김치에서 분리한 세균인 Lactobacillus plantarum YK-9의 식중독 원인세균에 대한 항균활성 및 특성 (Antibacterial Activity against Food-poisoning Causing Bacteria and Characterization of Lactobacillus plantarum YK-9 Isolated from Kimchi)

  • 본 연구는 김치에서 분리한 Lactobacillus plantarum YK-9의 다양한 특성을 조사하기 위하여 실시되었다. 형태학적 관찰 및 생리화학적 특성에 대하여 조사하였다. 16S rRNA 염기서열 분석을 통해 균주를 동정하였고, 그 결과 Lactobacillus plantarum로 확인되었고, L. plantarum YK-9으로 명명하였으며, 이 균주는 GenBank에 [FJ669130]로 등재하였다. 배양 시간에 따른 L. plantarum YK-9의 생장과 pH의 변화를 조사하였으며, 대사산물로서 유기산 (lactic acid와 acetic acid)의 생성은 HPLC를 통하여 확인하였다. 유기산의 생성은 L. plantarum YK-9의 생장에 따라 증가 하는 것을 확인하였다. 초기 pH 7.0은 배양 기간동안 3.6으로 감소하였고, 배양 72시간이 지난 후 생성된 lactic acid와 acetic acid의 농도는 각각 588.7 mM과 255.5 mM이었다. 식중독 원인균인 S. aureus, B. cereus, L. monocytogenes, E. coli, E. faecalis, S. enteritidis에 대하여 20배로 농축된 YK-9 배양 상등액에 처리하여 항균력을 조사하였으며, 실험에 사용된 6가지 식중독 원인세균 모두에 대한 항균효과가 있는 것이 관찰되었고, 이는 유기산 생성에 기인하는 것으로 나타났다.

Immunostimulatory Effect of Ovomucin Hydrolysates by Pancreatin in RAW 264.7 Macrophages via Mitogen-Activated Protein Kinase (MAPK) Signaling Pathway

  • Ovomucin (OM), which has insoluble fractions is a viscous glycoprotein, found in egg albumin. Enzymatic hydrolysates of OM have water solubility and bioactive properties. This study investigated that the immunostimulatory effects of OM hydrolysates (OMHs) obtained by using various proteolytic enzymes (Alcalase®, bromelain, α-chymotrypsin, Neutrase®, pancreatin, papain, Protamax®, and trypsin) in RAW 264.7 cells. The results showed that OMH prepared with pancreatin (OMPA) produced the highest levels of nitrite oxide in RAW 264.7 cells, through upregulation of inducible nitric oxide synthase mRNA expression. The production of pro-inflammatory cytokines such as tumor necrosis factor-α and interleukin-6 were increased with the cytokines mRNA expression. The effect of OMPA on mitogen-activated protein kinase signaling pathway was increased the phosphorylation of p38, c-Jun NH2-terminal kinase, and extracellular signal-regulated kinase in a concentration-dependent manner. Therefore, OMPA could be used as a potential immune-stimulating agent in the functional food industry.

양단탕 추출물의 항염증 효과에 대한 연구 (Studies on Anti-inflammatory Effects of Yangdan-tang Extracts)

  • Yangdan-tang (YD) is recorded as a treatment to treat exterior-related fever illness in the Korean medicine. In this study, we examined the anti-inflammatory effects of YD, using YD water extract and lipopolysaccharide (LPS)-induced RAW 264.7 cells. First of all, we measured the amount of nitric oxide (NO) and prostaglandin E2 (PGE2), the products of inflammatory metabolism. Also, we measured enzymes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), as well as cytokines such as tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6), interleukin 1 alpha (IL-1α), and interleukin 1 beta (IL-1β). YD suppressed the production of NO and PGE2 in a dose dependent manner and reduced the amount of protein and the mRNA expression of iNOS and COX-2. Also, YD reduced the mRNA expression of TNF-α, IL-6, IL-1α and IL-1β. In conclusion, YD decreased production of LPS-induced inflammatory factor, which could be a clinical basic subject for inflammatory diseases.

가미지황탕(加味地黃湯)이 천식모델 생쥐의 면역세포 및 사이토카인에 미치는 영향 (Effects of Kamijihwang-tang on Immune Cells and Cytokines in OVA-induced Asthmatic Mice)

  • Objectives: The purpose of this study wasto examine the effects of Kamijihwang-tang (KJHT) extracts on immune cells and cytokines in ovalbumin (OVA)-induced asthmatic mice. Methods: C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (four times a week) for asthma induction. Two experimental groups were treated with different concentrations of KJHT (400 mg/kg and 200 mg/kg) extracts and cyclosporine A (10 mg/kg) for the later 8 weeks. At the end of the experiment, the mice lung, PLN and spleen were removed and immune cells were analyzed by flow cytometer, IL-5, IL-13, eotaxin-2, $TNF-{\alpha}$ were analyzed by real-time PCR, serum histamine was analyzed by ELISA kit. Results: $CD3^+$, $CD3e^-/CCR3^+$, $CD3e^+/CD69^+$, $CD4^+/CD25^+$, $B220^+/IgE^+$, and $CD3e^+/DX5^+$ cells in lung, PLN, and spleen of the KJHT group (400 mg/kg) decreased compared with that of the control group. $CD3e^+/CD69^+$, $CD4^+/CD25^+$, and $CD3e^+/DX5^+$ cells in lung, PLN, and spleen of the KJHT group (200 mg/kg), CD3+, $CD3e^-/CCR3^+$ cells in lung and PLN of the KJHT group (200 mg/kg) and $B220^+/IgE^+$ cells in lung and spleen of the KJHT group (200 mg/kg) decreased compared with that of the control group. mRNA expression of IL-5, IL-13, eotaxin-2, and $TNF-{\alpha}$ in lung tissue of the KJHT groups (400 mg/kg and 200 mg/kg) decreased compared with that of the control group. Histamine in serum of the KJHT group (400 mg/kg) decreased compared with that of the control group. Conclusions: These results suggest that KJHT can be utilized effectively in treating asthma because it significantly reduces inflammatory cells and cytokines.

Effects of 2-deoxy-D-glucose and quercetin on the expression of osteonectin and osteopontin during the differentiation of irradiated MC3T3-El osteoblastic cells

  • Purpose : To characterize the effects of 2-deoxy-D-glucose (2-DG) and quercetin (QCT) on gene expression of osteonectin (ON) and osteopontin (OP) in irradiated MC3T3-El cells. Materials and Methods : When MC3T3-El osteoblastic cells had reached 70-80% confluence, cultures were transferred to a differentiating medium supplemented with 5 mM 2-DG or 10 ${\mu}M$ QCT and then irradiated with 2, 4, 6, and 8 Gy. At various times after irradiation, the cells were analyzed for the expression of bone mineralization genes such as ON and OP. Results : The mRNA expression of both ON and OP was increased according to the culture time in the differentiation medium, and the increase of the genes peaked at 14 days after the differentiation induction. In the case of OP, the increase of mRNA expression was maintained to 28 days after the differentiation, while the mRNA level of ON was reduced to the basal level at the same time. Irradiation adding 2-DG showed a significant peak value in the expression pattern of ON at 4 Gy 7 days after irradiation. Irradiation adding QCT increased the mRNA expression of ON and OP in a dose-dependant manner, but irradiation adding 2-DG did not show any differences between the control and experiments 14 days after irradiation. Irradiation adding QCT increased significantly the expression patterns of ON 21 days after irradiation. Conclusion : The results showed that QCT acted as a radiosensitizer in the gene expression of ON and OP during differentiation of the late stage of irradiated MC3T3-E1 osteoblastic cells in vitro.   • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. Magnesium vs. machined surfaced titanium - osteoblast and osteoclast differentiation

  • PURPOSE. This study focused on in vitro cell differentiation and surface characteristics in a magnesium coated titanium surface implanted on using a plasma ion source. MATERIALS AND METHODS. 40 commercially made pure titanium discs were prepared to produce Ti oxide machined surface (M) and Mg-incorporated Ti oxide machined surface (MM). Surface properties were analyzed using a scanning electron microscopy (SEM). On each surface, alkaline phosphatase (ALP) activity, alizarin red S staining for mineralization of MC3T3-E1 cells, and quantitative analysis of osteoblastic gene expression, were evaluated. Actin ring formation assay and gene expression analysis of TRAP and GAPDH performing RT-PCR were performed to characterize osteoclast differentiation on mouse bone marrow-derived macrophages (BMMs). RESULTS. MM showed similar surface morphology and surface roughness with M, but was slightly smoother after ion implantation at the micron scale. M was more hydrophobic than MM. No significant difference between surfaces on ALP activity at 7 and 14 days were observed. Real-time PCR analyses showed similar levels of mRNA expression of the osteoblast phenotype genes; osteopontin (OPN), osteocalcin (OCN), bone sialoprotein (BSP), and collagen 1 (Col 1) in cell grown on MM at 7, 14 and 21 days. Alizarin red S staining at 21 days showed no significant difference. BMMs differentiation increased in M and MM. Actin ring formation assay and gene expression analysis of TRAP showed osteoclast differentiation to be more active on MM. CONCLUSION. Both M and MM have a good effect on osteoblastic cell differentiation, but MM may speed the bone remodeling process by activating on osteoclast differentiation.

Antioxidative Action of Corni Fructus Aqueous Extract on Kidneys of Diabetic Mice

  • This study investigated the antioxidative action of Corni Fructus aqueous extract on kidneys of diabetic mice. The electron donating abilities of Corni Fructus aqueous extract and its antioxidant activities (XO, SOD, CAT, GST, eNOS) in kidneys of C57BL/6 or db/db mice were evaluated. For in vivo study, seven week-old male mice were divided into normal control group (NC, C57BL/6 mice), diabetic control group (DC, db/db mice) and Corni Fructus (500 mg/kg/day for 8 weeks) treated diabetic group (DCF, db/db mice). The electron donating abilities of Corni Fructus aqueous extract exhibited 7%, 24.4%, and 42.7% at concentrations of 100, 500, and $1000\;{\mu}g/ml$, respectively. The activity of XO in the DCF group was significantly lower than the DC group by 35% (p < 0.05). The SOD activity was significantly higher in the DCF group than the DC group by 26% (p < 0.05). The activities of CAT and GST were lowered in the DCF group than the DC group by 26% (p < 0.05) and 7.6%, respectively. The mRNA expression of eNOS in kidneys was lower in the DCF group than the DC group by 24%. These results indicate that Corni Fructus reduced oxidation stress as evidenced by the restoration of the enzymatic antioxidative defense system in renal tissues of db/db mice. It is suggested that these antioxidative actions of Corni Fructus on renal tissues in db/db mice could contribute to its renoprotective effects on diabetic nephropathy.

두개골 및 두개봉합부 초기발육과정에서의 전사조절인자인 Msx2와 Dlx5의 역할 (THE ROLE OF TRANSCRIPTION FACTOR MSX2 AND DLX5 IN CALVARIAL BONE AND SUTURE DEVELOPMENT)

  • 두개봉합부의 조기융합으로 일컬어지는 craniosynostosis는 두개봉합부에서의 골아세포의 조기분화 및 석회화의 결과로 나타나는 선천성 발육이상이다. 최근 유전학적 연구에 의하면 homeobox gene인 Msx2의 변이에 의해 Boston-type craniosynostosis가 야기되며, 또한 Dlx5 homozygote mutant mouse의 표현형에서 두개골의 골화지연을 포함한 다양한 두개안면부위의 이상을 발견하였다는 보고가 있었다. 게다가 Msx2와 Dlx5 homeodomain protein의 상호작용에 의해 성숙골아세포의 표지자인 osteocalcin의 전사를 조절할 수 있다는 사실이 알려져 있다. 이러한 일련의 결과들은 Msx2 Dlx5 및 osteocalcin 유전자들이 두개골의 골화과정과 두개봉합부의 형태발생에 중요한 역할을 담당하고 있음을 제시해주고 있다. 두개골의 성장과 두개봉합부의 형태발생시 이러한 유전자들의 기능을 알아보기위해 mouse의 태생기 (E15-E18) 동안 osteocalcin, Msx2, 및 Dlx5 유전자들의 발현양상을 조사하였다. Osteocalcin은 E15부터 두정골의 골막에서 관찰되었으며, 발생시기가 후기일수록 강한 발현양상을 나타내었다. Msx2는 시상봉합부의 미분화간엽조직과 osteogenic front에서 강하게 발현되었으며 경막과 hair follicle에서도 관찰되었다. Dlx5는 osteogenic front를 포함한 두정골의 골막에서 강하게 발현되었으나 시상봉합부의 미분화간엽조직 에서는 발현되지 않아, Msx2와는 발현양상의 차이를 나타내었다. 두개골과 두개봉합부의 발육과정에서의 Msx2와 Dlx5의 기능을 좀더 심도깊게 분석하기위해, 여러 가지 signaling molecule들의 protein을 사용하여 in vitro 실험을 시행하였다. BMP-2, -4 protein의 overexpression은 bead 주위로 Msx2 유전자의 발현을 유도하였으나, 다른 $TGF{\beta}$ superfamily인 $TGF{\beta}1$, GDF-6, -7 bead들 주위로는 Msx2를 관찰할 수 없었다. 또한 FGF, Shh protein 역시 bead주위로 Msx2의 발현을 유도하지않았다. 흥미롭게도 BMP-2, -4 protein의 overexpression은 bead 주위로 Dlx5 유전자의 발현을 유도하였으나, 다른 $TGF{\beta}$ superfamily, FGF, Shh bead주위로는 Dlx5를 관찰할 수 없어, Msx2와 동일한 결과를 나타내었다 이 결과들을 종합해볼 때, Msx2와 Dlx5 유전자는 두개골과 두개봉합부의 성장발육과정에 중요한 역할을 담당하고 있으며, BMP signaling은 이 두 전사조절인자들을 조절하므로써 두개골의 골화과정과 두개봉합부의 형태발생 및 유지에 관여하고 있음을 제시해주고 있다. 특히 BMP signaling에 specific downstream gene인 Msx2 및 Dlx5의 발현양상의 차이는 골아세포의 분화시 이들 유전자가 각각의 독특한 기능을 가지고 있음을 시사해주고 있다.

Cinnamic acid 유도체들의 SOS 반응을 지표로한 항돌연변이 효과에 관한 연구

  • 목적: Cinnamic acid의 유도체들중에서 항돌연변이 효과가 보고된 바 있어, 본 연구에서는 E. coil PQ37균주를 사용한 SOS chromotest를 이용하여 약 170개 cinnamic acid의 유도체들의 항돌연변이 효과를 확인하고자 하였고, 1차로 확인된 수종의 유도체들에 대한 결과를 보고하고자 한다. 방법: SOS chromotest는 sul A:: lacz fusion strain 인 E. coli PQ37 에서의 chemical로 인한 DNA damage에 대한 SOS response를 $\beta$-galactosidase enzyme activity level로서 측정하는 실험이며, m-RNA 또는 protein synthesis에 대한 test chemicals의 cytotoxic저해효과를 알아보기 위하여 alkaline phosphatase를 병행측정하여 보완하였다. 결과 및 고찰: (-)S-9 경우는 4-nitroquinoline 1-oxide(4-NQO)를 유도물질로 하였으며 대략 Induction factor가 9.7 정도였고, (+)S-9의 경우는 aflatoxinB$_1$을 유도물질로 하였고 이때의 Induction factor는 13.8 정도였다. 이결과 1차로 test된 cinnamic acid유도체 6개중에서 RK001, RK002, RK003, RK004, RK005는 거의 항돌연변이 효과를 나타내지 않았으나, RK006은 항돌연변이 효과를 보여주어 이들의 Structure-Activity Relationship (SAR) 및 Dose-response와 RK006의 항돌연변이 효과의 mechanism에 관해 M13 mp2 viral DNA의 유전자 염기서열을 이용하여 항돌연변이 기전연구를 수행중이다.
