수입식품에 대한 인식도 및 분별력과의 상관성 (Recognition Level of Imported Food and Its Correlation with Discrimination Ability)

  • This study used questionnaires to investigate the safety awareness for imported foods by 365 male and female adults in Taejon. The results of the study were as follow : By factor analysis, the subjects' behaviors and awareness of the imported food was grouped into 3 factors such as 'health and quality factor', 'purchasing factor' and 'contamination factor'. 'Health and quality factor' and 'purchasing factor' were not recognized negatively by the subjects, moreover' contamination factor' was recognized very highly. The subjects' concern and worry about the imported food was also very high. The marital status, education level, nutriton knowledge adn recognition level of contamination by pesticides and heavy metals of foods partially affected the recognitio level of imported foods. The major selection criteria of imported food were distribution period(36.3%), price(28.8%) and purchasing experience(17.3%). The imported food mean discrimination score was 8.4±3.1 out of 13. The worst discriminatio score was red pepper. The subjects' experiences with imported foods selection affected the most instead of education level or nutrition knowledge. The higher discrimination score group more negatively recognized imported food and contamination recognition level was higher whereas the lowerdiscrimination score group more positively recognized the purchasing frequency and with to buy more easily. But both groups desired to reinforce contamination control. The better discrimination score of imported food pooring recognized sanitation concerns(p<0.05), quality(p<0.05), cooking convenience(p<0.01), desire for more variety(p<0.05), and the higher contamination recognition level(p<0.05) and desire to reinforce contamination control(p<0.01).

배관 내부 방사능 오염도 측정용 ZnS(Ag)/BC-408 phoswich 검출기 개발 (Development of the ZnS(Ag)/BC-408 phoswich detector for monitoring radioactive contamination inside pipes)

  • 원자력 시설의 해체 시 발생되는 다양한 종류의 폐기물 중에서 배관 내부의 알파/베타선 오염 특성을 직접 평가하기 위해서는 소형의 방사선 검출 시스템이 요구된다. 본 연구에서는 배관 내부의 방사능 오염도를 직접 측정할 수 있는 장비 개발의 일환으로서 기존의 phoswich 검출기와 그 형태가 다른 ZnS(Ag)/플라스틱섬광체 조합의 알파/베타선 동시측정용 phoswich 검출기를 고안하였으며 섬광체 크기 및 검출 시스템의 최적 요건을 실험적으로 확인하고 알파/베타선 신호 분리도를 평가하였다. 그 결과 배관 내부의 오염도 측정에 적합한 phoswich 검출기의 최적 요건 및 배관 내부의 적용 가능성을 확인하였다.

  • 배관 내부의 방사성 오염도를 측정하기 위한 ZnS(Ag)/플라스틱섬광체 조합의 알파/베타선 동시측정용 phoswich 검출기를 개발하였다. 알파/베타선 동시측정용 phoswich 검출기의 오염위치에 따른 검출 성능을 PSD (Pulse shape discrimination) 방법을 이용하여 평가하였다. 또한, 검출기를 방사성 오염물질로부터 보호하기 위한 오염방지용 필름에 대한 방사선 감쇄 정도를 실험적으로 평가하였다. PSD 방법으로 알파/베타선 분리 정도를 측정한 결과 충분히 알파와 베타선이 분리되었으며 오염방지용 필름의 적용 가능성을 확인하였다.

Choosing Optimal STR Markers for Quality Assurance of Distributed Biomaterials in Biobanking

  • The quality assurance (QA) is of utmost importance in biobanks when archived biomaterials are distributed to biomedical researchers. For sample authentication and cross-contamination detection, the two fundamental elements of QA, STR genotyping is usually utilized. However, the incorporated number of STR markers is highly redundant for biobanking purposes, resulting in time and cost inefficiency. An index to measure the cross-contamination detection capability of an STR marker, the mixture probability (MP), was developed. MP as well as other forensic parameters for STR markers was validated using STR genotyping data on 2328 normal Koreans with the commercial AmpFlSTR kit. For Koreans, 7 STR marker (D2S1338, FGA, D18S51, D8S1179, D13S317, D21S11, vWA) set was sufficient to provide discrimination power of ${\sim}10^{-10}$ and cross-contamination detection probability of ${sim}1$. Interestingly, similar marker sets were obtained from African Americans, Caucasian Americans, and Hispanic Americans under the same level of discrimination power. Only a small subset of commonly used STR markers is sufficient for QA purposes in biobanks. A procedure for selecting optimal STR markers is outlined using STR genotyping results from normal Korean population.

전신계측기를 이용한 원전종사자 방사성오염 위치확인과 내부방사능 측정개선에 관한 연구 (A Study on the Verification and Improvement to Locate and Determine the Radioactive Contamination Using a Whole Body Counter)

  • 국내 원전에서는 원전종사자의 내부피폭 방사능을 측정하기위해 전신계측기를 이용하고 있다. 이 전신계측기는 Sodium Iodide를 이용한 섬광검출기로서 짧은 시간에 종사자가 보유한 방사성핵종과 방사능을 측정하는 기능을 가지고 있다. 그런데 종사자의 신체표면에 부착된 오염과 내부에 침적된 오염을 구분하지 못하기 때문에 방사선계측 과정에서 종종 오류를 범할 가능성이 있으며, 이 경우 내부피폭선량은 매우 보수적으로 과대평가된다. 이러한 문제점을 개선하고자 종사자의 인체 내부와 외부 표면오염을 구분하고, 보다 체계적으로 오염부위를 확인할 수 있도록 전신계측기와 인체모형 팬텀을 이용한 방사능 계측실험을 수행하였다. 또한 원전에서 발생하는 주요 핵종의 신체내 침적위치를 고려하여 전신계측기의 최적 방사능 측정모드를 결정하는 실험을 수행하였다. 이러한 방사능 측정 실험결과를 근거로 원전종사자의 내부방사능 측정과 선량평가 절차를 개선하였다. 이에따라 보다 정확한 전신계측프로그램의 적용으로 내부피폭선량의 보수적 평가를 방지할 수 있을 것으로 기대된다.

Comparison of an Immunochromatographic Assay Kit with DAS-ELISA for Large-Scale Diagnosis and Molecular Discrimination of Satsuma Dwarf Virus Collected from Citrus Orchards

  • Satsuma dwarf virus (SDV) seriously damages citrus production by reducing the quality and yield of fruit. To avoid contamination with SDV, mother trees are checked to be SDV-free in advance of nursery tree distribution. In this study, we compared an immunochromatographic assay (ICA) kit with double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) for large-scale diagnosis of SDV in orchardgrown trees in Shizuoka Prefecture, Japan. The two methods gave conflicting results for 11 of 1,705 samples, all of which were negative by DAS-ELISA but positive by ICA. The samples scored as positive by either DAS-ELISA or ICA were analyzed by reverse transcription polymerase chain reaction and all were confirmed to be positive. These results validate the use of ICA as a screening method for large-scale diagnosis. Strain discrimination revealed that 16 of 22 isolates belonged to SDV, while citrus mosaic virus (CiMV) infection only and co-infection (SDV and CiMV) were in a minority.

SELECTION OF VISIBLE/NIR WAVELENGTHS FOR CHARACTERIZING FECAL AND INGESTA CONTAMINATION OF POULTRY CARCASSES

  • Ingests and fecal contamination on a poultry carcass is a food safety hazard due to potential microbiological contamination. A visible/near-infrared (NIR) spectrometer was used to discriminate among pure ingesta and fecal material, breast skin contaminated with ingesta or fecal material and uncontaminated breast skin. Birds were fed isocaloric diets formulated with either maize, mile, or wheat and soybean meal for protein requirements. Following completion of the feeding period (14 days), the birds were humanely processed and eviscerated to obtain ingests from the crop or proventriculus and feces from the duodenum, ceca, and colon portion of the digestive tract. Pure feces and ingesta, breast skin, and contaminated breast skin were scanned from 400 to 2500 nm and analyzed from 400 to 900 nm. Principal component analysis (PCA) of reflectance spectra was used to discriminate between contaminates and uncontaminated breast skin. Results indicate that visible (400 to 760 nm) and NIR 760-900 nm spectra can detect contaminates. From PCA analysis, key wavelengths were identified for discrimination of uncontaminated skin from contaminates based the evaluation of loadings weights.

폐금속광산지역 토양오염정도와 혈 중 납 노출 수준의 상관성 (Association between Soil Contamination and Blood Lead Exposure Level in Areas around Abandoned Metal Mines)

  • Background: Abandoned metal mines are classified as vulnerable areas with the highest level of soil contamination among risk regions. People living near abandoned metal mines are at increased risk of exposure to toxic metals. Objectives: This study aimed to evaluate the correlation between soil contamination levels in areas around abandoned metal mine and the blood lead levels of local residents. Moreover, we assess the possibility of using soil contamination levels as a predictive indicator for human exposure level. Methods: Data from the Survey of Residents around Abandoned Metal Mines (2013~2017, n=4,421) and Investigation of Soil Pollution in Abandoned Metal Mines (2000~2011) were used. A random coefficient model was conducted for estimation of the lower level (micro data) of the local resident unit and the upper level (macro data) of the abandoned metal mine unit. Through a fitted model, the variation of blood lead levels among abandoned metal mines was confirmed and the effect of the operationally defined soil contamination level was estimated. Results: Among the total variation in blood lead levels, the variation between abandoned mines was 18.6%, and the variation determined by the upper-level factors such as soil contamination and water contamination was 8.1%, which was statistically significant respectively. There was also a statistically significant difference in the least square mean of blood lead concentration according to the level of soil contamination (p=0.047, low: 2.32 ㎍/dL, middle: 2.38 ㎍/dL, high: 2.59 ㎍/dL). Conclusions: The blood lead concentration of residents living near abandoned metal mines was significantly correlated with the level of soil contamination. Therefore, in biomonitoring for vulnerable areas, operationally defined soil contamination can be used as a predictor for human exposure level to hazardous substances and discrimination of high-risk abandoned metal mines.

Development of HRM Markers for Discrimination of Pyogo (Lentinula edodes) Cultivars Sanjo 701 and Chamaram

  • Pyogo (Shiitake, Lentinula edodes) is one of the most important edible mushrooms because of its outstanding nutritive and medicinal value. In the registration and protection procedure for newly developed mushroom cultivars, the application of molecular markers that can supplement the morphological characteristic-based distinction has been strongly requested. Sanjo 701 and Chamaram, newly developed at the Federation Forest Mushroom Research Center of Korea, have been characterized as innovative cultivars suitable for customer demands because of their high yields and cultivation rates. However, no technical tools can protect the rights to these important cultivars. In this study, using comparative genomic information from 23 commercially available pyogo cultivars, we identified single nucleotide polymorphisms (SNPs) that accurately differentiated Sanjo701 and Chamaram from the other cultivars. We also developed high-resolution melting analysis (HRM)-based SNP markers that discriminate among the tested 23 pyogo cultivars. The developed SNP markers can be utilized for rapid, accurate identification of pyogo cultivars with low genetic diversity and to prevent cultivar contamination caused by illegally distributed inocula. In addition, these markers can serve as a crucial scientific basis for securing the right to conserve new cultivars in international markets.

Plastic scintillator beta ray scanner for in-situ discrimination of beta ray and gamma ray radioactivity in soil

  • A beta ray scanner was proposed for in-situ discrimination of beta and gamma ray radioactivity. This scanner is based on the principle that gamma and beta rays experience different changes in detection efficiency in scintillators with different geometries, especially with regard to the scintillator thickness. The ratios of the counting rates of gamma rays (Rgamma), beta rays (Rbeta), and sample measurements (Rtotal) in a thick scintillator to those in a thin one are reported. The parameter Xthick, which represents the counting rate contributed by beta rays to the total counting rate in the thick scintillator, was derived as a function of those ratios. The values of Rgamma and Rbeta for 60Co and 90Sr sources were estimated as 3.2 ± 0.057 and 0.99 ± 0.0049, respectively. The estimated beta ray contributions had relative standard deviations of 2.05-4.96%. The estimated range of the beta rays emitted from 90Sr was 19 mm as per the Monte Carlo N-Particle simulation, and this value was experimentally verified. Homogeneous and surface contaminations of 60Co and 90Sr-90Y were simulated for application of the proposed method. The counting rate contributed by the beta rays was derived and found to be proportional to the concentration of 90Sr-90Y contamination.
