홍조류의 영양 성분과 항산화 활성 (Nutrients and Antioxidant Activity of Red Seaweeds)

  • 갈래곰보(Meristotheca papulosa (Montagne) Kylin), 진두발 (Chondrus ocellatus, Holmes), 꼬시래기(Gracilaria verrucosa (Hudson) Papenfuss) 및 돌가사리(Gigartina tenella, Harvey) 를 대상으로 영양성분을 분석하고, 에탄을 추출물의 항산화 및 아질산염 소거 활성을 실험함으로서 그 기능성을 규명하고자 하였다. 회분의 함량은 시료간의 차가 커 꼬시래기의 경우 $9.8{\pm}0.2g/100g$이었으나 돌가사리는 $17.8{\pm}0.6g/100g$으로 가장 높은 함량이었다. 조섬유의 함량은 $2.0{\pm}0.4{\sim}6.0{\pm}0.7\;g/100g$의 범위였다. 페놀화합물은 꼬시래기에서 $78.4{\pm}1.0mg/g$으로 가장 높은 함량이었으며, 총 플라보노이드의 함량은 진두발과 꼬시래기에서 각각 $14.9{\pm}0.5mg/g$$13.9{\pm}0.8mg/g$으로 정량되어 갈래곰보와 돌가사리에 비해 약 2배 정도 더 높은 함량이었다. 무기물의 총 함량은 갈래곰보에서 12,107.7 mg/kg으로 가장 높았다. 구성 아미노산은 $49.1{\sim}125.6mg/g$으로 시료에 따른 함량 차가 켰으며 대부분의 시료에서 glutamic acid의 함량이 가장 높게 정량되었다. SOD 유사활성은 시료의 농도가 증가함에 따라 유의적으로 증가하는 경향을 나타내었으나 전체적인 활성은 낮았다. 전자공여능이 가장 높은 돌가사리의 경우 $250{\mu}g/ml$농도에서 $53.96{\pm}0.98%$로 50% 이상의 활성을 보였으며 1000 ${\mu}g/ml$농도에서는 $70.52{\pm}1.09%$로 그 활성이 증가하였다. 100 ${\mu}g/ml$의 농도에서는 시료들의 hydroxy radical 소거활성은 $57.87{\pm}1.70{\sim}62.07{\pm}0.87%$의 범위로 ascorbic acid나 BHT에 비해 유의적으로 높은 활성을 나타내었다. 아질산염 소거능은 꼬시래기에서 가장 높았는데 $100{\sim}500{\mu}g/ml$ 농도에서는 $24.04{\pm}1.9{\sim}27.52{\pm}0.82%$의 범위였으나 1000 ${\mu}g/ml$ 농도에서는 $34.81{\pm}1.36%$로 증가하였다. 이상의 항산화 활성을 평가한 결과 본 실험에 사용된 홍조류 4종은 전자공여능과 hydroxy radical 소거활성이 우수하였다.

오디로부터 분리한 페놀성 물질의 항산화 효과 (Antioxidant Activity of Phenol Compounds from Mulberrry Fruits)

  • 뽕나무 열매(오디)를 이용하여 천연항산화제를 개발할 목적으로 오디로부터 페놀성 물질을 추출하여 항산화 활성을 측정하였다. 페놀성 물질의 추출은 60$\%$ ethanol을 사용하여추출하는 것이 수율이 가장 좋았으며, rutin과 isoquercetrin의함량이 높았다. 추출된 오디 추출물의 ABTS radical decolorization과 antioxidant protection factor(PF)를 살펴 본 결과 ABTS는 80 추출물이 약 98$\%$의 억제율로 최고의 저해율을 나타내었고, PF는 60$\%$ 추출물에서 1.25로 가장 높은 protection factor를 나타내었다. DPPH에 대한 전자공여능은 60$\%$ 에탄올 추출물이 물 추출물이나 다른 농도의 ethanol추출물의 전자공여능 보다 우수하였다. 활성산소 중 지방산화를 일으키는데 주요한 역할을 하는 hydroxyl radical에 대한 각 추출물들의 영향은 60$\%$ 에탄을 추출물이 다른 추출물에 비하여낮은 TBARS 값을 나타내었다. 이상의 결과로 볼 때 오디로부터 추출한 페놀성 물질은 천연 항산화 물질로 활용할 수있을 것으로 생각되었다.

염산 가수분해 추출법으로 조제된 자하거 약침액의 생리활성 효과 (Biological Activities of Hominis Placenta Herbal Acupuncture prepared by Hydrochloric Acid Hydrolysis)

  • 본 연구의 목적은 자하거약침액의 항산화기능에 관련한 생리활성을 보기 위하여 대한약침학회에서 제공받은 자하거 약침액을 시료로 사용하였다. 총 폴리페놀 함량은 자하거 약침액 원액에서 24.641 mg/L로 이는 산수유 등과 같은 약용식물과 비슷한 수준의 함량이다. 자하거약침액원액에서 DPPH radical 소거능과 Superoxide dismutase(SOD) 유사활성 측정은 각각 49.4%, 50.87%였다. 또한 ABTS radical 소거능과 Nitrite radical 소거능(pH1.5)에서는 각각 50.01%, 52.8%로 ABTS radical과 Nitrite radical 소거능이 우수하였다. 본 실험결과로볼 때, 자하거 약침액은 항산화능력은 뛰어난 것으로 사료된다. 하지만 현재 자하거 약침액과 관련된 실험이 부족한 실정이기 때문에 앞으로 이에 관련된 더 많은 연구가 필요하다고 생각된다.

상황버섯(Phellinus linteus) 추출물의 생리활성 (Biological Activities of Extracts from Phellinus linteus)

  • 상황버섯을 에탄올 농도별로 추출하여 항산화 효과와 생리활성을 측정해 보았다. 농도별로 추출한 상황버섯의 페놀 함량은 추출에탄올 농도가 짙어질수록 페놀함량이 높게 나타났으며 그 중 80%에탄올 추출물에서 28.36mg/100ml로 가장 높게 나타났다. 상황버섯 추출물의 수용성물질에 대한 항산화 효과는 DPPH radical소거활성이 전체적으로 80%가 넘는 높은 저해율을 나타났고, 80% 및 90%에탄올추출물에서는 각각 94.12, 94.14%로 가장 높은 저해율을 나타내었으며, ABTS radical cation decolorization에서는 80% 에탄올 추출물에서 96.02%로 가장 높은 저해율을 나타내었다. 지용성물질에 대한 항산화효과는 antioxidant protection factor(PF)는 80% 에탄올 추출물에서 1.69로 가장 높게 나타났으며, TBARS는 80%, 90%에탄올 추출물에서 $1.15{\times}100{\mu}M,\;1.21{\times}100{\mu}M$로 대조구인 $1.95{\times}100{\mu}M$보다 낮게 나와 산화촉진인자를 binding하는 능력이 좋은 것으로 나타났다. 고혈압에 관여하는 angiotensin converting enzyme과 gout에 관여하는 xanthine oxidase의 저해효과에서는 80% 에탄올 추출물에서 각각 95.10, 85.07%로 높은 저해율을 나타내었다. 농도별로 추출한 상황버섯을 HPLC로 분석한 결과 protocatecuic acid가 가장 많이 검출되었으며, 이외에도 cafferic acid, coumaric aicd 가 존재하는 것으로 나타났다.

동종골수이식 공여자 간호를 위한 표준임상지침서 개발 (Development of the Clinical Pathway for the Allogeneic Bone Marrow Transplantation Donor Nursing)

  • The purpose of this study was to develop a clinical pathway for the allogeneic bone marrow transplantation donor. For this study, a conceptual framework was developed through a review of the literature including six steps which are using in Jones Hopkins Hospital. USA. The researcher reviewed 129 medical re-cords of donor who had bone marrow donation between January 2002 to January 2004, to identify the overall service contents required by these patients and to make a preliminary clinical pathway. A content validity test was done for the preliminary clinical pathway, a professional group screened 51 medical re-cords and adopted with 3 hospitalization days as the clinical pathway framework. In the fifth step, clinical pathway test was also done to 7 donors from April 28th to July, 2004. After these processes the final clinical pathway was developed. The results of this study are as follows: 1. The vertical axis of the clinical pathway Includes the following 9 items: vital signs, nursing assessment, activity, diet, intervention, medication, test, consultation and patient teaching. The duration of the horizontal axis was 3days from admission to discharge 2. Analysis of the 129 medical records indicated that the average length of stay was 3 4 days. The medical performance according to the vertical axis in the preliminary clinical pathway consisted of 51 items After clinical validity test, it steel consisted of 51 items in the final form. 3. Clinical Validity test was done to 7 bone marrow donors. During these process, The first patient was deleted because he was out of the criteria the investigate set and 6 patients were used, finally The result of this study indicated all of 7 donors were discharged on expected day. 4. Clinical pathway enables to improve the quality of care, multidisciplinary team work It also helps nursing bone marrow donor, effective education to donor or medical member. The results of this study suggest that clinical pathway may be able to improve the quality of nursing care for bone marrow transplantation donors.

생강(生薑)의 Peroxynitrite 제거 기전 (Peroxynitrite Scavenging Mechanism of Zingiberis Rhizoma)

  • Objectives : Peroxynitrite($ONOO^-$), formed from the reaction of $O2^-$ and NO, is a cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA. It has been involved in the aging process and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate scavenging activities of $ONOO^-$ and its precursors, NO and $O_{2^-}$ and its scavenging mechanism of Zingiberis Rhizoma (ZR). Methods : To investigate scavenging activities of $ONOO^-,\;NO,\;O_{2^-}$ and its scavenging mechanism, we used fluorescent probes like DCFDA, DAF-2 and DHR 123. The $ONOO^-$ scavenging activity on ZR was assayed by measuring oxidized dihydrorhodamine 123 (DHR 123) by fluorometry. The scavenging efficacy was expressed as IC50, showing the concentration of each sample that is required to cause 50% inhibition of DHR 123 oxidation. In a separate study, the protective effect of ZR on $ONOO^-$-induced nitration of bovine serum albumin was investigated through immuno-assay with a monoclonal anti-nitryrosine antibody, and a horseradish peroxidase-conjugated anti-mouse secondary antibody from sheep. Results : ZR markedly scavenged authentic $ONOO^-,\;O_{2^-}$ and NO. It also inhibited $ONOO^-$ induced by $O_{2^-}$ and NO which are derived from SIN-1. The data demonstrated that ZR led to decreased $ONOO^-$ mediated nitration of tyrosine through electron donation. It also inhibited the nitration of bovine serum albumin induced by $ONOO^-$ in a dose-dependent manner. Furtheremore, it blocked LPS-induced ROS and RNS generation. Conclusions : These results suggest that ZR can be developed as an effective $ONOO^-$ scavenger for the prevention of aging process and age-related diseases.

호도(胡桃)의 활성산소 및 활성질소 제거 기전 (Scavenging Activities of Reactive Oxygen and Nitrogen Species by Junglans sinensis)

  • Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are widely implicated in the aging process and age-related diseases. The present study was carried out to investigate scavenging activities of Junglans sinensis extract and its subfraction using fluorescent probes, DCF-DA, DAF-2 and DHR 123. Jungians sinensis was washed and crushed. The crushed Junglans sinensis was extracted 3times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 16 g. Scavenging activities of $ONOO^-$ was measured by Kooy' method and ROS was measured by DCFDA assay. Junglans sinensis had the marked scavenging activites of $ONOO^-$, NO and $O_2^-$. Junglans sinensis scavenged $ONOO^-$ through electron donation and dose-dependently inhibited the nitration of bovine serum albumin by $ONOO^-$. Junglans sinensis also had ROS scavenging activity. Especially, ethylacetate fraction of Junglans sinensis showed the most effective scavenging activities for ROS and RNS. These results suggest that Junglans sinensis might be developed as an effective ROS and RNS scavenger Therefore, Junglans sinensis might be used as a preventive agent for the aging and relevant to aging of illness.

노인장기요양보험제도 실시에 따른 노인요양시설 종사자들의 운영환경변화 인식 (Recognition of Employees in Long-term Care Facilities on the Operating Environment Changes According to Introduction of Long-term Care Insurance)

  • This paper investigated the operating environment for the representative of each agency and the facility workers on the basis of analytical result of recognition changes of the operating environment changes under the operating the long-term care insurance. It was described plans to take positive effect on the operating as follows. The first, on the result of regression analysis, the service administrative range takes the biggest effect on the general recognition of executing the long-term care insurance off and on. The affirmative recognition of the service administrative range had the general recognition on the system be positive effect. But the operator of facility asserts that the care manager's professionalism related quality of service be strengthened. The second, on the result of regression analysis, in the financial accounting administrative it is revealed the more positive recognition it is, the more positive effects it has. From the difference verification of an operation size from operation subject, the small operation size and personal facility recognize the long term care insurance positively. On the other side the facilities where the operation size is big recognize the system negatively. The long-term care facility should rearrange a support program newly and the government needs to promote the donation activity, because it is needed to reduce the financial burden of facilities.

Substituent Effect on the Structure and Biological Property of 99mTc-Labeled Diphosphonates: Theoretical Studies

  • Theoretical calculations based on density functional theory (DFT) were performed to study the substituent effect on the geometric and electronic structures as well as the biological behavior of technetium-99m-labeled diphosphonate complexes. Optimized structures of these complexes are surrounded by six ligands in an octahedral environment with three unpaired 4d electrons ($d^3$ state) and the optimized geometry of $^{99m}Tc$-MDP agrees with experimental data. With the increase of electron-donating substituent or tether between phosphate groups, the energy gap between frontier orbitals increases and the probability of non-radiative deactivation via d-d electron transfer decreases. The charge distribution reflects a significant ligand-to-metal electron donation. Based on the calculated geometric and electronic structures and biologic properties of $^{99m}Tc$-diphosphonate complexes, several structure-activity relationships (SARs) were established. These results may be instructive for the design and synthesis of novel $^{99m}Tc$-diphosphonate bone imaging agent and other $^{99m}Tc$-based radiopharmaceuticals.

공공기관의 사회공헌 활동 현황 분석 연구 -혁신도시내 공기업을 중심으로- ([Analysis of Social Contribution Aactivities of Public Institutions] -Focused on Public Institutions in Wonju Innovation City-)

  • 본 연구는 사회공헌에 대한 기존 연구자료를 통해 사회공헌의 중요성과 그동안의 진전과정을 살펴보고 강원원주혁신 도시내에 입주해 있는 공공기관들의 사회공헌 활동 현황을 분석하였다. 총 14개의 공공기관 중에서 9개 기관의 사회공헌 활동에 대해 상세히 분석하였으며 이를 토대로 하여 공공기관의 사회공헌 활동의 올바른 방향을 제시하였다. 첫째, 최근의 경향은 단순 기부나 후원 형태의 사회공헌보다는 공익활동을 통해 지역사회와 상생 협력하는 방향으로 발전되어가고 있다. 둘째, 일회성 단발성 사회공헌보다는 지역사회와 파트너쉽을 가지고 체계적으로 접근하여야 시너지 효과가 극대화한다는 점이다. 셋째, 일방적 사회공헌보다는 지역시민사회와 협력하여 사회적 경제를 창출함으로써 중장기적으로 지역경제 활성화에 기여하여야 한다는 것이다.
