A Theoretical Analysis on Pressure Loss and Gas Volumetric Fraction of Gas-Liquid Two-Phase Flow (기액이상류의 압력손실과 가스상의 체적분율에 관한 이론적 해석)

  • Gas volumetric fractions and pressure loss are very important parameters in understanding and predicting gas-liquid two-phase flows. They are also essential to design large heat exchanging system in many industries, boiler and refrigerating systems mounted at ships. This paper therefore presents a theoretical method of predicting the pressure loss and gas volumetric fractions in gas-liquid two-phase flows for the whole range of pipe inclinations. The theoretical analysis is based on the two-fluid stratified flow model. It also provides the results of the comparisons between this theoretical analysis results and previous experimental results.

Fractionation and Characterization of Fractions with High Antioxidative Activity from the Gelatin Hydrolysates of Korean Rockfish Sebastes schlegelii Skin

  • The purpose of this study was to obtain a fraction with high antioxidative activity from second rockfish gelatin hydrolysates (SRSGHs), which were hydrolyzed with Alcalase and Flavourzyme through ultrafiltration membranes with serial digestions for 1 and 2 h, respectively, and to investigate the feasibility of this fraction as a potential functional food ingredient. Among various fractions that were ultrafiltered from the SRSGH with four types of membrane (1, 5, 10, and 30 kDa), the SRSGH-III fraction, which permeated the 10 kDa membrane but not the 5 kDa membrane, showed the highest antioxidant activity (protection factor=5.13) and angiotensin-I-converting enzyme-inhibiting activity ($IC_{50}$=0.82 mg/mL). These results suggest that the SRSGH-III fraction from the SRSGH can be used as a functional food ingredient. However, further studies examining its antioxidant activity in vivo as well as the different antioxidant mechanisms are needed.

Antioxidant and Anti-inflammatory Activities of the Halophyte Cyrtomium falcatum (염생식물 도깨비고비의 항산화 및 항염증 효과)

  • In the present study, the halophyte C. falcatum extract and its solvent fractions (n-hexane, 85% aqueous methanol, n-butanol, and water) were evaluated for antioxidant and anti-inflammatory activities. Antioxidative ability was measured by DPPH radical, intracellular reactive oxygen species (ROS) and peroxynitrite scavenging, DNA oxidation inhibition, and ferric reducing antioxidant power (FRAP). For DPPH radical and peroxynitrite scavenging, DNA oxidation inhibition, and FRAP, 85% aq.MeOH and n-BuOH fractions showed significant scavenging activity. For production of intracellular ROS in HT-1080 cells, 85% aq.MeOH fraction showed the highest scavenging activity. In addition, anti-inflammatory activity was also assessed by measuring the inhibitory effect against mRNA expression of pro-inflammatory factors (NO, IL-1β, IL-6 and COX-2) in LPS-stimulated Raw 264.7 macrophages. For NO production, crude extract exhibited a strong inhibitory effect at a concentration of 100 ㎍/ml. For mRNA expression of pro-inflammatory cytokines (IL-1β, IL-6, and COX-2), n-BuOH greatly suppressed expression levels of IL-1β and IL-6 at 100 ㎍/ml concentration while 85% aq. MeOH fraction significantly inhibited that of COX-2 even at 100 ㎍/ml. These results suggest that C. falcatum may be used as a potential source for the development of a natural antioxidant or anti-inflammatory agent.

Biochemical Characterization of a Putative Calcium Influx Factor as a Diffusible Messenger in Jurkat Cells, Xenopis Oocytes, and Yeast

  • Highly purified high performance thin layer chromatography (HPTLC) fractions containing a putative calcium influx factor (CIF) were prepared from the Jurkat cells and Xenopus oocytes in which $Ca^{2+}$ stores were depleted by thapsigargin treatment and from the yeast in which intracellular $Ca^{2+}$ stores were also depleted by genetic means. Microinjection of the fractions has been shown to elicit $Ca^{2+}$ dependent currents in Xenopus oocytes. The nature of the membrane currents evoked by the putative CIF appeared to be carried by chloride ions since the current was blocked by the selective chloride channel blocker 1 mM niflumic acid and its reversal potential was about -24 mV. Injection of the calcium chelator 1,2-bis(2-aminophenoxy)ethane-N, N, N',N'-tetraacetic acid (BAPTA) eradicated the current activities, suggesting the current responses are entirely $Ca^2$-dependent. Moreover, the currents were sensitive to the removal of extracellular calcium, indicating the dependence on calcium entry through the plasma membrane calcium entry channels. CIF activities were insensitive to protease, heat, and acid treatments and to Dische-reaction whereas the activities were sensitive to nucleotide pyrophosphatase and hydrazynolysis. The fraction might have a sugar because it was sensitive to Molisch test and Seliwaniff's resorcinol reaction. From the above results, CIF as a small and stable molecule seems to have pyrimidine, pyrophosphate, and a sugar moiety.oiety.

Enzymatic Milling Process for Barley Flour Preparation (보리의 효소적(酵素的) 제분(製粉) 및 이용(利用)에 관한 연구)

  • An enzymatic flour milling process for barley into three major fractions (barley flour, bran-crease-germ and water solubles) was studied. Carbohydrate and protein of barley endosperm could be efficiently solubilized by the digestion process of partially pearled barley with enzymes. Bran, crease and germ were removed from hydrolyzate by filtering through 30-40mesh sieves. And then filtered product was separated into fractions by sedimentation or centrifugation. The most effective digestion of the barley was obtained by the enzyme with higher activities of glucanase and protease under such conditions as barley-water ratio, 1:1.5(W/V) and temperature at $45^{\circ}C$. Total flour yield recovered was approximately 73-76% of the barley, and the portions recovered as bran-crease-germ and water solubles were about 3.6 and 15.8%, respectively.

Purification and Characteristics of New Biopolymer Produced by Alkaline-Tolerant Bacillus sp. (알칼리 내성 Bacillus sp.가 생산하는 생물 고분자의 정제 및 특성)

  • Biopolymer from alkaline-tolerant Bacillus so. was purified, and its physico-chemical and structural properties were investigated. Crude biopolymer, precipitated by acetone from culture broth was fractionated into two fractions by gel chromatography on Sephadex G-200. Among two fractions, one fraction(PS I), which an acidic biopolymer precipitated by the CPC(cetylpyridinium chloride) treatment was studied further. PS I fraction had carboxyl groups and was positive at color reaction of sugar. PS I fraction also showed UV absorbance at 190-225nm. The purified acidic biopolymer was composed of 4% glucose, 8% glucosamine and 88% glutamic acid. Sugar components of the purified acidic biopolymer seemed to be linked to PGA(polyglutamic acid) which existed in the from of ${\gamma}$-peptide bond. By the results of Smith degradation of sugar components, glucose and glucosamine was bound by 1,3 glocosidic linkage. Therefore, this biopolymer was a glycopeptide, oligosaccaride ${\gamma}$-PGA. We concluded that the equivalent weight and the molecular weight of this biopolymer were estimated as about 171 and 5x105 dalton, respectively.

Inhibition of α-Glucosidase by a Semi-Purified Ethyl Acetate Fraction from Submerged-Liquid Culture of Agaricus blazei Murill (신령버섯균사체 액체배양물의 α-glucosidase 저해 효과)

  • Natural anti-diabetic semipurified ethyl acetate fraction was isolated from the submerged-liquid culture of Agaricus blaze Murill (AB) in a medium containing soybean flakes. Hot-water extract of AB (HEAB) was prepared by extraction at $121^{\circ}C$ for 60 min, followed by filtering through a filter presser filled with diatomate. The ${\beta}$-glucan-free HEAB, which was a supernatant fraction from HEAB by precipitation in an 80% ethanol solution, was fractionated into hexane, chloroform, ethyl acetate, and butanol fractions. The inhibition of the ${\alpha}$-glucosidase activity by fractions was 59.0, 17.0, 61.6, and 37.9%, respectively, suggesting that ethyl acetate fraction was the most active. A subfraction having a strong ${\alpha}$-glucosidase inhibitory activity (80.4%) was isolated from the ethyl acetate fraction. This subfraction contained isoflavones (genistin and daidzin) and their conjugates with sugars as potent inhibiters of ${\alpha}$-glucosidase activity. These results suggest that the ethyl acetate fraction or HEAB containing isoflavones and their sugars conjugates could be useful sources for controlling blood sugar levels in humans.

Application of Sedimentary Neodymium Isotopes to the Reconstruction of the Arctic Paleoceanography (퇴적물의 네오디뮴 동위원소 비를 활용한 북극 고환경 복원)

  • Climate and environmental changes in the Arctic Ocean due to global warming have been linked to extreme climate change in mid-latitude regions, including the Korean Peninsula, requiring a better understanding of the Arctic climate system based on the paleo-analog. This review introduces three paleoenvironmental research cases using neodymium isotopes (143Nd/144Nd, εNd) measured on two different fractions of marine sediments: silicate-bound 'detrital' and Fe-Mn oxide-dominated 'authigenic' fractions. In the first case, detrital εNd in core HH17-1085-GC on the continental shelf off northern Svalbard was used for tracing changes in sediment provenance and associated glacier behavior over the last 16.3 ka. The second case showed the potential use of authigenic εNd as a quasi-conservative water mass tracer. Three prominent εNd peaks and troughs observed in core PS72/410-1 from the Mendeleev Ridge in the western Arctic Ocean over the past 76 ka suggested episodic meltwater discharge events during 51~46, 39~35 and 21~13 ka BP. The last case proposed the use of the difference between authigenic and detrital εNd as a proxy for reconstructing glacier fluctuation. The idea is based on the assumption that enhanced glacial erosion during glacier advances can supply sufficient freshly-exposed rock substrate for incongruent weathering, potentially leading to greater isotopic decoupling between bedrock and dissolved weathering products as recorded in detrital and authigenic εNd, respectively. Thus, it would be worthwhile to take advantage of sedimentary εNd to improve our understanding of past environmental changes in polar regions.

Isolation and Identification of Antioxidant Substances from the Stems of Butterbur (Petasites japonicus) (머위(Petasites japonicus) 엽병으로부터 항산화 물질의 분리 및 동정)

  • The stems of P etasites japonicus were extracted with ethanol and then partitioned with hexane, chloroform, ethyl acetate, n-butanol and water, successively. The antioxidant potency of five crude fractions were determined using (1) 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, (2) thiobarbituric acid reactive substances (TBARS) assay in the linoleic acid model system, and (3) lipoxygenase inhibition assay. Among the crude fractions, the ethyl acetate fraction exhibited the most potent antioxidant effect. By activity-guided fractionation, compound PJ-4 was isolated from the ethyl acetate fraction through the repeated silica gel open column chromatography. The chemical structure of the isolated compound was determined as kaempferol by $^1H-$ and $^{13}C$-NMR analysis and its antioxidative capacity was further investigated. DPPH radical scavenging activity of the compound was 65.76% at the concentration of $100 \;{\mu}g/mL$. The inhibitory activity of the compound against lipid peroxidation and lipoxygenase exhibited 43.47% and 58.60%, respectively at the concentration of $100\;{\mu}g/mL$. The result suggests that the compound may serve as a useful natural antioxidant and furthermore indicates the possibility of developing the stems of Petasites japonicus as a natural antioxidant source.

A Study on the Systematic Analysis of Lipids from Sweet Potatoes (고구마 지질성분(脂質成分)의 계통분석(系統分析))

  • Sweet potato lipids were isolated into free and bound lipid fractions and subjected to fractionation and quantitation by thin layer chromatography, followed by flavor evaluation. The results are summarized as follows: 1) Sweet potato flour contained 1.75% total lipids on a dry weight basis, of which free and bound forms were 0.95% and 0.80%, respectively. 2) TLC indicated the presence of 13 components in free lipids and 9 components in bound lipids, of which phospholipids accounted for 34.5% and free fatty acids, 17.2%, as the major components. 3) Free fatty acids were separated and quantitatively determined by reversed TLC to give 30.1% palmitic acid, 16.7% linoleic acid, 15.8% oleic acid and 9.8% linolenic acid as the main components. 4) The flavor of various lipid fractions were evaluated and the disagreeable taste and odor of sweet potato flour was mainly found in the bound lipid fraction.
