The Effects of CO2 Released from Deep Geological Formations on the Dissolution Process of Galena in Shallow Subsurface Environments (지중저장 이산화탄소의 누출이 천부환경에서 방연석의 용해 과정에 미치는 영향)

  • If $CO_2$ stored for geological sequestration escapes from deep formations and is introduced to shallow aquifers, it dissolves into groundwater, creates acidic environments, and enhance mineral dissolution from rocks and soils. Among these minerals, dissolution and spread of hazardous trace metals can cause environmental problems with detrimental impacts on groundwater quality. This study aims to investigate geochemical effects of $CO_2$ in groundwater on dissolution of galena, the main mineral controlling the mobility of lead. A series of batch experiments are performed with granulated galena in $CO_2$ solutions under various experimental conditions for $CO_2$ concentration and reaction temperature. Results show that dissolution of galena is significantly enhanced under acidic environments so that both of equilibrium concentrations and dissolution rates of lead increase. For thermodynamic analysis on galena dissolution, the apparent rate constants and the activation energy for galena dissolution are calculated by applying rate law to experimental results. The apparent rate constants are $6.71{\times}10^{-8}mol/l{\cdot}sec$ at $15^{\circ}C$, $1.77{\times}10^{-7}mol/l{\cdot}sec$ at $25^{\circ}C$, $3.97{\times}10^{-7}mol/l{\cdot}sec$ at $35^{\circ}C$ and the activation energy is 63.68 kJ/mol. The galena dissolution is suggested to be a chemically controlled surface reaction, and the rate determining step is the dissociation of Pb-S bond of surface complex.

  • The open-chain hexadentate N$_4$, O$_2$ ligands 1,13-bis(2-hydroxybenzyl)-2,5,9,12-tetraazatridecane-tetrahydrochloride (BSATD${\cdot}$4HCl) and 1,14-bis (2-hydroxybenzyl)-2,6,9,12-tetraazatetradecane-tetrahydrochloride (BSATED${\cdot}$ 4HCl) have been synthesized as their tetrahydrochloride salt and characterized by EA, IR, NMR and Mass. Their proton dissociation constants(log$K^{n}_{H}$) and stability constants(log$K_{ML}$) for Cu(II), Ni(II), Co(II), and Zn(II) ions were determined in aqueous solution by potentiometry and compared with those of analogous N$_4$, O$_2$ ligands contain ethylenic spacers or propylenic spacers, which make six-membered chelate rings between the aliphatic nitrogen atoms. Synthesis and characterization of [Cu(BSATD)]ClO$_4$ and [Cu(BSATED)](ClO$_4$)$_2$ complexes are described.

Liquid Phase Adsorption Equilibria of Amines onto High Silica Zeolite, Macroreticular Resin and Granular Activated Carbon (고시리카제올라이트, 거대망상수지 및 입상활성탄에 의한 아민류의 액상흡착평형)

  • Liquid phase adsorption equilibria of amines in an aqueous solution onto high silica zeolite pellets (HSZ), macroreticular resin particles (MR) and granular activated carbon (GAC) were determined using a batch bottle technique at 298K. The isotherm curves of HSZ-amines and GAC-amines indicate the nonlinear relationship of unfavorable adsorption type of HSZ-amines and favourable one of GAC-amines. However the curves of MR-amines represent the linear pattern of an adsorption isotherm. Among various equilibrium isotherms, the three parameters of the Redlich-Peterson equation and the two parameters of the Freundlich equation are found to be the most satisfactory within the range of this study. The two parameters of the Langmuir isotherm were not applicable to the present adsorption systems. The amines were adsorbed on the HSZ, MR and GAC in the following sequence of adsorptivity; aromatic amines > primary amines > secondary amines. The product of the Freundlich constants, k and n, proportionally increased with the boiling point, molar volume and dissociation constants of amines adsorbed on HSZ, MR and GAC.

Measurement and Analysis of the Dynamics of Peptide-Antibody Interactions Using an Ellipsometric Biosensor Based on a Silicon Substrate (실리콘 기판을 사용한 바이오센서와 회전 타원분광계를 이용한 펩타이드-항체 접합의 동특성 측정과 분석)

  • We precisely measured and analyzed the dynamics of peptide-antibody interactions, using an ellipsometric biosensor based on a silicon substrate. To reduce the signal error due to the imperfect flatness of the substrate for extremely low concentrations of peptide, we fabricated the biosensor with a silicon substrate coated with Dextran SAM, instead of a glass prism coated with a thin metallic thin film. At an injection speed of $100{\mu}l/min$ of buffer liquid, we detected the dynamics of antibody-Dextran SAM or peptide-antibody fixed on biosensor, respectively. We detected the dynamics of antibody-Dextran SAM interactions down to a low concentration of 5 ng per liter, and we precisely measured the dynamics of association and dissociation of peptide and antibody down to 100 nM of peptide. We obtained the rate constants for association and dissociation from fitting the data by using deduced dynamical equation. As a result, we obtained an equilibrium constant for dissociation of 97 nM of peptide-antibody complex, which belongs to Class I.

Non-Essential Activation of Co2+ and Zn2+ on Mushroom Tyrosinase: Kinetic and Structural Stability

  • Tyrosinase is a widespread enzyme with great promising capabilities. The Lineweaver-Burk plots of the catecholase reactions showed that the kinetics of mushroom tyrosinase (MT), activated by $Co^{2+}$ and $Zn^{2+}$ at different pHs (6, 7, 8 and 9) obeyed the non-essential activation mode. The binding of metal ions to the enzyme increases the maximum velocity of the enzyme due to an increase in the enzyme catalytic constant ($k_{cat}$). From the kinetic analysis, dissociation constants of the activator from the enzyme-metal ion complex ($K_a$) were obtained as $5{\times}10^4M^{-1}$ and $8.33{\times}10^3M^{-1}$ for $Co^{2+}$ and $Zn^{2+}$ at pH 9 and 6 respectively. The structural analysis of MT through circular dichroism (CD) and intensive fluorescence spectra revealed that the conformational stability of the enzyme in these pHs reaches its maximum value in the presence of each of the two metal ions.

  • The ultrasonic absorption has been measured in aqueous solution of dodecyl pyridinium chloride (DPC) at $20^{\circ}C$ over the frequency range of 0.1${\sim}$90 Mc.The excess absorption was observed only in solutions in which the concentration was higher than the critical micellar concentration(cmc). The mechanism for this feature was attributed to the reaction $M_2\;{\rightleftharpoons}\;M_1\;+\;1.2Cl^-$ Where $M_2$ and $M_1$ and M1 are two types of micelle. The rate constants of the forward and the reverse reactions were $6.6{\times}10^5 sec^{-1}\;and\;2.7{\times}10^11sec^{-1}mol^{-1.2}$respectively. Some kinetic charateristics including the free energy, enthalpy and entropy were calculated.

Influences of Free Fatty Acid on the Albumin Binding of Warfarin and Dansylsarcosine (Warfarin, Dansylsarcosine의 알부민결합에 미치는 유리지방산의 영향)

  • There are some reports about the influences of free fatty acids on the albumin binding of drugs. But they were concerned to the limited free fatty acids, mostly of azapropazone-warfarin bidning site bound drugs and determination of dissociation and association constants by stopped flow technique. These data were not enough to make conculsions for the general tendency of free fatty acid to albumin binding. Therefore the influence of various saturated fatty acids of $C_{10{\sim}20}$, oleic acid and linoleic acid as unsaturated fatty acids to albumin binding of warfarin and dansylsarcosine were studied by equilibrium dialysis. The concentration of free drug was determined by spectrophotometer according to the molar ratios of 0, 0.5, 1, 2 and 4 between free fatty acid and albumin. There were significant increasing in the free durg concentration of warfarin and dansylsarcosine when the molar ratio for capric acid, lauric acid and palmitic acid was 4. The free warfarin concentration was increased significantly at a molar ratio of 4 between oleic acid and albumin. Therefore the albumin binding of durgs can be variated significantly by increased free fatty acid of diabetics and cause to the pharmacokinetic variation between healthy and diabetics.

  • N-alkyl carboxy pyridinium chlorides such as N-lauryl carboxy-pyridinium chloride, N-myristyl carboxy pyridinium chloride, N-cetyl carboxy pyridinium chloride and N-stearyl carboxy pyridinium chloride were synthesized by the reaction of nicotinic acid and isonicotinic acid with long chain alkyl chlorides, and N-alkyl pyrinium carboxylates such as N-lauryl pyridinium carboxylate, N-myristyl pyridinium carboxylate. N-cetyl pyridinium carboxylate and N-stearyl pyridinium carboxylate were prepared from N-alkyl carboxy pyridinium chlorides. These reaction products could be separated by both column chromatogrphy, and paper chromatography, and there dissociation constants of N-alkyl pyridinium carboxylates were found to pKa $1.0{\times}10^{13}{\sim}6,31{\times}10^{14}$.

Label-Free Real-Time Monitoring of Reactions Between Internalin A and Its Antibody by an Oblique-Incidence Reflectivity-Difference Method

  • Surface protein internalin (InlA) is a major virulence factor of the food-borne pathogen L. monocytogenes. It plays an important role in bacteria crossing the host's barrier by specific interaction with the cell adhesion molecule E-cadherin. Study of this protein will help to find better ways to prevent listeriosis. In this study, a monoclonal antibody against InlA was used to detect InlA. The reaction was label-free and monitored in real time with an oblique-incidence reflectivity-difference (OI-RD) technique. The kinetic constants kon and koff and the equilibrium dissociation constant Kd for this reaction were also obtained. These parameters indicate that the antibody is capable of detecting InlA. Additionally, the results also demonstrate the feasibility of using OI-RD for proteomics research and bacteria detection.

Extraction of Water-Soluble Porphyrin and Metalloporphyrins into Acetonitrile by Salting-out

  • A cationic water soluble porphyrin (5,10,15,20-tetrakis (l-methyl-pyridinium-4-yl)porphyrin, $H_2tmpyp^{4+}$) and its metalloporphyrins (MP) were easily extracted into acetonitrile separated by addition of sodium chloride ($4mol\;dm^{-3}$) in the presence of sodium perchlorate, where M denotes $Zn^{2+}$, $Cu^{2+}$, $Co^{3+}$, $Fe^{3+}$, and $Mn^{3+}$ and $P^{2-}$ is porphyrinate ion. The extracted ion-pair complexes were completely dissociated to $[MP(ClO_4)_3]^+$, and $[MP(ClO_4)_2]^{2+}$. The extraction and the dissociation constants were determined by taking into account of the partition constant of sodium perchlorate ($K_D=1.82{\pm}0.01$). The chemical properties of the separated acetonitrile phase as $E_{T(30)}$ and $D_{II,I}$ were determined and compared with other water miscible solvents (acetone, actonitrile, 1,4-dioxane, tetrahydrofuran, 1-propanol and 2-propanol). Furthermore, a sensitive and selective method was proposed for the determination of a subnanogram amount of copper(II) in natural water samples by using the present salting-out method and the porphyrins.
