Detection and Distribution of the Pathogenic Bioagent Aeromonas (Gamma-Proteobacteria) in Water Supplies of Seoul (서울시 상수계통에서 병원성균 Aeromonas (감마-프로테오박테리아) 분포연구)

  • The detection and distribution of Aeromonas in water supplies were investigated by using the USEPA Method 1605. Water samples were collected from the Han River, finished waters and tap waters supplied from Water Treatment Plants in Seoul monthly from July 2002 to December 2003. Aeromonas species in each water sample were quantified based on the development of yellow colonies on the surface of membrane filter using a selective medium (Ampicillin-Dextrin Agar with Vancomycin). The Quality Control (QC) for this study met the acceptance criteria of Method 1605. The concentrations of Aeromonas species in surface water samples ranged from $1.0{\times}10^{0}\;to\;9.8{\times}10^{3}\;CFU/ml$. Aeromonas species were found only in one tap water sample with concentration of 1 CFU/500 ml. No Aeromonas species were found in any finished water samples. Aeromonas species detected here were identified as A. salmonicida(51%), A. caviae(4.7%), A. schubertti(3.4%), A. sobria(3.8%), A. hydrophila(2.1%), and A. ichithiosmia(0.4%). A. salmonicida was the dominant species, which is of no significance to human health. Chlorine resistance of A. salmonicida was evaluated and as a result, 99.99% of A. salmonicida decreased after 30 seconds exposure at residual free chlorine 0.2 mg/L. These suggest that the waters supplied in Seoul may be safe against the pathogenic agent Aeromonas.

Inclusion effect of soybean meal, fermented soybean meal, and Saccharina japonica in extruded pellet for juvenile abalone (Haliotis discus, Reeve 1846)

  • Inclusion effect of soybean meal (SBM) and fermented SBM (FSM) in extruded pellet for juvenile abalone (Haliotis discus) was compared in abalone farm. Dietary inclusion effect of the combined macroalgae (MA) (Undaria pinnatifida and Hizikia fusiforme) and a single Saccharina japonica on abalone was also compared. Three thousand six hundred juvenile abalone were purchased from a private hatchery and acclimated to the experimental conditions for 2 weeks. Six 5-ton flow-through raceway tanks were used, and abalone were randomly distributed into tanks (n = 600 per tank). Three experimental diets were prepared in duplicate. Fish meal, FSM, corn gluten meal, and shrimp meal and wheat flour and dextrin were used as the protein and carbohydrate sources, respectively, in the FSM diet. MA was also included in the FSM diet. FSM and MA in the FSM diet were substituted with SBM at the expense of wheat flour and S. japonica, referred to as the SBM and SJ diets. The experimental diets were pelletized by an extruded pelleter. Water stability of nutrients in the experimental diets was monitored at 12, 24, and 48 h after seawater immersion. The experimental diets were fed to abalone once a day to satiation with a little leftover for 120 days. The retained crude protein and lipid and ash content of the extruded pellets were changed over all period of time. Weight gain and specific growth rate (SGR) of abalone fed the SBM diet were greater than those of abalone fed the FSM and SJ diets. Weight gain and SGR of abalone fed the SJ diet were also greater than those of abalone fed the FSM diet. The longest shell length, widest shell width, highest shell height, and greatest soft body weight were obtained in abalone fed the SBM diet, followed by the SJ and FSM diets. Proximates of the soft body of abalone were not different among the experimental diets. In conclusion, SBM was a superior protein source to FSM in extruded pellet for growth performance of abalone. Dietary inclusion of a single S. japonica was superior to the combined inclusion of U. pinnatifida and H. fusiforme in the production of abalone.

Effects of Allium Hookeri on Glucose Metabolism in Type II Diabetic Mice (삼채(Allium Hookeri)의 급여가 당뇨마우스의 당질 대사에 미치는 영향)

  • This study was conducted to verify the potential of Allium hookeri to control glucose metabolism in a diabetes model. We fed the experimental diets (AL, AR, Dex) supplemented with the powder of leaf, root, or dextrin as a positive control, respectively at 3% of diet to the diabetic mice (C57BLKS/J, db/db) for 8 weeks. Control mice were fed with the diet supplemented with cornstarch (Cont) at 3% level of diet. At 8th week of feeding the diets, we measured body weight, blood glucose, HbA1c, and plasma insulin levels and conducted an oral glucose tolerance test (OGTT) and staining insulin immunoreactive cells in islets of pancreas. AL group treated with the leaf of A. hookeri showed significantly lower blood glucose and HbA1c levels, higher plasma insulin levels, and increased density of insulin immunoreactive cells compared with the Cont group. During the OGTT, AL group showed lower blood glucose levels than the Cont group for 120 min. Based on these results, leaf of A. hookeri is considered to be effective in improving glucose tolerance by partially affecting insulin secretion and it may be used to prevent and treat diabetic disease.

The Biological and Biochemical Characteristics of Staphylococcus epidermidis Isolated from Diseased Cultured Flounder, Paralichthys olivaceus (양식 넙치에서 분리한 Staphylococcus epidermidis의 생물학적 및 생화학적 특성)

  • A total of 8 strains of Staphylococcus epidermidis isolated in land-marine tank system of Kyongnam and Kyongbuk. Prefecture were tested for the biological and biochemical characteristics. These strains were isolated from pathologic specimens of cultured flounder. Paralichthys olivaceus. Growth of the isolates was good on BHIA, HIA. Staphylococcus No. 110 and ETGP. Growth was good at NaCl concentration between 2.0 and 3.0%, about $30^{\circ}C$ and at pH values about 7.0. DNase and coagulase production were negative, and all isolates except FSJ-2 strain were positive in hemolysis. Urease was positive reaction, and novobiocin resistance was negative. Acid was produced anaerobically from glucose and maltose. All isolates except FSJ-19 strain produced weakly were negative in anaerobic acid production from mannitol. Acid was produced aerobically from glucose, fructose, galactose, sucrose, maltose and dextrine. But all isolates were not gas production. In characterization of clinically isolates, four different biotype codes were obtained when the all isolated were tested simultaneously. Four different antibiotic susceptibility patterns were obtained. On the basis of these characteristics, the isolates were identified as Staphylococcus epidermidis.

  • Several bacteria are known as the causal agents of diseases of the cultivated button mushroom(Agaricus bisporus) and oyster mushroom(Pleurotus ostreatus). Pseudomonas tolaasii is the causal agent of brown blotch disease of commercial mushrooms. Pseudomonas sp. HC1 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This can markedly reduce the level of extracellular toxins (i.e., tolaasins) produced by Pseudomonas tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the Pseudomonas sp. HC1, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 5.0 and $20^{\circ}C$, respectively. The optimal culture medium for the growth of tolaasin inhibitor bacterium was determined as follows: 0.9% dextrin, 1.5% yest extract, 0.5% $(NH_4)_2HPO_4$, 4mM $FeCl_3$, and 3.0% cysteine.

Relationship between Molecular Structure of Rice Amylopectin and Texture of Cooked Rice (쌀의 아밀로펙틴 분자구조와 밥의 텍스쳐)

  • The relationship betwwen the molecular structure of amylopectin and the texture of cooked rice was investigated using Korean rice [3 varieties of Japonica type and 3 varieties of Tongil type(Japonica-Indica breeding type)]. The molecular structure of rice amylopectin was polymodal and distributed A chain of $\overline{DP}$ 12.4, short B chain of $\overline{DP}$ 20.6, B chain of $\overline{DP}$ 26.3, long B chain of $\overline{DP}$ 45 and super long chain of above $\overline{DP}$ 55. The super long chain of amylopectin was composed of long linear chain with poorly branched chain. Also, the super long chain of amylopectin showed positive correlated with average chain length, inherent viscosity and ${\beta}-amyloysis$ limit$({\%})$, but negative correlated with ${\lambda}max$ of iodine reaction of amylopectin. The structural properties of amylopectin in Japonica type were different from those of amylopectin in Tongil type. In relationship between molecular structure of amylopectin and texture of cooked rice, the average chain length, inherent viscosity, ${\beta}-amyloysis$ limit and super long chain of amylopectin was showed a positive correlation with hardness, but a negative correlation with adhesiveness of cooked rice. The long chain of rice amylopectin is the less, the eating quality of cooled rice was the better. These results suggest that the molecular structure of rice amylopectin could be responsible for the texture of cooked rice.

Structural Properties of Naked and Covered Barley Starches (쌀보리와 겉보리 전분의 분자구조적 성질)

  • The structural properties of covered and naked barley starches and those components were investigated. ${\beta}-amylosis\;limit(%)$ of covered and naked barley starches were 58.6 and 56.3%, respectively and those of their amyloses and amylopectins were 87.0, 77.7, 57.6 and 52.0%, respectively. The ratios of outer chain length ${\overline{OCL}}$ and inner chain length${\overline{ICL}}$ for amylopectins of covered and naked barley were about 2.2:1 and 2.0:1, respectively. The elution curves by Sephadex G-75 after debranching starches with pullulanase were similar patterns for two starches and yielded two peaks consisting of amylose component${\overline{dp}}>55$ and bimodal amylopectin components of ${\overline{dp}_(35-45)$ and ${\overline{dp}}(10-20)$ hydrolysates. Also, hydrolysis products by sequential hydrolysis with pullulanase and ${\beta}-amylase$ contained 0.03-0.5% non-hydrolyzed peak components of ${\overline{dp}}>55$. The elution profiles of ${\beta}-amylase$ hydrolysates yielded two peaks consisting of the inner components${\overline{dp}}>55$ and the outer chain components of amylopectin${\overline{dp}}>10$ regardless of samples. However, ${\overline{dp}}$ distributions of ${\overline{dp}>55}$ hydrolysates exhibited the significant differences due to the varieties.

The Effect of Bulking Agent on Quality of Kiwifruit Powder in the Process of Domestic Kiwifruit Tenderizer (국내산 키위연육제 제조과정 중 부형제의 첨가가 키위분말의 품질에 미치는 영향)

  • Development of tenderizer using domestic fruits was studied. Kiwifruit was dried using various methods, and the quality of kiwifruit powder was observed during 12 week storage. Frozen kiwifruit was prepared in paste, dice, and whole flesh. After drying, paste-type kiwifruit showed 2.0 and 1.3 times higher proteolytic activity than dice and whole flesh kiwifruits, respectively. Nine hour of hot-air drying or 46 h of freeze-drying eliminated more than 90% of water from kiwifruit, during which discoloring of kiwifruit occurred. Freeze-dried powder showed 6.6 times higher yield and proteolytic activity, and resulted in almost no discolorization than those of air-dried powder. Addition of bulking agent affected the quality of hot air-dried kiwifruit powder, except color, resulting in $3.2{\sim}3.6$ times higher proteolytic activity than that without bulking agent, which is comparable to 60% of the initial freeze-dried powder content. Moisture content of kiwifruit powder with bulking agent sustained consistently during 12 week storage, whereas proteolytic activity decreased for the first 4 weeks. Freeze-drying is a preferable method to produce kiwifruit powder for tenderizer, although hot air-drying with bulking agent treatment is more economical.

Effects of Chitosan on Cholesterol Level and Hepatic Morphology in Ethanol-treated Rats (키토산의 섭취가 에탄올을 급여한 흰쥐의 콜레스테롤농도 및 간조직 헝태에 미치는 영향)

  • This study was designed to determine the effect of chitosan on in vivo lipid metabolism in male Sprague-Dawley rats treated with ethanol. Rats were divided into four groups and reared for 6 weeks: E group ($35\%$ of total calories from ethanol), EC I group ($ethanol+0.5\%$ of chitosan), EC II group ($ethanol +1\%$ of chitosan) and control group (dextrin as much as ethanol treated). The levels of serum total cholesterol (TC) and LDL-cholesterol (LDL-C), GOT and GPT in plasma, and triglyceride (TG) in liver were remarkably increased in the rats treated with ethanol. However, the treatment of $1\%$ chitosan significantly lowered those parameter levels. In particular the values of r-HDL (the ratio of HDL-C to TC) in the rats fed in combination with ethanol and chitosan were relatively higher than that of the E group. The increased lipid droplets were observed in the hepatocytes of the rats treated with ethanol, but chitosan treatment reduced in the number and the size of the lipid droplets. These results suggest that chitosan improve in vivo lipid metabolism and Potentially protect hepatotoxicity of the rat liver treated with ethanol.

  • Allium hookeri is a plant species native to India, Sri Lanka, Myanmar, and China. The plant is widely cultivated in Korea lately as a medicinal food item. This study was conducted to evaluate the effects of Allium hookeri (A. hookeri) on lipid metabolism in Type II diabetic mice (n=8/group, 5 groups). High fat diets with dextrin as a positive control (Dex), leaf (AL), root (AR), and fermented root (FAR) at 3% of diet were fed to all experimental mouse, respectively for 8 weeks. Body weight gain, liver and epididymal fat weights, and excreted fecal lipid levels were measured. Serum and hepatic lipid profiles were analyzed, and fat accumulation in liver was evaluated. In this study, body weight gain and epididymal fat weight were lower in the FAR group, while serum HDL-cholesterol level and excreted fecal total lipid and triglyceride levels were higher in AL or FAR groups. These results suggest that A. hookeri, specially fermented root can be a useful food item to control lipid metabolism in diabetic mice.
