Pharmacognostical Studies on Korean Arisaema ringens $S_{CHOTT}$ (한국산 Arisaema ringens $S_{CHOTT}$의 생약학적 연구)

  • This paper attempts to observe the histological structure of Korean Arisaema ringens $S_{CHOTT}{'s}$ rhizome, to identify it's constituent and to investigate the pharmacological action with it's alcohol extract. The results are : 1) The inner structure of rhizome on the transverse section is divided into external and internal tissues by the ring of intercellular secretary sac. Raphides of calcium oxalate contained in mucilage cell, collateral vascular bundle, are extremely similar to those contained in Pinellia ternata. 2) The organs of the pistillate Arisaema ringens are larger and more plentiful than those of the staminate Arisaema ringens. The sexual identification is easy in the flowering season. 3) The alkaloid is identified by Meyer reagent as white p.p.t. at pH 2 of sulfuric acid. 4) The saponin is indentified remarkably by means of foaming reaction, Lieberman-Burchard reaction and hemolytic reaction. 5) The effect of alcohol extract on the relaxation of the isolated intestine of the rabbit is remarkably shown at the concentration of $10^{-3}g/ml$. 6) The effect of alcohol extract on the isolated ractus muscle of the frog increases the constructive action of acetylcholine at the concentration of $10^{-3}g/ml$. 7) The effect of alcohol extract on the isolated heart movement of the frog is decreased remarkably at the concentration of $10^{-3}g/ml$. 8) The effect of alcohol extract on the blood pressure of the rabbit is decreased by an interavenous injection of $10^{-3}g/kg$.

Effect of Ganoderma Lucidum on the Liver Function and Lipid Metabolism in Alcohol-Consuming Rats (영지가 알코올 섭취한 흰쥐의 간기능 및 지질대사에 미치는 영향)

  • The effects of Ganoderma Lucidum consumption (GL) on the liver function and lipid metabolic disorder induced by alcohol consuming were studied in rats using GL powder(1%, 2%, 4%) and 10% ethly alcohol. Thirty-five rats(Sprague-Dawley, male) were divided into five groups and fed experimental diets for seven weeks. The concentrations of serum cholesterol, HDL-cholesterol and triglyceride were the highest in the alcohol group, while the levels were apparently reduced by in groups fed GL. The atherogenic indices(AI) also tended to be higher in the alcohol group. Glutamic oxaloacetic transaminase(GOT) activities were significantly lower in body alcohol + 2% GL and alcohol + 4% GL groups, and glutamic pyruvic transaminase(GPT)activities lower only in the alcohol + 2% GL compared with the alcohol group. Alkaline phosphatase(ALP) activities were not significantly different among the groups. The elevated levels of liver cholesterol and triglyceride due to alcohol consumption, were apparently reduced by GL supplementation. On microscopic observation of liver tissues, fat droplets appeared extensively on the liver-lobule in the alcohol group, while fat droplets appeared only on the central vein in the alcohol + 1% GL group. However, in groups administered 2% and 4% GL, fat droplets appeared similar to the alcohol group. The concentrations of fecal bile acid were significantly increased in groups fed GL. These results indicate that GL exerted some beneficial effects on lipid metabolic disorder caused by long-term alcohol consumption. Thus, it seems that GL may be effective in preventing or curing some aspects of alcohol toxicity. More detailed studies are needed in order to determine proper dietary levels of GL for combating alcohol toxicity.

  • The effect of habitual alcohol consumption on the nutrient intake. body weight and serum values were studied in 194 upper middle class men aged 35-55yr. living in Seoul. Each subject was interviewed to get information of alcohol and nutrients intake. Blood samples were taken. Mean values for each variable were compared by fertile of alcohol intake. The average alcohol consumption of the subject was 32.2g/day. Most of times foods were taken along with drinks. The subjects preferred food items such as meat. fish, nuts. dry fishes when they drink. High alcohol consumers had a significantly higher intake of calorie. protein, fat. Fe, and riboflavin compared to nondrinkers and moderate drinkers. Body weight was significantly higher in high alcohol drinkers followed by moderate and nondrinker groups. They also showed the higher serum values of triglyceride and r-GTP. High alcohol consumption resulted in an increased intakes of most nutrients especially calorie and fat which leads to higher body weight and higher serum concentrations of triglyceride and r-GTP. Therefore it seems that heavy alcohol drinkers are more prone to obesity and related chronic diseases.

Effect of Daekumeumja Herb-acupuncture on c-Fos Expression in Hippocampus of Alcohol Intoxicated Rats (대금음자 약침이 알코올 독성 흰쥐의 해마에서 c-Fos 발현에 미치는 영향)

  • Objectives : The study was conducted to investigate the effect of Deakumeumja herb-acupuncture on c-Fos expression in each area of the hippocampus of Alcohol intoxicated rats. Methods : Experimental groups were divided into five groups ; normal group, the alcohol-treated (control) group, the alcohol- 1 mg/kg Daekumeumja treated (sample A) group, the alcohol- 5 mg/kg Daekumeumja treated (sample B) group, the alcohol- 10 mg/kg Daekumeumja treated (sample C) group (n = 6 for each group). Rats of normal group were injected intraperitoneally with saline once a day for 5 consecutive days, while animals of the alcohol-treated (control) group were injected once a day with 2 g/kg of alcohol for the same duration of time. Animals of the alcohol and Daekumeumja-treated (sample A, B, C) groups were acupunctured at Chung-wan$(CV_{12})$ with 2 g/kg of alcohol and the appropriate amount of Daekumeumja extract once a day for 5 days. Each groups was evaluated by the changes of c-fos-positive neurons in each area of the hippocampus by using an image analyzer and microscope. Results: 1. In the CAI region of the hippocampus, the number of Fos-positive cells in the sample B, C groups were significantly increased compared with the control group. 2. In the CA2-3 regions of the hippocampus, the number of Fos-positive cells of the sample B, C groups were significantly increased compared with the control group. 3. In the Dentate gyrus region of the hippocampus, the number of Fos-positive cells of the sample C group was significantly increased compared with the control group. Conclusion : c-fos expression in each area of the hippocampus was reduced in alcohol-intoxicated groups. Treatment of Daekumeumja increased this reduction. In conclusion, it can be suggested that Daedumeumja possesses protective effects of the amnesia and learning disability in alcoholism.

Effect of Pectin Degradation Enzyme during Alcohol Fermentation of Persimmon Pulp for Persimmon Vinegar Preparation (감식초 제조를 위한 감펄프의 알콜발효시 펙틴분해효소 처리의 효과)

  • This study was carried out for the purpose of improving the persimmon vinegar. Crushed persimmon(persimmon pulp) was used at alcohol fermentation using Saccharomyces bayanus for persimmon alcohol medium preparation. Glucose(8.39%) and fructose(7.96%) were the dominant free sugar in persimmon pulp before the at cohol fermentation. They decreased abruptly during alcohol fermentation and glucose was consumed more rapidly than fructose. Final alcohol concentration was finally reached to 8%(v/v) in 5 days for mentation of persimmon pulp. Pectinase pre-treatment of persimmon pulp resulted in tusker contents of galacturonic acid, galactose, methyl alcohol and isoamyl alcohol in main mash for alcohol fermentation than those in main mash prepared without pectinase pre-treatment. After alcohol fermentation tannin concentration was 350ppm and astringency was not perceived.

Development and Efficacy Evaluation of a Web-based Prevention Programs of Alcohol-related Problems for University Students (대학생 음주문제 예방 프로그램 개발 및 효능성 평가)

  • Objectives: This study is to develop a web-based prevention program of alcohol-related problem among university students and to examine its efficacy in changing drinking behaviors and reducing harms associated with alcohol use. Methods: An hour-length program was developed containing news paper articles, pictures, stories and questionnaires in a multimedia format such as flash, animation, and text. Survey data were analyzed to examine program efficacy from a convenient sample of 1,080 freshmen enrolled in a participating university. Results: Participation in the program has resulted in statistically significant changes in drinking behavior between pretest and follow-up, including reduction of frequency of drinking, heavy drinking, and intoxication, typical amount of drinking per occasion, and subjective norm of heavy drinking. Repeated ANOVA revealed significant increases in knowledge on alcohol related facts, including recommended drinking unit by WHO with substantial level of effect size and in making negative alcohol expectancy with very low effect size. Further studies are guaranteed with randomized controlled trial for the program effectiveness. Conclusions: A web-based prevention program of alcohol-related problems in this study was efficacious in changing drinking behavior, knowledge on alcohol-related fact, alcohol expectancy and subjective drinking norm.

Alcohol Drinking Status of Officers on a Ship and Drunk-navigation Experiments Using Ship Handling Simulator

  • Based on the previous studies on alcohol effects on human behaviour and performance in transportation system such as air-plane and car driving, the alcohol exposure before and on watch of a ship has a great influence on subsequent behaviour. In this paper, to examine the drinking status of officers on board multiple choice questionnaires are circularized under instruction and surveyed for 118 officers. According to the results of the questionnaire survey on alcohol dependency (Alcoholism) that was invented by WHO, over 27 % of those surveyed represented alcohol abuse symptoms. In addition to that, the existing state and awareness for on-board-drinking was summarized to make a scenario of drink-operation with a ship handling simulator to investigate the effect of alcohol (0.08 g% blood alcohol concentrations) on ship operational performance. A main effect for alcohol was found indicating that ship operational performance was comparatively impaired by this alcohol relative to performance in the non-alcohol condition The results of this research can be applied to minimize marine accidents as basic data.

Influence of Gigaltang on Alcohol Intake Amount in C57BL/6 mice (지갈탕(枳葛湯)이 C57BL/6형 생쥐의 알코올섭취량에 미치는 영향)

  • Objective : This study was designed to test the anti-craving effect of Gigaltang(extracts of Oriental medicinal herbs ; Hovenia dulcis and Puerariae flos) in C57BL/6 mice. Anti-craving effect was substituted by alcohol intake amount of C57BL/6 mice. Method : 60 C57BL/6 mice was randomized into vehicle(water) group and Gigaltang group. Only 39(vehicle group(21) and Gigaltang group(18)) was eligible for this study, and 21 C57BL/6 mice was dropped out. The procedure was composed of 5 days of adjustment period and 32 days of alcohol exposure and withdrawal period with limited access paradigm and 10 days of treatment period. Alcohol was offered only for 2 hours a day from 2 to 4PM and water was offered for 22 hours left. Food was offered for 24 hours a day. 1140mg/Kg/day of Gigaltang was administrated to 18 mice of Gigaltang group for 10 days whereas water to 21 mice of vehicle group. The amount of alcohol intake, water intake, food intake, body weight were measured every other day. Result : There weren't significant differences in 2 hours of alcohol intake, 22 hours of water intake, 24 hours of food intake and body weight for ten days between Gigaltang and vehicle group. Conclusion : Anti-craving effect of Gigaltang on C57BL/6 mice wasn't verified. To verify the anti-craving effect, further study with advanced animal model, various dose of Gigaltang and diverse period of drug administration should be taken.

  • The effects of benzyl alcohol on the properties of dyeing kinetic of silk fibroin were studied. The acid dye used was C.I. Acid Red 114. The half dyeing time is shorten by addition of benzyl alcohol. The diffusion activation energy is higher with the increase of the solvent. The rate of dyeing at benzyl alcohol addition to the purified silk fibroin is faster than that of the unpurified one.

Effects of Dried Persimmon Snacks on Alcohol Metabolism in Men

  • Persimmon has been known to help alcohol intoxication in Korea for a long time and is prepared as a processed food. The effect of dried persimmon snacks on alcohol metabolism was determined in vivo. Eight Korean men (25~27 years old) were administered 3.5 mL/kg of alcohol (22.5%, w/v) with or without a dried persimmon snack (2g/kg). The levels of alcohol in the blood and of acetaldehyde in the blood and the urine were determined by alcoholmeter and assay using alcohol dehydrogenase and HPLC, respectively. All subjects showed decreased levels of alcohol in blood, and six subjects showed a decrease of alcohol in urine after consumption of ethanol with dried persimmon snacks. Concentration of acetaldehyde in the blood an urine decreased significantly in three and five of the eight subjects, respectively. Reduction was more significant for alcohol than for alcohol than for acetaldehyde with administration of ethanol and dried persimmon snacks. This in vivo result suggests that dried persimmon snacks are effective in decreasing th concentration of alcohol after alcohol intake.
