Preimplantation Developmental Ability of Pig Embryos according to Embryonic Compaction Patterns (돼지수정란의 Compaction 양상에 따른 착상전 배발달 양상)

  • Embryonic compaction is essential for normal preimplantation development in mammals. The present study was to investigate the effects of compaction patterns on developmental competence of pig embryos. The proportion of blastocyst formation derived from compacted morula was higher than those of compacting and pre-compacting morula (P<0.01). Nuclei numbers of inner cell mass (ICM), trophectoderm (TE), and total of blastocysts derived from compacted group were also superior to those of compacting and pre-compacting groups (P<0.05). Then, compaction patterns, developmental ability and structural integrity were compared between mono- and poly-spermic embryos. The rate of compacted morula in mono-spermic embryos was higher than that of poly-spermic embryos (P<0.05). Especially, the rate of blastocyst formation derived from compacted embryos in mono-spermic embryo group was higher than that of poly-spermic embryo group (P<0.05), although no difference was detected between the two groups in the structural integrity. Finally, we confirmed that beta-catenin was differentially expressed according to compaction patterns in morula and blastocyst stage embryos. In conclusion, our results suggest that the compaction patterns during preimplantation development play a direct role in developmetal competence and quality of pig embryos.

Studies on cryopreservation of nuclear transplanted mouse embryos (마우스 핵이식란의 동결에 관한 연구)

  • The present study was carried out to investigate the developmental potency to blastocyst after freezing and thawing of nuclear transplanted 2-cell embryos. The nuclei from 2-, 4- and 8-cell mouse embryos were transferred into enucleated 2-cell embryos, and the reconstituted embryos were submitted to direct current(DC) pulse at output voltage of 2.0 kV/cm for $100{\mu}$ sec to induce cell fusion. The recovery rate and developmental potency to blastocyst after freezing and thawing of nuclear transplanted 2-cell embryos was investigated. 1. The recovery rate of nuclear transplanted 2-cell embryos in normal morphology after freezing and thawing was significantly higher in rapid freezing(DMSO 4.5M) than in slow cooling(p<0.01). 2. When the recovered embryos in normal morphology were cultured in vitro, there were no significant differences in the developmental potency to blastocyst between the freezing methods and the concentrations of cryoprotectant. In summary, these experiments have proved that rapid freezing method(DMSO 4.5M) is effective in nuclear transplanted 2-cell mouse embryos. If improved micromanipulation techniques and freezing are combined, nuclear transplantation technique will contribute to the improvement of productivity in livestock animals.

How to Investigate Students' Zone of Proximal Development (ZPD) (학생들의 근접발달영역(ZPD)에 대한 탐구)

  • This study investigates aspects of the zone of proximal development (ZPD), the distance between the actual development and the potential development. Out of 18 university students taking a geometry course, two students with the same actual developmental level in the van Hiele model in the pre-test and post-test were interviewed for measuring their potential developmental level. Based on the communicational approach to cognition, the characteristics of the two interviewees' discourse on 3D reflective symmetry were identified. There were considerable differences between the two interviewees in terms of their potential developmental level. Methodological implications for how to investigate students' ZPD in mathematics education research were addressed.

  • In this study, we describe the Bombyx mandarina hemolin cDNA. A sequence analysis of cDNA revealed a single open reading frame (ORF) of 1233 nucleotides. The deduced 410 amino acid sequence of B. mandarina hemolin contains 4 imunoglobulin (Ig) C-2 type domains. B. mandarina hemolin cDNA showed the highest sequence homology to known those of B. mori. The developmental profile in terms of expression level of hemolin mRNA was determined in the absence of a bacterial challenge. Hemolin mRNA was detected only in mid-gut, but not in hemocytes, fat body, testis, and silkglands. Hemolin mRNA in mid-gut was not detected until the spinning stage of the last instar larva, however, lit dramatically increased at the beginning of spinning and gradually decreased until pupal stage.

  • Motor delay, when present, is usually the first concern brought by the parents of children with developmental delay. Cerebral palsy that is the most common motor delay, is a nonspecific, descriptive term pertaining to disordered motor function that is evident in early infancy and is characterized by changes in muscle tone, muscle weakness, involuntary movements, ataxia, or a combination of these abnormalities. A wide range of causative disorders and risk factors have been identified for cerebral palsy, and broadly classified into 5 groups; perinatal brain injury, brain injury related to prematurity, developmental abnormalities, prenatal risk factors, and postnatal brain injury. Delay in attaining developmental milestones is the most distinctive presenting complaint in children with cerebral palsy. A detailed history and thorough physical and neurologic examinations are crucial in the diagnostic process. The clinician should be cautious about diagnostic pronouncement unless the findings are unequivocal. Several serial examinations and history review are necessary. All children with cerebral palsy should undergo a neuroimaging study, preferably MRI, because an abnormality is documented on head MRI(89%) and CT(77%). The high incidence rates for mental retardation, epilepsy, ophthalmologic defects, speech and language disorders and hearing impairment make it imperative that all children with cerebral palsy be screened for mental retardation, ophthalmologic and hearing impairments, and speech and language disorders; nutrition, growth, and swallowing also should be closely monitored.

Developmental characterization of embryo size mutant in rice (Oryza sativa L.)

  • In this experiment, three kinds of mutations(ge, re, and eml )relating to the size of embryos were used to study their generation, genetic mechanism and developmental characteristics, and the interactions between embryo and endosperm were also examined. Giant embryo mutation comprises 7 kinds including the already isolated ge, and ge-2, which share an identical gene site. The SAM and the size of radicule for the ge showed little difference compared to a normal type. The number of embryo cells did not increased as much as it would affect the size of embryo. Therefore, the enlargement of embryo was due to the enlargement of scutellum that originated from the corpulence of each cell. Both F$_1$' s of re ]and odm 49 formed reduce embryos, and other combinations of hybridization showed all wild type of embryo sizes. Accordingly, the odm 49 must have an identical gene site of re 1, while odm 48 and odm 62 have different gene sites. Their shoots and radicules also shrank by the same ratio, however no sign of physical change was noticed. The size of embryo cell showed no change, while the number of cells was the half of that of wild types. The three gene sites of re represent all of them control the size of the entire embryo forming organs. The eml 1 was defined to have temperature sensibilities that the generation of endosperms was active at a high temperature while that was hampered at a low temperature.

The Effects of Developmental Bibliotherapy in School : In the Case of the Model School (발달적 독서치료 프로그램 운영의 효과 - 시범학교의 사례연구 -)

  • This paper analyzed the effects of group bibliotherapy in junior high school assigned by Daegu Metropolitan Office or Education as a model school. Seventy seven students haying problems were selected and divided into the two different programs which developed for internalizing problems and externalizing problems respectively. Each bibliotherapy program has been carried out 15 times. The effects of two programs were proved positive and statistically significant. The effect of each program were differ from the other, owing to the purpose and reading materials. The study skewed bibliotherapy are more effective for students having internalizing Problems and female students. Bibliotherapy also affected student's reading attitude. In order to reach self bibliotherapy, developmental bibliotherapy should be connected to the reading guidance.

Stimulatory Effect of N-acetylcysteine on Odontoblastic Differentiation

  • Reparative dentine formation requires newly differentiated odontoblast-like cells. Therefore, identification of the molecule that stimulates the odontogenic differentiation of precursor cells in the tooth pulp will be helpful for the development of strategies to repair damaged pulp. In this study, we examined the effect of N-acetylcysteine (NAC) on the odontogenic differentiation of MDPC-23 cells, a mouse odontoblast-like cell line derived from dental papilla, and primary cultured rat dental papilla cells (RDPCs). NAC (1-30 mM) suppressed production of reactive oxygen species in MDPC-23 cells in a dose-dependent manner. Although 5 to 20 mM NAC did not alter MDPC-23 cell proliferation, 1 or 30 mM NAC significantly inhibited it. NAC enhanced mineralized nodule formation and the expression of several odontoblast differentiation-associated genes in both RDPCs and MDPC-23. This NAC stimulatory effect was significant, even at concentrations lower than 1 mM. However, NAC did not stimulate expression of bone morphogenetic protein-2, -4, or -7, which are known to enhance odontogenic differentiation. Since reactive oxygen species are also involved in the pulp toxicity of resin-based restorative materials, these results suggest that NAC may be a promising candidate for supplementation of dental restorative materials in order to enhance reparative dentine formation.

Effects of processed tobacco leaves for the development and emergence of cigarette beetle (담배 가공 원료엽의 궐련벌레 생육과 우화에 미치는 영향)

  • This study was conducted to determine the effects of processed tobacco leaves on the development, adult emergence and body weight of the cigarette beetle, Lasioderma serricorne Fabricius) (Coleoptera: Anobiidae) is serious insect pest of tobacco leaves and cigarette during storage. Developmental time, adult emergence rate and adult weight of the cigarette beetle, were evaluated on the cured tobacco and burley tobacco leaves at $30{\pm}1^{\circ}C$ with $70{\pm}5$ % RH under 12L:12D. The developmental time on all of the flue-cured tobacco leaves was about 61 days, but in the only CD3W and CD4TR grade burley tobacco, the developmental times ranged from 70 days to 74 days. Among the flue-cured tobacco leaves, the highest beetle emergence rate was 123 % on the CD3L grade, and the lowest was on the AB4OR grade. Adult body weights of the cigarette beetle reared on flue-cured tobacco were about 2.11~2.46 mg, and on the only CD3W and CD4TR grade burley tobacco were about 1.86~1.96 mg. Among the flue-cured tobacco leaves, the highest adult body weight(2.46 mg) of cigarette beetle was observed on the B1O grade flue-cured tobacco, whereas the lowest adult weight(2.11 mg) was observed on the CD4L grade flue-cured tobacco. The adult weight of cigarette beetle reared on whole meal was 2.04mg.

Effects of Cryoprotectants on In Vitro Development of Vitrified Immature Porcine Oocytes Following ICSI

  • In the present study, effects of concentration of cryoprotectant solutions on the nuclear maturation of vitrified-thawed porcine oocytes were examined. Also, the developmental capacity of vitrified-thawed immature porcine oocytes following ICSI was investigated. Oocytes were cultured in NCSU-23 medium supplemented with 5% FBS at $38^{\circ}C$ in 5% $CO_2$ and air. The in vitro maturation rate of vitrified-thawed oocytes ($24.1{\pm}2.5%$) was lower than that of the control ($46.0{\pm}3.2%$, p<0.05). The in vitro maturation rate of vitrified-thawed oocytes treated with $1.0{\sim}5.0\;ug$ CB + NCSU- 23 medium were $22.2{\pm}3.0%$, $30.7{\pm}3.2$, $46.3{\pm}3.1%$, $38.5{\pm}3.2%$, respectively. The in vitro maturation rate ($46.3{\pm}3.4%$) of the vitrified-thawed oocytes treated with $3.0\;{\mu}g$ CB for 30 min was the highest of all vitrification groups. When the in vitro developmental rates of the vitrified-thawed (with EDS and EDT) oocytes following ICSI were $18.5{\pm}2.5%$, $16.4{\pm}2.1%$, respectively. This results were lower than the control group ($24.0{\pm}2.5%$).
