Pharmacokinetic Comparison of Actonel and Risenel Tablet Containing Risedronate sodium in Healthy Volunteers (건강한 지원자에 있어서 리세드로네이트 35 mg 함유 악토넬정과 리세넬정의 약물 동력학적 비교)

  • The aim of this study was to evaluate the pharmacokinetic parameters of two risedronate preparations. The clinical assessment was conducted on 46 healthy volunteers who received one tablet (Risedronate sodium 35 mg/tablet) in the fasting state, in a randomized balanced $2{\times}2$ cross-over study design. After dosing of one tablet containing 35 mg risedronate sodium, blood samples were collected serially for a period of 48 hours. Plasma was analyzed for risedronate by using LC/MS/MS assay method. The analysis system was validated in specificity, accuracy, precision, and linearity. $AUC_t$, (the area under the plasma concentration-time curve from the zero-time to 48 hr) was calculated through the trapezoidal rule. $C_{max}$ (maximum plasma drug concentration) were compiled from the plasma risedronate concentration-time data of each volunteer. No significant sequence effect was found for the pharmacokinetic parameters indicating that the cross-over design was properly performed. The 90 % - Confidence intervals of the $AUC_t$ ratio and the $C_{max}$ were from log 0.8752 to log 1.1888 and log 0.8457 to log 1.1478, respectively. These values were within the acceptable intervals between 0.80 and 1.25. Therefore, this study demonstrated that no statistically significant difference was identified with respect to the rate and extent of absorption.

Enhanced Production of Anticancer Agent Camptothecin by Double Elicitors in Suspension cultures of Camptotheca acuminata (희수[Camptotheca acuminata] 현탁세포배양에서 복합 Elicitor에 의한 항암제 Camptothecin 생산 촉진)

  • Cell cultures of Camptotheca acuminata, which is known to produce the anticancer indole alkaloid camptothecin and its dervatives, were made to enhance the productivity of camptothecin. In suspension cultures, the maximum cell growth rate in exponential growth phase was $0.269day^{-1}$ which was correlated to 2.58days of cell doubling time. The production of camptothecin was non-growth associated. The camptothecin production was the highest at 11th day form inoculation and then it decreased. Various elicitors were applied to enhance the production of camptothecin. Both of jasmonic acid and cellulase increased the production of camptothecin. The optimal dosing time of elicitor was the beginning of the cultures. The combination of two elicitors was more effective to produce camptothecin than single applications. $20.42{\times}10^{-3}mg/\ell$ of camptothecin was obtained with combined application of two elicitors in two days.

  • Petrochemical complex has been a lot of research for the development of a more mature product and analysis for mid-process and finished products is essential in these process. But these analyzes are still by hand work samples being manufactured in many parts. Moreover they are exposed to hazardous chemical and such as the analysis is being made in a very poor working conditions. In this paper, in order to solve such problems the multi control system has been developed for the automated analysis. In addition, the organic behavior of these systems and the development of a program for the automated applied, and throughout the experiment to verify the reliability of this device for the accuracy of the dosing pumps for the standard solution prepared with a range of error of ${\pm}0.01m{\ell}$ was able to get a very good experimental results.

Anti-inflammatory Effect of Jungcheonhwadamgangki-tang in Formalin-Injection Mouse Paw Chronic Inflammation Model (정천화담강기탕(定喘化痰降氣湯)이 formalin으로 유발된 마우스의 만성 염증에 미치는 영향)

  • In the present study, the anti-inflammatory effect of Jungcheonhwadamgangki-tang(定喘化痰降氣湯) water extract was tested in formalin-injection mouse hind paw chronic inflammation model. The test articles were dosed once a day for 10 days, and changes on the body weight, paw weights were observed with histopathology of induced paw dorsum pedis. In addition, histomorphometry was also monitored at sacrifice. The increases of absolute and relative hind paw weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang. In addition, the increases of hind paw weight detected in the vehicle control, were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang-dosing groups. Base on these aforementioned results, it is concluded that Jungcheonhwadamgangki-tang have clear anti-inflammatory effect on the chronic inflammation induced by formalin-injection.

Evaluation of Maternal Toxicity in Rats Exposed to Multi-Wall Carbon Nanotubes during Pregnancy

  • Objectives: The present study investigated the potential adverse effects of multi-wall carbon nanotubes (MWCNTs) on pregnant dams and embryonic development following maternal exposure in rats. Methods: MWCNTs were orally administered to pregnant rats from gestational day (GD) 6 through 19 at dose levels of 0, 8, 40, 200, and 1000 mg/kg/day. During the test period, clinical signs, mortality, body weights, food consumption, serum biochemistry, oxidant-antioxidant status, gross findings, organ weights, and Caesarean section findings were examined. Results: All animals survived to the end of the study. A decrease in thymus weight was observed in the highest dose group. However, maternal body weight, food consumption, serum biochemical parameters, and oxidant-antioxidant balance in the kidneys were not affected by treatment with MWCNTs. No treatment-related differences in gestational index, embryo-fetal mortality, or fetal and placental weights were observed between treated and control groups. Conclusions: The results show that 14-day repeated oral dosing of MWCNTs during pregnancy induces minimal maternal toxicity at 1000 mg/kg/day in rats. Under these experimental conditions, the no-observed-adverse-effect level of MWCNTs is considered to be 200 mg/kg/day for dams and 1000 mg/kg/day for embryonic development.

The Estimating an Effect of Rapid Flux Increase to a Membrane in the Intermittent Aeration MBR Process Using Alum Treatment (응집제를 활용한 간헐포기 MBR공정에서 순간플럭스 증가가 분리막에 미치는 영향 평가)

  • By supplying air intermittently in various mode, the effects of oxic/anoxic time ratio and air scrubbing in aeration condition on the membrane flux and permeability were investigated. When suction pump stops, vacuum pressure remains inside the suction pump. Therefore, the effect of remaining vacuum pressure in the suction pump on fouling of membrane was investigated. The effect of EPS (Extra cellular Polymeric Substance) which is generated due to the long SRT and high concentration of MLSS and the dose of coagulant on the membrane were also investigated. The suitable oxic/anoxic time ratio for the best removal efficiency of organic matter and nitrogenous matter was 40 minutes (Oxic) : 20 minutes (Anoxic). At this time ratio, alum was dosed into the aeration tank. The result of dosing alum was that the concentration of alum solution might affect nitrification and denitrification. To remove 1 mg/L of phosphorus in MBR process, it needs 0.75 mg/L of alum solution.

The Experimental Model on Morphological Changes of the Liver Cell Treated with Ethanol (에타놀 처치 간세포(肝細胞)의 형태학적 변화에 대한 실험모델)

  • In order to prepare an experimental model on the basis of morphological changes of liver, 90 rabbits were allocated into four groups and 5, 15, 30 and 50% of ethanol (5ml/day) were dosed to each group for 5, 10, 20, 30, 60 and 90 days, and observed the livers with light and electron microscope. The results obtained are summarized as follows: 1. Light microscopically, the hydropic changes were found in the liver from 10th day after 5% and 15% ethanol ingestion, and these findings were remarkable as time goes by. Necrosis of hepatocytes, however, was most remarkable on 60th and 90th day after ingestion. The proliferation of interlobular connective tissue was found from 30th day in this group. 2. The hydropic necrosis of liver cells was found from 5th day in 30% and 50% ethanol dosing groups. The proliferation of interlobular connective tissues and infiltration of the lymphocytes were observed from 30th day in 30% ethanol ingestion group, however, it was found from 20th day in 50% ethanol ingestion group. 3. Electron microscopically, the appearance of various lipid droplets and dilated rER, slightly increased sER and glycogen field, dense mitochondria with scanty cristae and hydropic change were observed on 5th day after 5% ethanol ingestion. These findings were more remarkable on 30th and 90th day. Especially, disorganized outer and inner membrane of mitochondria and autophagic vacuoles were appeared on 90th day. The similar findings were also found in 15%, 30% and 50% ethanol ingestion groups, however, the appearance of fibrillar and myelin-like structures were observed from 30th day after 15% ethanol ingestion. The severe hydropic change and increased sER were observed on 90th day after 15% ethanol ingestion. From 5th day after 30% and 50% ethanol ingestion these findings were more remarkable.

Anti-Inflammatory Effect of Haepyoejin-tang plus Antler water extract in Xylene-Application Mouse Ear Acute Inflammation Model (해표이진탕가녹용(解表二陳湯加鹿茸)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향)

The test articles were once dosed before Xylene-Application, and the changes on Dody weight and weights and histopathological observation of induced ear were conducted with ear histomorphometry, The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Haepyoejin-tang plus Antler in the present study, A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Haepyoeiin-tang plus Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control, were also dose-dependently decreased in the all Haepyoeiin-tang plus Antler-dosing groups. Base on these results, it is concluded that Haepyoeiin-tang plus Antler water extracts have a clear anti-inflammatory effect on the acute inflammation. However, somewhat lower anti-inflammatory effects were detected in Haepyoeiin-tang plus Antler water extracts compare to that of Dethamethason and Dicrofenac. About 500 mg/kg of Haepyoeiin-tang plus Antler water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.

Four-Week Repeated Oral Toxicity Study of Clean Natural in Sprague-Dawley Rats (Clean Natural의 Sprague-Dawley 랫드를 이용한 4주 반복경구투여 독성시험)

  • This study was performed to investigate repeated-dose toxicities of Clean natural, a new disinfectant, in Sprague-Dawley(SD) rats. In the 4-week repeated oral toxicity study, Clean Natural was orally administered once daily via gavage to male and female rats at dose levels of 0, 500, 1,000 and 2,000 mg/kg body weight for 4-weeks. There were no deaths and clinical signs during the dosing period. In both sexes, there were no statistically significant differences between the administered and control groups in urinalysis indicators and hematological parameters. In serum biochemistry, aspartate aminotransferase(AST) was significantly decreased and sodium content was increased in the 2,000 mg/kg male group, while chlorine was significantly decreased in the 2,000 mg/kg female group. Also, albumin, total cholesterol and total bilirubin were significantly increased in the 2,000 mg/kg male and female group. In histopathological examinations, centrilobular hepatocellular hypertrophy in the liver was observed in the 2,000 mg/kg male and female groups. And pigmentation in the spleen was observed in the 2,000 mg/kg male group. In conclusion, four-week repeated oral dose of Clean Natural to rats did not cause apparent toxicological change at the dose less than 2,000 mg/kg body weight. Thus it is suggested that no-observed adverse-effect level(NOAEL) for Clean Natural in rats was considered to be 1,000 mg/kg/day.
