국내 유통식품에서 분리된 Verotoxin 생성 Escherichia coli의 특성 (Characterization of Verotoxin-producing Escherichia coli Isolated from Domestic Foods)

  • 국내 유통식품에서 verotoxin(VT)을 생성하는 대장균의 오염도를 조사하고 이로 인한 식중독의 발생을 사전에 예방하기 위하여 1997년부터 1999년 사이 햄버거. 식육 및 채소류에서 VT생성 대장균의 분포조사를 실시하였으며, 이 결과 분리된 대장균에 대한 분자생물학적 특성을 조사하였다 식육, 햄버거 및 채소류 총 1,700건의 식품 중에서 VT를 생성하는 대장균이 검출된 것은 3건으로 매우 낮은 검출율을 보였는데, 분리주의 혈청형은 각각 026 : H4, 0157 : H7 및 055 : Hl2였다. 혈청형 026 : H4 분리주는 VT I과 VT II를 생성하였고,055 : Hl2주는 VT I을 각각 생성하였으며, 두 분리주에서 eae유전자와 60 MDa plasmid DNA는 검출되지 않았다. 혈청형 0157 : H7주는 VT II를 생성하였고 60 MDa plasmid DNA는 검출되었으나 eae유전자는 확인되지 않았다. 따라서 분리된 VT생성 대장균들은 eae 유전자가 검출되지 않아 질병유발 가능성은 낮은 것으로 추정되었다. 항생제 내성능에서 혈청형 0157 : H7 분리주는 ampicillin과 streptomycin에 내성을 나타낸 반면, 혈청형 026 : H4주와 혈청형 055 : Hl2주는 ampicillin, carbenicillin, cephalothin, trimethoprim/sulfamethoxazole과 tetracycline에 내성을 보여 분리주의 다재내성능을 확인하였다 세포배양액으로 실시한 vero cell과 HeLa cell에 대한 세포독성능은 3주의 분리주에서 확인되었다.

신선 과채류에 존재하는 박테리오신 유사 활성을 지니는 유산균의 분포와 다양성 (Distribution and Biodiversity of Lactic Acid Bacteria Having Bacteriocin-like Activity from Fresh Fruits and Vegetables)

  • 25 종류의 신선 과채류로부터 MRS 한천 고체배지에서 생육하는 1,250주의 유산균을 분리한 후 식중독 세균인 Escherichia coli 0157:H7, Listeria monocytogenes, Bacillus cereus와 유산균인 Lactobacillus plantarum, L. casei, Lactococcus lactis subsp. lactis에 대한 생육저해활성을 조사하였다. 조사한 1,250 균주 중에서 23 종류의 과채류에 존재하는 607 균주(총 조사균주의 49%)가 E. coli 0157:H7, L. monocytogenes 또는 B. cereus에 대한 항균활성을 나타내었다. 이 균주들의 L. plantarum, L. casei, Lactococcus lactis subsp. lactis에 대한 항균활성을 측정한 결과, 7 종류의 과채류에 존재하는 24 균주(총 균주의 3%)가 박테리오신 유사활성을 나타내었으며, 이 균주들의 생리적 형태학적 특징을 조사한 결과 전형적인 유산균의 특징을 지니고 있었다. 24 균주 중에서 3 균주는 nisin을 생산하는 것으로 판단되었으며, RAPD-PCR 분석을 통하여 동일한 과채류에 존재하는 균주들은 유사한 분류학적 특징을 지니고 있는 것으로 확인되었다.

  • Three and 2 strains of E coli O157 were isolated from fecal materials of cattle (390) and pigs (420) in Korea, respectively. One strain of O157:H7 isolated from cattle and 2 strains of O157:H7 isolated from pigs were identified as verotoxin-1 (VT-1) produing strains and 2 strains (O157:H7 and O157:H-) isolated from cattle were identified as verotoxin-2 (VT-2) producing strains by neutralization test on HeLa and Vero cells. Culture supernatants of the isolates were cytotoxic to HeLa and Vero cells. The levels of cytotoxin produced by isolates were $10^2{\sim}10^4$ cytotoxic dose($CD_{50}$)/ml. Also, VT-2-converting bacteriophage was isolated from KSC109 strain which had been isolated from cattle. Molecular weight of the phage DNA was determined as approximately 45 Kb in 0.8% agarose gel electrophoresis, and morphology of the phage stained with phosphotungstic acid was observed by transmissible electron microscopy.

Preparation of a Silk Fibroin Film Containing Catechin and Its Application

  • Silk fibroin (SF) film containing catechin was prepared and the antimicrobial activity as well as physical property of the film was examined. Tensile strength of the SF film decreased with increasing concentration of catechin, and water vapor permeability of the film decreased. The film's antimicrobial activity against Escherichia coli O157:H7 increased with increasing catechin concentration. Sausage samples were inoculated with E. coli 0157:H7 and Listeria monocytogenes, and the sausage packaged with the SF film containing catechin had a decrease in the populations of E. coli O157:H7 and L. monocytogenes by 0.83 and 0.85 log CFU/g after 12 days of storage, respectively, compared to the control. In addition, the sausage had a better quality than the control regarding lipid oxidation. Our results indicate that sausages can be packed with the SF film containing catechin to extend shelf life.

  • 본 연구는 E. coli O157:H7에 대한 LP system(lactoperoxidae/thiocyanate/hydrogen peroxide)의 항균효과를 측정하기 위해 수행되었다. 초기 접종수준($10^{2},\;10^{4},\;10^{7}cfu/ml$), LP의 농도 (10ppm, 20ppm, 30ppm), 배지종류 (TSB, UHT milk, raw milk), 배양온도 ($5^{\circ}C,\;10^{\circ}C,\;15^{\circ}C$) 등에 따라 E. coli O157:H7에 대한 항균효과를 측정, 비교한 결과, 초기 접종수준을 $10^2/ml$으로 하였을 때와 LP의 농도를 10ppm 및 $5^{\circ}C$ 배양에서 항균력이 가장 높게 나타났다.

유산균 발효유의 이용과 건강증진 (Utilization of Fermented Milk and It's Health Promotion)

  • This study was designed to investigate the health promotion effect of fermented milk and historical story of Korean dairy products from the ancient period to present. Although the origin of fermented milk is Europe, the recede of fermented milk was founded in far-east and middle east areas at BC 4C. After the spread of fermented milk to Korea and Japan. The consumption of fermented milk in Korea was dramatically increased to 14.2 kg per person in 1997. Health promotion effect of fermented milk can be devided to 5 major effected improvements of intestinal microflora, anticancer, cholesterol assimilation anti-pathogenic activity. Fermented milk reduced the level of ${\beta}$-glucornidase and nitroreductase to 50% and it provides anticancer activity by cell wall an polysaccharides. Fermented milk has cholesterol assimilation activity ca. 54${\sim}$40% (B. longum, Str. thermophillus). Anti-pathogenic activity of fermented milk was significant. It appeared that Sal. ser. typhimurium was more susceptible than 5. coli 0157 at low pH fermented milk. Viable cells of E. coli 0157 were not dramatically decreased in most of fermented milks tested, but in general, Sal. ser. typhimurium was drastically decreased in most of the fermented milks.

  • OK serogroups of 268 cultures of Escherichia coli isolated from piglets with colibacillary diarrhea were determined by the use of the simplified routine diagnostic procedures of Sojka. The results obtained are summarised as follows: 1. Of 268 cultures of Escherichia coli tested, 190 cultures were classified into 15 OK groups and the remaining 78(29.1%) were untypable. 2. The most frequently isolated enteropathogenic E. coli in order of prevalence were 0157 : K 'Vl7' (14.2%), 0149 : K91, K88a, c (13.7%), 064 : K 'V142' (11.6%), 08 : K87, K88a, b (10.5%) and O141 : K85a, b, K88a, b (7.9%).

장조림 원료육의 미생물 분포 및 분리 병원성 세균의 동정

  • 장조림 원료 쇠고기 우둔살과 가열 처리한 쇠고기 우둔살에서 중온성 세균, 저온성 세균, 혐기성 세균, 포자형성균 및 대장균군 등과 같은 미생물 균수의 변화를 측정하였고, B. cereus, C. botulinum, C. perfringens, L. monocytogenes 등과 같은 9종류의 주요 병원성 세균의 분리를 시도하였다. 원료육에서 중온균, 저온균, 혐기성 균 등은 대부분 높은 분포를 보였으나 가열 처리육에서는 급격히 감소하였으며, 대장균군, E. coli, Enterpbacteriacea, C. perfringens, S. aureus 등은 원료육과 가열 처리한 쇠고기 우둔살에서 모두 검출되지 않았다. 식중독균인 B. cereus, C. perfringens 및 L. monocytogenes등 3균주가 원료육에서 분리된 반면, C. botulinum, E. coli 0157:H7, Salmonella spp., Shigella spp., S. aureus 및 Y. enterocolitica는 분리되지 않았다.

설사자돈으로부터 병원성대장균, 캠필로박터속균 및 살모넬라속균의 분리동정 (Isolation of Enteropathogenic Escherichia coli, Thermophilic Campylobacter and Salmonellae from Scouring Piglets)

  • This study was conducted to isolate etiological agents from the 103 scouring piglets in Gyeongnam area and also carried out antimicrobial drug susceptibility test and epidemiogical served. The incidence of scouring piglet was most prevalent as 81.6% in the age of 2 to 4 weeks after birth, while the rate was less than 10% in the age of 5 to 6 weeks and under 1 week after birch. When compared the isolation frequency of the each etiological agent, enteropathogenic E. coli was most prevalent as 46.6%, thermophilic Campylobacter 26.2% and Salmonellae was 8.7% in order. In the OK serotyping for 117 isolates of enteropathogenic E. coli, type 0141 : K85 (20.5%), 0157:K88ac(14.5%), 0138:K81 and 0149:K91 (13.3%) were encountered most frequently. In the biotyping for 27 isolates of thermophilic Campylobacter, most strains of C. jejuni were belong to type I (50.0%) and II (25.0%), and most strains of C. coli were belong to biotype I (78.9%). In the serotyping for 9 strains of Salmonellae, 3 strains were grouped as D, 2 strains as C. and each 1 strain was group B and E. The other 2 strains were untypable. The 117 isolates of enteropathogenic E. coli were resistant more than 90% to erythromycin, penicillin, tetracycline and streptomycin, wherease about 90% of the isolates were sensitive to kanamycin and gentamicin. In the case of Salmonellae, all of the isolates were resistant to penicillin, but about 89% of the isolates were sensitive to gentamicin and colistin. All of C. jejuni and C. coli isolates were resistant to cephalothin, but more than 89% of C.jejuni and C. coli were sensitive to kanamycin and gentamicin.
