  • Sister chromatid exchange(SCE) has recently become a common cytogenic assay system for detecting exposure to chemical mutagens and carcinogens. One application of SCE is the monitoring of populations believed to be exposed to such agents. A cross-sectional study of SCE frequency in peripheral blood lymphocytes from 40 college students aged 18 to 26 years was conducted. The effects of cigarette smoking alcohol and coffee consumption, dietary and environmental factors on SCE were assessed. A mean spontaneous SCE per cell for the smokers(4.88$\pm$0.17). The SCE levels of the smokers were associated with the personal smoking amount ; the observed increase in the SCE frequency correlated with the number of cigarettes smoked per day (P<0.05). There was no effect of age on SCE. There were positive linear relationship between SCE and food frequency score of meat and fish group (P<0.05) or instant food group(P<0.01) in non-smokers. But in smokers, a significant inverse association between SCE and food frequency score of green and yellow vegetable group(P<0.05). Alcohol intake produced a significant increase(P<0.01) of SCE in comparison with the mean SCE for those not drinking alcohol in combine subjects. Other dietary parameters, including coffee intake, use of artificial sweetners and processed foods, did not show any increase in SCE. SCEs were inversely related to blood glucose and serum cholesterol levels of the combine subjects. No significant correlations were found between SCE frequencise and any other hematological parameters of the subjects.

  • This study was designed to demonstrate the antigenotoxic potential of methyl alcohol extracts from Auricularia mesenterica and Gyrophora esculenta against the frequency of micronucleate polychromatic erythrocyte(MNPCE) produced by benzo($\alpha$) pyrene in vivo. We used the mouse bone marrow test system to measure the effect of single and multiple treatments of each sample. Genotoxicity of benzo ($\alpha$) pyrene(150mg/kg, i.p.) as positive control was the highest at 36 hours. However, each sample per dose was not genotoxic, showing MNPCE values in the range of the control level. Treatments of methyl alcohol extracts both of Auricularia mesenterica and Gyrophora esculenta showed significant decreased frequencies of NMPCE induced by benzo($\alpha$) pyrene within 12 hours by single treatment(100mg/kg, oral). And also, the MNPCE level produced benzo($\alpha$) pyrene was decreased by the treatment of benzo($\alpha$) pyrene(5 to 200mg/kg, oral) of each sample, but significantly different redults were obtained with 100mg/kg. In the multiple treatment, the highest antigentoxic effects were demonstrated with 20mg/kg in the each sample, a range which induced inhibition indices of 54.2 and 56.3%, respectively.

  • Phytochemical investigation of the methanolic extract of Diospyros kaki leaves led to the isolation of osmanthuside H (1) and a new phenol glycoside, named gamnamoside [4-(3-hydroxypropyl)-2-methoxyphenol $\beta$-D-apiofuranosyl( 1 $\rightarrow$ 6)$\beta$-D-glucopyranoside] (2) along with (-) catechin (3) through a series of reversed phase column chromatography and preparative C18 HPLC. The structures of the isolates were determined by spectroscopic methods including IR, UV, HRTOFMS, and 2D NMR. Compounds 1, 2, and 3, showed good inhibitory activities ($IC_{50}$) of 175.4, 94.4, and 126.6 ${\mu}g/mL$ respectively, whereas a reversible ADH inhibitor, 4-methylpyrazole, showed the $IC_{50}$ of 326.6 ${\mu}g/mL$ against alcohol dehydrogenase (ADH).

  • Drinking excessive alcohol has been recognized as a risk factor for hypertension. However, the mechanism by which alcohol intake causes hypertension still remains elusive. We tested the hypothesis that ethanol itself acts as a stress factor on vasculature and indirectly modulates vascular contractility. After end of exposure to 1, 2.5 and 5% ethanol for 45 min, rat aortic strips were subjected to contractile responses, immunoblot for Hsp70 and the measurement of levels of myosin light chain phosphorylation. Exposure to 5% ethanol not only augmented contractions to KCl or phenylephrine, but also increased expression of Hsp70 and the levels of myosin light chain phosphorylation. There were no significant differences in contractions produced by $1\;{\mu}mol/L$ phorbol 12,13-dibutyrate, a protein kinase C activator, whether the tissues were exposed to 5% ethanol or not. This is the first report to show that even short exposure to ethanol has a delayed effect to increase vascular smooth muscle contractility through a modulation of thick filament regulation. It may be a mechanism by which ingestion of alcohol induces hypertension.

  • Fermentation characteristics of Baikhaju (a typical Korean traditional alcoholic beverage) were investigated during fermentation with differently treated rice samples, which were steamed rice (SR), plain steamed rice bread (RB), rice porridge (RP) and roasted rice (RR). The RB sample showed an excellent fermentation efficiency with the highest alcohol production (20.84%). The SR and RR samples showed 17.79% and 17.31% of alcohol production, respectively. The RP sample was the lowest alcohol production (11.47%). The pH values of all the beverage samples were similar and were ranged from 3.6 to 3.9. The fermentation periods of RB and RR samples were longer than the SR and RP. The inner temperature and microbial growth in all the beverages increased at each of input steps of the rice and Nuruk (a Korean-style bran koji). Fusel oil content of the SR and RP samples (613.6 ppm) was higher than that of RR (482.7 ppm). The RB sample with the best fermentation efficiency had the lowest fusel oil content (341.8 ppm). The RP sample showed the highest score in sensory evaluation and RB sample showed the lowest. The results on sensory evaluation were contrary to those on the fermentation efficiency.

  • Poly(vinyl alcohol) (PVA) composites with various graphite oxide (GO) contents (0 to 10 wt%) were prepared by sonicating the mixture of PVA and GO, followed by crosslinking with glutaraldehyde. GO was pre-treated with oxyfluorination ($O_2:F_2$ = 8:2) in order to modify the surface of GO to allow it to carry hydrophilic functional groups. PVA/GO composite hydrogels were characterized by scanning electron microscopy and Fourier-transform infrared spectrometer (FT-IR). The morphology of the PVA/GO composite hydrogels and the variations in soluble gel portion were investigated under various GO contents and UV irradiation doses. The variation in the chemical structure of photo degraded PVA/GO composite hydrogels was studied by FT-IR. The photochemical stability of PVA/GO composite hydrogels under UV irradiation was found to improve noticeably with increasing content of uniformly dispersed GO.

  • The objective of the current study was to evaluate the quality characteristics (alcohol contents and antioxidant capacities) and sensory characteristics of the Korean traditional rice-based wine, Makgeolli supplemented with 5% and 10% Enteromorpha prolifera (Ep) during an 8-days cold storage at 4℃. The alcohol content in the unsupplemented Makgeolli (control) and Ep Makgeolli gradually increased from day 0 to day 8 (control, 2.05-8.03; 5% Ep, 2.15-9.03; 10% Ep, 3.15-10.07). The DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activities was significantly higher in the 10% Ep Makgeolli (75.87%) than 5% Ep Makgeolli (57.34%) and control (21.91%). The ABTS [2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical scavenging activities were also significantly higher in the 10% Ep Makgeolli (90.85%) than 5% Ep Makgeolli (75.94%) and control (47.21%). Makgeolli supplemented with 10% Ep had the highest color, smell, taste, and overall acceptance scores. The current study suggest that Makgeolli supplemented with 10% Ep could be potentially developed because of its high antioxidant properties and good consumer acceptance.

  • This study was performed to see the effect of nutritional status on fatty liver. Subjects were 219(male : 174, female : 45) fatty liver patients living in cities, and 10-60yrs old. Nutrient and alcohol intakes, health state and medical history were investigated by individual interview, and blood sample was taken. Results were as follow; Most subjects were obese an overweighing and suffering from hepatitis. Calorie and carbohydrate intakes of male and females patients were lower than mean intakes of normal citizens. Protein and fat intakes of male patients were higher than mean intakes of normal citizens. Proportions of protein and fat to total calorie intake in patients were higher than those of normal citizens. Alcohol intake was markedly higher in patients than normal group, and increased with increasing body weight. Serum total cholesterol, HDL-cholesterol, total lipid, total protein and albumin conc.s and Hb and Hct levels were in normal ranges, But serum TG level of patients was higher than normal level. Serum total fatty acid level, $\omega$-3/$\omega$-6 and P/S FA ratios were higher in patients than normals. Serum total EAAs, NEAAs and all AAs levels of patients were higher than normals. It is plausible that high animal food and alcohol intakes, excess body fat and hepatitis were associated with fatty liver.
