Treatment for heart failure is directed to reduce atrial volume overload by diuretic agents, to lessen ventricular pressure overload by vasodilatory agents and to increase myocardial performance through inotropic agents. Of those cardiac therapeutics, diuretic agents are the most important to control heart failure in dogs, although long-term use often causes detrimental side-effects such as acute renal failure and electrolyte abnormalities. Thus, this study was designed to find a new diuretic agent from medicinal herbs which has better diuretic effect and less unfavorable complications in dogs. In a preliminary study performed with 5 normal healthy dogs, the extract from Akebia quinata Decaisne showed mild to moderate diuretic effect (0.3-0.5 potency of furosemide 2 mg/kg) and minimal changes in serum chemistry and electrolyte. Although the study population was not large enough and study period was not sufficient enough, this study found a good alternative diuretic agent which can replace or reduce the use of furosemide in dogs with heart failure.
Captopril, angiotensin converting enzyme (ACE) inhibitor, when given intravenously in dog, elicited the diuretic action along with the increases of glomerular filtration rates (GFR), renal plasma flow (RPF) and osmolar clearances (Cosm) with no changes of free water clearnces ($C_{H_2O}$), and then captopril produced the enlargement of excretion rates of electrolytes in urine and the reduction of reabsorption rates of electrolytes in renal tubles. Captopril, when given into a renal artery, exhibited no changes of renal function in the experinental kidney, whereas diuretic action with the same mechanism as shown in intravenous captopril in control kidney. Captopril, when injected into a carotid artery, showed increases in rates of urine flow in a small does which did not affect on renal action when it was administered intravenusly. Diuretic action induced by captopril was not influenced by renal artery denervation, propranolol and angiotensin II inhibiters. Above results suggest that captopril produced diuretic action along with renal hemodynamic changes by slight contraction of vas efferense and reduction of reabsorption rate of electrolytes in renal tubules, especilly distal tubules, that may be mediatedby endogenous substances.
Angiotensin II, adminstered (infused or injected) intravenously, elicited the antidiuretic action with the decreased parameters of renal function at a small dose ($0.01\;{\mu}g/kg/min$), whereas, at a large dose (0.03, $0.1\;{\mu}g/kg/min$ and $5.0\;{\mu}g/kg$), it produced the diuretic action accompanied the increased amounts of sodium and potassium excreted in urine ($E_{Na}\;and\;R_K$). At this time, glomerular filtration rates (GFR) were weakened slightly and renal plasma flows (RPF) were reduced markedly, and then filtration fractions (FF) were increased. Angiotensin II, infused into a renal artery, exhibited antidiuretic action at a small dose ($0.003\;{\mu}g/kg/min$), and diuretic action at a large dose ($0.01\;{\mu}g/kg/min$), only in infused (experimental) kidney. The mechanism of the action was similar to the cases of the intravenous angiotensin II. The above results suggest that angiotensin II of a large dose produced diuretic action due to mechanism inhibiting reabsorption of electrolytes in renal tubules, mainly in proximal tubule in dog.

Objective : Diuretics are effective on the patients with hypertension and heart failure, as well as edema by regulating the function of kidney, which is key organ to maintain the balance of the different electrolytes in the body. The present review article is designed to review the diuretic effect of Korean medicinal prescription base on the experimental studies.Method : For this purpose, every article related to ‘diuresis’ and 'Korean medicinal prescription' were analysed from articles which published at domestic and international journals.Results : 1. Representative Korean medicine prescriptions showed diuretic effect along with electrolyte excretion were Oryeong-san, Paeryung-tang, Hwangryungbokryung-tang and Daeganghwal-tang. 2. Some Korean medicinal prescription including Oryeong-san, Jueryeong-tang, Sipcho-tang distribute diuresis through the inhibition of renin-angiotensin-system. 3. Oryeong-san, Bojungchiseup-tang had a diuretic effect on the down-regulation of aquaporin water channel in the renal collecting duct.Conclusion : Korean medicinal transcriptions have a diuretic effect through several types of mechanism such as along with electrolytes excretion, inhibition of renin-angiotensin system, and down-regulation of water channels.
The essential oil fraction of Leonurus sibiricus was analyzed by TLC and gas chromatography. By utilizing silica gel column, a ketone compound, m.w. 167, was isolated from the essential oil. The essential oil showed considerably the diuretic action, but the water extract exhibited weak action. This diuretic action of the water extract was potentiated by combined administration of essential oil. On the isolated rabbit's uterus the essential oil decreased spontanous movement and showed relaxation.
The diuretic action of guanethidine was investigated in the dogs by means of the stop-flow technique. Guanethidine increased the rejection of sodium in the ascending limb of Henle's loop, as well as in the proximal and distal tubules, resulting in the decrease of the concentrating ability of the kidney, in marked natriuresis and diuresis. It was also effective during an osomotic diuresis, which was induced by infusing 10% mannitol can exhibit its effect even under the diuretic action of mercurophylline, suggesting a different mechanism from that of mercuric iuretics.
To observe the diuretic action in mice, the anti-inflammatory action by carrageenin method and the anti-pyretic action by yeast method in rats, the Bittern was administered. Diuretic effort of Bittern was studied by measuring the urine flow, sodium, potassium in urinary excretion. The results in this work were summarized as follows: 1. The Bittern showed significant diuretic action. 2. The Bittern showed significant anti-inflammatory effect. 3. The Bittern showed significant anti-pyretic effect. According to the above results, the Bittern seems to be applicable to the treatment of edema.

In the present study, Pisonia grandis leaves were extracted with chloroform and methanol. The extracts were vacuum dried to yield the respective chloroform (CE) and methanol extract (ME). CE and ME were evaluated for analgesic, anti-inflammatory (acute and chronic) and diuretic activity at 2 dose levels (250 and 500 mg/kg). Significant analgesic and anti-inflammatory activities were associated with CE and ME. CE at the dose level of 500 mg/kg was found to exhibit equivalent chronic anti-inflammatory activity as diclofenac at 50 mg/kg dose level. Significant diuretic activity was exhibited by ME. Graded dose response for all the activities were observed for the extracts.
This study was performed to elucited the mechanisms of the antidiuretic action by SKP-450, a $K^+$ channel opener, given into the vein, and of the diuretic action observed only in the ipsilateral kidney, when given into a renal artery, in dog. The antidiuretic action of SKP-450 was not affected by renal denervation or pretreatment with glibenclamide, a ATP-dependent $K^+$ channel blocker. The diuretic action of SKP-450 was inhibited by renal denervation or pretreatment with glibenclamide. SKP-450 given into carotid artery had little effect on renal function. These results suggest that the antidiuretic action of SKP-450 given into the vein is caused by some endogenous substances probably not related to $K^+$ channel, whereas the diuretic action of SKP-450 observed only in ipsilateral kidney, when given into a renal artery, is provoked through $K^+$ channel related to renal nerves.

The pharmacological interest coupled with traditional uses (antidiarrhoeal, antiseptic, analgesic etc) prompted us to test for anti-inflammatory, antinociceptive and diuretic activitities of Trema (T.) orientalis Linn. The crude methanolic leaves extract of T. orientalis was investigated for its possible anti-inflammatory activities using carrageenin induced rat paw edema model and cotton pellet implantation method in mice. Then the extract analyzed for its antinociceptive activities by acetic acid induced writhing model in mice. The extract possessed significant anti-inflammatory activity in both models at the doses of 200 and 400 mg/kg body weight of mice. Moreover, the extract showed significantly reduced the number of acetic acid-induced abdominal constriction in mice of 200 and 400 mg/kg body weight. The extract also showed positive diuretic activity in albino mice.
