939kg\;CO_2\;ha^{-1}$ at 8 cm depth plot, $6,431kg\;CO_2\;ha^{-1}$ at 4 cm depth plot and $5,222kg\;CO_2\;ha^{-1}$ at water saturated condition. The GWPs without rice straw application were $4,449kg\;CO_2\;ha^{-1}$ at 8 cm depth plot, $3,702kg\;CO_2\;ha^{-1}$ at 4 cm depth plot and $4,579kg\;CO_2\;ha^{-1}$ at water saturated condition.

Effect of Day Length and Temperature on the Diapause Termination of Riptortus pedestris (Hemiptera: Alydidae) Male Adults (일장과 온도 처리가 톱다리개미허리노린재 수컷 성충의 휴면종료에 미치는 영향)

  • The bean bug, Riptortus pedestris (=clavatus) Fabricius (Hemiptera: Alydidae) is a serious pest of soybean in many countries. It enters a reproductive diapause in the winter by short day length (<13.5 h.) in Korea and Japan. The combined effects of temperatures ($25^{\circ}C$ and $8^{\circ}C$) and day lengths (10L:14D and 14L:10D) upon the termination of the male's diapause were studied. The combinations are (1) HTLD = $25^{\circ}C$, 14L:10D treatment for 1, 2, 3 weeks and 30 days; (2) HTSD = $25^{\circ}C$, 10L:14D treatment for 1, 2, and 3 weeks; (3) LTLD$\rightarrow$HTLD = $8^{\circ}C$, 14L:10D treatment for 1, 2, and 3 weeks followed by HTLD treatment; (4) LTSD$\rightarrow$HTLD = $8^{\circ}C$, 10L:14D treatment for 1, 2, and 3 weeks followed by HTLD. The amount of aggregation pheromone components secreted was adopted as the criteria for diapause termination in males. Males did not secrete aggregation pheromone under HTSD conditions. However, the males treated with HTLD for more than 21 days secreted significantly higher amounts of aggregation components, (E)-2-hexenyl (Z)-3-hexenoate and (E)-2-hexenyl (E)-2-hexenoate, compared to the control or HTSD treatments. The three components of the aggregation pheromone were secreted by the 14%, 29%, and 100% males treated with HTLD for 7 days, respectively. However more than 83% of the HTLD-treated males secreted all three components by the $30^{th}$ day under the treatment. In comparison, the pheromone amounts secreted by the males treated with LTLD$\rightarrow$HTLD or LTSD$\rightarrow$HTLD were equal or less than those secreted by males with continuous HTLD treatment.

  • Hypothermia during lung preservation decreases metabolic processes. After the rabbit lung was flushed with modified Euro-Collins solution, heart-lung block was harvested and the left lung was assessed after ligation of the right pulmonary artery and right main-stem bronchus. Heart-lung block was immersed in the same solution for 6 hours. The modified Euro-Collins solution and storage temperature of group 1(10 cases) was 4t, roup 2(10 cases) was l$0^{\circ}C$. On completion of the storage period, the left lung was ventilated and reperfused with blood u:high used a cross-circulating paracorporeal rabbit as a "biologic deoxygenator" for 60 minutes. Pulmonary artery pressure, airway pressure, difference in oxygen tension between mow and outflow perfusate and degree of pulmonary edema were assessed at 10-minute intervals while the left lung was ventilated at 0.8 of the inspired oxygen fraction. The mean pulmonary venous oxygen tensions at 10 and 60 minutes after reperfusion were 209.52$\pm$42.46 and 103.48$\pm$ 15.96 mmHg in group I versus 247.78$\pm$36.19 and 147.91 $\pm$ 11.07 mmHg in group II(p=0.049, (0.0001). The mean alveolar-arterial oxygen differences at 20 and 60 minutes after reperfusion were 357. 95$\pm$ 12.84 and 437.31 14.26 mmHg in group I versus 310.88$\pm$3).47 and )90.93$\pm$ 15.86 mmHg in group II (p=0.0092, (0.0001). The mean pulmonary arterial pressures at 10 and 60 minutes after reperfusion were 40.56$\pm$ 18.66 and 87. 2$\pm$ 17.22 mmHg in group I versus 31.22$\pm$6.84 and 65.78$\pm$ 11.02 mmHg in group rl (p : 0.048, 0.0062). The mean pulmonary vascular resistances at 10 and 60 minutes after reperfusion were 2.69$\pm$0.85 and 4.36$\pm$0.86 mmHg/ml/min in group I versus 1.99$\pm$0.39 and 3.29$\pm$0.55 mmHg/ml/min in group II(p : 0.0323, 0.0062). There were no difference between groups in peak airway pressure, lung compliance and degree of pulmonary edema. In conclusion that preservation of lung at l$0^{\circ}C$ was superior to preservation at 4$^{\circ}C$.}C$.

Quality Characteristics of Semi-dried Red Pepper During Frozen Storage (반건조 고추의 냉동저장 중 품질 특성)

  • We investigated the effect of freezing on changes in the chemical components of semi-dried red pepper (SDRP). We used storage temperatures of $0^{\circ}C,\;-10^{\circ}C,\;-20^{\circ}C,\;and\;-70^{\circ}C$. After 30 days of storage, capsaicin content had decreased by 40% at $0^{\circ}C$ and by 21% at $-20^{\circ}C$. Initial vitamin C content was 1,358.02 mg%. Compared with control, the $0^{\circ}C$ storage group showed a significant decrease in vitamin C content but no such decrease was noted in the $-20^{\circ}C$ and $-70^{\circ}C$ storage groups after 30 days. ASTA values were not influenced by storage temperature or period, in agreement with previous results. We concluded that storage was effective at temperatures of less than $-20^{\circ}C$. Next, both dried red pepper (DRP) and SDRP were stored at $-20^{\circ}C$ for 12 months. DRP had the lower level of capsaicinoids (55.01 mg%) owing to the long drying time. After 12 months, SDRP capsaicinoid had decreased by 30-33%, compared with a decrease of 54% in DRP. Initial vitamin C contents were 721.48 and 955.25 mg% in DRP and SDRP, respectively, and, after 12 months, vitamin C loss in the SDRP group (37%) was less than that in fresh red pepper (FRP) samples (45%). Initial $\beta$-carotene content was greatest in the FRP group (259.82 mg%), and that of DRP decreased by 20% after 12 months. The color a/b value of SDRP (1.40) was greater than that of DRP (1.00).
