Effect of Probability Distribution of Coefficient of Consolidation on Probabilistic Analysis of Consolidation in Heterogeneous Soil (비균질 지반에서 압밀계수의 확률분포가 압밀의 확률론적 해석에 미치는 영향)

  • In this study, a simple probabilistic approach using equivalent coefficient of consolidation ($c_e$) was proposed to consider the spatial variability of coefficient of vertical consolidation ($c_v$), and the effect of the probability distribution of coefficient of consolidation on degree of consolidation in heterogeneous soil was investigated. The statistical characteristics of consolidation coefficient were estimated from 1,226 field data, and four probability distributions (Normal, Log-normal, Gamma, and Weibull) were applied to consider the effect of probability distribution. The random fields of coefficient of consolidation were generated based on Karhunen-Loeve expansion. Then, the equivalent coefficient of consolidation was calculated from the random field and used as the input value of consolidation analysis. As a result, the probabilistic analysis can be performed effectively by separating random field and numerical analysis, and probabilistic analysis was performed using a Latin hypercube Monte Carlo simulation. The results showed that the statistical properties of $c_e$ were changed by the probability distribution and spatial variability of $c_v$, and the probability distribution of $c_v$ has considerable effects on the probabilistic results. There was a large difference of failure probability depend on the probability distribution when the autocorrelation distance was small (i.e., highly heterogeneous soil). Therefore, the selection of a suitable probability distribution of $c_v$ is very important for reliable probabilistic analysis of consolidation.

Relationship between inflammation biomarkers, antioxidant vitamins, and bone mineral density in patients with metabolic syndrome

  • Few studies have shown the correlation between metabolic syndrome and bone mineral density (BMD). The main pathogenic mechanisms of metabolic syndrome rely on chronic low-level inflammatory status and oxidative stress. There are few studies that examine the gender-specific effects of inflammation and antioxidants on BMD. In this study, we evaluated the relative contribution of these factors in patients with metabolic syndrome. We conducted a cross-sectional study of 67 men and 46 postmenopausal women with metabolic syndrome; metabolic syndrome was defined as having three or more metabolic syndrome risk factors. BMD, body fat mass, and lean body mass were evaluated. We also examined the levels of high sensitive C-reactive protein (hs-CRP), interleukin-6 (IL-6), adiponectin, vitamin E, and C in serum. Log-transformed hs-CRP levels were significantly higher in lumbar spine osteoporotic subjects than in normal subjects for women but not for men. There was no significant difference between the normal group and the osteoporotic group in other inflammatory markers. Stepwise regression analyses for BMD of the lumbar spine showed that lean body mass and vitamin E were significant determinants in men. Lean body mass and log-transformed hs-CRP were significant determinants in women Analysis for BMD of the femoral neck showed that lean body mass was a significant determinant for both men and women. There was no significant factor among the inflammatory markers or antioxidant vitamins affecting the femoral neck BMD for either gender. In conclusion, while hs-CRP is an independent predictor of the BMD of the lumbar spine in women, vitamin E showed profound effects on BMD in men but not women with metabolic syndrome.

Antimicrobial resistance and transfer of R plasmid of pathogenic Eseherichia coli isolated from poultry in Korea (가금 유래 병원성 대장균의 항균제 내성 및 R plasmid 전달 양상)

  • Antimicrobial drugs are widely used in poultry industry as growth promoters or to control infectious diseases. However, this practice is reported to have caused high resistance to antimicrobial drugs in normal chicken flora and pathogens. Antimicrobial resistance to Escherichia coli (E. coli) from chicken has been mainly reported in normal flora, but rare in pathogenic organism in Korea, recently. Therefore, this study was conducted to investigate prevalence of antimicrobials resistance, transfer of R plasmid, and association between antimicrobial drug resistance and O serotype of 203 pathogenic E. coli from poultry in Korea during the period from April 2003 to December 2005. These isolates showed a high resistance to tetracycline (Tc, 93.6%), nalidixic acid (Na, 92.6%), streptomycin (Sm, 81.8%), ampicillin (Ap, 77.3%), ciprofloxacin (Ci, 70.9%), sulfisoxazole (Su, 66.5%), and trimethoprim (Tp, 58.1%). Two hundred-one (99.0%) of the isolates were resistant to one or more drugs. They showed 57 different resistant patterns, and the most prevalent resistant pattern among them was Tc, Sin, Su, Ap, Tp, Ci, Na. Sixty-eight (33.8%) of the isolates transferred all or a part of their antimicrobial resistant pattern to the recipient strain by R plasmid. The most common antimicrobial resistant pattern was Tc, Sm, Su, Ap, Tp, Ci, Na in serotype O78, O88 and O15, respectively. These results exhibit high individual and multiple resistance to antimicrobials of pathogenic E. coli from poultry in Korea. They also suggest the needs for surveillance to monitor antimicrobial resistance in pathogenic bacteria that can be potentially transmitted to humans from food animals and to regulate the abuse of antimicrobials on food-producing animals in Korea.

Free vibration analysis of a non-uniform beam with multiple point masses

  • The natural frequencies and the corresponding mode shapes of a non-uniform beam carrying multiple point masses are determined by using the analytical-and-numerical-combined method. To confirm the reliability of the last approach, all the presented results are compared with those obtained from the existing literature or the conventional finite element method and close agreement is achieved. For a "uniform" beam, the natural frequencies and mode shapes of the "clamped-hinged" beam are exactly equal to those of the "hinged-clamped" beam so that one eigenvalue equation is available for two boundary conditions, but this is not true for a "non-uniform" beam. To improve this drawback, a simple transformation function ${\varphi}({\xi})=(e+{\xi}{\alpha})^2$ is presented. Where ${\xi}=x/L$ is the ratio of the axial coordinate x to the beam length L, ${\alpha}$ is a taper constant for the non-uniform beam, e=1.0 for "positive" taper and e=1.0+$|{\alpha}|$ for "negative" taper (where $|{\alpha}|$ is the absolute value of ${\alpha}$). Based on the last function, the eigenvalue equation for a non-uniform beam with "positive" taper (with increasingly varying stiffness) is also available for that with "negative" taper (with decreasingly varying stiffness) so that half of the effort may be saved. For the purpose of comparison, the eigenvalue equations for a positively-tapered beam with five types of boundary conditions are derived. Besides, a general expression for the "normal" mode shapes of the non-uniform beam is also presented.

C-terminal Truncation Mutant of the Human ${\beta}_2$-adrenergic Receptor Expressed in E. coli as a Fusion Protein Retains Ligand Binding Affinity

  • To investigate whether human $\beta$$_2$-adrenergic receptor devoid of the C-terminal two transmembrane helices retain its ligand binding activity and specificity, 5'780-bp DNA fragment of the receptor gene which encodes amino acid 1-260 of human $\beta$$_2$-adrenergic receptor was subcloned into the bacterial fusion protein expression vector and expressed as a form of glutathione-S-transferase (GST) fusion protein in E. coli DH5$\alpha$. The receptor fusion protein was expressed as a membrane bound form which was verified by SDS-PAGE and Western blot. The fusion protein expressed in this study specifically bound $\beta$-adrenergic receptor ligand [$^3$H] Dihydroalprenolol. In saturation ligand binding assay, the $K_{d}$ value was 7.6 nM which was similar to that of intact $\beta$$_2$-adrenergic receptor in normal animal tissue ( $K_{d}$=1~2 nM) and the $B_{max}$ value was 266 fmol/mg membrane protein. In competition binding assay, the order of binding affinity of various adrenergic receptor agonists to the fusion protein was isoproterenol》epinephrine norepinephrine, which was similar to that of intact receptor in normal animal tissue. These results suggest that N-terminal five transmembrane helices of the $\beta$$_2$-adrenergic receptor be sufficient to determine the ligand binding activity and specificity, irrespective of the presence or absence of the C-terminal two transmembrane helices.s.s.s.

Effects of Sagunjatang and Samultang on the Immunosuppression induced by Methotrexate in Rats (사군자탕(四君子湯) 및 사물탕(四物湯)이 methotrexate로 유도(誘導)된 흰쥐의 면역기능저하(免疫機能低下)에 미치는 영향(影響))

  • The effects of Sagunjatang and Samultang on the immunosuppression induced by methotrexate(MTX) in rats were investigated in this study. The multiple parameters of immunity assessed in each rats included leukocyte count, lymphocyte rate, the number of lymphocyte in tibial bone marrow, contact hypersensitivity to DNFB, morphological change of thymocyte and IgG antibody on SDS-PAGE. Sprague-Dawley male rats were used and divided into five groups at random. Group A was normal control. Group B, the MTX treatment control, was injected i.v. with 2mg/kg of on days 9, 11 after sensitization with SRBC on 5th day. Group C, the experimental control, was treated Sagunjatang for 18days and MTX. Group D was treated Samultang for 18days and MTX. Group E was treated Sagunjatang-Samultang for 18days and MTX. The dosage of Sagunjatang and Samultang was $1m{\ell}/day$ respectively. In the case of Group E, rats Were fed Sagunjatang $1m{\ell}$ in the morning and Samultang $1m{\ell}$ in the afternoon. The results are summarized as follows: 1. Leukocyte count in rats induced by intravenous sensitization with SRBC was decreased significantly in Group E. 2. Leukocyte counts of 2weeks later after being treated MTX were increased significantly in Groups C and D. 3. Lymphocyte rate in rats induced by intravenous sensitization with SRBC wasn't changed significantly in all the experimental groups. 4. Lymphocyte rate of 2weeks. later after being treated MTX was increased significantly in Group D. 5. The number of lymphocyte in tibial bone marrow was incereased significantly in Group C. 6. Contact hypersensitivity wasn't changed significantly in all the experimental groups. 7. Morphological finding of thymocyte in group C was similar to normal group as compared with control group. 8. Purified IgG of all the experimental groups showed two bands of 50,000 and 25,000 on SDS-PAGE. But there was no difference among experimental groups.

THE CONCENTRATIONS OF PROSTAGLANDIN E2, 6-KETO-PROSTAGLANDIN F1α, AND LEUKOTRIENE B4 IN PULPAL AND PERIAPICAL LESIONS (치수 및 치근단병소에서 Prostaglandin E2, 6-keto-Prostaglandin F1α, Leukotriene B4의 분포에 관한 연구)

  • Prostaglandins (PGs) and Leukotrienes (LTs) have been implicated in the genesis of pulpal and periapical inflammation. In this study, the relationships among $PGE_2$, 6-keto-PG $F_1{\alpha}$ (a stable metabolite of $PGI_2$) and $LTB_4$ concentrations in inflamed pulp and periapical lesions were discussed. Pulp tissue were obtained in routine endodontic treatment and periapical lesions in periapical surgery after clinical diagnoses were made. These specimens were divided into four groups as normal pulp group (Control group), acute pulpitis group, chronic pulpitis group, and periapical lesion group. Pulp tissue and periapical lesions were stored in liquid nitrogen. The concentration of $PGE_2$, $PGI_2$ and $LTB_4$ were measured with ELISA. The data were analyzed by one-way ANOVA. Significantly higher levels of $PGE_2$, 6-keto-PG $F_1{\alpha}$ a and $LTB_4$ were found in acute pulpitis group than chronic pulpitis group and periapical lesion group(p<0.05). Periapical lesion group showed significantly higher mean concentrations of $PGE_2$ and $LTB_4$ than chronic pulpitis group. In control and chronic pulpitis group, significant higher levels of $PGI_2$ than $PGE_2$ and $LTB_4$ were found. These results suggested that the high levels of $PGE_2$ and $LTB_4$ in periapical lesions may be due to rich endothelium., fibroblast and lymphocyte known as the main producers of $PGE_2$ and $LTB_4$. $PGI_2$ may be thought to one of the most abundant PGs in normal pulp tissue.

Analysis of organizational variables related to nursing between the doctors' strike and normal periods (의료파업시기와 정상시기간의 간호조직 내 제요인 분석)

  • Purpose : To analyze organization variables related to nursing between the doctors' strike and normal periods. Methods : The sample consisted of 1,220(600 for doctors' strike and 620 for normal periods) nurses working in 5 hospitals. Six instruments were used for data collection including Job Activity Scale(JAS; Laschinger, 1994), Organization Relationship Scale(ORS; Laschinger, 1994), Interpersonal Support Scale(developed by Firth et al., 1986 and revised by Ahn(2000), Self-efficacy Scale(Riggs & Knight, 1994),Job Satisfaction Scale(Slavitts et al., 1978), and Organizational Commitment Questionnaire(OCQ; Mowday et al., 1979). Results : There were significant differences among variables(i.e., formal power, interpersonal support, self-efficacy and organizational commitment)between doctors' strike and normal periods. The formal power was high during the normal period, however interpersonal support, self-efficacy and organizational commitment were high during the doctors' strike. Conclusion : Interpersonal support, self-efficacy and job satisfaction were change by working conditions, but organizational commitment was not changed by working conditions.

A study on the nonlinearity in bio-logical systems using approximate entropy and correlation dimension (근사엔트로피와 상관차원을 이용한 비선형 신호의 분석)

  • We studied how linear and nonlinear heart rate dynamics differ between normal fetuses and uncomplicated small-forgestational age (SGA) fetuses, aged 32-40 weeks' gestation. We analyzed each fetal heart rate time series for 20 min and quantified the complexity (nonlinear dynamics) of each fetal heart rate (FHR) time series by approximate entropy (ApEn) and correlation dimension (CD). The linear dynamics were analyzed by canonical correlation analysis (CCA). The ApEn and CD of the uncomplicated SGA fetuses were significantly lower than that of the normal fetuses in all three gestational periods (32-34, 35-37, 38-40 weeks). Canonical correlation ensemble in SGA fetuses is slightly higher than normal ones in all three gestational periods, especially at 35-37 weeks. Irregularity and complexity of the heart rate dynamics of SGA fetuses are lower than that of normal ones. Also, canonical ensemble in SGA fetuses is higher than in normal ones, suggesting that the FHR control system has multiple complex interactions. Along with the clear difference between the two groups' non-linear chaotic dynamics in FHR patterns, we clarified the hidden subtle differences in linearity (e.g. canonical ensemble). The decrease in non-linear dynamics may contribute to the increase in linear dynamics. The present statistical methodology can be readily and routinely utilized in Obstetrics and Gynecologic fields.

Association of Bone Mineral Density and Blood Pressure, Calcium Intake among Adult Women in Seoul.Kyunggi Area - Based on 2011 KNHANES - (서울.경기지역 성인여성의 골밀도와 혈압, 칼슘섭취의 상관성 연구 -2011년 국민건강영양조사를 중심으로-)

  • This study was carried out to investigate bone mineral density (BMD), blood pressure, calcium, sodium intake and related biochemical variables, and to analyze the relationship between BMD, and blood pressure, physical characteristics, nutrient intakes of Korean adult women. The study subjects were 30~80years old, 513 women living in Seoul and Kyunggi area who participated in 2011 KNHANES. The study subjects were divided into three BMD groups; normal, n = 259 (50.4%), osteopenia, n = 169 (32.9%), and osteoporosis, n = 85 (16.7%). Average height and weight and fat free mass decreased with a decrease in BMD. Average BMD of normal, osteopenia, osteoporosis were 0.87, 0.79, $0.70g/cm^2$, and T-score were 0.42, -0.66, -1.62, respectively. Higher systolic blood pressure, alkaline phosphatase, and triglyceride were significantly associated with a decrease in the BMD. The rates of hypertension were significantly increased from normal to osteoporosis. i.e. 18.5% in normal, 34.3% in osteopenia and 63.5% in osteoporosis. Average nutrientintakes such as protein, fat, calcium, phosphorus, iron, sodium, potassium, vitamin A, vitamin B, vitamin B2, and vitamin C were significantly lower in subjects with lower BMD.n. Average calcium intake of normal, osteopernia and osteoporosis were 528.50, 416.96, 389.56 mg, respectively. There were significant negative correlations between calcium, phosphorus and age, systolic blood pressure, triglyceride. Also, there were negative correlation systolic blood pressure (incomplete sentence). In this study, we found low BMD according to low calcium intake increase the risk of osteoporosis and hypertension rather than sodium intake, also calcium intake decreased with increase age. Therefore, in order to prevent osteoporosis and hypertension, subjects need to be educated regarding the importance of calcium nutrients in diet.
