Distribution of Multidrug Efflux Pump Genes in Enterococci spp. Isolated from Bovine Milk Samples and Their Antibiotic Resistance Patterns (원유 시료에서 분리한 장알균속 세균의 다중약물 유출 펌프(Multidrug Efflux Pump) 유전자의 분포도와 항생제 내성 패턴)

  • The major aim of this study was to investigate the distribution of genes that encode multidrug efflux pumps in Enterococci spp. isolates from bovine milk samples and antibiotic resistance patterns of these strains. Of the 245 isolates, 44.1% showed ampicillin resistance, 79.2% showed erythromycin resistance, 76.3% showed tetracycline resistance and 36.3% showed chloramphenicol resistance. In case of vancomycin and ciprofloxacin, all of the isolates were susceptible to these antibiotics. Of the 245 enterococcal isolates, 82.1% have MFS type eme(A) gene, 72.7% have ABC type efr(A) gene, 77.1% have ABC type efr(B) gene, and 71.8% have ABC type lsa gene. In case of Enterococcus faecalis, the original strain for these genes, 92.5% have eme(A), 87.4% have efr(A), 88.4% have efr(B), and 88.4% have lsa. Interestingly, in case of different species of Enterococci, eme(A) was also detected in four strains of E. faecium, seven strains of E. avium, four strains of E. durans and two strains of E. raffinosus. efr(A) was also detected in two strains of E. faecium and two strains of E. durans and efr(B) was also detected in four strains of E. faecium, five strains of E. avium and four strains of E. durans. This means the possibility of co-transfer of resistance genes between Enterococci species in natural environment. These results are the first report describing the presence of same multidrug efflux pumps in different species of Enterococci in Korea.

  • Bioassay-guided isolation of the dried roots of Aralia continentalis led to the isolation of (-)-pimara-8(14), 15dien-19-oic acid (continentalic acid) and $(24E)-Stigmasta-5,22-dien-3{\beta}-ol$ (stagmasterol). Their structures were elucidated using $^1H-NMR$, $^{13}C-NMR$, UV and mass spectra analyses. The gram-positive bacterial, including methicilline-resistant (MRSA), were more sensitive to the continentalic acid and stagmasterol than gram-negative bacterial.

  • 최근 몇년간 국립수의과학검역원에서 조사한 바에 따르면, 국내 젖소에서 유방염을 일으키는 주된 원인균은 황색포도상구균을 제외하고는 거의 대부분 젖소의 환경으로부터 유래하는 환경성 세균에 의한 것으로 확인된 바 있다. 국내 젖소의 유방염 원인균으로 작용하고 있는 이러한 환경성 세균 중 coagulase-음성 포도상구균 (Coagulase-negative Staphylococci, CNS)과 E. coli 등의 그람 음성균이 가장 많은 비율을 차지하였지만 그 외에 Streptococcus uberis를 비롯한 환경성 연쇄상 구균과 장구균도 상당한 비율을 차지하는 것으로 조사되었으며 이에 대해서는 지난 9월호에서도 상세히 기술한 바 있다. 환경성 연쇄상구균과 장구균은 젖소에서 유선감염 및 임상형 유방염의 중요한 원인이며 환경성 연쇄상구균 중 가장 흔히 발견되는 유방염 원인체는 Streptococcus uberis 로서, 이는 국내에서뿐만 아니라 미국, 유럽, 뉴질랜드, 호주 등에서 수행된 많은 연구결과에서도 마찬가지로 나타났다. 장구균 중 가장 흔히 분리되는 균종은 E. faecium과 E. faecalis로서 검역원에서 지난 몇 년간 조사된 결과에서도 이 두 가지 균 종이 유방염우유에서 분리된 장구균의 90% 이상을 차지하였다. 이러한 환경성 연쇄상구균과 장구균은 사람과 동물의 장관, 분변, 감염된 유방 및 전체적인 목장 환경에서 분리되고 있다. 장구균은 일반적으로 환경성 연쇄상구균/장구균 유방염 감염증 중의 일부일뿐이고 장구균에 의한 유방염이 목장의 주된 문제가 되는 일은 비교적 드물다. 현재, 장구균에 의해 발생하는 유방염은 S. uberis를 통제하기 위해 사용되는 방법과 동일한 방법으로 통제할 수 있다고 여겨진다. 원인체에 상관없이 유방염을 통제하기 위한 기본적인 원칙은 유방염을 일으킬 수 있는 병원체에 유두끝이 노출되는 기회를 줄이거나 감염에 대한 젖소의 저항성을 증가시키는 두 가지 방법이다. 역으로 얘기하면, 병원체에 대한 유두끝 노출을 증가시키거나 감염에 대한 젖소의 저항성을 감소시키는 요인들은 우군 내에서 더 많은 유방염을 발생시킬 가능성이 높기 때문에 위험요인으로 간주된다. 본 고에서는 환경성 연쇄상구균에 의한 유방염과 관련이 있는 것으로 알려진 이러한 여러 가지 위험요인들에 대해 알아보고 그러한 위험요인을 줄일 수 있는 방법에 대해서도 고찰하고자 한다.

Detection of Enterococci and their Vancomycin Resistance in Drinking Spring-Water (먹는물 약수터 장구균의 검출 특성과 반코마이신 내성 현황)

  • This study was performed to detect enterococci strain as an indicator of faecal contamination, to identify of 16S rDNA sequence and vancomycin resistance by MIC (Minimum Inhibitory Concentration) test from drinking spring-water samples in Seoul. The detection frequency of enterococci was 42 (19.8%) among 212 samples, and its concentration was ranged from 0 to 110 CFU/100 mL. These results were confirmed the possibility as an indicator microorganisms that similar to the frequency of E. coli detection (t test p-value 0.268, significant level 0.05). Isolated 56 enterococci samples were identified by 16S rDNA sequence data and their NCBI BLAST searching. They were identified to Enterococcus faecalis of 24 samples, E. faecium (10), E. casseliflavus (10), E. gallinarum (3), E. hirae (2), E. durans (2), E. sanguinicola (1). E. faecalis was dominant species that clinical case report of a domestic was similar. Vancomycin resistant enterococci (VRE) of 53 samples showed that vanB and vanC1/C2 type with 2 and 12 case, respectively. These results indicated that the drinking spring-water quarantined to fecal pollution for block out outbreak of gastrointestinal symptom with using such as disinfection process.

Analysis of Genotype and Phenotype of Erythromycin Resistance in Enterococci spp. Isolated from Raw Milk Samples (원유시료에서 분리한 장구균의 에리스로마이신 내성 유전자형 및 표현형 분석)

  • The aim of this study was to investigate the erythromycin resistance patterns of Enterococci sp. present in cow milk. A total 110 erythromycin resistant Enterococci were isolated from milk samples; E. faecalis (n=101), E. avium (n=7), and E. durans (n=2). The minimum inhibitory concentration of erythromycin against 110 Enterococci were determined. The 66.3% of Entercocci (n=73) showed high level resistance (${\geq}64$ mg/ml). Among 110 isolates, 86.3% (n=95) showed $cMLS_B$ phenotype and 13.6% (n=15) showed $iMLS_B$ The aim of this study was to investigate the erythromycin resistance patterns of Enterococci sp. present in cow milk. Physiological Characteristics and Anti-Obesity Effect of Enterococcus faecalis MD366 isolated from Raw Milk (원유에서 분리한 Enterococcus faecalis MD366의 생리적 특성 및 비만 억제 효과)

  • The aim of this study was to investigate the physiological characteristics and anti-obesity effects of E. faecalis MD366 isolated from raw milk. E. faecalis MD366 inhibited lipase activity ($65.0{\pm}0.9%$) and differentiation of 3T3-L1 adipocytes ($27.4{\pm}1.4%$) at a concentration of $100{\mu}g/mL$ ($10^8CFU/g$ E. faecalis MD366). The optimum growth temperature of E. faecalis MD366 was $37^{\circ}C$. Among 16 tested antibiotics, E. faecalis MD366 demonstrated the highest sensitivity to novobiocin and the highest resistance to neomycin, kanamycin, and vancomycin. The strain also showed high acid phosphatase activity. Moreover, E. faecalis was relatively tolerant to bile juice and acid, and displayed high resistance to Escherichia coli, Salmonella Typhimurium, and Staphylococcus aureus (80.4%, 60.2%, and 65.4%, respectively). These results demonstrate that E. faecalis MD366 can be potentially used as a probiotic with anti-obesity effects.

  • The gene for a L-aspartate $\beta-decarboxylase$ (ADC) from Enterococcus faecalis was cloned and sequenced. The gene comprised an open reading frame of 1,611 base pairs, which encodes a protein of 58,960 Da consisting of 536 amino acid residues. The gene was subcloned into an expression plasmid for overexpression of the ADC. The recombinant ADC was produced using E. coli as the host and purified to homogeneity. Our result showed that the ADC may be obtained from bacteria known nucleotide sequence. Thus, we suggest that high value L-alanine might be produced by low value aspartate.

Prevalence and Antimicrobial Resistance of Enterococus faecalis and Enterococcus faecium Isolated from Beef, Pork, Chicken and Sashimi (시판 축산물 및 수산물에서 Enterococcus faecalis와 Enterococcus faecium 분포 및 항생제 감수성에 관한 연구)

  • In this study, a total of 256 samples of retail raw meats (beef, pork and chicken) and sashimi were investigated for the presence of Enterococcus faecalis and Enterococcus faecium. We isolated a total of 117 E. faecalis and E. faecium from the samples, with contamination rates ranging from 18.8% for sashimi samples to 68.8% of chicken samples. E. faecalis was the predominant species recovered from all of the retail raw meats beef (42.2%), pork (42.2%), chicken (65.6%) and sashimi (12.5%). Among 117 isolates, 61 isolates (52.1%) were resistant to tetracycline, 32 isolates (27.4%) were resistant to erythromycin, 23 isolates (19.7%) were resistant to chloramphenicol, 16 isolates (13.7%) were resistant to ripampin, 10 isolates (8.5%) were resistant to gentamycin, 9 isolates (7.7%) were resistant to ciprofloxacin and 1 isolate (0.9%) was resistant to ampicillin and penicillin G. No resistance to amoxicillin + clavulanic acid and vancomycin was observed. Although no strain was resistant to vancomycin, the vanB gene was observed in 9 of 117 of Enterococcus (7.7%) demonstrating potential risk of vancomycin-resistant Enterococcus (VRE). Our results indicate that E. faecalis and E. faecium were highly prevalent in retail raw meats, but most strains were sensitive to tested antibiotics.

Analysis of virulence gene profiles of Salmonella spp. and Enterococcus faecalis isolated from the freshly slaughtered poultry meats produced in Gyeong-Nam province (경남지역 가금류 도축장 신선육에서 분리한 Salmonella spp.와 Enterococcus faecalis의 독성인자 보유 패턴 분석)

  • In order for monitoring of pathogenic bacterial contamination in the freshly slaughtered poultry meats produced in Gyeong-Nam province, we first isolated 4 strains of Salmonella spp. and 32 strains of Enterococcus faecalis from the total 164 samples, then we analyzed potential virulence gene profiles of the bacterial isolates by PCR using species-specific primer. The potential virulence genes we selected in this study were stn, invA, fimA, spvR, and spvC for the isolates of Salmonella spp. and those of esp, cylM, cylA, cylB, gelE, fsrA, fsrB, and fsrC were for the isolates of E. faecalis. The PCR results showed that all 5 virulence genes were detected simultaneously in the all isolates of Salmonella spp. However, there was a diverse occurrence pattern of the virulence genes in the case of E. faecalis. The gene for enterococcal surface protein (esp) was not detected among the isolates (0/32), and the haemolysin gene prevalence rate of cylA, cylB, and cylM were 3.1% (1/32), 9.3% (3/32), and 9.3% (3/32), respectively. Moreover, the genes of gelE, fsrA, fsrB, and fsrC that associated with gelatinase activity were detected in the rate of 53.1% (17/32), 53.1% (17/32), 53.1% (17/32), and 53.1% (17/32), respectively. In conclusion, in the isolates of Salmonella spp., all possessed 5 virulence genes tested, suggesting that they are all related with each other clonally. However, in the case of E. faecalis isolates, the occurrence of the haemolysin genes (cylM, cylA, cylB) and the gelatinase genes (gelE, fsrABC) was highly variable among the isolates.

Antimicrobial Susceptibility of Several Bacterial Species Isolated from Human Blood Culture (혈액배양에서 분리된 일부세균의 항균제 감수성)

  • This study was undertaken to determine the antimicrobial susceptibilities of 716 bacterial strains among 1,104 microbes which were isolated in blood culture from 13,595 in-patients and out-patients in K hospital during the pe-riod of 1993 to 1996. The results obtained were as follows : The isolation rate of microbes from the blood culture for 3 years was 8.1% (1,104/13,595). Among 716 bacte-rial strains, Escherichia coli and Staphylococcus epidermidis were 29.8% (213) and 19.5% (140) respectively. The bacterial isolation frequency according to ages was high at fifties, sixties and two or less years old groups, especially the isolation rate of S. epidermidis was 47% at two or less years old group and that of Salmonella typhi was 36% at thirties years old group. The seasonal isolation rate of the bacterial species except E. coli was high during May to September. Gram positive cocci were resistant to penicillin-G except Enterococcus faecalis, but susceptible to vanco-mycin and teicoplanin. Gram negative bacilli were susceptible to amikacin, ciprofloxacin, and imipenem, but resistant to ampicillin except S. typhi. These results indicate that bacterial strains isolated from bacteremia patients were resis-tant to penicillins, but susceptible to vancomycin, teicoplanin, amikacin, ciprofloxacin and imipenem.
