Phenotypic characterization of pre-harvest sprouting resistance mutants generated by the CRISPR/Cas9-geminiviral replicon system in rice

  • Pre-harvest sprouting is a critical phenomenon involving germination of seeds in the mother plant before harvest under relative humid conditions and reduced dormancy. In this paper, we generated HDR mutant lines with one region SNP (C/T) and an insertion of 6 bp (GGT/GGTGGCGGC) in OsERF1 genes for pre-harvest sprouting (PHS) resistance using CRISPR/Cas9 and a geminiviral replicon system. The incidence of HDR was 2.6% in transformed calli. T1 seeds were harvested from 12 HDR-induced calli and named ERF1-hdr line. Molecular stability, key agronomic properties, physiological properties, and biochemical properties of target genes in the ERF1-hdr line were investigated for three years. The ERF1-hdr line showed significantly enhanced seed dormancy and pre-harvest sprouting resistance. qRT-PCR analysis suggested that enhanced ABA signaling resulted in a stronger phenotype of PHS resistance. These results indicate that efficient HDR can be achieved through SNP/InDel replacement using a single and modular configuration applicable to different rice targets and other crops. This work demonstrates the potential to replace all genes with elite alleles within one generation and greatly expands our ability to improve agriculturally important traits.

  • Electro-theological fluid is recently used for the micro polishing of 3-dimensional micro-aspherical lens. It's also used for polishing small area defects on the wide flat wafer. Since ER fluid shows a behavior of viscosity changing under certain electric fields. micro polishing efficiency may be enhanced for certain cases. In this paper, a perfluorinated carbonyl fluoride oil based ER fluids was used to improve surface polishing rate and submicron-scale accuracy. As the polishing electrodes, micro size cylindrical tools had been used for maximizing the electric field. An experimental device, which was applied for micro polishing a number of wafers of 4inches in size and other workpiece. was made on a precision polishing system. It consisted of a steel electrode. a wafer fixture. l0㎃ current and DC 5㎸ power supply unit, and a controller unit. From the Experiments. the ER fluid is applicable for micro polishing of small parts.

  • This paper presents a proof-concept investigation on the active vibration control of two hybrid smart structures (HSSs). The first one is consisting of a piezoelectric film (PF) actuator and an electro-rheological fluid(ERF) actuator, and the other is featured by a piezoceramic (PZT) actuator and a shape memory alloy (SMA) actuator. For the PF/ERF hybrid smart structure, both the increment of the damping ratios and the suppression of the tip deflections are evaluated in order to demonstrate control effectiveness of the PF actuator and ERF actuator and the hybrid actuation. For the PZT/SMA hybrid smart structure, the PZT actuator takes account of the high frequency excitation, while the SMA actuator exerts large vibration control force. The experimental results exhibit superior abilities of the hybrid actuation systems to tailor elastodynamic responses of the HSS rather than a single class of actuation system alone.

Advances in modelling the mechanisms and rheology of electrorheological fluids

  • An electrorheological fluid (ERF) is typically a suspension of semi-conducting solid particles dispersed in an insulating carrier fluid, and shows a dramatic change in rheological properties when an external electric field is applied. This rapid and reversible change in flow properties has potential application in many electronically controlled mechanical devices, but the development of efficient devices and optimal materials for ERF is still hindered by incomplete understanding of the fundamental physical mechanisms involved. In recent years there have been considerable advances In relating microstructural models to the rheological behaviour, and these will form the basis of this review. Results of the theoretical calculations and simulations will be compared to the key experimental evidence available. An overview of the fundamental physical concepts behind electrorheological fluid behaviour will also be presented.

Experimental response function of a photoelectron spectrometer

  • We developed the experimental function (ERF) which can be used for the numerical curve fitting analysis in photoelectron spectroscopy (PES). We selected the core-levels of Ag 3d5/2 and Au 4f7/2 to obtain the ERF from the measured core-level spectra. For the numerical fourier transformation we applied the fast transform (FFT) algorithm. we considered optical (Wiener) filtering with the FFT due to noise and used Hann window function to remedy the information leakage in frequency domain due to discrete and finite sampling of measurement. The comparison of the curve fitting results using the ERF obtained in this work and the mathematical response function with a gaussian in the conventional approach shows clearly the improvement of the curve fitting analysis.

  • The development of a robot system being able to work instead of human in the hazardous environment have been conducted for many year. In this study, the new design of controllers for the Master-Slave system is discussed. The Master-Slave system, force, velocity and torque signals are communicated between a master and a slave system. the conventional requires the enhancement of characteristics of tactility for minute force, precision signals and mechanical abrasion of loader. It is possible b controlling the viscosity of ERF(Electro-rheological fluid) since it varies with the electric field.   • This paper describe the vibration characteristics of cantilevered beam filled with ERF subjected to variousr electrode charge. The Proposed ER fluids used in the present study consists of starch particles and silicon oil. The ER fluids undergo a phase-change when subjected to an external electric filed.   • In this work, an experimental type of torque converter has been constructed and its characteristics have been evaluated by adapting an electrorheological fluid(ERF) as an operating medium. The device was designed by using the equations which were proposed by Carlson et al. The correlation between the rheological behaviour of an ERF and mechanical parameters of the clutch has been investigated. The torque generated by an ERF in this device is sum of one due to the yield strength by polarizing dispersed particles in dielectric oil and one due to the viscous drag. The experimental results are presented in terms of torque and current density as a function of rotational speed at various electric field strength applied. Experimental results showed that the measured torque was rapidly increased with the increase of the electric field, generally being proportional to the rotational speed of the motor. The measured current was shown to be increased with the increased electric field. Also, the current was decreased with the increase of increased with the increased electric field. Also, the current was decreased with the increase of the rotational speed of the motor and reached plateau region after f = 5 Hz.

  • An experimental investigation was performed to study the characteristics of ER(Electro-Rheological) fluid flow in a horizontal rectangular tube with or without D.C voltage. To determine some characteristics of the ER flow, 2D PIV(Particle Image Velocimetry) technique is employed for velocity measurement. This research found the mean velocity distribution with 0kV/mm, 1.0kV/mm and 1.5kV/mm for Re = 0, 0.62, 1.29 and 2.26. When the strength of the electric field increased, the cluster of ERF are clearly strong along the test tube and the flow rate decreased. In this study, the rheology of ER fluid stagnating or flowing through a dispersion meter will be investigated by PIV method. And then the ER effect, which appears at the ER valves and their appliance will be visualized.

  • In this study, ER fluids were working fluid and yield shear stress of attained dispersive ER fluids made by analyzed to adapt effectively at the practical equipment like three port ER valve. In addition, movement of ER particle was observed by using a microscope the same as the strength of electric field with Bingham characteristic. Basis material was presented to develop 3 port ER-Valve by quantitatively comparing and analyzing entrance, load and outlet flow's differences of 3 port rectangular tubes. This study, after designing and producing the 3port ER-Valve, considered the characteristic of pressure drop, outlet and load flow rate by changing the strength of electric field on ER fluids flowing between electrodes.
