Quinetides: diverse posttranslational modified peptides of ribonuclease-like storage protein from Panax quinquefolius as markers for differentiating ginseng species

  • Background: Peptides have diverse and important physiological roles in plants and are ideal markers for species identification. It is unclear whether there are specific peptides in Panax quinquefolius L. (PQ). The aims of this study were to identify Quinetides, a series of diverse posttranslational modified native peptides of the ribonuclease-like storage protein (ginseng major protein), from PQ to explore novel peptide markers and develop a new method to distinguish PQ from Panax ginseng. Methods: We used different fragmentation modes in the LTQ Orbitrap analysis to identify the enriched Quinetide targets of PQ, and we discovered Quinetide markers of PQ and P. ginseng using ultrahigh-performance liquid chromatography-quadrupole time-of-flight mass spectrometry analysis. These "peptide markers" were validated by simultaneously monitoring Rf and F11 as standard ginsenosides. Results: We discovered 100 Quinetides of PQ with various post-translational modifications (PTMs), including a series of glycopeptides, all of which originated from the protein ginseng major protein. We effectively distinguished PQ from P. ginseng using new "peptide markers." Four unique peptides (Quinetides TP6 and TP7 as markers of PQ and Quinetides TP8 and TP9 as markers of P. ginseng) and their associated glycosylation products were discovered in PQ and P. ginseng. Conclusion: We provide specific information on PQ peptides and propose the clinical application of peptide markers to distinguish PQ from P. ginseng.

Effect of Next-Generation Exome Sequencing Depth for Discovery of Diagnostic Variants

  • Sequencing depth, which is directly related to the cost and time required for the generation, processing, and maintenance of next-generation sequencing data, is an important factor in the practical utilization of such data in clinical fields. Unfortunately, identifying an exome sequencing depth adequate for clinical use is a challenge that has not been addressed extensively. Here, we investigate the effect of exome sequencing depth on the discovery of sequence variants for clinical use. Toward this, we sequenced ten germ-line blood samples from breast cancer patients on the Illumina platform GAII(x) at a high depth of ${\sim}200{\times}$. We observed that most function-related diverse variants in the human exonic regions could be detected at a sequencing depth of $120{\times}$. Furthermore, investigation using a diagnostic gene set showed that the number of clinical variants identified using exome sequencing reached a plateau at an average sequencing depth of about $120{\times}$. Moreover, the phenomena were consistent across the breast cancer samples.

  • 기존에 이동 Ad Hoc On-demand 방식의 대표적인 DSR과 AODV 프로토콜 라우팅 알고리즘은 단지 경로의 손실이 발생할 경우에만 경로 발견을 시작하며, 단절된 경로를 발견하고 새로운 경로 확립하는데 막대한 경비와 시간이 소요된다. 본 논문에서는 기존의 대표적인 DSR과 AODV 프로토콜 라우팅 알고리즘에서 제안하는 견고한 On-demand 방식의 사전 활성화 경로 선택과 관리유지 알고리즘의 추가시키는 측면을 연구한다. 본 연구의 핵심 아이디어는 수신되는 패킷의 신호파워 세기가 손실되기 전의 최적 임계치 신호파워 세기까지 근접하게 되면 경로는 손실될 경향이 높다고 간주하는 것과 수신되는 패킷의 신호파워 세기가 최적 임계치 이하로 떨어졌을 경우, 사전경고 패킷을 발생하는 것이다. 사전경고 패킷을 발생 후에, 송신 노드는 계속적으로 패킷이 전송하는 동안 사전에 경로 발견을 시작하기 때문에, 모든 경로의 단절에 대한 잠재적인 가능성을 피할 수 있다. 성능평가 결과에 의하면, 제안하는 견고한 On-demand 방식의 사전 활성화 경로 선택과 관리유지 알고리즘은 기존의 DSR과 AODV 프로토콜보다 패킷 전달율과 평균 지연시간 그리고 오버헤드 측면에서 성능이 우수한 경향을 나타낸다.

Liquid chromatography-tandem mass spectrometric analysis of oleracone D and its application to pharmacokinetic study in mice

  • We have demonstrated a sensitive analytical method of measuring oleracone D in mouse plasma using a liquid chromatography-tandem mass spectrometry (LC-MS/MS). Oleracone D and oleracone F (internal standard) in mouse plasma samples were processed using a liquid-liquid extraction method with methyl tertbutyl ether, resulting in high and reproducible extraction recovery (80.19-82.49 %). No interfering peaks around the peak elution time of oleracone D and oleracone F were observed. The standard calibration curves for oleracone D ranged from 0.5 to 100 ng/mL and were linear with r2 of 0.992. The inter- and intra-day accuracy and precision and the stability fell within the acceptance criteria. The pharmacokinetics of oleracone D following intravenous and oral administration of oleracone D at doses of 5 mg/kg and 30 mg/kg, respectively, were investigated. When oleracone D was intravenously injected, it had first-order elimination kinetics with high clearance and volume of distribution values. The absolute oral bioavailability of this compound was calculated as 0.95 %, with multi-exponential kinetics. The low aqueous solubility and a high oral dose of oleracone D may explain the different elimination kinetics of oleracone D between intravenous and oral administration. Collectively, this newly developed sensitive LC-MS/MS method of oleracone D could be successfully utilized for investigating the pharmacokinetic properties of this compound and could be used in future studies for the lead optimization and biopharmaceutic investigation of oleracone D.

  • The 'discovery' or 'conquest' of the New World in 1492 was the starting point of world history that irrevocably changed the fate of the Latin American continent. The global stream known as 'Columbian Exchange', which was the widespread inter-continental contacts, inter-civilizational conflicts, or bilateral communication, has rendered multifarious effects throughout many historical periods up to the very contemporary time. The propagation of Catholicism initiated along with this 'discovery' transformed Latin America of nowadays in the region that has the biggest Catholic population in the world. The previous studies in Korea regarding Latin American Catholicism has focused on the spread of Catholicism in relation to the European colonization, rather than analyzing the concrete and detailed ways in which Catholicism exerted tremendous influences in the whole continent. They were less attentive to various historical contexts in which the diffusion of Catholicism differed greatly according to cultural landscapes and political specificities. Thus, this essay attempts to examine the diffusion of Catholicism from the perspective of confrontation between royal authorities and the power of church. The essay points out that the royal communities and institutional authorities which facilitated the intial process of Catholic evangelization maintained antagonistic relationship with ecclesial powers. By delving into the gradual transformation of church systems, it reveals that Catholicism in Latin America became a major field for conquerors in power to attain economic and political dominance. And unlike the initial submission and hospitality, the religious convert of the indigenous people attested to the violent inhumanity and opposition. Therefore, the essay aims to pave a clearer way to the understanding of complicated dynamics and conflicts between Catholicism in Latin America and the establishment of Spanish colonization.

무선네트워크에서 존 마스터 기반의 링크 안정성 해석 (Analysis of Link Stability Based on Zone Master for Wireless Networks)

  • 무선 네트워크에서 토폴로지의 빈번한 변화로 노드 간 링크 단절과 경로재설정이 발생하여 네트워크 내에 제어메시지 과부하와 같은 문제점들이 발생한다. 본 연구에서는 링크 단절과 제어 메시지 과부하와 같은 문제점을 해결하기 위하여 무선 네트워크 환경에서 주변 노드 탐색과정, 경로 탐색과정, 경로 관리과정의 3단계로 경로 설정을 수행하고 경로 관리 과정에서 존 마스터가 수집한 라우팅 테이블의 정보를 이용하여 링크 안정성 값을 산출한다. 그리고 산출 된 링크 값을 존 마스터가 모니터링 하여 임계 값 이하로 되면 링크 단절로 예측하고 해당 송수신 노드에게 경로 재설정을 수행하게 된다. 제안한 기법은 이동 노드 수 변화에 따른 데이터 처리량, 평균 경로 설정 시간 및 이동 노드의 속도 변화에 따른 데이터 처리량에서 기존의 OLSR 프로토콜보다 성능 향상을 나타내었다.

  • 오랜 기간과 막대한 비용에도 성공 확률이 낮은 제약·바이오 산업의 생산성 위기를 해결하기 위한 전략으로 전 세계적으로 인공지능과 빅데이터를 활용하려는 사례가 증가하고 있고 가시적인 성과가 나오고 있지만 국내에서는 신약연구에 인공지능 플랫폼 도입에는 관망하는 상황이다. 본 연구는 신약개발을 지원하는 인공지능 플랫폼의 사용과 확산을 촉진하기 위해 도입 및 수용을 견인하는 지각된 가치와 변화에 대한 저항, 수용의도 관계를 검증할 가치기반수용모형과 혁신저항모형 결합 연구모형을 제시하였다. 인공지능 신약개발 플랫폼 사용의도의 연구모형은 지각된 편익으로 유용성, 지식풍부성을, 지각된 희생으로 복잡성, 알고리즘 불투명성을 채택하였고 지각된 가치, 혁신저항의 매개변수로 구성되었다. 실증 결과, 유용성, 지식풍부성, 복잡성, 인공지능 알고리즘의 불투명성이 지각된 가치에 유의미한 영향을 미치고, 유용성, 지식풍부성, 알고리즘의 불투명성, 시험가능성, 인공지능 기술지원환경이 플랫폼 도입에 따른 혁신저항에 유의미한 영향을 미치는 것으로 나타났다.

Vascular rinsing and chilling carcasses improves meat quality and food safety: a review

  • Rinse & Chill® technology (RCT) entails rinsing the vasculature using a chilled isotonic solution (3℃; 98.5% water and a blend of dextrose, maltose, and sodium phosphates) to rinse out the residual blood from the carcass. Infusion of pre-chilled solutions into intact animal carcasses immediately upon exsanguination is advantageous in terms of lowering the internal muscle temperature and accelerating chilling. This technology is primarily used for purposes of effective blood removal, favorable pH decline, and efficient carcass chilling, all of which improve meat quality and safety. Although RCT solution contains some substrates, the pre-rigor muscle is still physiologically active at the time of early postmortem and vascular rinsing. Consequently, these substrates are fully metabolized by the muscle, leaving no detectable residues in meat. The technology has been commercially approved and in continuous use since 2000 in the United States and since 1997 in Australia. As of January 2022, 23 plants have implemented RCT among the 5 countries (Australia, US, Canada, New Zealand, and Japan) that have evaluated and approved RCT. All plants are operating under sound Sanitation Standard Operation Procedures (SSOP) and a sound Hazard Analysis Critical Control Point (HACCP) program. No food safety issues have been reported associated with the use of this technology. RCT has been adapted by the meat industry to improve product safety and meat quality while improving economic performance. Therefore, this review summarizes highlights of how RCT technically works on a variety of animal types (beef, bison, pork, and lamb).

Can Artificial Intelligence Boost Developing Electrocatalysts for Efficient Water Splitting to Produce Green Hydrogen?

  • Water electrolysis holds great potential as a method for producing renewable hydrogen fuel at large-scale, and to replace the fossil fuels responsible for greenhouse gases emissions and global climate change. To reduce the cost of hydrogen and make it competitive against fossil fuels, the efficiency of green hydrogen production should be maximized. This requires superior electrocatalysts to reduce the reaction energy barriers. The development of catalytic materials has mostly relied on empirical, trial-and-error methods because of the complicated, multidimensional, and dynamic nature of catalysis, requiring significant time and effort to find optimized multicomponent catalysts under a variety of reaction conditions. The ultimate goal for all researchers in the materials science and engineering field is the rational and efficient design of materials with desired performance. Discovering and understanding new catalysts with desired properties is at the heart of materials science research. This process can benefit from machine learning (ML), given the complex nature of catalytic reactions and vast range of candidate materials. This review summarizes recent achievements in catalysts discovery for the hydrogen evolution reaction (HER) and oxygen evolution reaction (OER). The basic concepts of ML algorithms and practical guides for materials scientists are also demonstrated. The challenges and strategies of applying ML are discussed, which should be collaboratively addressed by materials scientists and ML communities. The ultimate integration of ML in catalyst development is expected to accelerate the design, discovery, optimization, and interpretation of superior electrocatalysts, to realize a carbon-free ecosystem based on green hydrogen.

패션 큐레이션(curation) 쇼핑에 영향을 미치는 컨텍스트 특성과 소비자 특성에 관한 연구 (A Study on the Context Characteristics and Consumer Characteristics Affecting Fashion Curation Shopping)

  • This study attempted to analyze the context characteristics and consumer characteristics that affect fashion curation shopping. The data used for this study were 223 questionnaires targeting male and female college students in their 20s in Busan and South Gyeongsang Province who had had the curated shopping experience in the latest three months. The SPSS program was used for the data analysis, and a reliability measurement, factor analysis, multiple regression analysis, T-test, and one-way ANOVA were conducted. The results were as follows. First, fashion curation shopping exhibited three factors: product subscription, marketing use, and product recommendation shopping. Furthermore, the context characteristics had sub concepts of five factors: selection, sharing, experience, discovery, and storage. Second, the context characteristics (selection, sharing, experience, discovery, and storage) had a significant influence on product subscription, marketing use, and product recommendation, which belong to the curation shopping category. Third, the fashion consumers' price sensitivity, trend sensitivity, and product knowledge had a deep impact on the marketing use and product recommendation. Fourth, there was no difference in the fashion curation shopping by male and female consumers and the average monthly fashion shopping frequency, and there were differences in shopping cost and time. This study can analyze the context and consumer characteristics that affect fashion curation shopping to establish an efficient fashion curation shopping system in practical terms. Additionally, academically, it can be proposed as basic data on the development of measurement tools for analyzing consumer behavior that prefers fashion curation shopping.
