  • Shale gas is in the limelight as one of the new energy sources under the circumstances of unstable energy supply and high energy consumption. It is expected to change dynamics of global energy markets due to its abundant resources and global distribution. Shale gas extraction process consists of drilling, fracturing and production. We have surveyed the technologies required for shale gas developments such as a horizontal drilling, a hydraulic fracturing and so on, the environmental issues occurred during the development, the additional technologies to solve the environmental problems and the current research and developments status.

Differential Protein Expressions in Virus-Infected and Uninfected Trichomonas vaginalis

  • Protozoan viruses may influence the function and pathogenicity of the protozoa. Trichomonas vaginalis is a parasitic protozoan that could contain a double stranded RNA (dsRNA) virus, T. vaginalis virus (TVV). However, there are few reports on the properties of the virus. To further determine variations in protein expression of T. vaginalis, we detected 2 strains of T. vaginalis; the virus-infected ($V^+$) and uninfected ($V^-$) isolates to examine differentially expressed proteins upon TVV infection. Using a stable isotope N-terminal labeling strategy (iTRAQ) on soluble fractions to analyze proteomes, we identified 293 proteins, of which 50 were altered in $V^+$ compared with $V^-$ isolates. The results showed that the expression of 29 proteins was increased, and 21 proteins decreased in $V^+$ isolates. These differentially expressed proteins can be classified into 4 categories: ribosomal proteins, metabolic enzymes, heat shock proteins, and putative uncharacterized proteins. Quantitative PCR was used to detect 4 metabolic processes proteins: glycogen phosphorylase, malate dehydrogenase, triosephosphate isomerase, and glucose-6-phosphate isomerase, which were differentially expressed in $V^+$ and $V^-$ isolates. Our findings suggest that mRNA levels of these genes were consistent with protein expression levels. This study was the first which analyzed protein expression variations upon TVV infection. These observations will provide a basis for future studies concerning the possible roles of these proteins in host-parasite interactions.

Improvement of heat and drought photosynthetic tolerance in wheat by overaccumulation of glycinebetaine

  • Within their natural habitat, crops are often subjected to drought and heat stress, which suppress crop growth and decrease crop production. Causing overaccumulation of glycinebetaine (GB) has been used to enhance the crop yield under stress. Here, we investigated the response of wheat (Triticum aestivum L.) photosynthesis to drought, heat stress and their combination with a transgenic wheat line (T6) overaccumulating GB and its wild-type (WT) Shi4185. Drought stress (DS) was imposed by controlling irrigation until the relative water content (RWC) of the flag leaves decreased to between 78 and 82%. Heat stress (HS) was applied by exposing wheat plants to $40^{\circ}C$ for 4 h. A combination of drought and heat stress was applied by subjecting the drought-stressed plants to a heat stress as above. The results indicated that all stresses decreased photosynthesis, but the combination of drought and heat stress exacerbated the negative effects on photosynthesis more than exposure to drought or heat stress alone. Drought stress decreased the transpiration rate (Tr), stomatal conductance (Gs) and intercellular $CO_2$ concentration (Ci), while heat stress increased all of these; the deprivation of water was greater under drought stress than heat stress, but heat stress decreased the antioxidant enzyme activity to a greater extent. Overaccumulated GB could alleviate the decrease of photosynthesis caused by all stresses tested. These suggest that GB induces an increase of osmotic adjustments for drought tolerance, while its improvement of the antioxidative defense system including antioxidative enzymes and antioxidants may be more important for heat tolerance.

D3 when it was irradiated to the fabrics which had different UV8 transmittance, production of vile. Ds by UVB(Ultraviolet B) and inhibition from formation of vile. D3 by fabrics were absorbed and followings are the results. As the amount of irradiated compared UVB increased, the amount of the production of vile. D3 produced by UVB irradiation from 7-DHC was increased. After treatment of 7-DHC by UVB irradiation and incubated respectively for 24hr, 48hr and 72hr at 36.5$^{\circ}C$ The amount of lit. D3 was increased as incubating time passed. When irradiated UVB on 7-DHC, intermediate of vile. D3, lumisterol, tachysterol and previt. D were showed and those materials were seemed to be changed to vile. D3 as incubation time passed. The amount of vile. D3 which was produced by irradiation 7-DHC showed close relation with UVB transmittance rate of summer fabrics (r= 0.987). Clothes, hats, and sun screen cream reduce the amount of vile. D3 Produced naturally in human skin and it result the decrease of calcium in blood which is absorbed through vit. D. Those all can cause or worsen osteomalacia especially to women and the aged people. Therefore, it is necessary to research and to develop function oriented clothing which can transmit UV which produce vile. D3 at the same time which can protect toxical UVB.

Evaluation of Drought Tolerance using Anthesis-silking Interval in Maize

  • We screened the drought tolerant maize using seventeen maize genotypes from different sources, nine inbred genotypes from United States Department of Agriculture (USDA) (B73, CML103, CML228, CML277, CML322, CML69, Ki3, Ki11, and NC350), three Southeast Asian genotypes (DK9955, LVN-4, and 333), and five Korean hybrids (Cheongdaok, Gangdaok, Ilmichal, Kwangpyeongok, and Pyeonganok). We evaluated anthesis-silking interval (ASI), leaf senescence (LS), ears per plant (EPP), tassel length (TL), and fresh weight (FW) at silking date. According to ASI and LS examination, CML103 and Kill were drought tolerant genotypes, wheareas Ki3 and 333 were drought susceptible. EPP, TL, and FW differed according to drought resistance. Grain yield was correlated strongly with ASI, but moderately with LS. Difference in ASI between drought-stressed (DS) and well-watered (WW) conditions was less than three days in CML228, CML103, Cheongdaok, NC350, B73, Ki11, CML322, and Kwangpyeongok, whereas that of Ki3, Pyeonganok, and Gangdaok was more than 6.5 days. We concluded that CML228, CML103, Cheongdaok, NC350, B73, Ki11, CML322, and Kwangpyeongok are drought tolerant genotypes, whereas Ki3, Pyeonganok, and Gangdaok are drought susceptible.

Improving Cellulase Production in Trichoderma koningii Through RNA Interference on ace1 Gene Expression

  • Ribonucleic acid interference (RNAi) inhibits the expression of target genes in a sequence-specific manner, and shows potential for gene knockdown in filamentous fungi, in which the locus-specific gene knockout occurs in low frequency. In this study, the function of the repressor of cellulase expression I (ACEI) was verified in Trichoderma koningii (T. koningii) YC01 through RNAi, and ace1-silenced strains with improved cellulase productivity were obtained. An expression cassette that transcribed the interfering double-stranded RNA (dsRNA) of ace1 was constructed and transformed into T. koningii, and the transformants, in which the expression of ace1 was successfully silenced, were selected. As a result of the ace1 gene silencing, the expression levels of the main cellulase and xylanase genes were elevated, and the enhanced production of total proteins, cellulase, and xylanase was observed in the cultivation. In addition, the down-regulation of ace1 resulted in an increasing expression of xyr1, but no clear variation in the expression of cre1, which suggested that ACEI acted as a repressor of the xyr1 transcription, but was not involved in the regulation of the cre1 expression. The results of this work indicate that ace1 is a valid target gene for enhancing enzyme production in T. koningii, and RNAi is an appropriate tool for improving the properties of industrial fungi.

Evaluation of Radio-Frequency Performance of Gate-All-Around Ge/GaAs Heterojunction Tunneling Field-Effect Transistor with Hetero-Gate-Dielectric by Mixed-Mode Simulation

  • In this work, the frequency response of gate-all-around (GAA) Ge/GaAs heterojunction tunneling field-effect transistor (TFET) with hetero-gate-dielectric (HGD) and pnpn channel doping profile has been analysed by technology computer-aided design (TCAD) device-circuit mixed-mode simulations, with comparison studies among ppn, pnpn, and HGD pnpn TFET devices. By recursive tracing of voltage transfer curves (VTCs) of a common-source (CS) amplifier based on the HGD pnpn TFET, the operation point (Q-point) was obtained at $V_{DS}=1V$, where the maximum available output swing was acquired without waveform distortion. The slope of VTC of the amplifier was 9.21 V/V (19.4 dB), which mainly resulted from the ponderable direct-current (DC) characteristics of HGD pnpn TFET. Along with the DC performances, frequency response with a small-signal voltage of 10 mV has been closely investigated in terms of voltage gain ($A_v$), unit-gain frequency ($f_{unity}$), and cut-off frequency ($f_T$). The Ge/GaAs HGD pnpn TFET demonstrated $A_v=19.4dB$, $f_{unity}=10THz$, $f_T=0.487$ THz and $f_{max}=18THz$.

Changes of Soil Physico-chemical Properties by Repeated Application of Chicken and Pig Manure Compost (계분 및 돈분퇴비의 연용에 의한 토양의 물리화학성 변화)

  • The changes of soil physico-chemical properties were investigated in the sandy loam soil amended with various application rates of chicken and pig manure composts. After repeated application of the composts for 3 years, total nitrogen content in soil treated with applied 120Mg/ha of chicken and pig manure composts was 2.1 g/kg equally. Organic matter content was 38.8 and 39.1 g/kg, available phosphate content was 602 and 585 mg/kg, and cation exchange capacity(CEC) was 10.1 and 12.4 cmol/kg in chicken and pig manure compost 120 Mg/ha treatment, respectively. Exchangeable K, Ca, Na, Mg contents, and electrical conductivity(EC) increased with the amount of applied compost. Also, with increased amount of applied compost, porosity of soil increased, but hardness, bulk density and Y value decreased.

The Requirement of Ruminal Degradable Protein for Non-Structural Carbohydrate-Fermenting Microbes and Its Reaction with Dilution Rate in Continuous Culture

  • A continuous culture study was conducted to determine the impact of ruminal degradable soy protein (S-RDP) level and dilution rate (D) on growth of ruminal non-structural carbohydrate-fermenting microbes. Corn starch, urea and isolated soy protein (ISP) were used to formulate three diets with S-RDP levels of 0, 35 and 70% of total dietary CP. Two Ds were 0.03 and $0.06h^{-1}$ of the fermenter volume in a single-effluent continuous culture system. As S-RDP levels increased, digestibilities of dietary dry matter (DM), organic matter (OM) and crude protein (CP) linearly (p=0.001) decreased, whereas digestion of dietary starch linearly (p=0.001) increased. Increasing D from 0.03 to $0.06h^{-1}$ resulted in decreased digestibilities of dietary DM and OM, but had no effect on digestibilities of dietary starch (p=0.77) and CP (p=0.103). Fermenter pH, the concentration of volatile fatty acids (VFA) and daily VFA production were unaffected (p=0.159-0.517) by S-RDP levels. Molar percentages of acetate, propionate and butyrate were greatly affected by S-RDP levels (p=0.016-0.091), but unaffected by D (p=0.331-0.442). With increasing S-RDP levels and D, daily bacterial counts, daily microbial N production (DMNP) and microbial efficiency (MOEFF; grams of microbial N produced per kilogram of OM truly digested) were enhanced (p=0.001). The increased microbial efficiency with increasing S-RDP levels is probably the result of peptides or amino acids that served as a stimulus for optimal protein synthesis. The quantity of ruminal degradable protein from soy proteins required for optimum protein synthesis of non-structural carbohydrate-fermenting microbes appears to be equivalent to 9.5% of dietary fermented OM.

Transdermal Delivery of FITC-Ovalbumin with Microneedle System (마이크로 피부침을 이용한 FITC-OVA의 경피흡수)

  • For transdermal delivery of large molecular drugs such as vaccine and protein drugs, novel microneedle treatment device with roll was designed. The roll dimension is 1.43 cm diameter and 2.8 cm perimeter. Total number of microneedle on the roll is 3,360 with $230\;{\mu}m$ height and $740\;{\mu}m$ distance. The pore with $150\;{\mu}m$ depth and $35\;{\mu}m$ diameter on the skin was made by the designed microneedle device. This system could be achieved without pain. The permeation rates of FITC labelled ovalbumin (FITC-OVA, molecular weight: 45,000 g/mol) as a model protein were determined by modified Franz diffusion cells using skins of hairless mice or SD rats which were treated by using microneedle device two or four times. The average penetration fluxes of model protein increased from 674 to $872\;{\mu}g/cm^{2}{\cdot}hr$ as the number of treatment to make pore increased from two to four times. In conclusion, we confirmed the possibility of using the designed microneedle treatment device for transdermal delivery of the large molecular drugs.
