Effects of the Feeding Mixed Oils of the Butter, Sardine and Safflower Oils on Fatty Acid Metabolism of Serum and Liver in Rats (버터, 정어리유 및 홍화유를 혼합 급이한 흰쥐의 혈청 및 간장의 지방산 대사에 미치는 영향)

  • This study was designed to observe the effects of the feeding mixed oils of the sardine oil containing n-3 EPA(eicosapentaenoic acid) and DHA (docosahexaenoic acid) and the safflower oil in which were plenty of n-6 linoleic acid, on the fatty acid metabolism of serum and liver in the dietary hyperlipidemic rats. Experimental oils mixed with 16% butter(control group), 8% butter + 8% olive oil and 8% butter with various level of sardine and safflower oils were administered to the male rats of the Sprague Dawley for 4 weeks. In the fatty acid compositions of serum phospholipid, triglyceride and cholesteryl ester, according as the contents of mixed safflower oil increased, n-3 PUFA(polyunsaturated fatty acid) contents and ratio of EPA/AA(arachidonic acid) tended to be decreased, but n-6 PUFA contents and ratio of AA/PUFA tended to be increased. In the fatty acid composition of triglyceride component in liver lipid, DHA contents were most in the liver lipid. And components of cholesteryl ester, n-6 linoleic acid contents were high percentage. Fatty acid compositions of serum and liver lipids were influenced from the fatty acid composition of the test lipids.

  • In order to investigate of the Influence of $Mg^{2+}$, $Ca^{2+}$ on ${\alpha}$-linolenic acid converted into the eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) forming in plasma lipid and in liver microsomes of rabbit, the animals were fed on the perila oil rich ${\alpha}$-linolenic acid or sardine oil rich EPA and DBA diet for 4 weeks were examined. In plasma, liver lipid, $Mg^{2+}$ was influenced on arachidonic acid(AA), EPA, DHA formative from ${\alpha}$-linolenic acid in perilla oil, but stearic acid was increased, $Ca^{2+}$ was Influenced on stearic acid increased and DHA was decreased. In phospholipid, $Mg^{2+}$, $Ca^{2+}$ was influenced on stearic acid increased and DHA was decreased in perilla oil.

The Growth and EPA Synthesis of Shewanella oneidensis MR-1 and Expectation of EPA Biosynthetic Pathway

  • Shewanella oneidensis MR-1 has the ability to inhale certain metals and chemical compounds and exhale these materials in an altered state; as a result, this microorganism has been widely applied in bioremediation protocols. However, the relevant characteristics of cell growth and biosynthesis of PuFAs have yet to be thoroughly investigated. Therefore, in this study, we have attempted to characterize the growth and fatty acid profiles of S. oneidensis MR-1 under a variety of temperature conditions. The fastest growth of S. oneidensis MR-1 was observed at $30^{\circ}C$, with a specific growth rate and doubling time of $0.6885h^{-1}\;and\;1.007 h$. The maximum cell mass of this microorganism was elicited at a temperature of $4^{\circ}C$. The eicosapentaenoic acid (EPA) synthesis of S. oneidensis MR-1 was evaluated under these different culture temperatures. S. oneidensis MR-1 was found not to synthesize EPA at temperatures in excess of $30^{\circ}C$, but was shown to synthesize EPA at temperatures below $30^{\circ}C$. The EPA content was found to increase with decreases in temperature. We then evaluated the EPA biosynthetic pathway, using a phylogenetic tree predicted on 16s rRNA sequences, and the homology of ORFs between S. oneidensis MR-1 and Shewanella putrefaciens SCRC-2738, which is known to harbor a polyketide synthase (PKS)-like module. The phylogenetic tree revealed that MR-1 was very closely related to both Moritella sp., which is known to synthesize DHA via a PKS-like pathway, and S. putrefaciens, which has been reported to synthesize EPA via an identical pathway. The homology between the PKS-like module of S. putrefaciens SCRC-2738 and the entire genome of S. oneidensis MR-1 was also analyzed, in order to mine the genes associated with the PKS-like pathway in S. oneidensis MR-1. A putative PKS-like module for EPA biosynthesis was verified by this analysis, and was also corroborated by the experimental finding that S. oneidensis MR-1 was able to synthesize EPA without the expression of $dihomo-{\gamma}-linoleic$ acid (DGLA) and arachidonic acid (AA) formed during EPA synthesis via the FAS pathway.

Effects of the Feeds Mixed with Various Level of Lard , Perilla Oil and Evening Primrose Oil on Fatty Acid Compositions of Liver and Brain Tissue in Rats (돈지, 들깨유 및 달맞이꽃 종자유의 혼합급이가 흰쥐의 간장 및 뇌조직의 지방산 조성에 미치는 영향)

  • In order to observe the effects of the fees mixed with the lard and two vegetable seed oils on the fatty acid compositions of liver and brain tissue, the oils mixed with 2.5% lard and various levels of perilla oil and evening primrose oil were administered to the male rats of the Sprague-Dawley for 4 weeks . In the fatty acid composition of liver lipid, saturated fatty acid (SFA) contents were rich in the phopholiipide and cholesteryl ester fraction. Monounsaturated fatty acid (MUFA) contents were rich in the triglyceride fraction and polyunsaturated fatty acid (PUFA) contents were rich in the phospholipid fraction. In the fatty acid composition of liver lipid fractions, according as the contents of mixed perilla oil decreased and the contents of mixed evening primrose oil increased , n -3 PUFA contents tended to decrease and n-6 PUFA contents tended to increase. Fatty acid composition of liver lipid fractions were influenced from the fatty acid composition of the test lipids. In the fatty acid composition of brain phospholipd, PUFA contents (40%) were rich and according as the contents of mixed evening primrose oil increased, the ratio on n-3/n-6 PUFA and eicosapentaenoid acid (EPA) /arachidonic acid (AA) tended to slightly decrease.

  • This study was performed to investigate the effect on polyunsaturate fatty acid diets and feeding periods on the antithrombosis. the hematological changes in the blood and fatty acid compositions of platelets in rats. Each group of rats was fed a diet containing 20%(W/W) corn oil beef tallow sardine oil and the general stock diet for 10, 20. 40 and 80 days. Rats fed sardine oil diet showed significantly longer bleeding time than any other diet groups after 20 days feeding The whole blood clotting time of sardine oil group fed for 80 days was increased significantly. The number of platelet and the concentration of hemoglobin showed no significant difference among all groups. The number of white blood cell was decreased continously in sardine oil group after 10 days feeding. The level of malondialdehyde generation during thrombin-induced aggregation of platelets was decreased continously in sardine oil grou after 20 days feeding. With regard to the composition of platelet fatty acid the ratio of eicosapentaenoic acid(EPA 20: 5 $\omega$-3) to arachidonic acid(AA 20:4 $\omega$-6) was increased in sardine oil group but decreased in corn oil groups and beef tallow groups with days. In conclusion the rats fed sardine oil diet for more than 20 days showed the fact that EPA induced the antithrombosis. the changes in number of white blood cell and the fatty acid composition of platelets.

  • The three analysis methods, EPA method 3050, the method offered by Ministry of Environment in Korea, and modified method corrected in this laboratory, were studied to investigate the effect of matrix on the analysis of inorganic priority pollutants. 7 inorganic priority pollutants(Ni, Cr, Cu, Zn, Pb, Cd, Hg) were spiked to the plating, leather, paper, electric, and dye sludges. Mean recovery of the elements except Hg was 95.5% when the procedure of EPA method was applied. However, recovery by the two other extraction methods showed 11.1% and 27.7%, respectively. Digestions were done by MDS (microwave digestion system) and $HNO_3+HClO_4$ methods. To study organic and inorganic matrix effect, samples were made by adding triethanol amine as a organic matrix and $FeCl_3{\cdot}6H_2O$+$AlCl_3{\cdot}6H_2O$ as a inorganic matrix, respectively. The extracts were analyzed by AAS and HG-AAS. Mean recovery of the elements by the $HNO_3+HClO_4$ procedure, except Hg, gave better result than that of the MDS method. Mean recovery of elements was decreased when organic and inorganic matrices were added in the sludge samples. The procedure of MDS and $HNO_3+HClO_4$ digestion gave higher recoveries than that of direct analysis. In general, the results of the studies showed a significant matrix effect on the inorganic priority pollutants analysis in sludges.

Effects of $\omega$3 and $\omega$6 Polyunsaturated Fatty Acid Diets on Fatty Acid Composition and Vitamin E Levels in Milk and Serum of the Rat ($\omega$3계 및 $\omega$6계 지방산 식이가 흰쥐의 모유와 혈청의 지방산 성분 및 비타민 E 수준에 미치는 영향)

  • The effects of feeding diets with different fatty acids on the composition of fatty acids and vitamin E status in maternal milk & serum and pup's serum were studied. Dietary fats(10% by wt) include on oil(CO), soybean oil(SO), perilla seed oil(PO : about 60% , C18 : 3 $\omega$3) and fish oil(FO : rich in C20 : 5$\omega$3, eicosapentaenoic acid = EPA & 22 : 6$\omega$3, docosahexaenoic acid = DHA), Sprague-Dawley rats weighing 200-250g, were fed experimental diets from pregnancy through lactation period. Maternal milk was obtained by gentle squeezing after 30 minutes of oxytocin(0.2 IU, intraperitoneal) injection. The fatty acid compositions of milk and serum were analyzed at day-2 and day-15. The concentrations of vitamin E in maternal milk and serum and pup's serum were also analyzed. The groups of CO, SO and PO which had no DHA in their diet, contained DHA in their milk, The rations of EPA+DHA/arachidonic acid(AA) were higher in PO group than those in either CO or SO group. This seemed to be due not only to more conversion from C18 : 3$\omega$3 to C20 : 5$\omega$3 and C22 : 6$\omega$3 but also to inhibition of C18 : 2$\omega$6 conversion to C20 : 4$\omega$6. More DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA was found in day-2 milk than in day-15 milk. It was also noted that milk contained more DHA than serum and this difference was larger in day-2 than in day-15 milk. Even though the concentrations of vitamin E both in maternal serum and milk were lower in PO and FO groups fed highly unsaturated fat than in CO or SO groups, pup's serum did not show a significant difference among all the experimental groups indicating that the pups man secure their essential nutrients by the biomagnification mechanism.

Effects of Dietary Fatty Acids on Serum Lipids and Fatty Acid Composition of Serum Phospholipids in Men

  • Serum lipid levels and fatty acid composition of serum phospholipids(PL) were investigated in 24 healthy male subjects who consumed either corn oil(CO) rich In linoleic acid(LA), perilla oil(PO) rich in $\alpha$-linolenic acid($\alpha$-LNA), or canola oil acid(OA) as a major fat source fir 5 weeks. The PO and the CNO groups showed significant(P <0.05) increases in serum high density lipoprotein-cholesterol(HDL-C) levels and in ratios of HDL- C/total cholesterol(TC) compared with initial values measured at the beginning of the study. Significantly(p <0.05) increased concentrations of serum triglycerides(TG) ware observed after 5 weeks of the CO based diet compared with both its intial value and the concentration observed after 5 weeks with the PO-based diet. fatty acid composition of serum PL reflected changes in dietary fatty acid composition and metabolism. Compared with the initial levels, significandy increased contents of eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) were observed in serum PL of the PO group and significantly increased contents of $\alpha$-LNA and EPA were observed in the CNO group. Arachidonic acid(AA) content of serum PL did not change in the CO group during the study period, although, the increase in LA was significant(P <0.05). Compared with the CO-based diet, both the PO and the CNO-based diets seem to have beneficial effects on atherosclerosis by influencing the serum lipid profile and fatty acid composition of serum PL.

  • A gas chromatography-mass spectrometric (GC-MS) method was developed for determining 25 semivolatile organic compounds in water. A 1.0 L water sample was placed in a separatory funnel and saturated with NaCl, and the solution was extracted two times with 40 mL of methylene chloride. Under the established condition, the linear quantification range was 0.02-800 ng/L and the relative standard deviation was less than 15%. The method was used to analyze 16 surface water samples collected from various regions in Gum-River. The samples revealed SVOC concentrations in the range of 0.02-96.8 ng/L. Maximum concentrations of VOCs detected were not exceeded the EPA or Germany guidelines in any of the samples. The developed method may be valuable for monitoring SVOCs in water.
