Water Quality Assessment in Small Streams by Epilithic Diatoms and DAIpo (부착조류와 유기오탁지수에 의한 소하천의 수질평가)

  • In this study, a method for river water quality evaluation by algal examination was investigated. For development of this method, algae living in small rural streams in Gwangcheon-Cheon were collected and observed by microscope. Samples were collected at six stations from May 2008 to May 2009. To elucidate the relationship between pollution and the algal species, diatom grasps were collected from the streams and used to determine the water quality grade. The numerical EC value was compared with the value of DAIpo (Diatom Assemblage Index of Organic Water Pollution), and the results showed a tendency to a mutually inverse relationship. Further, the numerical BOD and COD values were compared to the DAIpo value. The results showed that, various diatoms reside in Gwangcheon-Cheon, and water quality was found to be second-grade at all the examined points. These results indicate pollution of streams by livestock wastewater; however continuous monitoring will be necessary to more definitely determine the reason for the Gwangcheon-Cheon pollution.

Species Composition of the Epiphytic Diatoms on the Leaf Tissues of Three Zostera Species Distributed on the Southern Coast of Korea

  • The epiphytic diatom flora living on leaf tissues of Zostera marina, Z. japonica and Z. caespitosa at three study sites (Dadae, Gabae, and Jangmok) located in Geoje Island has been examined from July 2001 to March 2002 bimonthly. Total 81 species in 26 genera of diatoms have been observed on leaf tissues of the 3 Zostera species throughout the present study. Cocconeis spp. were predominant on Z. japonica, whereas Navicula spp. and Nitzschia spp. were predominant on Z. marina and Z. caespitosa. The community structure of the diatom assemblages varied according to the seagrass species. Species compositions of epiphytic diatoms appear to be closely related to morphology and lifespan of seagrasses.

Isolation and Physiological Characterization of a Novel Algicidal Virus Infecting the Marine Diatom Skeletonema costatum

  • Diatoms are a major component of the biological community, serving as the principal primary producers in the food web and sustaining oxygen levels in aquatic environments. Among marine planktonic diatoms, the cosmopolitan Skeletonema costatum is one of the most abundant and widespread species in the world's oceans. Here, we report the basic characteristics of a new diatom-infecting S. costatum virus (ScosV) isolated from Jaran Bay, Korea, in June 2008. ScosV is a polyhedral virus (45-50 nm in diameter) that propagates in the cytoplasm of host cells and causes lysis of S. costatum cultures. The infectivity of ScosV was determined to be strain- rather than species-specific, similar to other algal viruses. The burst size and latent period were roughly estimated at 90-250 infectious units/cell and <48 h, respectively.

Assessment of Water Quality in the Sum-river and the Dal-stream using Epilithic Diatom-based Indices (부착규조류를 이용한 달천과 섬강의 생물학적 수질평가)

  • To evaluate the biological water quality, diatom-based biological indices (DAIpo and TDI) were examined in the Sum-River and the Dal-stream. Having BOD concentrations below $2mg{\cdot}L^{-1}$ in most sites, water quality of both rivers was good. The DAIpo and TDI values ranged from 29.8 to 91.4 and from 38.7 to 93.0 respectively. From the biological water quality assessment, DAIpo and TDI for both rivers displayed fair to fairly poor water quality levels. However, the two indices showed more polluted conditions than expected from the BOD vlaues. In addition, DAIpo, having wider range of differences, appears to be more sensitive to the change in water quality when compared to TDI values. Statistical analysis using principal component analysis showed that Nitzschia palea and Diatoma vulgare might not be the appropriate indicators due to their low correlations with other indicators.

Description of an unrecorded diatom Fragilaria saxoplanctonica Lange-Bertalot & Ulrich (Bacillariophyceae) from Paldang Reservoir in Korea

  • Diatoms are unicellular eukaryotic microalgae, and they are highly diversified in aquatic environments. We describe an unrecorded diatom species Fragilaria saxoplanctonica Lange-Bertalot & Ulrich (Bacillariophyceae) collected from Paldang Reservoir, Korea, on 4 April 2022. The valve was needle shaped and narrowly rectangular, its ends were rounded, and no spines were found on the outline of their valves. The valve was 67.2-70.2㎛ in length and 1.4-2.9㎛ in width. The ratio of width-to-length was 1 : 23.2-50.1. The pattern of striation was alternate or opposite, and the number of striae in 10㎛ was 24-26. Molecular comparisons of the 18S rDNA and rbcL sequences showed that it belonged to the genus Fragilaria. These morphological and phylogenetic results confirmed that our species was F. saxoplanctonica, and it was the first record in Korea.

The biofilm removal effect of MnO2-diatom microbubbler from the dental prosthetic surfaces: In vitro study (치과 보철 재료 표면에서 MnO2-diatom microbubbler의 세균막 제거 효과 연구: In vitro study)

  • Purpose: The aim of this study is to evaluate the effectiveness of MnO2-diatom microbubbler (DM) on the surface of prosthetic materials as a mouthwash by comparing the biofilm removal effect with those previously used as a mouthwash in dental clinic. Materials and methods: DM was fabricated by doping manganese dioxide nanosheets to the diatom cylinder surface. Scanning electron microscopy (SEM) was used to observe the morphology of DM and to analyze the composition of doped MnO2. Stereomicroscope was used to observe the reaction of DM in 3% hydrogen peroxide. Non-precious metal alloys, zirconia and resin specimens were prepared to evaluate the effect of biofilm removal on the surface of prosthetic materials. And then Streptococcus mutans and Porphyromonas gingivalis biofilms were formed on the specimens. When 3% hydrogen peroxide solution and DM were treated on the biofilms, the decontamination effect was compared with chlorhexidine gluconate and 3% hydrogen peroxide solution by crystal violet staining. Results: Manganese dioxide was found on the surface of the diatom cylinder, and it was found to produce bubble of oxygen gas when added to 3% hydrogen peroxide. For all materials used in the experiments, biofilms of the DM-treated groups got effectively removed compared to the groups used with chlorhexidine gluconate or 3% hydrogen peroxide alone. Conclusion: MnO2-diatom microbubbler can remove bacterial membranes on the surface of prosthetic materials more effectively than conventional mouthwashes.

Analysis of the Epilithic Diatom Community and Comparison of Water Quality in the Kumho River (금호강의 부착규조 군집분석과 수질 변화비교)

  • In order to assess the water quality of Kumho river using Diatom Assemblage Index to organic water pollution (DAIpo) and to campare from the previous water quality, 5 sites in Kumho river were selected and assessed total of 6 times, every April and September from April 2010 to September 2012. As a result, 116 taxa were found in 2 Order, 3 Suborder, 7 Families, 26 Genera, 103 species and 13 varieties. Saprophilous species of 10 dominant species including Achnanthes minutissima, were increased as going to downstream sites. Correlation coefficients (r) between epilithic diatom communities and physicochemical factors were from - 0.62 to 0.71. Correlation coefficients (r) between TN and diatom indices (DAIpo, TDI) were 0.57 and 0.65, respectively. Therefore, epilithic diatom communities were greatly influenced by TN. The Correlation between DAIpo and TDI measured to be high in correlation coefficient (r=0.62) from the result of correlation analysis. Mean of DAIpo values ranged between 37.24~74.98 and decreased as going to downstream sites. Saprobic level was estimated as ${\beta}$-oligosaprobic at st. 1, ${\alpha}$-oligosaprobic at st. 2 and ${\beta}$-mesosaprobic at st. 3. But st. 4 and 5 which were ${\alpha}$-mesosaprobic and polysaprobic in the previous results, were improved as ${\beta}$-mesosaprobic. The RPId, general assessment of the water quality using DAIpo, was 52.27, which means the water quality of Kumho river was middle level.

Relative Immigration Activity of Epilithic Diatom in Sum River : Comparison of Natural and Artificial Substrate (섬강수계 부착규조의 유입능 특성: 자연기질과 인공기질의 비교)

  • Effects of substrates on the relative immigration activities (RIAs) of epilithic diatoms were examined in Sum River, a tributary of South Han River, Korea. Two hundreds of tile substrates coated with 5% agar were deposited at seven study stations (30 tiles per site) for one month from March 7 to April 7, 2008. Water sampling, physico-chemical factor measurement, and diatom analysis were performed in the field and laboratory. Over the study, major epilithic diatom species were Nitzschia amphibia, Navicula subminuscula, Cymbella minuta in both the natural and artificial substrates. Two dominant species, Nitzschia amphibia and Cymbella minuta often found at seven stations. Among all observed taxa, Nav. subminuscula and Caloneis silicula showed the highest RIA, with the value over 15 throughout the study. Regarding the algal morphology, the biraphe type species showed higher RIA than any other morph, while they comprised over 55% of total biomass, indicating a density effect. Additionally, a significant relationship between RIA and 20-day deposited substrate (p<0.05) showed in two diatom species, Nitzschia fonticola, Gomphonema quadripunctatum, which showed relatively high RIAs. These results indicate that the relative immigration activities of epilithic diatom species can be influenced by algal density in the water and substrate, nutrients, and deposited time at least 20 days.

Annotation and Expression Profile Analysis of cDNAs from the Antarctic Diatom Chaetoceros neogracile

  • To better understand the gene expression of the cold-adapted polar diatom, we conducted a survey of the Chaetoceros neogracile transcriptome by cDNA sequencing and expression of interested cDNAs from the Antarctic diatom. A non-normalized cDNA library was constructed from the C. neogracile, and a total of 2,500 cDNAs were sequenced to generate 1,881 high-quality expressed sequence tags (ESTs) (accession numbers EL620615-EL622495). Based on their clustering, we identified 154 unique clusters comprising 342 ESTs. The remaining 1,540 ESTs did not cluster. The number of unique genes identified in the data set is thus estimated to be 1,694. Taking advantage of various tools and databases, putative functions were assigned to 939 (55.4%) of these genes. Of the remaining 540 (31.9%) unknown sequences, 215 (12.7%) appeared to be C. neogracile-specific since they lacked any significant sequence similarity to any sequence available in the public databases. C. neogracile consisted of a relatively high percentage of genes involved in metabolism, genetic information processing, cellular processes, defense or stress resistance, photosynthesis, structure, and signal transduction. From the ESTs, the expression of these putative C. neogracile genes was investigated: fucoxanthin chlorophyll (chl) a,c-binding protein (FCP), ascorbate peroxidase (ASP), and heat-shock protein 90 (HSP90). The abundance of ASP and HSP90 changed substantially in response to different culture conditions, indicating the possible regulation of these genes in C. neogracile.
