The Effect of Rutin on Antioxidant and Anti-inflammation in Streptozotocin-induced Diabetic Rats

  • The study examined the antioxidant activities and anti-inflammatory effects of rutin from the streptozotocin (STZ)-induced diabetic rats. Results revealed that the levels of plasma glucose and serum glucose were remarkably higher in the STZ-treated group compared to other groups and were significantly reduced in the STZ+rutin treated group compared to the STZ-treated group. In terms of weight, it significantly increased in all experimental groups during the experiment period except for STZ-induced diabetic group. The weight of the STZ-treated group was remarkably reduced compared to other groups. Regarding the weight of each body organ, the STZ-treated group showed higher organ weight compared to the other groups while STZ+rutin-treated group showed significantly reduced kidney and liver weights compared to those of STZ-treated group. In the pancreas tissue of the STZ-treated group, ${\beta}$-cell destruction and vacuolization were observed. Inflammation in the heart, liver, kidney, and retina tissues were also vividly recorded. In the STZ+rutin administered group, the heart and retina tissues were shown to be preserved normally while the liver and kidney tissues showed reduced histopathology in general compared to the STZ-treated group. Conclusionally, the rutin has the effect on the antioxidant activities and anti-inflammation in the STZ-induced diabetic rats.

Anti-diabetic Effects of Mixed Water Extract from Ginseng Radix Rubra, Acanthopanacis Cortex, and Cordyceps (홍삼.가시오가피.동충하초 혼합수엑스의 항당뇨작용)

  • The effect of water extract composed of panax ginseng radix rubra, acanthopanacis cortex, and cordyceps (PAC) on diabetic animal models were investigated in two different diabetic animal models. FAC water extract significantly reduced the plasma glucose levels on day 30 as compared with the diabetic control group in $KKA^Y$ obese, hyperglycemic and hyperglycemic and hyperinsulinemic mice, and also reduced the plasma glucose levels as well as total cholesterol in multiple low dose (MLD) strep tozotocin-induced diabetic SD rats. PAC water extract also showed an inhibitory effect on reduction of body weight and on development of MLD STZ-induced diabetic state. Elevated kidney hypertrophy, which is a characteristic feature shown in early stage of diabetic nephropathy and calculated as the ratio of kidney mass (g) relative to the body weight (g), was also markedly improved in PAC water extract- treated group as compared to the diabetic control group. Taken together, these data suggest that PAC water extract may have a potential as a antidiabetic agent in type 2 diabetes mellitus.

Effects of Ginseng Saponin on the Lysosomal Enzyme Activities in Streptozotocin-induced Diabetic Mice (인삼 Saponin이 Lysosome 효소 활성에 미치는 영향)

  • Lysosomal enzymes might play a most important role in the pathogenesis od diabetic microangiopathy. Some glycosidases, which participate in the catabolism of glycoprotein, are significantly decreased in diabetic mice. In search of new potential lysosomal enzyme inducers, we examined the effects of crude red-ginseng saponin fraction on N-acetyl-$\beta$-D-glucosaminidase, $\beta$-D-galactosidase and $\alpha$-D-mannosidase activities in the liver and kidney of normal and streptozotocin induced diabetic mice. It was found that i.p. administration of ginseng saponin produced the induction of lysosomal enzymes in the kidney more intensively than in the liver. The obtained results suggest the possibility that ginseng saponin might prevent the diabetic microangiopathy.

Effect of the New Diabetic Formula on Streptozotocin-induced Diabetic Rats (Streptozotocin 당뇨모델에 대한 신당뇨방의 효과)

  • The present study evaluated the beneficial effect of new diabetic formula(NDF) in diabetic rats. Adult Sprague-Dawley rats weighing 250-300 g were used. Diabetes mellitus was induced by intramuscular injections of streptozotocin(STZ, 50㎎/㎏). The extracts of NDF were orally administered at low or high dose two times a day to fasted diabetic rats for 3 weeks. Adminstration of NDF alliviated a significant reduction in weight gain in rats with STZ-induced diabetes. Following acute and repeated treatment, low dose of NDF suppressed the blood glucose concentrations of fasted diabetic rats. Repeated adminstration of NDF for 21 days improved liver and kidney functions in diabetic rats, as indicated by decline of serum alanine aminotransferase(AL T), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine level and kidney weights. The present study showed that NDF exerted antihyperglycemic effects and alliviated liver and renal damages caused by streptozotocin-induced diabetes.

Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

Renoprotective Effect of Hominis Placenta Aqua-acupuncture in Streptozotocin Induced Diabetic Rats (자하차약침(紫河車藥鍼)이 당뇨유발(糖尿誘發) 흰쥐의 신장보호기능(腎臟保護機能)에 미치는 영향(影響))

  • Objective : This study was undertaken to determine whether Hominis Placenta(HP) aqua-acupuncture exerts renoprotective effect on diabetic nephropathy induced by streptozotocin(STZ) in rats. Methods : In order to study the renoprotective effect of HP aqua-acupuncture, experimental animals were divided into 5 groups and treated for 2 weeks as follows: control group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 5% HP aqua-acupuncture group was injected subcutaneously by 5% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, 10% HP aqua-acupuncture group was injected subcutaneously by 10% HP aqua-acupuncture into Sinsu(BL23) in STZ induced diabetic rats, normal group was injected subcutaneously by saline aqua-acupuncture into Sinsu(BL23) in normal rats, and Captopril group was administrated with captopril at a dose of 50mg/kg in STZ induced diabetic rats. Results : While HP aqua-acupuncture did not reduce any body weight, index of kidney hypertrophy, the plasma glucose concentration and BUN respectively, HP aqua-acupuncture showed lowering urinary albumin excretion rate and serum creatinine as compared with the control group. Gene and protein expressions of $TGF-{\beta}1$ and fibronectin in kidney, one of the extracellular matrix proteins were investigated. There were significant differences in expression levels in HP aqua-acupuncture group as compared with the control group, and $100{\mu}g/m{\ell}$ and $500{\mu}g/m{\ell}$ of HP depressed apoptosis, showing in a dose dependent manner. In the HE staining, HP aqua-acupuncture inhibited the injury of glomerulus and proximal convoluted tubule. Conclusions : HP aqua-acupuncture showed the renoprotective effect possibly through suppressions of $TGF-{\beta}1$ and fibronectin expressions in kidney.

Effects of Vitamin E on the Microstructural Changes of Renal Tissue in Streptozotocin-Induced Diabetic Rats (식이 Vitamin E가 Streptozotocin 유발 당뇨쥐 신장조직에서의 병리조직학적 변화에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on the histochemical change of kidney tissue in diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40mg per kg diet. Diabetes was exper imentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. The contents of thiobarbituric acid(TBARS) in kidney were increased 119%, 84% and 33% in DM 0E, DM 40E and DM 400E groups, respectively, compared to normal group. That of DM 400E group was decreased 39% compared to DM 0E group. Content of 2 microglobulin in urine in DM 0E, DM 40E, and DM 400E groups were increased by 248%, 181%, and 164%, respectively, compared to normal group. The diabetic groups showed the regressive lesion such as renal tubule, intumescence of epithelial cell, vacuolization. The results of the observation through electronic microscope showed the mitochondria shape of proximal tubule epithelial cell, irregular array, increase of ribosome, and irregular arrangement of small villosity, etc. These types of changes appeared severer in DM 0E group than in DM 400E group. These results indicate that the TBARS productions on kdney in STZ induced diabetic rats were increased, consequently those leaded to damage of renal tubule and minuteness structure. But a large quantity vitimin E supplementation was suppressed in TBARS production and improved in peroxidative damage of renal tissue so that relieved degenerative changes of renal tubule epithelial cell.

Effect of the root extract of Pueraria thunbergiana Bentham on high fat/high sucrose diet and single low-dose streptozotocin-induced diabetic mice (갈근이 고지방·고탄수화물식이와 저용량 streptozotocin-유도 당뇨병 마우스에 미치는 효능 연구)

  • Objectives : This study was performed to investigate the effect of root extract of Pueraria thunbergiana Bentham (Puerariae Radix, PR) in diabetic mice as similar as emaciation-thirst disease in Oriental medicine. Methods : C57BL/6 mice were fed high fat (HF) and high sucrose (HS) for 8 weeks, and then administrated with 90 mg/kg body weight (bw) of streptozotocin (STZ) for induction of diabetes which is similar to the middle emaciation stage. After 5 days, blood glucose levels were measured, and selected the mice with ranges above $250mg/d{\ell}$. PR water extract was administrated orally once a day for 4 weeks with high fat and high sucrose. The levels of glucose, insulin, total cholesterol, triglyceride, ${\gamma}glutamyl$ transpeptidase (${\gamma}GTP$), glutamic oxaloacetic transaminase (GOT) and glutamate pyruvate transaminase (GPT) were analysed in the serum. Also, observed their histological changes by hematoxylin and eosin (H&E) of different organs, lung, heart, pancreas, stomach, liver, and kidney. Results : PR extract significantly decreased the levels of serum glucose and insulin in diabetic mice. PR extract significantly increased the levels of triglyceride, total cholesterol, GOT and GPT in diabetic mice. In H&E stain, PR extract inhibited the histopathological changes of lung (as a channel of the upper emaciation stage in the channel-tropism theory), pancreas (as a channel of the middle emaciation stage) and kidney (as a channel of the lower emaciation stage) in diabetic damage. Conclusions : PR extract has an anti-diabetic effect in HF/HS and low-dose STZ-induced diabetic mice. This result suggests that PR follows the channel-tropism theory in the emaciation-thirst disease through the protection of lung, pancreas and kidney.

  • Objectives : This study aimed to evaluate the hypoglycemic, hepatoprotective, and nephroprotective effects of 'Saengjinyanghyul-tang($Sh\={e}ngj\={i}ny\v{a}ngxu\`{e}-t\={a}ng$ : SJYHT)' in streptozotocin (STZ)-induced diabetic SD rat model. Methods : SJYHT extracts were once a day dosed for 28 days at a dosage 1000, 500, and 250mg/kg/5ml from 25 days after STZ-dosing, and the changes on blood glucose levels, liver and kidney weight, serum AST, ALT, BUN and creatinine levels were observed with histological changes on the liver and kidney. Results : Increased blood glucose levels, and diabetic hepatopathy & nephropathy including increased serum AST, ALT, BUN and creatinine levels, histological changes after STZ-dosing were dose-dependently reduced by SJYHT extract-dosing. Conclusions : 'SJYHT' water extracts have favorable hypoglycemic, hepatoprotective, and nephroprotective effects on the STZ-induced diabetic SD rats. Therefore, it is expected that 'SJYHT' has potential for use in the management of diabetes and diabetic complications.

Effect of the Lycii Fructus on multiple low-dose streptozotocin-induced diabetic rats (Streptozotocin 저용량 반복투여로 유도한 당뇨 흰쥐에서 구기자 추출물의 효능연구)

  • Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats.Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured.Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage.Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.
