  • A random study of 574 dairy farms in Gyeongnam area was designed to determine 1) management factors that may be associated with the occurrence of drug residues; 2) the dairy farmer's attitudes and knowledge about residues; 3) how these variables influence the occurrence of residues in dairy cattle. Management factors perceived as having the greatest influence on drug residues in milk were insufficient knowledge about withdrawal periods, errors due to hired help, insufficient identification and record of animals treated for mastitis, dry cow treatment for mastitis, and metritis treatment. Seventy-one percent of farms with residues problem used mixed own feeds compared with twenty-nine percent of farms with premedicated feeds. Factors significantly associated with the occurrence of residues were herd size, increased number of hired persons, increased frequency of use of mixed-own feeds, category of medicated feed, and producer's attitude toward the public health significance of residues. Our findings suggest that residue occurrence was mainly associated with errors due to hired help, insufficient knowledge about withdrawal periods, poor animal identification and records of treatment animals and use of medicated feeds. Any residue avodiance educational program needs to stress how to deal with these factors. This educational program should be directed to dairy farmers and employees, especially temporary employees. In addition, Dairy farmer's attitudes and knowledge about drug residues need to be improved. More evidence on the public healths significance of residues should be available to them. Because belife in importance of public healths concerns was related to successful residue avidance and because 81.3% of the dairy farmers with residue problem thought public healths concerns were less important than economic ones, it would be helpful to provide educational programs specifically directed to this issue. it may be useful to provide programs not only for the dairy farmers but also change of their concerns about on the public healths.

시설재배 근대 중 Neonicotinoid계 살충제 Acetamiprid 및 Thiamethoxam의 생산단계 잔류특성 (Residue Dissipation Patterns of Neonicotinoid Acetamiprid and Thiamethoxam in Swiss Chard for the Harvest Periods under Greenhouse Conditions)

  • BACKGROUND: Dissipation of acetamiprid and thiamethoxam in greenhouse grown chard samples was evaluated at 5 intervals including the pre-harvest interval after application. This study was conducted to determine the residue levels, the biological half-lives and dissipation rate of acetamiprid and thiamethoxam in chard under controlled conditions. METHODS AND RESULTS: Acetamiprid and thiamethoxam were applied in accordance with good agricultural practices for chard. Chard samples were collected at 0, 1, 2, 3, 5, 7, 10 and 14 days after application. Quantitaion was performed by HPLC-DAD system with C18 column. Limit of quantification (LOQ) of acetamiprid and thiamethoxam were both 0.02 mg/kg for chard. The recovery of acetamiprid and thiamethoxam were 77.8~107.5% and 94.3~108.6% at two concentration levels. The half-lives of pesticide dissipation in chard for two fields were 11.9 and 8.2 days for acetamiprid and 3.6 and 3.3 days for thiamethoxam respectively. The dissipation rate of acetamiprid and thiamethoxam were estimated according to the statistics method with a 95% confidence. CONCLUSION: Dissipation of acetamiprid and thiamethoxam in chard were determined under greenhouse. The concentration of acetamiprid and thiamethoxam in chards at 0 days after application were below specified by Korean MRL. Dissipation rate constant will be useful to set the pre-harvest residue limit for public health and consumer protection.

다공성 규조토 컬럼을 이용한 한약재 중 잔류농약 분석법 개선 (Improvement of Analytical Method for Residue Pesticides in Herbal Medicines Using Macroporous Diatomaceous Earth Column)

  • 식품의약품안전청에서 고시한 한약재 중 잔류농약 분석법에 따라 천궁 중 bifenthrin, chlorfenapyr 및 chlorpyrifos와 황기 중 acetamiprid, azoxystrobin을 분석한 결과 크로마토그램 상에 많은 간섭물질이 나타나고 회수율이 낮은 문제점이 나타났다. 이를 개선하기 위한 방법으로 액-액 분배 과정 대신 macroporous diatomaceous earth(MDE) 컬럼을 적용하고 정제 시 용출용매 조건을 최적화함으로써 회수율은 황기에서 acetamiprid 89.6~94.1%, azoxystrobin 86.8~94.4%, 천궁에서 bifenthrin 83.6~88.4%, chlorfenapyr 77.4~83.8%, chlorpyrifos 82.6~84.3%, 변이계수(CV)는 전체시료에서 0.5~1.7%로 나타나 잔류농약 분석기준인 70~120% 의 회수율과 10% 이내의 변이계수를 만족하였다. 액-액 분배 과정을 대신하여 MDE 컬럼을 적용함으로써 발암가능 물질로 분류된 methylene chloride 사용을 배제할 수 있어 분석자의 안전성을 증대시킬 수 있었다. 또한 발생하는 폐액의 감소와 전처리 시 노동력 및 시간의 절감, 분석자간의 재현성 양호 등의 이점도 얻을 수 있었다.

수출입 원예작물의 검역을 위한 살균제 Fluazinam의 공정 잔류분석법 개발 (Development of an Official Method for Measurement of Fluazinam Residues for Quarantine of Imported and Exported Horticultural Products)

  • 본 연구는 국내 및 수출입 원예작물의 일상적 잔류농약 검사에 활용 가능한 dinitroaniline계 광범위 살균제 fluazinam의 GC-ECD/MS 잔류 분석법을 확립하고자 하였다. 원예작물 중 채소류에서는 청양고추와 결구배추를, 과일류에서는 감귤과 사과를 대표 시료로 선정하였고, 아세톤으로 추출된 fluazinam은 식염수로 희석하고, 수용액층을 n-hexane 액-액 분배법으로 회수하였으며, 회수된 추출액으로부터 fluazinam의 최종적 정제를 위하여 florisil 칼럼 크로마토그래피법으로 추가 정제하여 GC-ECD/MS 분석용 시료로 사용하였다. Fluazinam의 정량적 분석을 위하여 DB-17 capillary column을 적용한 GC-ECD 분석 조건을 확립하였으며, 각 작물별 4수준의 농도로 fluazinam을 인위 첨가하여 회수율을 분석하였다. 각 대표 원예작물에 대해 정량한계, 정량한계의 10, 50, 및 100배 수준에서 회수율을 검토한 결과 모든 처리농도에서 82.5-99.9% 수준을 나타내었으며, 정량한계(LOQ)는 $0.004mg{\cdot}kg^{-1}$이었다. 반복 간 변이계수(CV)는 최대 6.6%를 나타내어 국제적 잔류분석 기준인 회수율 70-120% 및 분석오차 10% 이내를 충족시키는 만족한 결과를 도출하였다. 또한 GC/MS SIM을 이용하여 실제 각 작물 시료에 적용하여 재확인 하였다. 이상의 결과로 신규 fluazinam의 GC-ECD/MS 분석법은 검출한계, 회수율 및 분석오차 면에서 국제적 분석기준을 만족하는 신뢰성이 확보된 정량 분석법으로서 국내 및 수출입 검역을 위한 원예작물의 공정 잔류분석법으로 사용 가능할 것이다.

원유내 약물잔류의 원인에 관한 연구 (The reasons of drug residues in bulk milk)

  • The 102 farms received a positive result of the bulk milk drug residue test were selected to investigate the reasons of drug residues in bulls milk. The most frequent causes of drug residues were milker or producer mistakes (28.4%), failure to observe withdrawal time (21.5%), and withholding milk from treated quarters only (19.6%). Milker or producer mistakes occurred high at the farms having a parlor system (4 cases out of 11 farms), and related to the inadequate records and marking of treated cows. The lack of knowledge on the absorption of antibiotic from treated quarters and its excretion from untreated quarters caused mainly withholding milk from treated quarters only. Among the 91 farms identified the cause of drug residues, most of the route of drug administration was intramammary infusion (81.3%), and mostly drug used for the treatment of cows was ${\beta}$-lactam antibiotic (57.1%)

한국의 잔류농약 모니터링 프로그램 현황과 개선 (Present Status on the Pesticide Residue Monitoring Program of South Korea and Its Improvement)

  • 이 연구는 식품 중 잔류농약에 대한 국내 모니터링 프로그램의 전반적인 현황을 이해하기 위해 수행되었으며 더 나아가 개선이 필요한 사항이 제안되었다. 이 연구로 부터 국내 잔류농약 모니터링 프로그램 현황은 다음과 같이 요약될 수 있었다. 국내에서는 식품의약품안전처가 잔류농약 모니터링을 총괄한다. 그리고 모니터링 시점(유통 또는 생산단계에서 시료수집)에 따라 모니터링 책임 기관이 다른데, 유통단계의 식품에 대해서는 식품의약품안전처, 지방식품의약품안전청, 지방자치단체가, 생산단계에서는 국립농산물품질관리원(NAQS)과 지방자치단체가 모니터링을 실시한다(부분적으로 판매와 유통단계에서도 실시). 국내의 모니터링 프로그램을 목적에 따라 구분하면 위해평가 모니터링(monitoring for risk assessment)으로 MFDS의 "잔류실태조사"와 NAQS의 "국가잔류조사"가 있고, 지방식품의약품안전청과 지방자치단체에서는 주로 규제 모니터링(monitoring for regulation)을 실시하고 있었다. 수입식품의 경우 통관단계(지방식품의약품안전청 책임)와 유통단계 모두에서 모니터링이 실시되어야 한다. 유통단계 수입식품 모니터링은 MFDS, 지방식품의약품안전청, 지방자치단체가 담당하고 있는 데 아직 체계적이고 지속적인 국가수준의 모니터링 프로그램이 실시되고 있지 않는 것으로 보인다. 국내 잔류농약 모니터링 프로그램과 관련하여 앞에서 기술한 내용과 더 상세한 내용을 토대로 모니터링 프로그램의 개선을 위해 i) 모니터링 프로그램의 목적에 대한 명확성 제고, ii) 수입식품에 대한 모니터링 프로그램의 강화 iii) 일반국민에게 모니터링 결과의 공개(연간보고서와 데이터베이스 발간)를 제안하였다. 식품의 안전성을 확보고 시행하기 시작한 농약 허용물질목록 관리제도(positive list system, PLS)가 성공을 거두기 위해서는 잔류농약 모니터링 프로그램에 대한 철저한 검토와 개선을 위한 노력이 필요한 것으로 생각된다.

Development and validation of an analytical method for the quantification of 2,6-diisopropylnaphthalene in agricultural products using GC-MS/MS

  • An analytical method was developed and optimized for the quantification of a plant growth regulator, 2,6-diisopropylnaphthalene (2,6-DIPN), in agricultural products using gas chromatography-tandem mass spectrometry. The samples were extracted, partitioned, and were purified using a Florisil® cartridge. To validate the analytical method, its specificity, linearity, limit of detection (LOD) and limit of quantification (LOQ) of the instrument, LOQ of the analytical method (MLOQ), accuracy, and repeatability were considered. The method displayed excellent results during validation, and is suitable for the determination and quantification of the low residual levels of the analyte in the agricultural samples. All of the results with the optimized method were satisfactory and within the criteria ranges requested in the Codex Alimentarius Commission guidelines and the Ministry of Food and Drug Safety guidelines for pesticide residue analysis. The developed method is simple and accurate and can be used as a basis for safety management of 2,6-DIPN.

  • Food safety is a term broadly applied to food quality that may adversely affect human health. These include zoonotic diseases and acute and chronic effects of ingesting natural and human-made xenobiotics. There are two major areas of concern over the presence of residues of antibiotics in animal-derived foodstuffs with regard to human health. The first is allergic reactions. Some antibiotics, such as penicillins can evoke allergic reactions even though small amounts of them are ingested or exposed by parenteral routes. The second is development of antibiotic resistance in gut bacteria of human. Recently multi-resistant pneumococcal, glycopeptide-resistant enterococci and gram negative bacteria with extended-spectrum $\beta$-lactamases have spread all over the world, and are now a serious therapeutic problem in human. Although it is evident that drugs are required in the efficient production of meat, milk and eggs, their indiscriminate use should never be substituted for hygienic management of farm. Drug should be used only when they are required. In addition to veterinary drugs, environmental contaminants that were contaminated in feed, water and air can make residues in animal products. Mycotoxins, heavy metals, pesticides, herbicides and other chemicals derived from industries can be harmful both to animal and human health. Most of organic contaminants, such as dioxin, PCBs and DDT, and metals are persistent in environment and biological organisms and can be accumulated in fat and hard tissues. Some of them are suspected to have endocrine disrupting, carcinogenic, teratogenic, immunodepressive and nervous effects. The governmental agencies concerned make efforts to prevent residue problems; approval of drugs including withdrawal times of each preparation of drugs, establishment of tolerances, guidelines regarding drug use and sanitation enforcement of livestock products. National residue program is conducted to audit the status of the chemical residues in foods. Recently HACCP has been introduced to promote food safety from farm to table by reducing hazardous biological, chemical and physical factors. Animal Production Food Safety Program, Quality Assurance Programs, Food Animal Residue Avoidance Databank are para- or non-governmental activities ensuring food safety. This topic will cover classification and usage or sources of chemical residues, their adverse effects, and chemical residue status of some countries. Issues are expanded to residue detection methodologies, toxicological and pharmacokinetic backgrounds of MRL and withdrawal time establishments, and the importance of non-governmental activities with regard to reducing chemical residues in food.

Multi-class, Multi-residue Analysis of 59 Veterinary Drugs in Livestock Products for Screening and Quantification Using Liquid Chromatography-tandem Mass Spectrometry

  • BACKGROUND: The objective of this study was to develop a comprehensive and simple method for the simultaneous determination of 59 veterinary drug residues in livestock products for safety management. METHODS AND RESULTS: For sample preparation, we used a modified liquid extraction method, according to which the sample was extracted with 80% acetonitrile followed by incubation at -20℃ for 30 min. After centrifugation, an aliquot of the extract was evaporated to dryness at 40℃ and analyzed using liquid chromatography combined with tandem mass spectrometry. The method was validated at three concentration levels for beef, pork, chicken, egg, and milk in accordance with the Codex Alimentarius Commission/Guidelines 71-2009. Quantitative analysis was performed using a matrix-matched calibration. As a results, at least 52 (77.6%) out of 66 compounds showed the proper method validation results in terms of both recovery of the target compound and coefficient of variation required by Codex guidelines in livestock products. The limit of quantitation of the method ranged from 0.2 to 1119.6 ng g-1 for all matrices. CONCLUSION(S): This method was accurate, effective, and comprehensive for 59 veterinary drugs determination in livestock products, and can be used to investigate veterinary drugs from different chemical families for safety management in livestock products.
