Background: Prenatal exposure to infectious and/or inflammatory insults can increase the risk of developing neuropsychiatric disorder such as bipolar disorder, autism, and schizophrenia later in life. We investigated whether Valeriana fauriei (VF) treatment alleviates prepulse inhibition (PPI) deficits and social interaction impairment induced by maternal immune activation (MIA). Methods and Results: Pregnant mice were exposed to polyriboinosinic-polyribocytidilic acid (5 mg/kg, viral infection mimic) on gestational day 9. The adolescent offspring received daily oral treatment with VF (100 mg/kg) and injections of clozapine (5 mg/kg) for 30 days starting on the postnatal day 35. The effects of VF extract treatment on behavioral activity impairment and protein expression were investigated using the PPI analysis, forced swim test (FST), open field test (OFT), social interaction test (SIT), and immunohistochemistry. The MIA-induced offspring showed deficits in the PPI, FST, OFT, and SIT compared to their non MIA-induced counterparts. Treatment with the VF extract significantly recovered the sensorimotor gating deficits and partially recovered the aggressive behavior observed in the SIT. The VF extract also reversed the downregulation of protein expression induced by MIA in the medial prefrontal cortex. Conclusions: Our results provide initial evidence of the fact that the VF extract could reverse MIA-induced behavioral impairment and prevent neurodevelopmental disorders such as schizophrenia.
Cordycepin은 Cordyceps militaris에서 처음 유래된 nucleoside adenosine 유도체의 일종으로 면역증강 및 항암활성을 포함한 다양한 약리 기능이 있는 것으로 알려져 있다. 본 연구에서는 LNCap 인체 전립선 암세포 모델을 이용하여 cordycepin에 의한 항암활성 기전을 연구하였다. Cordycepin 처리에 따라 LNCap 세포는 처리 농도 의존적으로 증식이 억제되었으며, 이는 apoptosis 유발과 연관성이 있음을 poly ADP-ribose polymerase의 단편화 현상과 Annexin V 염색에 의한 정량적 분석으로 확인하였다. Cordycepin 처리에 따른 flow cytometric analysis 결과로서 cordycepin이 세포주기 G2/M기 정체 현상을 유발하였음을 알 수 있었으며, 이는 cyclin B1 및 cyclin A의 발현 감소와 연관성이 있었다. 또한 cordycepin이 처리된 LNCap 세포에서 cyclin-dependent kinase (CDK) inhibitor p21Waf1/Cip1의 발현이 증가되었지만, CDK2, CDC2 및 Cdc25C의 발현에는 큰 영향을 미치지 않았으며, cordycepin에 의하여 증가된 p21 단백질은 CDK2 및 CDC2와의 복합체를 형성하고 있었다. 본 연구의 결과는 LNCap 전립선 암세포에서 cordycepin에 의한 G2/M 및 apoptosis 유발은 p53 비존적인 CDK inhibitor p21의 발현 증가가 중요한 역할을 하고 있음을 보여주는 것이다.
Anisomycin은 Streptomyces griseolus에 의하여 생성되는 항생제의 일종으로 flagecidin으로도 알려져 있으며, ribosomal 28S subunit에 결합함으로서 단백질의 생성을 억제하는 것으로 알려져 있다. TRAIL은 ligand로서의 death receptor와의 결합을 통하여 세포의 apoptosis를 유발하는 것으로 알려져 있으나, 많은 암세포에서는 이미 TRAIL에 대한 저항성을 획득하여 TRAIL 유도 apoptosis를 회피하는 능력을 가지고 있다. 본 연구에서는 TRAIL 저항성 Hep3B 간암세포를 대상으로 anisomycin이 TRAIL 매개 apoptosis를 촉진 시킬 수 있는지의 여부를 조사하였다. 본 연구의 결과에 의하면, 단독 처리 조건에서 Hep3B 세포의 증식에 유의적인 영향을 미치는 않았던 anisomycin과 TRAIL의 동시 처리는 anisomycin 처리 농도 의존적으로 세포의 증식을 시켰으며, 이는 caspase 활성화를 통한 apoptosis 유발 증가와 연관성이 있음을 확인하였다. 특히 siRNA를 이용한 Hep3B 세포의 인위적인 Bid 발현의 차단은 anisomycin과 TRAIL 동시 처리군에 비하여 apoptosis 유발능이 더욱 증대시켜 TRAIL 연관 Bid의 truncation을 통한 미토콘드리아 의존적 apoptosis 유발 과정을 anisomycin이 효과적으로 촉진시켰음을 보여주었다. 따라서 본 연구의 결과는 anisomycin과 TRAIL의 동시 처리는 TRAIL 저항성 암세포의 사멸을 촉진시킬 수 있는 효과적인 방법임을 의미한다.
본 연구에서는 장수풍뎅이 유충 추출물을 이용하여 알레르기 및 염증 반응에 미치는 효과를 확인하였다. 장수풍뎅이 유충 추출물의 항알레르기와 항염증 효능은 Compound 48/80에 의해 활성화된 비만세포(RBL-2H3)와 lipopolysaccharide(LPS)로 활성화된 대식세포(Raw 264.7)로부터 분비 또는 발현되는 ${\beta}-hexosaminidase$, $TNF-{\alpha}$, IL-4, IL-6, COX-2, nitric oxide, 및 iNOS를 측정하여 관찰하였다. 비만세포에 장수풍뎅이 유충 추출물과 Compound 48/80을 함께 처리한 경우 비만세포의 탈과립에 의해서 분비되는 ${\beta}-hexosaminidase$의 양이 현저히 감소하였으며 $TNF-{\alpha}$, IL-4, COX-2의 발현 또한 효과적으로 감소시킴을 확인할 수 있었다. 또한 LPS에 의해 활성화된 대식세포(Raw 264.7)는 장수풍뎅이 유충 추출물 처리에 의해 염증반응인자인 IL-6와 nitric oxide의 분비량이 현저히 감소함을 확인할 수 있었다. 이상의 결과는 장수풍뎅이 유충 추출물이 알레르기와 염증반응의 원인 물질인 ${\beta}-hexosaminidase$ 억제 효과뿐만 아니라, 염증 cytokine의 발현을 저해하는 것으로 보아 알레르기와 염증 질환의 예방과 치료에 효과적으로 이용될 수 있음을 확인할 수 있었다.
본 연구에서는 청국장의 항비만 및 항염증 효과를 알아보고자 C57BL/6 마우스에 고지방식이를 13주감 섭취시켜 비만을 유도한 후에 고지방 식이에 청국장을 10% 첨가하여 항비만과 항염증 활성을 확인하였다. 고지방식이를 통해 비만이 유도된 mice (HFD)에 고지방식이와 함께 청국장을 첨가하여 섭취한 그룹(CGJ)의 체중 증가율은 고지방식이군과 비교하였을 때 현저하게 감소하였을 뿐만 아니라 정상식이군보다도 유의적으로 감소하였으며 혈중 TG, TC, glucose, insulin, AST와 ALT 분비는 감소하였고 adiponectin의 수치는 유의적으로 증가하였다(p<0.05). 또한 H&E와 Oil red O staining을 통해 13주 동안 고지방식이를 통해 유도된 지방 간의 심각정도가 청국장의 섭취로 인하여 감소한 것을 확인하였다(p<0.05). 간세포는 대식세포, T세포와 같은 면역시스템의 세포들을 포함하고 있는데 비만으로 유도된 간 조직의 염증반응에 청국장이 미치는 영향을 조사하기 위해 FACS 분석을 수행하였다. CGJ군은 HFD군에 비하여 T cell의 침윤과 대식세포의 축적, 호중구의 유입이 유의적으로 감소하였다(p<0.05). 간 조직으로 침윤된 면역세포의 증가가 간 조직 내 염증반응을 유도하는지 확인하기 위해 염증성 cytokine의 수준을 조사한 결과 간과 지방 조직에서 염증성 cytokine인 MCP-1과 $TNF-{\alpha}$의 mRNA 발현이 감소하였으며 지방합성에 관여하는 $PPAR-{\gamma}$, SREBP-1와 $CEBP/{\alpha}$의 mRNA 발현 역시 감소하였다. 이러한 결과들을 통해 청국장이 면역 시스템과 지방생성 전사 인자들의 발현에 관여함으로써 adipogenesis를 조절함과 동시에 염증 억제 효과를 가진다는 사실을 증명하였다.

Excessive microglial activation and subsequent neuroinflammation lead to synaptic loss and dysfunction as well as neuronal cell death, which are involved in the pathogenesis and progression of several neurodegenerative diseases. Thus, the regulation of microglial activation has been evaluated as effective therapeutic strategies. Although dieckol (DEK), one of the phlorotannins isolated from marine brown alga Ecklonia cava, has been previously reported to inhibit microglial activation, the molecular mechanism is still unclear. Therefore, we investigated here molecular mechanism of DEK via extracellular signal-regulated kinase (ERK), Akt and nicotinamide adenine dinuclelotide phosphate (NADPH) oxidase-mediated pathways. In addition, the neuroprotective mechanism of DEK was investigated in microglia-mediated neurotoxicity models such as neuron-microglia co-culture and microglial conditioned media system. Our results demonstrated that treatment of anti-oxidant DEK potently suppressed phosphorylation of ERK in lipopolysaccharide (LPS, $1{\mu}g/ml$)-stimulated BV-2 microglia. In addition, DEK markedly attenuated Akt phosphorylation and increased expression of $gp91^{phox}$, which is the catalytic component of NADPH oxidase complex responsible for microglial reactive oxygen species (ROS) generation. Finally, DEK significantly attenuated neuronal cell death that is induced by treatment of microglial conditioned media containing neurotoxic secretary molecules. These neuroprotective effects of DEK were also confirmed in a neuron-microglia co-culture system using enhanced green fluorescent protein (EGFP)-transfected B35 neuroblastoma cell line. Taken together, these results suggest that DEK suppresses excessive microglial activation and microglia-mediated neuronal cell death via downregulation of ERK, Akt and NADPH oxidase-mediated pathways.
Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.
Curcumin has diverse anticancer activities that lead to tumor growth inhibition of cancer cells and induction of apoptosis. Curcumin is involved in the regulation of multiple genes via transcription factors including NF-${\kappa}B$, STATs, AP1, and SP. Notch signaling plays critical roles in maintaining the balance between cell proliferation, differentiation and apoptosis, and thereby may contribute to the development of various cancers involving breast cancer. This study was to investigate the effects of curcumin on Notch1 gene expression and to explore the underlying mechanism. Here, we found that curcumin decreased the levels of Notch1 mRNA and protein in MDA-MB-231 human breast cancer cells, along with the downregulation of Sp family genes (Sp1, Sp2, Sp3, and Sp4). The repressive effect of curcumin on Notch1 gene transcription was confirmed by performing Notch1 promoter-driven reporter assay and three Sp-binding sites were identified on Notch1 promoter that may act as curcumin-respose elements. Moreover, treatment with mitramycin A, a specific Sp inhibitor, decreased the levels of Notch1 mRNA and protein in human breast cancer cells. Taken together, our results indicate that Notch1 gene expression is downregulated by curcumin, at least in part, through the suppression of Sp family, which may lead to apoptosis in human breast cancer cells.
Chunghyul-dan (CHD) is a combinatorial drug known to exert anti-inflammatory effects in endothelial cells. In this study, we employed global transcriptional profiling using cDNA microarrays to identify molecular mechanisms responsible for the anti-inflammatory activity of CHD in endothelial cells. An analysis of the microarray data revealed that transcript levels of monocyte chemotactic protein-1 (MCP-1), vascular cell-adhesion molecule-1 (VCAM-1) and activated leukocyte cell-adhesion molecule were dramatically altered in CHD-treated endothelial cells. These changes in gene expression were confirmed by RT-PCR, Western blotting and ELISA. Chronic CHD treatment also appeared to decrease MCP-1 secretion, probably as a result of decreased MCP-1 expression. In addition, we determined that chronic CHD treatment inhibited lipopolysaccharide-stimulated adhesion of THP-1 leukocytes to endothelial cells. The inhibitory effect of CHD on LPS-stimulated adhesion resulted from downregulation of VCAM-1 expression. Transmigration of THP-1 leukocytes through endothelial cells was also inhibited by chronic CHD treatment. In conclusion, CHD controls a variety of inflammatory activities by regulating MCP-1 and VCAM-1 gene expression.
본 연구에서는 새로운 미백소재를 개발하기 위해 적양 에탄올추출물을 효소처리 후 EtOAc 분획물(AJE)을 준비하여 in vitro 상에서 이들의 tyrosinase 저해활성과 세포 수준에서의 멜라닌 합성 저해효과를 측정하였다. AJE는 mushroom tyrosinase의 활성에는 영향을 미치지 않았으나 B16-F1 melanoma cell을 이용한 멜라닌 합성 저해효과에 있어서 농도 의존적으로 멜라닌 합성을 저해하여, $40{\mu}g/mL$의 농도에서 52% 이상의 저해효과를 나타내었다. 이러한 멜라닌 합성 저해효과에 대한 작용 기전을 확인하기 위해 western blot을 통해 멜라닌 합성 경로에 관련된 단백질의 발현을 측정하였다. 그 결과 멜라닌 합성에 관여하는 효소인 tyrosinase related protein 1 (TRP-1)의 발현을 억제하였고, 이를 조절하는 전사인자인 microphthalmia associated transcription factor (MITF) 발현 역시 효과적으로 억제하였다. 또한 extracellular signal-regulated kinase (ERK) pathway를 활성화시킴으로써 phosphorylated extracellular signal-regulated kinase (p-ERK)의 발현을 상당히 증가시키는 것을 확인할 수 있었다. 이러한 결과는 AJE가 멜라닌 합성의 신호전달 경로 중 ERK pathway의 활성화를 통해 MITF의 분해를 촉진시키고 이로 인해 MITF의 발현을 감소시키며, 그 결과 멜라닌 합성에 관여하는 효소 중 TRP-1의 발현을 감소시킴으로써 멜라닌 합성을 저해하는 것으로 사료되며, 따라서 AJE는 미백용도의 기능성 원료로서의 가능성이 큰 것으로 판단된다.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
