Analysis of the Researches on Stress and Immune Responses (스트레스와 면역반응에 대한 국내 논문분석)

  • This study was aimed to analyze the variables measuring stress and immune responses, to identify the relationship between stress and immune responses, and to find out the effect of nursing interventions associated with stress and immune responses by reviewing thirty-four published articles since 1970 in Korea. The articles were selected in the field of nursing, stress management, and masters or doctoral dissertations and limited to human subject. Among these, the thirty-one articles were published since 1996 and mainly distributed in nursing (44.1%) and medicine(44.1%). The prevailing research design was nonequivalent control pre-post experimental design(41.1%). The research subjects were 55.9% for patients and 44.1% for healthy general persons including 20.6% of university students. To evaluate stress, both physiologic and psychosocial measures were adapted together in 35.3% of the articles. The most frequent two variables measuring stress and immune response were cortisol level(15.9%) and number or activity of natural killer cell(25.9%). The relation between stress and immune responses was positive in 4 articles, negative in 9 cases, and none in 12 cases. Decreased stress and enhanced immune function have been found when massage, abdominal breathing, exercise, relaxation, and touch were provided as nursing interventions. The articles to investigate the relationship between stress and immune function were limited and the tested variables were diverse. Also there was no consistent evidence to correlate the stress and immune function at present. Further studies are needed to construct a valid research design and to investigate the relationship between stress and immune responses. Nursing interventions to decrease stress should be developed to result in the increased immune function and the effect of these interventions would be verified.

Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii (Phellinus baumii으로부터 세포외 다당체 생산의 최적화)

  • The polysaccharide isolated from Phellinus species has been known as a folk remedy, including antitumor and immune-stimulating activities. However, there are lacks of knowledge about mycelial growth and exopolysaccharide (EH) production in its submerged culture. We investigated the optimal conditions on mycelial growth and EPS production in Phellinus baumii. The optimal temperature and initial pH for mycelial growth and EPS production in shake flask culture of P. baumii were proved to be 3$0^{\circ}C$ and pH 5.0, respectively. In case of carbon source, cellobiose and maltose were highly efficient for mycelial growth and fructose and mannitol were also relatively favorable for EPS production. Yeast extract was the most suitable nitrogen source for mycelial growth and EPS production. The composition of optimal culture medium was determined to be fructose 20 g/L, yeast extract 20 g/L, and $CaCl_2$ 0.55 g/L, respectively. Under the optimal culture condition, the maximum mycelial biomass and EPS achieved in a 5-L stirred-tank fermenter were 17.43 g/L and 3.6 g/L, respectively. It was found that the EPS was a glycoprotein onsisted of mainly arginine (14.1%) and glycine (12.0 %) in protein moiety and mainly mannose (48.7%) and arabinose (38.4%) in carbohydrate moiety.

Effects of Ara-C on UV and MMS-induced Excision Repair, Chromosome Aberrations, Sister Chromatid Exchanges and Replication Inhibition (자외선과 MMS에 의한 절제회복, 염색체이상, 자매염색분체 교환 및 복제억제 현상에 미치는 Ara-C의 영향)

  • Unscheduled DNA synthesis, chromosome aberrations, sister chromatid exchanges and DNA replication inhibition induced by the combined treatments with ara-C and UV-light or MMS in $HF_1$, CHO and $HelaS_3$ cells were studied, and the results obtained were as follows: (1) Ara-C was found to inhibit UV-or MMS-induced unscheduled DNA synthesis and the inhibitory effect of ara-C was more remarkable in its post-treatment. (2) Ara-C enhanced the rate of chromosome aberrations induced by MMS or UV-light. Post-treatment with ara-C exhibited the synergistic effect on MMS-induced chromosome aberrations mainly by increases of chromatid deletions. (3) Contrarily, ara-C did not increase the rate of sister chromatid exchanges, particularly in the pre-treatment with MMS, although it was found to induce sister chromatid exchanges. (4) The rate of DNA synthesis was declined immediately after are-C treatment and then recovered. The combined treatments with ara-C and UV-light or MMS showed that the initial response on replication inhibition was similar to that of ara-C, but later responses were similar to that of UV-light or MMS treated group.

  • Objective : The aim of this study is to evaluate internal reliability and correlation among assessment instruments of knee OA. Methods : Data were obtained from 85 patients with knee OA. They were asked to answer VAS pain scale, two disease-specific questionnaires (WOMAC and LFI) and one instrument about health-related quality(KHAQ). Reliability was assessed using cronbach`s alpha. Correlation was assessed by examining the pearson`s correlation coefficients. Results : Cronbach`s alpha of all Index except WOMAC stiffness subscale.(6679) exceeded the value of 0.70. A good correlation was obtained between WOMAC Subscales (especially Pain Subscale) and LFI (.726

Changes in Phosphatase Activity of the Mouse Uterus during the Estrous Cycle (發情週期에 EK른 생쥐子宮의 Phosphatase 活性의 變化에 관하여)

  • Quantitative analysis of the activities of transport ATPases as well as alkaline phosphatase of the mouse uterus was carried out during the estrous cycle. Even though the proportional patterns of the enzyme activities were similar each another between the stages of estrous cycle, the absolute activities of the enzymes except $K^+$-dependent and $Na^+$, $K^+$-activated ATPases at the time of estrus were significantly (p<0.025) higher than that at any other time of the estrous cycle. That is, the activities of $K^+$-dependent and $Na^+$, $K^+$-activated ATPases were negligible during the period of time from diestrus to estrus while the little activities (0.04 $\\sim$ 0.05$\\mu$M/mg protein/hr in average, $6\\sim7$% of the total enzyme activity) of these enzymes appeared at the time of metaestrus. On the other hand, at the time of estrus, the activities of $Mg^++$-dependent phosphatase, transport ATPase and alkaline phosphatase were rapidly and tremendously increased to be 0.69 (35%), 0.42 (21%) and 1.58 (79%), respectively. The activity of alkaline phosphatase was in the range of 0.60 $\\sim$ 1.58 (79 $\\sim$ 90%) and predominant throughout the estrous cycle. The activity of $Mg^++$-dependent alkaline phosphatase was estimated as 12 $\\sim$ 16% of the total enzyme activity. Therefore, it is assumed likely that $K^+$-dependent and $Na^+$, $K^+$-activated ATPases are not the main factors to control the fluid accumulation at the time of estrus, but may be the factors to reabsorb the luminal fluid into the uterine epithelium at the time of metaestrus, and that $Mg^++$-dependent phosphatase, transport ATPase and alkaline phosphatase must be closely involved in the secretion of luminal fluid from the epithelial cells of the mouse uterus.

Characteristics of Groundwater Pollution and Contaminant Attenuation at Waste Disposal Sites (폐기물 매립지 주변의 지하수 오염과 오염물질의 지연 특성)

  • The objectives of this study are to investigate the groundwater and surface water contamination, to interpret the attenuation mechanism of contaminant transport, and to find the appropriate contamination indicator. at the two big landfill sites : Nanjido Landfill and Hwasung Landfill. Leachate from the Nanjido, th, Hwasung and the Kimpo waste disposal sites is characterized by high temperature (31.7-40.1$^{\circ}C$), high electric conductivity (14,650-32,800 ${\mu}$S/cm), somewhat higher pH(7.58-8.45) and low Eh (-119.4-20.4 mV), and is enriched in both major (Na$^{+}$, K$^{+}$, Ca$^{2+}$, Mg$^{2+}$, HC $O_3$$^{-}$, Cl$^{-}$) and minor (Mn, Sr$^{2+}$, Ba$^{2+}$, Li$^{+}$, F$^{-}$, Br$^{-}$) ions. Municipal solid waste leachate and industrial waste leachate are effectively discriminated by the content of S $O_4$$^{2-}$, Fe, and heavy metals. The attenuation mechanism of each component was assessed using the chemical analysis. Cl-normalizing process, WATEQ4F simulation, and preceding flownet analysis. Based on the calculation of Contamination Factor, K, Na, Ca, Mg, B, Zn, HC $O_3$, Cl, F, Br and TOC are effective contamination indicators in the Nanjido landfill site, and K, Na, Ca, Mg, B, S $O_4$, HC $O_3$, Cl, F, Br and TOC in the Habsburg landfill site Particularly, TOC is the best contamination indicator in landfill sites influenced by sea water.

Flavor Improvement of a Complex Extract from Poor-quality, Individually Quick-frozen Oysters Crassostrea gigas (IQF 굴(Crassostrea gigas) 복합엑스분의 추출 및 풍미개선)

  • To develop an effective use for poor-quality individually quick-frozen (IQF) oysters Crassostrea gigas stored for a long period, the extract conditions, quality characteristics, and optimum reaction flavoring (RF) conditions of a complex extract from these IQF oysters were investigated. The moisture, pH, and volatile basic nitrogen contents of IQF oysters stored for 18 months (18M-IQFO) were 77.9%, 6.32, and 17.9 mg/100 g, respectively. Three different kinds of extract were prepared from 18M-IQFO: a hot-water extract (HE), scrap enzymatic hydrolysate (EH), and complex extract (CE). The respective extracts contained 5.5, 8.6, and 6.6% crude protein and 281.7, 366.0, and 343.0 mg/100 g amino nitrogen, and had 811, 359, and 1,170 mL/kg extraction yields. The CE was superior to the traditional HE in terms of the extraction yield, amino-nitrogen content, and organoleptic qualities, except for the odor. To improve flavor via the Maillard reaction, the reaction system used to produce a desirable flavor comprised CE (Brix $30^{\circ}$), 0.4 M glucose, 0.4 M glycine, and 0.4 M cysteine solution (4:2:1:1, v/v). The reaction time and pH were the independent variables, and the sensory scores for baked potato odor, masking shellfish odor, and boiled meat odor were the dependent variables. The surface response methodology (RSM) analysis of the multiple responses optimization gave a reaction time of 120.6 minutes and pH 7.33 at $120^{\circ}C$. The reaction improved the flavor of CE considerably, as compared to that of the unreacted extract.

Production of Enantioselective Lipase from Acinetobacter sp. SY-01 (Acinetobacter sp. SY-01로부터 Enantioselective Lipase의 생산)

  • Lipase from Acinetobacter sp. SY-01 plays an important role enzyme that products chiral drug. We investigated optimum condition for mass production of Acinetobacter sp. SY-01 lipase. Addition of among the different oils to medium. olive oil was optimal for enzyme production. When 0.2% olive oil was added as a carbon source, the production of lipase was increased to a maximum. The optimum pH and temperature were pH 7 and $30^{\circ}C$. In the presence of $Fe^{2+}$ and $Ca^{2+}$, the lipase activity was dramatically enhanced by 280% and 160%, respectively. SY-01 lipase was stable in the most of the DMSO among organic solvents. The addition of triton-X 100 increased the SY-01 lipase by 100-fold. The optimum composition of medium for production of the enzyme was 0.8% yeast extract, 0.2% olive oil, 0.4% triton X-100+40% DMSO. 0.1% $NH_4Cl$, 0.4% $K_2HPO_4$ 3.9% $NaH_2PO_4$, 0.03% $CaCl_22H_2O$, 0.01% $FeSO_4$$7H_2O$(pH 7.0).

Study on the Oxidation Process of Potential Acid Sulfate Soil (잠재 산성황산염토양의 산화과정에 대한 연구)

  • To find out the oxidation process of potential acid sulfate soil(PASS) along with time. the PASS were treated with lime and ammonia water to adjust soil pH in laboratory column condition. pH range of PASS showed 6.5 to 7.5. however, complete oxidized PASS by $H_2O_2$ showed 2.1 to 2.5. After pilling the PASS under the natural condition. oxidation occured slowly from surface of the pilled soil. The oxidation of PASS proceeded slowly when the soil was in submerged condition. but quickly in dried condition. The content of sulfide-sulfur in PASS sharply decreased after exposing to the air and the decreasing rate was greater in dried than in submerged condition. The content of sulfate-sulfur continuously decreased in submerged condition. but increased in dried condition. Contents of $Fe^{+{+}}$ and $Al^{+{+}}$ in PASS were generally increased with time and the increasing rate was greater in submerged than in dried condition. Liming to PASS was slowly acting to pH change and ammonia water caused fast pH change within a short period of time. The contents of sulfate-sulfur and exchangeable aluminum in drainage water decreased with time and the contents of sulfide-sulfur and ferrous iron were increased.

A Biogeochemical Study on the Heavy Metal Leaching from Coal Fly Ash Disposed by Dangjin Fire Plant in the Coastal Environment (당진화력발전소의 석탄회 연안매립과 중금속 원소의 용출에 대한 생지화학적 연구)

  • It is known that coal-derived fly ashes have the unique chemical composition and mineralogical characteristics. Since iron oxides in coal fly ash are enriched with heavy metals, the subsurface media including soils, underground water, and sea water are highly likely contaminated with heavy metals when the heavy metals are leached from fly ashes by water-fly ash interactions. The purpose of this study was to investigate how indigenous bacteria affect heavy metal leaching and mineralogy in fly ash slurry during the fly ash-seawater interactions in the ash pond located in Dangjin seashore, Korea. The average pH of ash pond seawater was 8.97 in nature. Geochemical data showed that microbial activity sharply increased after the 7th day of the 60-day course batch experiments. Compared with other samples including autoclaved and natural samples, ${SO_4}^{2-}$ was likely to decrease considerably in the fly ash slurry samples when glucose was added to stimulate the microbial activity. Geochemical data including Eh/pH, alkalinity, and major and trace elements showed that the bacteria not only immobilize metals from the ash pond by facilitating the chemical reaction with Mn, Fe, and Zn but may also be able to play an important role in sequestration of carbon dioxide by carbonate mineral precipitation.
