메톡시 폴리(에틸렌 글리콜)/폴리에스테르 블록공중합체의 합성 및 미셀 특성 비교 (Synthesis of Methoxy Poly(ethylene glycol)/Polyesters Diblock Copolymers and Evaluation of Micellar Characterization as Drug Carrier)

  • 메톡시 폴리(에틸렌 글리콜)(MPEG)과 생분해성 폴리에스테르 계열의 폴리카프로락톤(PCL), 폴리발레로락톤(PVL), 폴리락타이드(PLLA) 및 폴리 (락타이드-co-글리콜라이드) 공중합체(PLGA)로 이루어진 블록공중합체의 합성 및 특성을 비교하였다. MPEG-PCL과 MPEG-PVL 블록공중합체는 단량체 활성화제로서 $HCl{\cdot}Et_2O$의 존재 하에 상온에서 카프로락톤(${\epsilon}-CL$)과 발레로락톤(${\delta}-VL$)의 개환중합에 의해 합성하였으며, MPEG-PLLA와 MPEG-PLGA 블록공중합체는 $Sn(Oct)_2$의 존재 하에 $130^{\circ}C$에서 락타이드(L-LA)와 PLGA의 개환중합에 의해 합성하였다. 합성된 블록공중합체는 $^1H-NMR$, GPC, DSC 및 XRD의 측정을 통해 특성을 분석하였다. 합성된 NPEG-폴리에스테르 블록공중합체의 수용액 상에서의 미셀 특성은 $^1H-NMR$, 광산란기, 원자 현미경 및 형광 측정기를 이용하여 확인하였다. 원자 현미경을 통해 관찰된 미셀의 형태는 대부분의 블록공중합체에서 구형으로 존재함을 확인할 수 있었다. 본 연구에서는 미셀의 특성 비교를 통해 폴리에스테르 블록의 종류에 따라 열적 특성, 결정화도, 임계 미셀 농도 및 미셀의 직경이 다르게 나타남을 확인할 수 있었으며, MPEG-폴리에스테르 블록공중합체의 미셀내부에 선택적으로 소수성의 약물을 분포시킬 수 있으므로 소수성 약물 전달체로서의 가능성을 확인하였다.

Effects of Different Dietary Cadmium Levels on Growth and Tissue Cadmium Content in Juvenile Parrotfish, Oplegnathus fasciatus

  • This feeding trial was carried out to evaluate the effects of different dietary cadmium levels on growth and tissue cadmium content in juvenile parrotfish, Oplegnathus fasciatus, using cadmium chloride ($CdCl_2$) as the cadmium source. Fifteen fish averaging $5.5{\pm}0.06$ g (mean${\pm}$SD) were randomly distributed into each of twenty one rectangular fiber tanks of 30 L capacity. Each tank was then randomly assigned to one of three replicates of seven diets containing 0.30 ($C_0$), 21.0 ($C_{21}$), 40.7 ($C_{41}$), 83.5 ($C_{83}$), 162 ($C_{162}$), 1,387 ($C_{1,387}$) and 2,743 ($C_{2,743}$) mg cadmium/kg diet. At the end of sixteen weeks of feeding trial, weight gain (WG), specific growth rate (SGR) and feed efficiency (FE) of fish fed $C_{21}$ were significantly higher than those of fish fed $C_{83}$, $C_{162}$, $C_{1,387}$ and $C_{2,743}$ (p<0.05). Weight gain, SGR and FE of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Protein efficiency ratio of fish fed $C_0$, $C_{21}$ and $C_{41}$ were significantly higher than those of fish fed $C_{1,387}$ and $C_{2,743}$. Average survival of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{162}$ were significantly higher than that of fish fed $C_{2,743}$. Tissue cadmium concentrations increased with cadmium content of diets. Cadmium accumulated the most in liver, followed by gill and then muscle. Muscle, gill and liver cadmium concentrations of fish fed $C_0$, $C_{21}$, $C_{41}$ and $C_{83}$ were significantly lower than those of fish fed $C_{162}$, $C_{1,387}$ and $C_{2,743}$. Based on the ANOVA results of growth performance and tissue cadmium concentrations the safe dietary cadmium level could be lower than 40.7 mg Cd/kg diet while the toxic level could be higher than 162 mg Cd/kg diet.

고려인삼과 미국삼의 품질요인별 수삼 및 홍삼등급 비교 (Comparison of Grade of Raw and Red Ginseng on each Factor of Quality in Korean and American Ginseng)

  • 고려인삼과 미국삼에 있어 원료수삼의 품질 외적요인과 홍삼품질과의 관계를 조사하여 종간특성을 비교하였다. 원료수삼의 편급과 등급을 결정하는 품질요인은 체형계수, 동장 그리고 근중을 적용하였으며 이들 요인과 홍삼 품질관계를 구명하였다. 1. 편급의 분포는 고려인삼의 경우 중편과 대편급이 많았으나 미국삼은 중편과 소편급이 많았으며 미국삼은 고려인삼에 비해 동장이 짧고 동직경이 굵고 부정형 지근이 많았다. 2. 원료삼 품질은 고려인삼이 미국삼에 비하여 양호하였고 홍삼품질은 고려인삼이 지삼 이상의 고급홍삼이 많았으나 미국삼은 양삼과 같이 저급홍삼이 많았다. 3. 원료수삼 품질요인중 중편삼과 대편삼의 경우 고려인삼은 체형계수 0.5 이하였으나 미국삼은 0.5를 상회하여 미국삼은 홍삼품질에 적합하지 않았다. 4. 고려인삼과 미국삼 모두 품질요인으로 동장과 근중은 홍삼품질과의 관계에서 비슷한 경향이나 체형계수가 특히 홍삼의 품질을 좌우하는 요인이었다. 5. 따라서 고려인삼과 미국삼 모두 원료삼과 홍삼품질 향상을 위해 근중, 체형계수 그리고 동장의 조정이 요망되었다.

누에나방(Bombyx mori L.) 종령유충일용 전환기(轉換期)의 전흉선(前胸腺) 분필세포(分泌細胞)의 미세구조적 변화 (Ultrastructural Changes on the Secreting Cells of the Prothoracic Gland During the Larva-pupal Molt of Bombyx mori L.)

  • In order to define the morphological changes of the secreting cells of prothoracic gland during larva-pupal molt, ultrastructural observations were carried out using Bombyx mori L. as the experimental material. At first stage of present experiment, 4 day old 5th instar larva, the polyhedral secreting cells were centrally located in the prothoracic gland surrounded by the connective sheath. The secreting cells were attached to the neighboring cells by the prominent desmosomes, and the plasma membrane contacted with connective sheath were highly infolded. In cytoplasm, the most of the cell organelles, such as rod-like mitochondria, rough surfaced endoplasmic reticulum, ribosome were developed. As the stages advance from larva to pupa, general feature of the secreting cells were retained, but structural changes of the various cytoplasmic organelles-ribosome, rough surfaced endoplasmic reticulum, mitochondria, Golgi apparatus, lamellar body, and vesicle-were noted. In the perinuclear cytoplasm of the secreting cells at the stage of 6 day old 5th instar larva, it is peculiar that only a large amount of ribosomes were distributed and the other organelles were retreated from the juxtanuclear region. Just before and after spining cocoon, these features were more remarkable. Rough surfaced endoplasmic reticulum were gradually increased from the stage just before spining cocoon to the pharate pupa. Rod-like mitochondria with irregular cristae and the matrix showing low density were distributed throughout the cytoplasm in the secreting cells of the 4 day old 5th instar larva. Sometimes, longitudinally distended and curved mitochondria were observed. At the stage of pharate pupa, most of mitochondria were deformed. The rod-like mitochondria of the secreting cells of pupal prothoracic gland were narrower than those of 4 day old 5th instar larva, and the electron density of the mitochondrial matrix is increased in pupa. Golgi apparatus were a few in number in both stages, last instar larva and spining cocoon. In stages of the pharate pupa, the Golgi apparatus were frequently observed. Cytoplasmic vesicles were observed for the first time in the secreting cells of one day after spining cocoon, and the number and the size of cytoplasmic vesicles were distinctly increased inpharate pupa and just after pupation. In the secretory cells of the PG, it in concluded that the RER was closely related to syntheting the enzymes seem to produce the ecdysone.

Phenolic Compounds and Triterpenes from the Barks of Diospyros burmanica

  • Diospyros burmanica Kurz. is an evergreen deciduous tree distributed in Mandalay of Myanmar, which belongs to the family of Ebenaceae. In Myanmar, it has been used to treat diarrhea, diabetes, diabetes and also as lumbers. In this study, seven flavonoids (1 - 7), a phenolic compound (8), and five triterpenes (9 - 13) were isolated from the barks of D. burmanica and their chemical structures were elucidated. Isolates were identified to be (+)-catechin (1), (+)-catechin 3-O-$\alpha$-L-rhamnopyranoside (2), (+)-catechin 3-O-gallate (3), (-)-epicatechin (4), (-)-epicatechin 3-O-gallate (5), (+)-afzelechin 3-O-$\alpha$-L-rhamnopyranoside (6), (+)-2,3-trans-dihydrokaempferol 3-O-$\alpha$-L-rhamnopyranoside (7), methyl gallate (8), lupeol (9), methyl lup-20(29)-en-3-on-28-oate (10), $\beta$-amyrin (11), $\alpha$-amyrin (12), $3\beta$-hydroxy-D:B-friedo-olean-5-ene (13) through MS, 1H NMR and 13C NMR spectroscopic evidences.

Performance of Male Crossbred Calves as Influenced by Substitution of Grain by Wheat Bran and the Addition of Lactic Acid Bacteria to Diet

  • To study the effect of wheat bran and lactic acid producing bacteria (LAB) on the performance of calves, 20 crossbred male cattle calves (day old), distributed into two groups were fed on calf starters containing 50 or 0% maize grain, along with green berseem ad libitum and milk as per body weight. Each group was further divided into two sub groups and one subgroup of each group was supplemented with mixed culture of LAB (Lactobacillus acidophilus L. casei, L. Jugarti). Milk feeding was discontinued after 8 weeks of age. The addition of culture increased (p<0.05) DM intake in calves receiving grainless diet from eighth week to the thirteenth one. There was about 21% higher body weight gain and 14% lower feed : gain ratio in culture supplemented calves. DM digestibility was significantly lower (p<0.05) in calves getting grain without culture. The crude protein NDF and ADF digestibility was higher (p<0.05) in grainless than the grain fed group. No major change on rumen fermentation pattern among different treatments was found. The concentration of total volatile fatty acids (TVFA) and protozoa count was higher (p<0.05) in grain fed group. However, lactic acid concentration was higher and rumen pH was lower due to culture feeding. The incidence as well as severity of diarrhoea was reduced in culture supplemented group. The results indicate that crossbred calves can be reared successfully on grainless diet and berseen fodder. The performance of calves was also improved by LAB supplementation.

한국산(韓國産) 느릅나무속 3수종(樹種)의 종자저장성(種子貯藏性) 및 발아특성(發芽特性) (Germination Behaviors and Seed Longevities of Three Ulmus Species in Korea)

  • 우리나라에 자생하는 느릅나무속 3수종[느릅나무(Ulmus davidiana var. japonica(Rehder) Nakai), 비술나무(U. pumila L.), 참느릅나무(U. parvifolia Jacq.)]의 온도조건(4개)에 따른 발아특성(발아율, 평균발아소요일수, 발아속도) 및 건조기간에 따른 종자 활력의 변화를 분석하였다. 종자 발아에 미치는 온도조건의 영향을 살펴보면 3수종 모두 평균발아일수에서 차이를 보였으나, 발아율에서는 참느릅나무에서만 통계적으로 고도의 유의성이 인정되었다. 느릅나무와 비술나무는 모든 온도조건에서 비교적 고른 발아율을 보여주었으며, 특히 비술나무는 저온(10~15$^{\circ}C$)에서도 높은 발아율을 나타냈다. 참느릅나무는 저온(10~20$^{\circ}C$)에서는 거의 발아가 되지 않으며 25$^{\circ}C$ 이상의 높은 온도에서 발아가 되는 독특한 특성을 나타냈다. 비술나무와 참느릅나무 종자는 4~8% 수분함수율 조건에서 시설저장이 가능한 저장성 종자(Orthodox seed)로 판명되었으며, 느릅나무 종자는 장기저장이 어려운 난저장성 종자(Recalcitrant seed)로 나타났다. 결론적으로 느릅나무속 3수종의 발아특성 및 저장성의 차이는 수종별 분포특성에 따른 상이한 환경 조건에 적응하기 위한 종 특유의 생존 전략으로 판단된다.

한반도 자생식물 헐떡이풀 종자의 휴면유형과 발아특성 (Seed Dormancy Type and Germination Characteristics in Tiarella polyphylla D. Don Native to Korea)

  • 헐떡이풀은 우리나라에서 울릉도에서만 분포하는 자생식물로 전통적으로 약용으로 쓰여왔으며, 관상식물로의 활용도 기대되는 자원식물이다. 본 연구는 헐떡이풀 종자의 휴면타파와 발아를 위한 조건을 확립하고 종자휴면 유형을 분류하고자 수행되었다. 실험은 헐떡이풀 종자에 저온층적처리($5^{\circ}C$에서 0, 12주)와 고온층적처리($23^{\circ}C$에서 0, 4, 8, 12주 처리 후에 $5^{\circ}C$에서 8주, 다시 $23^{\circ}C$에서 배양함), $GA_3$ (0, 10, 100, 1000 mg/L)처리를 수행하였다. 이렇게 처리한 종자는 무균배지에 소독하여 파종하고 온도 $23^{\circ}C$, 광도 $40{\mu}mo{\ell}{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD, 일장16시간 조건의 배양실에서 배양하였다. 헐떡이풀 종자는 자연상태에서 미숙배인 상태로 탈리되며, 종자 내부로의 수분흡수에 대한 물리적인 장벽은 없었다. 그러나 아무 처리 없이 파종하였을 때 30일이 지나도록 전혀 발아하지 않았다. 따라서 헐떡이풀 종자는 형태적 휴면(MD)과 생리적 휴면(PD)을 가지고 있는 것으로 판단하였다. 저온층적처리(0, 12주) 실험의 최종 발아율은 각각 66.7%, 45.6%로 나타났다. 저온처리는 오히려 발아를 약 3주 정도 지연시켰다. 고온층적처리(0, 4, 8, 12주) 실험에서는 최종 발아율이 각각 13.3%, 24.4%, 20.0%, 51.1%로 나타났다. 헐떡이풀 종자의 배는 상대적 고온에서 발달하였다. $GA_3$ 처리는 종자의 휴면을 극복하고 발아를 촉진하였다. $GA_3$ 처리에서의 최종 발아율은 0, 10, 100, 1000 mg/L 처리구에 대하여 각각 33.3%, 45.0%, 42.5%, 72.5%로 나타났다. 위의 결과를 종합하여 헐떡이풀 종자는 non-deep simple 형태생리적 휴면(MPD)을 가지고 있으며, $GA_3$ 처리를 통해 고온을 대체하고 종자의 휴면을 타파할 수 있었다. 이는 한국에 자생하는 헐떡이풀속의 종자휴면에 대해 처음으로 보고한 논문이다.

인체 상피세포주와 암세포주에 대한 소목 추출물의 증식억제 효과 (Antiproliferative Effects of Caesalpinia sappan Extract on Human Epithelial Cell Line HaCaT and Cancer Cell Lines)

  • Caesalpinia sappan L. (Leguminosae) is an oriental medicinal herb distributed in China and Taiwan, and its heartwood has been traditionally used as an analgesic, a therapy for thrombosis or tumor. This study was to investigate the proliferation and inhibition effects of Caesalpinia sappan extracts against human epithelial cell and cancer cell lines. The methanol extract of dried C. sappan heartwood was evaporated (KS-6), and then sequentially extracted by hexane (KS-01), chloroform (KS-02), ethyl acetate (KS-03), n-butanol (KS-04), and water (KS-05). After 48 hr of exposure, these fractions at a concentration of $50{\mu}g/ml$ significantly increased, and reduced cell proliferation in both human normal epithelial and cancer cell lines. The ethyl acetate fraction (KS-03) among organic solvent fractions was 120.2% of the most proliferation activity ($50{\mu}g/ml$) against HaCaT human epithelial cell. However, fractions from chloroform, butanolic and methanolic extract had 7.2, 28.7 and 20.8% of antiproliferative effect on HaCaT cell, respectively. In cell proliferation effects of C. sappan extract on HeLa, SiHa and C33A human cervical cancer cells, chloroform fraction (KS-2) was the most antiproliferative activity, its antiproliferative rate (dosage, $50{\mu}g/ml$) relative to control was 25.8, 12.2 and 17.4% for SiHa, HeLa and C33A, respectively. The results indicated that the six extract fractions could induce cell cycle stimulate or arrest in some way. Finally, further investigation is needed to assess the molecular mechanisms mediated anticancer activities of this plant.

콩첨가에 따른 증편의 품질과 표면구조 변화 (Changes in Adding Soybean on Quality and surface structure of Korean Rice Cake(Jeung-Pyun))

  • Jeung-Pyun is a traditional fermented Korean food with rice flour, water, sugar, salt and unrefined rice wine(Takju). In order to investigate how the addition of soybean has an influence on Jeung-Pyun fermentation, changing to adding amount of soybean 0%, 5%, l0%. 15%, 20% based on rice weight, we carried out sensory evaluation and measured the physicochemical properties, instrumental characteristics and degree of gelatinization. The specific volume of Jeung-Pyun was increased as the more soybean was added. The pH of Jeung-Pyun batter was decreased as the fermentation time was longer, but it was increased as more soybean was added. The degree of gelatinization of Jeung-Pyun was decreased as storage day was longer, but it was high as more soybean was added compared to control(0%). In sensory evaluation, Jeung-Pyun added 5∼10% of soybean was good generally. Correlation coefficient between hardness and degree of gelatinization was high. In instrumental characteristics, hardness was increased only in control for room temperature storage(20$^{\circ}C$), but at 4$^{\circ}C$ it was less increased than control(0%) as more soybean was added. Cohesiveness was decreased generally as storage day was longer at 4$^{\circ}C$, but it was less decreased as more soybean was added. In brittleness at 4$^{\circ}C$, Jeung-Pyun of adding soybean was higher compared to control. In surface structure of Jeung-Pyun observerd by SEM. air pore size was small and distributed regularly as more soybean added. In conclusion. it can be suggested that the addition of soybean improve the quality of Jeung-Pyun.
