Effect of Phytoecdysteroid on Disease Incidence, Melting and Economic Characters of the Mulberry Silkworm

  • A study has been made to know the effect of a phytoecdysteroid 'Sampoorna' on uniform maturation of silkworms during spinning and its effect on diseased silkworms infected by major silkworm disease viruses, Bombyx mori nuclear polyhedrosis virus (BmNPV) and Bombyx mori infectious flacherie virus (BmIFV). In the present investigation, the effect of the phytoecdysteroid "Sampoorna" on Grasserie disease caused by BmNPV have shown an average cocoon melting of 11.91% with a disease incidence of 5.83%. The values of 't' test for different treatments of BmNPV indicated low survival rate and cocoon traits were drastically reduced. Another major disease Flacherie caused by BmIFV has shown considerable levels of larval disease incidence (22-32%) and cocoon melting (3-7.67%) with an average melting of 12.95% and 20.24% disease incidence. There is a drastic reduction in survival rate, cocoon yield and other economic traits. The control batches were indicated negligible values for disease incidence and cocoon melting with Sapoorna application and without the inoculation of the two disease-causing viruses. The application of Sampoorna on already infected batches with major pathogens triggered high mortality and disease incidence and melting percentage was also significantly increased with reduced economic traits. Hence, it is suggested that application of Sampoorna in infected batches should be done only in the extreme conditions of rearing. Application of Sampoorna on healthy batches led to uniform maturation and improvement in productivity with the added advantage of better quality cocoons and labour saving.

  • Foot-and-mouth disease (FMD) virus exists in seven serotypes and is known to be a highly contagious disease that is hard to eradicate from the world. The O, A, Asia1 and SAT2 serotypes commonly infected cattle, sheep and goats during 2007~2009 throughout the world. In particular, the outbreak of the Asia1 serotype in China appeared in all areas from 2005 and is still present. Surprisingly, in 2009, Taiwan reported the first outbreak of the type O serotype since 2001. Then type A appeared in China for the first time since the early 1960s. The virus shows a close relationship to the viruses from Southeast Asia suggesting one or more recent introductions into China in the OIE reports. Recently the subtype of A/Iran05 spread to nearby countries exhibiting genomic evolution. The use of molecular epidemiology is an important tool in understanding and consequently controlling the FMD virus. The phylogenetic analysis with VP1 gene was especially useful for molecular epidemiological studies and showed the same pattern which matches with serotype classification. This paper describes basic information about the disease, and the serotype-specific characteristics and evolution to perform molecular epidemiological analysis. Furthermore, we show the importance of the genetic evolution on the FMD serotypes in global surveillance and molecular epidemiology of FMD for outbreak investigation.

Development and Characterization of Monoclonal Antibodies against Nucleoprotein for Diagnosis of Influenza A Virus

  • Influenza, which is a highly contagious disease caused by the influenza A virus, continues to be a major health concern worldwide. Although the accurate and early diagnosis of influenza virus infection is important for controlling the spread of this disease and rapidly initiating antiviral therapy, the current influenza diagnostic kits are limited by their low sensitivity. In this study, we developed several new influenza nucleoprotein (NP)-specific monoclonal antibodies (mAbs) and compared their sensitivity and specificity of those with commercially available anti-NP mAbs. Three mAbs, designated M24.11, M34.3, and M34.33, exhibited higher reactivities to recombinant NPs and A/Puerto Rico/8/1934 (H1N1) viral lysates compared with the commercial mAbs, as assessed using enzyme-linked immunosorbent assays. M34.3 and M34.33 showed higher reactivities with A/California/04/09 (pandemic H1N1) and A/Philippines/2/82 (H3N2) viral lysates than the commercial mAbs. In contrast, M24.11 had marked reactivity with H3N2 but not with pandemic H1N1. Immunofluorescent confocal microscopy showed that the three mAbs effectively detected the presence of influenza virus in lung tissues of mice infected with A/Puerto Rico/8/1934. These results indicate that the newly developed M34.3 and M34.33 mAbs could be useful for the development of influenza diagnostics.

  • In korean society, the chronic life style diseases are increasing. It is caused by the approach of the aged society and the highly increasing income. Accordingly the preventive side of the health promotion and management is very important. In the oriental medicine, the general disease pre-estimate and the management program are necessary. On this point, the sign forecast is very significant in connection with the disease pre-estimate in the preventive disease management side. The sign forecast according to the human shape type diagnosis is mainly the sign of the super-early stage, The difference of the shape type has the difference of the special affinity about the disease. Accordingly we can find the sign forecast from the latent disease in the early stage. In NAE-GYEONG(內經), the theory of "five body form's type" can be pre-estimated the latent tendency and the clue of the disease and the growing tendency of disease in relation to the Differentiation of Syndrome, In the disease pre-estimate side, the graspe and management of the sign forecast from the latent disease will be the part of the new development.

Development of Nucleic Acid Lateral Flow Immunoassay for Rapid and Accurate Detection of Chikungunya Virus in Indonesia

  • Chikungunya fever is an arboviral disease caused by the Chikungunya virus (CHIKV). The disease has similar clinical manifestations with other acute febrile illnesses which complicates differential diagnosis in low-resource settings. We aimed to develop a rapid test for CHIKV detection based on the nucleic acid lateral flow immunoassay technology. The system consists of a primer set that recognizes the E1 region of the CHIKV genome and test strips in an enclosed cassette which are used to detect amplicons labeled with FITC/biotin. Amplification of the viral genome was done using open-source PCR, a low-cost open-source thermal cycler. Assay performance was evaluated using a panel of RNA isolated from patients' blood with confirmed CHIKV (n = 8) and dengue virus (n = 20) infection. The open-source PCR-NALFIA platform had a limit of detection of 10 RNA copies/ml. The assay had a sensitivity and specificity of 100% (95% CI: 67.56% - 100%) and 100% (95% CI: 83.89% - 100%), respectively, compared to reference standards of any positive virus culture on C6/36 cell lines and/or qRT-PCR. Further evaluation of its performance using a larger sample size may provide important data to extend its usefulness, especially its utilization in the peripheral healthcare facilities with scarce resources and outbreak situations.

  • Although there has been a considerable research in psychoneuroendocrinology, particularly in relation to the biology of mood disorders, there has been a paucity of research regarding the effects of psychological factors on endocrine diseases. Of the existing literature, the most research is focused on diabetes mellitus, graves' disease. Especially diabetes appears to be the only endocrine disease that has been the focus of recent research studies. Most of the studies had retrospective designs or several methodological flaw. There has been no consistent results demonstrating that psychological factors affect the onset of diabetes mellitus. Several studies have suggested that psychological factors affect the course of diabetes. A small number of studies showed some evidence that psychological stress was associated with changes in glucose regulation in a subset of diabetic patients. There was also evidence to suggest that some temperament and coping skill influence glycemic control in child, adolescent diabetics. Studies of behavioral or psychosocial intervention on diabetic control have been small and have produced mixed results. There was a few controlled study suggested that stressful life events might be a causal role in Cushing's disease and Graves'disease, but there is no presently sufficient evidence to suggest that psychological factors affect the onset and the course of Cushing's disease and Graves' disease. In future, there is need for a well-designed biopsychosocial research with prospective designs, well defined patient population and control groups to study the relationship between psychological factors and endocrine diseases.

Establishment and application of a solid-phase blocking ELISA method for detection of antibodies against classical swine fever virus

  • Background: Classical swine fever (CSF) is a severe infectious disease of pigs that causes significant economic losses to the swine industry. Objectives: This study developed a solid-phase blocking enzyme-linked immunosorbent assay (spbELISA) method for the specific detection of antibodies against the CSF virus (CSFV) in porcine serum samples. Methods: A spbELISA method was developed based on the recombinant E2 expressed in Escherichia coli. The specificity of this established spbELISA method was evaluated using reference serum samples positive for antibodies against other common infectious diseases. The stability and sensitivity were evaluated using an accelerated thermostability test. Results: The spbELISA successfully detected the antibody levels in swine vaccinated with the C-strain of CSFV. In addition, the detection ability of spbELISA for CSFV antibodies was compared with that of other commercial ELISA kits and validated using an indirect immunofluorescence assay. The results suggested that the spbELISA provides an alternative, stable, and rapid serological detection method suitable for the large-scale screening of CSFV serum antibodies. Conclusions: The spbELISA has practical applications in assessing the vaccination status of large pig herds.

  • Legg-Calve-Perthes (LCP) disease is characterized by idiopathic avascular osteonecrosis of the epiphysis of the femur head. The main factor that plays a role in the etiology of the disease is decreased blood flow to the epiphysis. Many predisposing factors have been suggested in the etiology of LCP disease, and most have varying degrees of effects. Here we present the case of a boy aged 4 years and 10 months with complaints of short stature and a diagnosis of multiple hypophyseal hormone deficiency, in whom LCP disease and difficult birth-related pituitary stalk interruption syndrome were identified by anamnesis. The present case revealed that LCP disease and hypophyseal hormone deficiency could be secondary to difficult birth and that LCP disease could be secondary to insulin-like growth factor 1 deficiency. Additionally, to the best of our knowledge there is no published case on the relation between LCP disease and insulin-like growth factor 1 deficiency. Therefore, we believe that this case is worthy of presentation.

  • Cat scratch disease is a zoonotic disease usually caused by the gram-negative bacterium Bartonella henselae. It is transmitted commonly by scratch or bite from cats or kitten. Cat scratch disease typically affects children and young adults, who develop regional lymphadenopathy. In contrast, in immunocompromised hosts, bacteremia may occur, bacillary angiomatosis and bacillary peliosis hepatitis or splenitis are the most common manifestations. Bartonella henselae was detected in three of thirty veterinarians and Bartonella clarridgeiae was detected in one of thirty veterinarians by a novel nested polymerase chain reaction. Cat scratch disease will not be neglected, and it needs continuous studies as well as observation and prevention of this disease.

Alzheimer's disease recognition from spontaneous speech using large language models

  • We propose a method to automatically predict Alzheimer's disease from speech data using the ChatGPT large language model. Alzheimer's disease patients often exhibit distinctive characteristics when describing images, such as difficulties in recalling words, grammar errors, repetitive language, and incoherent narratives. For prediction, we initially employ a speech recognition system to transcribe participants' speech into text. We then gather opinions by inputting the transcribed text into ChatGPT as well as a prompt designed to solicit fluency evaluations. Subsequently, we extract embeddings from the speech, text, and opinions by the pretrained models. Finally, we use a classifier consisting of transformer blocks and linear layers to identify participants with this type of dementia. Experiments are conducted using the extensively used ADReSSo dataset. The results yield a maximum accuracy of 87.3% when speech, text, and opinions are used in conjunction. This finding suggests the potential of leveraging evaluation feedback from language models to address challenges in Alzheimer's disease recognition.
