Effects of Self-foot Reflexology on Fatigue and Sleep States in Women Nurses (자가 발반사마사지가 여성간호사의 피로와 수면상태에 미치는 효과)

  • Purpose: This study examined the effects of Self-foot reflexology(SFR) on fatigue and sleep states in clinical women nurses. Method: This study was a nonequivalent pretest-posttest quasi-experimental. Women nurses were assigned to an experimental group (EG, n=20) or a control group (CG, n=20). The EG participated in SFR for a total of 40 minutes, 2 times per week during 4 weeks on their Rt & Lt feet. The CG did not receive SFR during the research period. The EG & CG had never before received SFR, and they had no open wounds or fractures. Data was analyzed using the $x^2$-test, and t-test by the SPSS version 12.0 program at a 5% significant level. Results: The score of fatigue in the EG was significantly lower than that of the CG and the score of sleep states in the EG was also significantly higher than that of the CG. Conclusion: The findings show that the score of fatigue decreased and sleep states increased in the study. Therefore, we should consider SFR as an intervention on clinical nurses. However, it is still needed to verify its effects through more intensive study.

  • Effect of Mylabris sidae(MS) and Epicauta gorhami(EG) polysaccharide fractions on the inflammation and immune responses were studied in vivo. MS and EG contained cantharidin about 0.61 and 0.65%, respectively. It was shown that MS and EG polysaccharide fractions at a oral dose of 100 mg/kg have the significant anti-inflammatory and analgesic activity; They inhibited significantly the carrageenin-induced inflammation and acetic acid-induced writhing syndrome. They accelerated significantly the carbon clearance and the phagocytosis of colloidal carbons by Kupffer cells in liver, but they at a oral dose of 100 mg/kg suppressed significantly the Arthus reaction in the sheep red blood cell(S-RBC)-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer and plaque-forming cells. On the other hand, they at a oral dose of 200 mg/kg accelerated slightly the oxazolone-induced dermatitis in rats and delayed hypersensitivity in the S-RBC-challenzed mice in consistent with the increase of rosette forming cells. As the above results, it exhibited that MS and EG polysaccharide fractions inhibited the humoral immune responses, but they accelerated the function of macrophages and cellular immune responses. EG polysaccharide fraction had more active than MS polysaccharide fraction.

Enhanced Skin Permeation of a New Capsaicin Derivative (DA-5018) from a Binary Vehicle System Composed of Isopropyl-myristate and Ethoxydiglycol

  • DA-5018, a recently synthesized capsaicin analog, appears to possess potent analgesic activity when administered topically. The objective of this study is to test the feasibility of the topical administration of this compound. Specifically, our goal was to identify vehicle system that permit a reasonable transdermal permeation of the compound in mice. Among the vehicles examined, isopropyl myristate (IPM) showed the largest in vitro permeability across the intact skin (83.6 ${\pm}$ 5.42${\mu}$l/$\textrm{cm}^2$/h ). However, due to the limited solubility of DA-5018 in IPM (0.53 mg/ml), the maximal flux from the IPM medium remained at only 44.3 ${\pm}2.87{\mu}$g/$\textrm{cm}^2$/hr. In order to increase the flux, addition of better solvents for DA-5018 was attempted, under the assumption that flux is the result of both solubility and permeability. Ethoxydiglycol (EG) and oleic acid (OA) were selected as examples of food solvents. The addition of IC or OA to IPM at a 1:1 volume ratio resulted in a comparable increase in the solubility of the compound (i.e., to 61.1 and 50.2 mg/ml for EG and OA, respectively). However, the addition of EG at a 1:1 volume ratio, for example, increased the flux 6.3 fold (i.e., $279{\mu}$g/$\textrm{cm}^2$/hr), while OA, at a 1:1 volume ratio, decreased the flux 5 fold (i.e., $9.26{\mu}$g/$\textrm{cm}^2$//hr). The mechanism of this discrepancy between EG and OA was investigated by measuring the permeabilty of DA-5018 across the stratum corneum-removed skin of the mouse, under the hypothesis that the viable skin layer may serve as a barrier for the permeation of lipophilic substances such as DA 5018. The permeability of DA-5018, from the medium of EG or OA, across the viable skin differed greatly for EG ($0.41{\mu}$l/$\textrm{cm}^2$/hr) and OA ($0.086{\mu}$l/$\textrm{cm}^2$/hr), suggesting that a higher permeability across the viable skin layer is needed for the second solvents. The maximum flux across the intact skin was achieved for DA-5018 when EG was added to IPM at a 1:1 volume ratio. Thus, the use of a binary system appears to be the best approach for realizing the transdermal delivery of DA-5018 at a reasonable rate.

  • Eugenol is a volatile compound synthesized by eugenol synthase in various plants and belongs to phenylpropene compounds. However, characteristics of eugenol synthase in tomato has not been known. Therefore, we cloned a full length cDNA of a putative eugenol synthase from tomato 'Micro-Tom' using rapid amplification of cDNA ends (RACE) technique and named a clone SlEGS. Open reading frame of SlEGS was 921bp long and its deduced amino acid sequence was 307bp. The BLAST analysis indicated that SlEGS shared high similarity with PhEGS1 (67.1%) and CbEGS2 (69.4%). Amino acid composition of SlEGS was determined by CLC genomics workbench tool and 3D structure of SlEGS was constructed by homology modeling using Swiss-PDB viewer and validated using PROCHECK and ProSA-web tool. In addition, the physiochemical properties of SlEGS was evaluated using ExPASy's ProtParam tool. Molecular weight was 33.93kDa and isoelectric point was 5.85 showing acidic nature. Other properties such as extinction coefficient, instability index, aliphatic index, and grand average hydropathy was also analyzed.

Development of Effective Cryopreservation Method for Mammalian Embryo (포유류 초기 배아의 효율적인 동결 보존 방법에 관한 연구)

  • This study was carried out to evaluate the effects of embryonic stage, cryoprotectant, and freezing-thawing method on the rates of survival and development of the cryopreserved mouse early embryo and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology (ART). Two to eight cell embryos were obtained from oviducts of mated $F_1$ hybrid female mice superovulated by PMSG and hCG. Two-step EG, DMSO and 4-step EG, DMSO were used as cryoprotectant and dehydration and rehydration method of embryos, and slow-cooling or rapid-cooling method was used as frozen program. The survival rates of embryos were measured after thawing and rehydration, and the developmental rates of embryos were compared and observed during culturing embryos for 24, 48, 72, 96 hrs. As for the survival and development rates of embryos according to embryonic stage, the survival rate of 2 cell stage in EG and DMSO was significantly higher than 4~8 cell (65.4% versus 61.2%, 81.1% versus 72.5%) (p<0.01, p<0.01), but the development rates of 4~8 cell embryos in EG and DMSO were significantly higher than 2 cell embryos for whole culture period (p<0.01) and the development rates of 4~8 cell embryos in EG were significantly higher than 2 cell embryos in DMSO (p<0.01). As for the survival and development rates of embryos according to cryoprotectant, the survival rate of 2 cell embryo in DMSO was significantly higher than that in EG(77.0% versus 64.4%) (p<0.01), whereas the development rate of embryos was not differ till 24 hrs. The development rate from morular to hatching blastocyst, however, was sinificantly higher in EG than in DMSO during 48 hr (p<0.01). The survival rate of 4~8 cell embryo was 62.5% in EG and 73.3% in DMSO. The development rates of embryo in EG were significantly higher for whole culture periods (p<0.01, 0.05). In respect to the effect of freezing and thawing program on the survival and development rates of embryos, method of slow cooling and rapid thawing was more effective than that of rapid cooling and rapid thawing. The survival rate of embryo in 2 cell stage was higher than in 4~8 cell stage, and EG appears more effective cryoprotectant than DMSO because EG showed better development rates of embryos in 2 and 4~8 cell stage. Moreover, slow cooling and rapid thawing method was considered as the best cryopreservation program.

Effects of Meiotic Stages, Cryoprotectants, Cooling and Vitrification on the Cryopreservation of Porcine Oocytes

  • Different factors may affect the sensitivity of porcine oocytes during cryopreservation. The effect of two methods (cooling and vitrification), four cryoprotectants [glycerol (GLY), 1, 2-propanediol (PROH), dimethyl sulfoxide (DMSO) or ethylene glycol (EG)] and two vitrification media (1 M sucrose (SUC)+8 M EG; 8 M EG) on the developmental capacity of porcine oocytes at the germinal vesicle (GV) stage or after IVM at the metaphase II (M II) stage were examined. Survival was assessed by FDA staining, maturation and cleavage following IVF and IVC. A toxicity test for different cryoprotectants (GLY, PROH, DMSO, EG) was conducted at room temperature before cooling. GV and M II-oocytes were equilibrated stepwise in 1.5 M cryoprotectant and diluted out in sucrose. The survival rate of GV-oocytes in the GLY group was significantly lower (82%, p<0.01) than that of the other group (92 to 95%). The EG group achieved a significantly higher maturation rate (84%, p<0.05) but a lower cleavage rate (34%, p<0.01) than the DMSO group and the controls. For M II-oocytes, the survival rates for all groups were 95 to 99% and the cleavage rate of the GLY group was lower than the PROH-group (21 vs 43%, p<0.01). After cooling to $10^{\circ}C$, the survival rates of GV-oocytes in the cryoprotectant groups were 34 to 51%, however, the maturation rates of these oocytes were low (1%) and none developed after IVF. For M II-oocytes, the EG group showed a significantly higher survival rate than those of the other cryoprotectant groups (40% vs 23-26%, p<0.05) and the cleavage rates of PROH, DMSO and EG group reached only 1 to 2%. For a toxicity test of different vitrification media, GV and M II-oocytes were equilibrated stepwise in 100% 8 M EG (group 1) and 1 M SUC + 8 M EG (group 2) or equilibrated in sucrose and then in 8 M EG (SUC+8 M EG, group 3). For GV-oocytes, the survival, maturation and cleavage rates of Group 1 were significantly lower than those in group 2, 3 and control group (p<0.05). For M II-oocytes, there were no differences in survival, maturation and cleavage rates between groups. After vitrification, the survival rates of GV and M II-oocytes in group 2 and 3 were similarly low (4-9%) and none of them matured nor cleaved after in vitro maturation, fertilization and culture. In conclusion, porcine GV and M II-oocytes do not seem to be damaged by a variety of cryoprotectants tested, but will succumb to a temperature decrease to $10^{\circ}C$ or to the process of vitrification, regardless of the cryoprotectant used.

Effects of Kimchi Supplementation and/or Exercise Training on Body Composition and Plasma Lipids in Obese Middle School Girls (운동 및 김치 보충제 섭취가 비만 여중생의 신체조성 및 혈중지질에 미치는 영향)

  • The effects of aerobic exercises and/or supplementation of kimchi on changes of the body composition and plasma lipids of obese middle school girls were studied. Thirty eight girls, 28 obese girls and 10 normal weighed girls, were paricipated. Among obese girls, 8 were assigned to exercise group (FG) 12 were grouped as kimchi group (KG) , and 8 were asked to practice excercise and to take kimchi simultaneously(excercise kimchi group, EKG). Ten girls whose weight is normal asked to remain o their own diet during 6 weeks of experiment (control group, CG)/ EG practiced jogging and rope-jumping for 60 minutes four times a week and KG took 3 g of freeze-dried kimchi packed in a 500 mg capsule daily which is equivalent to 30 g of fresh kimchi, EKC, EG and KG showed beneficial effects on changes of the body composition and plasma lipids compared to those of CG, EG showed greater effect than KG in reducing body fat resulted decrease in BMI, fat mas,. abdominal fat, and triglyceride concentration and increase in HDL-cholesterol. KG seemed to have greater effect on lowering plasma cholesterol and LDL-cholesterol than EG/ But the greatest effects in terms of reduction in weight, BMI fat mass, abdominal fat, total cholesterol, LDL-cholesterol, triglyceride, and increase in HDL-cholesterol were observed from EKG. These results indicate that kimchi supplemenation while practicing excercise might improve the obese state by reducing body fat content as well as reducing plasma lipids.

  • The objective of this study is to examine an effective cryopreservation method and various vitrification containers on the survival vates of embroys. For the vitrification, in vitro produced embryos at blastocyst stage were exposed to ethylene glycol 5.5 M freezing solution (EG5.5) for 20 sec, loaded on containers such as grid, straw and paper and then immediately plunged into - 196$^{\circ}C$ L$N_2$. The blastocysts were thawed serially in 0.5, 0.25 and 0.125%; P < 0.05). Therefore, this study suggests that bovine embryos can be easily, effectively and successfully cryopreserved by grid, straw, and paper in the presence of freezing solution. Furthermore, vitrification using paper may be used as a no M sucrose in CR1aa, each for 1 min, and cultured in CR1 an medium supplemented with 10% FBS. After thawing, there were not significant differences in recovery rates of EM grid, straw and paper as 84.6, 88.3, and 93.7%, respectively (Table 1). However, survival rates of EM grid (78.1%) and paper (77.1%) showed significantly higher than straw (52. 1w method for bovine embryos.

Efficient Cryopreservation of Hanwoo (Korean Cattle) Blastocysts Derived from Nuclear Transfer with Somatic Cell Using Vitrification

  • Cryopreservation by vitrification of Hanwoo blastocysts derived from nuclear transfer with Hanwoo adult ear cell was compared with that of in vitro fertilized blastocysts. For vitrification, day 7 or day 8 blastocysts were serially exposed in glycerol (G) or/and ethylene glycol (EG) mixtures (10% G for 5 min, 10% G plus 20% EG for 5 min, and 25% G plus 25% EG for 30 sec) which was diluted in 10% FBS added D-PBS. Thawing of straw was carried out in air for 10 sec and then in water bath of $25^{\circ}C$ for 20 sec. The contents of the straw (0.2 ml) was expelled into a culture dish contained with 0.5M sucrose and 10% FBS(S-DPBS) by cutting the cotton plug and then the recovered embryos were put into fresh 0.25M and 0.125M S-DPBS for 30 sec, respectively, The embryos were transferred into D-PBS with 10% FBS for 5 min. and were cultured in a 10u1 droplet of co-culture environment (cumulus cell monolayer + 10% added CRl medium) for 24 h. In the result, survival rates were 88.9% and 85.4% for nuclear transfer embryos and in vitro fertilized embryos, respectively. After transfer of vitrified-thawed blastocysts produced from nuclear transfer, 4 of 5 total recipients did not return to the subsequent estrus cycle at 30 days. It is concluded that the Hanwoo blastocysts derived from nuclear transfer can be successfully cryopreserved using simple and efficient vitrification method.

Effect of Groove Conditions on the Mechanical Properties of Welds Produced by the Combined Welding Process of Flux Cored Arc and Electro Gas in EH36 TMCP Steel Plate for Hull Structures (선체구조용 EH36 TMCP 후판의 FCAW 및 EGW 조합 용접부 기계적 성질에 미치는 개선조건의 영향)

  • Characterization of microstructures and mechanical properties of 83mm thickness EH36-TM welds produced by the combined flux cored arc (FCA) and electro gas (EG) welding processes has been studied with the two different groove conditions, single-V (SV) and double-V (DV) bevels. The welding consumables used for FCA and EG welding processes were ASME/AWS A5.29 (E81T1-K2) and A5.26 (EG72T), respectively. Experimental results showed that all the mechanical properties of welds such as tensile property, CVN toughness and Vickers hardness met IACS requirements. The tensile strength of EG welded plates were reduced by approximately 4% (DV: 3.8%, SV: 4.2%) compared to the base metal. The hardness value of SV-beveled weld metal was slightly lower than that of DV-beveled one. There were no significant differences as per welding groove conditions except for the weld metal. In addition, at the fusion line, the toughness of SV condition was 20J lower and the weld metal was 40J lower than DV condition, respectively. On the basis of microstructural analysis, grain boundary ferrite (GBF) structures for SV condition were 2 times higher volume fraction than for DV condition and their packet sizes were coarsened to almost double. It was thus suggested that the GBF volume fractions and packet sizes in the weld metal of EH36-TM steel plates are the most important factors affecting the mechanical properties of the combined FCA and EG welded joint. Nevertheless, all the results of welds with both DV and SV conditions were found to be excellent.
